The release of growth factors from tissue engineering scaffolds provides signals that influence the migration, differentiation, and proliferation of cells. The incorporation of a drug delivery platform that is capable of tunable release will give tissue engineers greater versatility in the direction of tissue regeneration. We have prepared a novel composite of two biomaterials with proven track records - apatite and poly(lactic-co-glycolic acid) (PLGA) – as a drug delivery platform with promising controlled release properties. These composites have been tested in the delivery of a model protein, bovine serum albumin (BSA), as well as therapeutic proteins, recombinant human bone morphogenetic protein-2 (rhBMP-2) and rhBMP-6. The controlled release strategy is based on the use of a polymer with acidic degradation products to control the dissolution of the basic apatitic component, resulting in protein release. Therefore, any parameter that affects either polymer degradation or apatite dissolution can be used to control protein release. We have modified the protein release profile systematically by varying the polymer molecular weight, polymer hydrophobicity, apatite loading, apatite particle size, and other material and processing parameters. Biologically active rhBMP-2 was released from these composite microparticles over 100 days, in contrast to conventional collagen sponge carriers, which were depleted in approximately 2 weeks. The released rhBMP-2 was able to induce elevated alkaline phosphatase and osteocalcin expression in pluripotent murine embryonic fibroblasts. To augment tissue engineering scaffolds with tunable and sustained protein release capabilities, these composite microparticles can be dispersed in the scaffolds in different combinations to obtain a superposition of the release profiles. We have loaded rhBMP-2 into composite microparticles with a fast release profile, and rhBMP-6 into slow-releasing composite microparticles. An equi-mixture of these two sets of composite particles was then injected into a collagen sponge, allowing for dual release of the proteins from the collagenous scaffold. The ability of these BMP-loaded scaffolds to induce osteoblastic differentiation in vitro and ectopic bone formation in a rat model is being investigated. We anticipate that these apatite-polymer composite microparticles can be extended to the delivery of other signalling molecules, and can be incorporated into other types of tissue engineering scaffolds.

Bone morphogenetic protein-2 (BMP-2) has the ability to induce osteoblast differentiation of undifferentiated cells, resulting in the healing of skeletal defects when delivered with a suitable carrier. We have applied a versatile delivery platform comprising a novel composite of two biomaterials with proven track records – apatite and poly(lactic-co-glycolic acid) (PLGA) – to the delivery of BMP-2. Sustained release of this growth factor was tuned with variables that affect polymer degradation and/or apatite dissolution, such as polymer molecular weight, polymer composition, apatite loading, and apatite particle size. The effect of released BMP-2 on C3H10T1/2 murine pluripotent mesenchymal cells was assessed by tracking the expression of osteoblastic makers, alkaline phosphatase (ALP) and osteocalcin. Release media collected over 100 days induced elevated ALP activity in C3H10T1/2 cells. The expression of osteocalcin was also upregulated significantly. These results demonstrated the potential of apatite-PLGA composite particles for releasing protein in bioactive form over extended periods of time.

Chitosan, which is a non-toxic, biodegradable and biocompatible biopolymer, has been widely researched for several applications in the field of biomaterials. Calcium phosphate ceramics stand out among the so-called bioceramics for their absence of local or systemic toxicity, their non-response to foreign bodies or inflammations, and their apparent ability to bond to the host tissue. Hydroxyapatite (HA) is one of the most important bioceramics because it is the main component of the mineral phase of bone. The aim of this work was to produce chitosan membranes coated with hydroxyapatite using the modified biomimetic method. Membranes were synthesized from a solution containing 2% of chitosan in acetic acid (weight/volume) via the solvent evaporation method. Specimens were immersed in a sodium silicate solution and then in a 1.5 SBF (simulated body fluid) solution. The crystallinity of the HA formed over the membranes was correlated to the use of the nucleation agent (the sodium silicate solution itself). Coated membranes were characterized by means of scanning electron microscopy - SEM, X-ray diffraction - XRD, and Fourier transform infrared spectroscopy - FTIR. The results indicate a homogeneous coating covering the entire surface of the membrane and the production of a semi-crystalline hydroxyapatite layer similar to the mineral phase of human bone. Foram estudadas as características de floculação da hidro-xiapatita, calcita e quartzo com poliacrilamidas aniônicas. Tanto as suspensões de hidroxiapatita como a calcita foram completamente floculadas enquanto que o quartzo foi refratário a floculação com esses polimeros. Entretanto, observou-se que a floculação da calcita foi impedida com a utilização de um poliacrilato de sódio, enquanto que a hidroxiapatita continuou a ser floculada pela poliacrilamida num intervalo de concentração de O a 1000 g/ton de poliacrilato. Estes resultados permitiram estabelecer condições para a floculação seletiva de hidroxiapatita em misturas sintéticas com quartzo e calcita. Nestes sistemas foram estudados diversos parâmetros operacionais, tais como tipo de separação, efeito do teor da alimentação, e influência da moagem. Onde a separação estagiada apresentou melhor resultado, uma moagem adicional no sistema não favoreceu a seletividade. Por outro lado, o teor de P205 no concentrado é proporcional ao teor presente na alimentação. A hidroxiapatita, em estudo, de alta pureza (de acordo com técnicas de análise química, espectrofotometria de infravermelho e difração de raios X) apresentou um parto de carga zero em pH 6,77 na presença de diversos eletrólitos. A aplicação do sistema de floculação seletiva a fraçoes ultrafinas industriais foi estudada em detalhe e comparada com processos de flotação e floco-flatação. 0s resultados obtidos mostraram que a floculação seletiva somente foi eficiente nos sistemas sintéticos. A flotação, embora o minério apresente uma granulometria muito fina, foi mais eficiente que a floculação seletiva e a floco-flotação nos sistemas reais, em termos de taxa de enriquecimento e percentual de rejeição de massa. Entretanto com a floco-flotação foi possivel obter altas recuperações(-90%) para os minérios de Araxá e Itataia. Para o minério de Tapira, um aumento do teor de P2O5 foi obtido com a flotação da fração seletivamente floculada. As taxas de enriquecimento obtidas foram baixas (entre 1,35 e 2,101 em função das associações entre a apatita do minério com óxido de ferro e argila, o que revelou a caracterização mineralógica. Conclue-se que a concentração de minerios fosfatados de baixo teor é uma operação onerosa, onde o fator predominante é a absorção seletiva de reagentes e atransferência não específica de material de ganga nos respectivos concentrados.

The fission-track method (FTM) in apatite was applied to 45 samples collected in the Serra da Mantiqueira (Mantiqueira mountain range), the Serra do Mar (Mar mountain range), regions next to these mountain ranges and the coastal region between Ubatuba and Santos in the State of São Paulo, Brazil, to study the thermochronology of the South American Platform in southeast Brazil and its influence on Santos and Campos basins. The data presented in this work complement the previously presented data on the same region (Tello Saenz et al., 2003. J. S. Am. Earth Sci. 15, 765-774) with 31 new samples analyzed. The weighted mean of the corrected ages from high Mantiqueira (around 1000 m), (121 +/- 6) Ma, coincides with the South Atlantic opening. The fact that its thermal history starts at a relatively low temperature (similar to 80 degrees C) suggests that the age of similar to 120 Ma would be the formation age of Serra da Mantiqueira due to a rapid pulse, in which tracks had no time to be retained at the closure temperature, that is similar to 120 degrees C. The Serra do Mar presents a more complicated thermal history, with several reactivations indicated by the changes in the slope of its cooling curve. The thermal histories obtained in the regions next to these mountain ranges are compatible with the results mentioned above. The Santos Basin has unconformities that agree with changes in the slope thermal histories of the studied region. (c) 2005 Elsevier Ltd. All rights reserved.

In this work a software developed in the Instituto de Fisica Gleb Wataghin, IFGW, UNICAMP, Campinas, SP, Brazil for obtaining thermal histories using apatite fission track analysis is presented. This software works in Microsoft-Windows environment. It will be freely disposable in the web site of the Departamento de Raios Cosmicos, IFGW, UNICAMP. Thermal histories obtained through this software are compared with those deduced using Monte Trax the software compatible with Apple Macintosh developed by Gallagher. (C) 2001 Elsevier B.V. Ltd. In this work, we present an approach for neutron fluence measurements based on natural thorium thin films and natural uranium-doped glasses calibrated through natural uranium thin films to be used for dating with the Fission-Track Method (FTM). This neutron dosimetry approach allows the employment of FTM even when dating is carried out using low neutron themalization facilities. Besides, it makes possible the determination of the Th/U ratio of the mineral to be dated. Durango apatite which is often employed in FTM as an age standard was analyzed. This apatite presented a fairly high Th/U ratio, 29.9 +/- 1.7. Th fissions were 18%, 12% and 10% of the total for irradiations where thermal to fast neutron flux ratios were 2.4, 4.4 and 5,2, respectively. These results show that Th fission must be considered for this apatite, when not well-thermalized irradiation facilities are used. The ratio between spontaneous and induced track length, L(S)/L(1), close to 0.89, indicates a certain amount of rejuvenation of the age of Durango apatite. Therefore, its apparent age should be corrected, the application of a technique based on track-length measurements produced a corrected age of 29.7 +/- 1.1 Ma, consistent with the independent reference age of this apatite (31.4 +/- 0.5 Ma). This result represents a support for viability of the neutron dosimetry approach studied in this work for FTM.(C) 2002 Elsevier B.V. B.V. All rights reserved.

An apatite fission track study of crystalline rocks underlying sedimentary basins in northeastern Brazil indicate that crustal blocks that occur on opposite sides of a geological fault experienced different thermal histories. Samples collected on the West block yielded corrected fission-track ages from 140 to 375 Ma, whereas samples collected on the East block yielded ages between 90 and 125 Ma. The thermal models suggest that each block experienced two cooling events separated by a heating event at different times. We concluded that the West block moved downward relative to the East block ca. 140 Ma ago, when sediments eroded from the East side were deposited on the West side. This process represents the early stage of sedimentary basin formation and the opening of the South Atlantic Ocean in the region. Downward and upward movements related to heating and cooling events of these crustal blocks at different periods until recent times are proposed. (c) 2005 Elsevier Ltd. All rights reserved.
