Background Very few articles have been written about the expression of kallikreins (KLK4 and KLK7) in oral cancers. Therefore, the purpose of this study was to examine and report on their prognostic potential. Methods Eighty archival blocks of primary oral cancers were sectioned and stained for KLK4 and KLK7 by immunohistochemistry. The percentage and the intensity of malignant keratinocyte staining were correlated with patient survival using Cox regression analysis. Results Both kallikreins were expressed strongly in the majority of tumor cells in 68 of 80 cases: these were mostly moderately or poorly differentiated neoplasms. Staining was particularly intense at the infiltrating front. Patients with intense staining had significantly shorter overall survival (p < .05). Conclusion This is the first observation on the patient survival influenced by kallikrein expression in oral carcinoma. The findings are consistent with those for carcinomas at other sites, in particular the prostate and ovary. KLK4 and/or KLK7 immunohistochemistry seems to have diagnostic and prognostic potential in this disease.

In his 2007 PESA keynote address, Paul Smeyers discussed the increasing regulation of child-rearing through government intervention and the generation of “experts,” citing particular examples from Europe where cases of childhood obesity and parental neglect have stirred public opinion and political debate. In his paper (this issue), Smeyers touches on a number of tensions before concluding that child rearing qualifies as a practice in which liberal governments should be reluctant to intervene. In response, I draw on recent experiences in Australia and argue that certain tragic events of late are the result of an ethical, moral and social vacuum in which these tensions coalesce. While I agree with Smeyers that governments should be reluctant to “intervene” in the private domain of the family, I argue that there is a difference between intervention and support. In concluding, I maintain that if certain Western liberal democracies did a more comprehensive job of supporting children and their families through active social investment in primary school education, then both families and schools would be better equipped to deal with the challenges they now face.

INTRODUCTION: Galectin family members have been demonstrated to be abnormally expressed in cancer at the protein and mRNA level. This study investigated the levels of galectin proteins and mRNA expression in a large cohort of patients with papillary thyroid carcinoma and matched lymph node metastases with particular emphasis on galectin-1 and galectin-3. METHODS: mRNA expression of galectin family members (1, 2, 3, 4, 7, 8, 9, 10 and 12) were analysed by real-time polymerase chain reaction in 65 papillary thyroid carcinomas, 30 matched lymph nodes with metastatic papillary thyroid carcinoma and 5 non-cancer thyroid tissues. Galectin-1 and 3 protein expression was determined by immunohistochemistry in these samples. RESULTS: Significant expression differences in all tested galectin family members (1, 2, 3, 4, 7, 8, 9, 10 and 12) were noted for mRNA in papillary thyroid carcinomas, with and without lymph node metastasis. Galectin-1 protein was more strongly expressed than galectin-3 protein in papillary thyroid carcinoma. Galectin-1 protein was found to be overexpressed in 32% of primary papillary thyroid carcinomas. A majority of lymph nodes with metastatic papillary thyroid carcinoma (53%) had significantly increased expression of galectin-1 protein, as did 47% of primaries with metastases. Galectin-1 mRNA levels were decreased in the vast majority (94%) of primary thyroid carcinomas that did not have metastases present. Galectin-3 protein levels were noted to be overexpressed in 15% of primary papillary thyroid carcinomas. In primary papillary thyroid carcinoma with lymph node metastases, 32% had over expression of galectin-3 protein. Overexpression of galectin-3 mRNA was noted in 58% of papillary thyroid carcinomas and 64% of lymph nodes bearing metastatic papillary thyroid carcinoma. Also, primary papillary thyroid carcinoma with lymph node metastases had significantly higher expression of galectin-3 mRNA compared to those without lymph node metastases. CONCLUSION: Galectin family members show altered expression at the mRNA level in papillary thyroid cancers. Overexpression of galectin-1 and 3 proteins were noted in papillary thyroid carcinoma with lymph node metastases. The results presented here demonstrated that galectin-1 and galectin-3 expression have important roles in clinical progression of papillary thyroid carcinoma.

The aims of the present study are to quantitatively analyze survivin expression, its clinicopathologic roles, and correlation with telomerase activity in a large cohort of patients with colorectal adenocarcinoma. Real-time polymerase chain reaction was used to quantitate expression level of survivin messenger RNA and human telomerase reverse transcriptase messenger RNA (telomerase activity) in 51 patients with colorectal adenocarcinomas. The findings were correlated with the clinicopathologic features of patients, which were prospectively collected into a computerized database. Survivin messenger RNA was expressed in all tumor samples. The level of expression in tumor tissues was increased in comparison with matched nontumor mucosa in the same patient (P = .01). The level of expression of survivin was significantly correlated with the level of human telomerase reverse transcriptase expression (P = .008) and size of the colorectal adenocarcinomas (P = .004). Survival of the patients with colorectal adenocarcinoma was associated with the TNM stages (P = .001) and not with the level of expression of survivin. Thus, survivin activity was altered in colorectal adenocarcinoma. The high prevalence of survivin expression and correlation with telomerase activity are important factors for consideration in gene targeting therapy for colorectal adenocarcinoma.

The venture, 23andMe Inc., raises a host of issues in respect of patent law, policy, and practice in respect of lifestyle genetics and personalised medicine. The company observes: ‘We recognize that the availability of personal genetic information raises important issues at the nexus of ethics, law, and public policy’. 23andMe Inc. has tested the boundaries of patent law, with its patent applications, which cut across information technology, medicine, and biotechnology. The company’s research raises fundamental issues about patentability, especially in light of the litigation in Bilski v. Kappos, Mayo Collaborative Services v. Prometheus Laboratories Inc. and Association for Molecular Pathology v. United States Patent and Trademark Office and Myriad Genetics Inc. There has been much debate and controversy over 23andMe Inc. filing patent applications – particularly in respect of its granted patent on ‘Polymorphisms associated with Parkinson’s Disease’. The direct-to-consumer marketing of genetic testing by 23andMe Inc. has also raised important questions of bioethics and human rights. It is queried whether the terms of service for 23andMe Inc. provide adequate recognition of the concepts of informed consent and benefit-sharing, especially in light of litigation in this area in the United States. Given the patent thickets surrounding genetic testing, the case study of 23andMe Inc. also highlights questions about patent infringement and patent exceptions. The future reform of patent law, policy, and practice needs to take into account new developments in lifestyle genetics and personalised medicine – as exemplified by 23andMe Inc.

In an exploration of intellectual property and fashion, this article examines the question of the intermediary liability of online auction-houses for counterfeiting. In the United States, the illustrious jewellery store, Tiffany & Co, brought a legal action against eBay Inc, alleging direct trademark infringement, contributory trademark infringement, false advertising, unfair competition and trademark dilution. The luxury store depicted the online auction-house as a pirate bazaar, a flea-market and a haven for counterfeiting. During epic litigation, eBay Inc successfully defended itself against these allegations in a United States District Court and the United States Court of Appeals for the Second Circuit. Tiffany & Co made a desperate, unsuccessful effort to appeal the matter to the Supreme Court of the United States. The matter featured a number of interventions from amicus curiae — Tiffany was supported by Coty, the Fashion Designer's Guild, and the International Anticounterfeiting Coalition, while eBay was defended by publicly-spirited civil society groups such as Electronic Frontier Foundation, Public Citizen, and Public Knowledge as well as Yahoo!, Google Inc, Amazon.com, and associations representing telecommunications carriers and internet service providers. The litigation in the United States can be counterpointed with the fusillade of legal action against eBay in the European Union. In contrast to Tiffany & Co, Louis Vuitton triumphed over eBay in the French courts — claiming its victory as vindication of the need to protect the commercial interests and cultural heritage of France. However, eBay has fared somewhat better in a dispute with L’Oréal in Great Britain and the European Court of Justice. It is argued that, in a time of flux and uncertainty, Australia should follow the position of the United States courts in Tiffany & Co v eBay Inc. The final part examines the ramifications of this litigation over online auction-houses for trade mark law reform and consumer rights; parallel disputes over intermediary liability and safe harbours in the field of copyright law and the Anti-Counterfeiting Trade Agreement 2010. The conclusion calls for a revision of trade mark law, animated by a respect for consumers’ rights and interests in the electronic marketplace.

Genital tract carriage of group B streptococcus (GBS) is prevalent among adult women; however, the dynamics of chronic GBS genital tract carriage, including how GBS persists in this immunologically active host niche long term, are not well defined. To our knowledge, in this study, we report the first animal model of chronic GBS genital tract colonization using female mice synchronized into estrus by delivery of 17β-estradiol prior to intravaginal challenge with wild-type GBS 874391. Cervicovaginal swabs, which were used to measure bacterial persistence, showed that GBS colonized the vaginal mucosa of mice at high numbers (106–107 CFU/swab) for at least 90 d. Cellular and histological analyses showed that chronic GBS colonization of the murine genital tract caused significant lymphocyte and PMN cell infiltrates, which were localized to the vaginal mucosal surface. Long-term colonization was independent of regular hormone cycling. Immunological analyses of 23 soluble proteins related to chemotaxis and inflammation showed that the host response to GBS in the genital tract comprised markers of innate immune activation including cytokines such as GM-CSF and TNF-α. A nonhemolytic isogenic mutant of GBS 874391, Δcyle9, was impaired for colonization and was associated with amplified local PMN responses. Induction of DNA neutrophil extracellular traps, which was observed in GBS-infected human PMNs in vitro in a hemolysin-dependent manner, appeared to be part of this response. Overall, this study defines key infection dynamics in a novel murine model of chronic GBS genital tract colonization and establishes previously unknown cellular and soluble defense responses to GBS in the female genital tract.

Tutkimuksen tavoitteena on hahmottaa 1800-luvulla tapahtunutta keskiluokan sosiaalista nousua pohjalaissyntyisen papin, opettajan, liikemiehen ja valtiopäiväedustajan Alfred Kihlmanin (1825-1904) elämänvaiheiden kautta. Kyseessä ei ole perinteinen elämäkerta, vaan työn pyrkimyksenä on mikrotason tarkan analyysin kautta päästä käsiksi laajempiin yhteiskunnallisiin muutosprosesseihin. Erityisen kiinnostuksen kohteena ovat erilaiset sosiaaliset verkostot sekä niiden merkitys Kihlmanin kaltaisen keskiluokkaisen yksilön sosiaaliselle nousulle. Tutkimus jakautuu kahteen pääosaan, joista ensimmäinen käsittelee heränneiden pappien verkostoja 1840-luvulla ja 1850-luvun alussa ja jälkimmäinen liikemiesverkostoja 1850-luvun jälkipuolelta 1870-luvun alkuun. Pääasiallisena lähdeaineistona on Alfred Kihlmanin laaja kirjeenvaihto. Alfred Kihlman liittyi kouluvuosinaan 1840-luvun alussa nuorison piirissä nopeasti levinneeseen herännäisyyteen. Nuoret heränneet papit muodostivat tiiviin sosiaalisen verkoston, joka perustui jaetulle uskonnolliselle ideologialle sekä pappisperheiden välisille avioliittositeille. Lisäksi verkostoa pidettiin yllä yhteisillä kokoontumisilla, vierailuilla, kirjeenvaihdolla sekä palvelusten vaihtamisella. Verkoston välityksellä saatiin tehokkaasti mm. informaatiota sekä rahalainoja. Herännäisyys nousi nuoren keskiluokkaisen papiston omaa yhteiskunnallista voimattomuuttaan kohtaan tuntemasta tyytymättömyydestä. Kun sivistyneistön mahdollisuudet menestyä perinteisillä foorumeilla olivat autonomian alkupuolella heikot, nuoret heränneet kielsivät uskonnollisessa ideologiassaan arvon maalliselta menestykseltä ja rakensivat oman verkostonsa, jonka sisällä he saattoivat toteuttaa pätemisentarvettaan. Yhteiskunnallisen tilanteen muututtua verkosto muuttui 1850-luvun alussa hyödystä rasitteeksi, minkä seurauksena heränneiden pappien rivit hajaantuivat. Irtauduttuaan herännäisyydestä Alfred Kihlman siirtyi opettajan uralle. Heränneeltä vaimoltaan perimänsä varallisuuden sekä nuoruus- ja herännäisvuosinaan solmimiensa ystävyyssuhteiden ansiosta hän ajatui 1850-luvun kuluessa yhä enemmän myös liike-elämään. Hänen merkittävimmäksi toimintakentäkseen muodostuivat Suomen Yhdyspankki sekä Tampereen Pellava- ja Rautateollisuus, jonka johtajana hän toimi vuodesta 1866 lähtien. 1860-luvulla liikemiesverkostot perustuivat suurelta osin sukulaisuuteen, mutta yhä enenevässä määrin myös ystävyys-, kollega- ja liikekumppanuussuhteisiin. Kun pankkilaitos oli vielä hyvin kehittymätön, verkostot olivat etenkin pääomien hankinnassa avainasemassa. Tiiviit lainaverkostot olivat huonojen suhdanteiden aikana toisaalta myös vaaraksi, sillä yksi suuri konkurssi saattoi vetää mukanaan joukoittain liikemiehiä. Verkostojen kautta voitiin välittää ja saada myös suosituksia sekä sisäpiiritietoa, jota ei ollut saatavissa muista lähteistä. Yhteiskunnan muuttuminen ja henkilösuhteet mahdollistivat Alfred Kihlmanin sosiaalisen nousun. Teollistuminen ja liike-elämän vapautuminen tarjosivat uusia mahdollisuuksia keskiluokan yritteliäille yksilöille. Ylempien yhteiskuntaluokkien välisten säätyjen välisten raja-aitojen tasaantuminen tekivät 1850-luvun puolivälissä mahdolliseksi myös papin osallistumisen liike-elämään. The Catherwood Library, which serves Cornell's School of Industrial and Labor Relations, houses a complete set of the studies either published by or produced under the auspices of the Work in America Institute, Inc. These volumes were donated to the Catherwood Library by the Institute's Board of Directors through the initiative of Jay W. Waks, ILR '68, who succeeded Mr. Rosow as Chair of the Institute and who, for many years, sat on the Institute's Executive Committee with Mr. Rosow and Thomas R. Donahue, former Secretary-Treasurer and Interim President of the AFL-CIO. Each volume bears a bookplate with this message: "This volume was donated by the Work in America Institute, Inc. in honor of its founder, Jerome M. Rosow, 1919-2002."
