La consecución de tolerancia aloespecífica es de mucha relevancia en trasplante. Las células dendríticas (DC) son las principales responsables de la inducción de la respuesta inmune frente a las moléculas de histocompatibilidad (MHC) del donante, provocando el rechazo del injerto. Sin embargo las DC son también responsables de la inducción de tolerancia. Diversos modelos animales de alotrasplante han mostrado la tolerización del injerto mediante DC diferenciadas in vitro en condiciones tolerogénicas (tDC). En humanos, las fuentes de aloantígenos potencialmente utilizables en terapia son, entre otras, los cuerpos apoptóticos y los exosomas. Éstos expresan antígenos MHC de forma abundante y su composición es relativamente uniforme, lo que supone una ventaja frente a otras fuentes. En este proyecto, se ha evaluado la obtención de exosomas secretados por una línea de linfocitos T y por células dendríticas derivadas de médula ósea. Se ha caracterizado la captura de exosomas derivados de linfocitos T por células dendríticas humanas derivadas de sangre periférica y su presentación a linfocitos T autólogos. Por otra parte, se ha comenzado a desarrollar los experimentos para estudiar la inducción de tolerancia en un modelo de trasplante renal en rata. Se han generado células dendríticas tolerógenicas derivadas de médula ósea (tolDC), en presencia de dexametasona. Las tolDC expresan menos moléculas de histocompatibilidad y de coestimulación e inducen una menor proliferación en reacciones mixtas leucocitaras, comparadas con las células dendríticas maduras. Por último, se han caracterizado los exosomas de plasma humano con el fin de estudiar su posible uso como aloantígenos. El análisis proteómico revela la presencia de proteínas relacionadas con el sistema inmune, la coagulación, la señalización celular y moléculas implicadas en el transporte y metabolismo de nutrientes. El estudio de la captura por diferentes líneas celulares sugiere que deben existir mecanismos específicos para su internalización.

A adrenoleucodistrofia ligada ao cromossomo X (X-ALD) é uma doença peroxissomal bioquimicamente caracterizada pelo acúmulo de ácidos graxos de cadeia muito longa (“Very Long Chain Fatty Acids” - VLCFA), principalmente os ácidos hexacosanóico (C26:0) e tetracosanóico (C24:0), em diferentes tecidos e fluidos orgânicos. A doença é clinicamente caracterizada por progressiva desmielinização central e periférica e insuficiência adrenal. Os mecanismos exatos do dano cerebral na X-ALD são pouco conhecidos. O tratamento usual para X-ALD com a mistura gliceroltrioleato/gliceroltrierucato na proporção 4:1, conhecido como óleo de Lorenzo (OL), em combinação com uma dieta de restrição dos VLCFA normaliza os níveis de VLCFA, mas em pacientes sintomáticos os sintomas neurológicos persistem ou progridem. Os radicais livres parecem estar envolvidos em um grande número de enfermidades no ser humano, tais como nas doenças neurodegenerativas (como doença de Parkinson, doença de Alzheimer e esclerose múltipla), nas doenças crônico-inflamatórias, nas doenças vasculares e no câncer. Considerando que a geração de radicais livres está envolvida em várias desordens neurodegenerativas, no presente estudo foram avaliados vários parâmetros de estresse oxidativo em plasma, eritrócitos e fibroblastos de pacientes sintomáticos com X-ALD. Também foi avaliado o efeito do tratamento com OL sobre diferentes parâmetros de estresse oxidativo em plasma e em eritrócitos de pacientes sintomáticos e assintomáticos com X-ALD tratados e não tratados com OL. Considerando que não é possível estudar estresse oxidativo em córtex cerebral de pacientes com X-ALD, foi avaliado o efeito in vitro da mistura VI ácido oleico (C18:1)/ácido erúcico (C22:1) sobre diversos parâmetros de estresse oxidativo em plasma e em eritrócitos humanos normais e em córtex cerebral de ratos adicionados dos ácidos hexacosanóico(C26:0) e tetracosanóico (C24:0). Foi verificado um aumento significativo da quimiluminescência e das espécies reativas do ácido tiobarbitúrico (TBA-RS), refletindo uma indução da peroxidação lipídica, bem como uma diminuição da reatividade antioxidante total (TAR) medida em plasma de pacientes sintomáticos, indicando uma capacidade deficiente em rapidamente combater um aumento das espécies reativas. Também foi observado um aumento da atividade da glutationa peroxidase (GPx) em eritrócitos e das atividades da catalase (CAT) e da superóxido dismutase (SOD) em fibroblastos dos pacientes sintomáticos estudados. Estes dados sugerem que o estresse oxidativo pode estar envolvido na fisiopatologia da X-ALD. Verificamos também que o OL não reverteu o aumento do TBA-RS no plasma de indivíduos X-ALD. A determinação de TAR não apresentou alterações no plasma de pacientes X-ALD sintomáticos e assintomáticos antes e após o tratamento com OL. A atividade das enzimas antioxidantes CAT, GPx e SOD não se mostrou alterada em eritrócitos destes pacientes tratados ou não com OL. Nos experimentos in vitro, foi constatado um aumento significativo da quimiluminescência e de TBA-RS em plasma humano e em córtex cerebral de ratos adicionados dos ácidos C26:0C24:0, porém a mistura C18:1/C22:1 não reverteu este efeito. A medida de TAR não se mostrou alterada em córtex cerebral de ratos adicionado de C26:0C24:0, bem como não foi modificada pela mistura C18:1/C22:1. Por outro lado, a adição de C26:0C24:0 em plasma humano diminuiu a medida de TAR, porém a mistura C18:1/C22:1 não alterou esse efeito. As atividades das enzimas antioxidantes CAT e GPx não foram alteradas em eritrócitos humanos e em córtex cerebral de ratos pela adição de C26:0C24:0 , nem pela mistura C18:1/C22:1. O mesmo foi observado na medida de SOD em eritrócitos humanos adicionados de C26:0C24:0. A medida da atividade da SOD em córtex cerebral de ratos apresentou um aumento significativo induzido por C26:0C24:0 , porém a presença da mistura C18:1/C22:1 não modificou este efeito. Estes resultados sugerem fortemente que o estresse oxidativo possa estar envolvido na fisiopatologia da X-ALD e que o tratamento com óleo de Lorenzo não modifica estes parâmetros de estresse oxidativo. Assim, novas estratégias terapêuticas deveriam ser investigadas para pacientes de X-ALD.

Since the first description of sulfated polysaccharides from seaweeds, the biological activities of these compounds have been evaluated under different aspects and experimental procedures. Among the broad biological activities presented by seaweed polysaccharides, anticoagulant action appears as a promising function. In this present study we have obtained sulfated polysaccharides from the green seaweed Codium isthmocladium by proteolytic digestion, followed by separation into five fractions (0.3, 0.5, 0.7, 0.9 and 1.2) by sequential acetone precipitation. The chemical analyses have demonstrated that all fractions are composed mainly by sulfated polysaccharides. The anticoagulant activity of these fractions was determined by activated partial thromboplastin time (aPTT) and prothrombin time test (PT) using citrate normal human plasma. None fraction has shown anticoagulant activity by PT test. Furthermore, all of them have shown anticoagulant activity by aPTT test. These results indicated that the molecular targets of these sulfated polysaccharides are mainly in the intrinsic via of the coagulation cascade. Agarose gel electrophoresis in 1,3-diaminopropane acetate buffer, pH 9.0, stained with 0.1% toluidine blue showed the presence of two or three bands in several fractions while the fraction 0.9 showed a single spot. By anion exchange chromatography, the acid polysaccharides from the 0.9 acetone fraction were separated into two new fractions eluted respectively with 2.0 and 3.0 M NaCl. These compounds showed a molecular weight of 6.4 and 7.4 kDa respectively. Chemical analyses and infrared spectroscopy showed that Gal 1 and Gal 2 are sulfated homogalactans and differ one from the other in degree and localization of sulfate groups. aPPT test demonstrated that fractions 2,0 and 3,0M (Gal1 and Gal 2, respectively) have anticoagulant activity. This is the first time that anticoagulant sulfated homogalatans have been isolated from green algae. To prolong the coagulation time to double the baseline value in the aPTT, the required amount of sulfated galactan 1 (6,3mg) was similar to low molecular heparin Clexane®, whereas only 0,7mg of sulfated galactan 2 was needed to obtain the same effect. Sulfated galactan 2 in high doses (250mg) induces platelet aggregation. These results suggest that these galactans from C. isthmocladum have a potential application as an anticoagulant drug

Fucans are a family of sulfated homo and teropolysaccharides respectively, composed mainly of a- (1®2) and a- (1®3) linked by L-fucose residues. Properties such as the ability to act as an anti-contraceptive, to reduce cholesterol levels, and to act as an anti-tumor agent are much related. We have focused our attention on the anticoagulant properties, platelet aggregation, hemorrhagic activity and complement system in vitro of commercial fucoidan (F) and their purified fractions (F1, F2 and F3) from Fucus vesiculosus obtained from fractionation of the fucoidan with different concentrations of acetone 1, 2 and 3v. These compounds were chemically characterized and the fucoidan (F) was modified by desulfation. The anticoagulant activity of the compounds was assessment by activated partial thromboplastin time (APTT) and prothrombine time assay (PT) using citrated normal human plasma. The results of APPT test showed that F, F1 and F2 have high anticoagulants activities 240.0 s (5 µg). The F3 showed 73.7 s in the same concentrations. The results obtained with PT test to F, F1, F2 and F3 were 81.5 s, 120.0 s, 57.1 and 32.5 s respectively with 50 µg. The dessulfated polymer showed a decrease in the anticoagulant activity in these two tests. Platelet aggregation assay was measured turbidimetrically with platelet aggregometer by method of Born. The aggregation platelet with F and fractions F1, F2 and F3 exhibited a two-phase answer in the concentration of 5 mg/mL with maximum aggregation of 76.36 ± 10.3% ; 69.54 ± 9.40%; 75.94 ± 9.01%; 51.13 ± 9.59% respectively. However, was observed a hipoaggregate profile F (15.17 ± 5.2%), F1 (7.40 ± 3.04 %), F2 (19.1 ± 5.41%) and F3 (5.09 ± 3.02%) at 0.1 mg/mL. The hemorrhagic activity assay was carried in Wistar rats and showed that these compounds have low hemorrhagic effect when compared to heparin. The complement system ( alternative pathway was made using non-sensibilized rabbit red blood cells The results of complement system essay showed that F , F2 and F3 have action inhibitory in relation to the group control 0.544, 0.697, 0.622 and 0.958 respectively The results showed that these compounds have action on this system. Interaction of the polisaccharides with proteins C3 and C4 showed that the fraction F1 stimulated the activity assay hemolytic using red blood cells

The bone repair process is controlled by complex molecular mechanisms that involve systemic and local factors. Fibrin glue is derived from human plasma and mimics the final pathway of coagulation network. Tranexamic acid inhibits fibrinolysis and prevents or decreases the formation of degradation products of fibrin and fibrinogen. The purpose of this study was to evaluate histologically in rats the effect of tranexamic acid associated with the fibrin glue on bone healing. The experiment used 60 (n = 5) male rats in: GI: Control, GII: fibrin glue, GIII tranexamic acid and GIV /fibrin glue/tranexamic acid. Bone defect (2.5mm diameter) was created in right tibia. The animals were euthanized at 7,14 and 30 days postoperatively, and the pieces were processed with hematoxylin and eosin. The results showed at 7 days post-operative surgical cavity filled with dense connective tissue rich in fibroblasts, permeated by delicate neoformed bone trabeculae in percentage of 70-80% for GI, GII and GIII and GIV to 94.8%. At 14 days post-operative newly formed bone was found between 75-85% for GI, GII and GIII and percentage above 95% for GIV. At 30 days postoperative GI and GIV showed 95-100% of mature bone tissue; GII and GIII in percentage close to 80-90%. Based on the results and methodology is concluded that fibrin / tranexamic acid glue association has positive action on bone repair.

CHAPTER II: Snake venoms are a complex mixture of organic and inorganic compounds, proteins and peptides such as aminotransferases, acetylcholinesterase, hyaluronidases, L-amino acid oxidase, phospholipase A2, metalloproteases, serine proteases, lectins, disintegrins, and others. Phospholipase A2 directly or indirectly influence the pathophysiological effect on envenomation, as well as their participation in the digestion of the prey. They have several other activities such as hemolytic indirect action, cardiotoxicity, aggregating of platelets, anticoagulant, edema, myotoxic and inflammatory activities. In this work, we describe the functional characterization of BaltMTx, a PLA2 from Bothrops alternatus that inhibits platelet aggregation and present bactericidal effect. The purification of BaltMTx was carried out through three chromatographic steps (ion-exchange on a DEAE-Sephacel column, followed by hydrophobic chromatography on Phenyl–Sepharose and affinity chromatography on HiTrap™ Heparin HP). The protein was purified to homogeneity as judged by its migration profile in SDS–PAGE stained with coomassie blue, and showed a molecular mass of about 15 kDa under reducing conditions and approximately 25 kDa in non-reducing conditions. BaltMTx showed a rather specific inhibitory effect on platelet aggregation induced by epinephrine in human platelet-rich plasma in a dose-dependent manner, whereas it had little or no effect on platelet aggregation induced by collagen or adenosine diphosphate. BaltMTx also showed antibacterial activity against Staphylococcus aureus and Escherichia coli. High concentrations of BatlMTx stimulated the proliferation of Leishmania (Leishmania) infantum and Leishmania (Viania) braziliensis. BaltMTx induced production of inflammatory mediators such as IL-10, IL-12, TNF-α and NO. BaltMTx could be of medical interest as a new tool for the development of novel therapeutic agents for the prevention and treatment of thrombotic disorders as well as bactericidal agent.

The plasma represents a average of the information referring biochemists to the physiology of the organism as a whole, therefore it indirectly or directly interacts with all tissues of the body. In such a way the plasma can be considered as a metabolic “soup”. Using the Nuclear Magnetic Resonance Spectroscopy sanguineous plasma spectra had been generated and using deconvolution techniques it was possible to know the contribution of the albumin for the formation of the spectra of the sanguineous plasma

Para el diagnóstico de la tuberculosis se realizó un estudio clínico y de laboratorio con el objeto de determinar la sensibilidad y especificidad en la detección de anticuerpos en IGG. Se clasificaron en tres grupos las personas : el primero, 50 pacientes con baciloscopía positiva en esputo, el segundo con 50 aparentemente sanas; y, un tercero con 50 pacientes, clinicamente sugestivos de la enfermedad. El presente estudio se llevó a cabo en los hospitales José Carrasco Arteaga del Instituto Ecuatoriano de Seguridad Social y Vicente Corral Moscoso de ministerio de Salud Pública, en la ciudad de Cuenca. Se aplicó para la búsqueda de anticuerpos en IGG, el método inmunológico Enzimoinmunoanálisis [EIA], utilizado el Immuzyme Myco que detecta anticuerpos para micobacterias en general y el Immuzyme TB Complex, un antígeno recombinante de gran especificidad para mycobacterium tuberculoso [38KDa]. En los pacientes baciloscópicos positivos, la sensibilidad al Immuzyme Myco fue del 70 por ciento con una especificidad del 74 por ciento y la prueba positiva en el 42 por ciento de aquellos que tubieron sintomatología sugestiva de tuberculosis. En cuanto al Immuzyme TB Complex se obtuvo una sensibilidad del 26 por ciento y una especificidad del 94 por ciento en el grupo de baciloscópicos positivos y en los clinicamente sugestivos fue positiva en el 8 por ciento de los casos, [4 pacientes]. Al igual que en la bibliografía consultada, la sensibilidad es baja, por lo que el test no es recomendable en población abierta. La especificidad es alta en ambas pruebas, siendo más relevante el Immuzyme Myco, lo que permitiría hacer confirmación diagnóstica en personas seleccionadas, altamente sospechosas de la enfermedad y que no tiene ni baciloscopía ni cultivo positivo.
