This paper investigates whether and to what extent a wide range of actors in the UK are adapting to climate change, and whether this is evidence of a social transition. We document evidence of over 300 examples of early adopters of adaptation practice to climate change in the UK. These examples span a range of activities from small adjustments (or coping) to building adaptive capacity, implementing actions and creating deeper systemic change in public and private organisations in a range of sectors. We find that adaptation in the UK has been dominated by government initiatives and has principally occurred in the form of research into climate change impacts. These actions within government stimulate a further set of actions at other scales in public agencies, regulatory agencies and regional government (or in the devolved administrations), though with little real evidence of climate change adaptation initiatives trickling down to local government level. The water supply and flood defence sectors, requiring significant investment in large scale infrastructure such as reservoirs and coastal defences, have invested more heavily in identifying potential impacts and adaptations. Economic sectors that are not dependent on large scale infrastructure appear to be investing far less effort and resources in preparing for climate change. We conclude that while the government-driven top-down targeted adaptation approach has generated anticipatory action at low cost, it may also have created enough niche activities to allow for diffusion of new adaptation practices in response to real or perceived climate change. These results have significant implications for how climate policy can be developed to support autonomous adaptors in the UK and other countries.

Fluorescence amplified fragment length polymorphism (fAFLP) was used to assess the genetic relatedness of 40 Staphylococcus aureus strains isolated from human and animal skin samples in seven dairy farms with manual milking. S. aureus was isolated from 11 out of 30 (36%) human skin samples and from 29 out of 100 (29%) teat skin samples from apparently healthy cows. Genomic DNA from each isolate was double-digested with EcoRI and MseI and complementary oligonucleotide adaptors were ligated to the restriction fragments. Pre-selective and selective, amplification reactions were performed, the amplified fragments were separated by electrophoresis in an ABI377 sequencer and analysed using GeneScan 3.1 and Genotyper 2.5. Three single isolates (a-c), a predominant cluster with 35 isolates (d) and another cluster with two isolates (e) were identified. Both clusters d and e included human and animal isolates genetically related, because the profiles had 90-100% homology. Since no cluster was comprised uniquely of human or animal isolates and given the close genetic relatedness among human and animal samples in the farms, the present findings support the. hypothesis that dairy workers can spread S. aureus through manual milking. The present study aims at analyzing how dark humour as a cinematic genre travels cross-culturally through a specific mode of audiovisual translation, i.e. dubbing. In particular, it takes into consideration the processes involved in dubbing humour from English into Italian as observed in the English- and Italian-language versions of ten British and American dark comedies from the 1940s to the 2000s. In an attempt to identify some of the main mechanisms of the dark humour genre, the humorous content of the films was analyzed in terms of the elements on which specific scenes are based, mainly the non-verbal and verbal components. In the cases in which verbal elements were involved, i.e. the examples of verbally expressed humour, the analysis was concerned with whether they were adapted into Italian and to what effect. Quantification of the different kinds of dark humour revealed that in the sample of dark comedies verbal dark humour had a higher frequency (85.3%) than non-verbal dark humour (14.7%), which partially disconfirmed the first part of the research hypothesis. However, the significance of contextual elements in the conveying of dark humour, both in the form of Nsp VEH (54.31%) and V-V (V+VE) (21.68%), provided support for the hypothesis that, even when expressed verbally, dark humour is more closely linked to context-based rather than purely linguistic humour (4.9%). The second part of the analysis was concerned with an investigation of the strategies adopted for the translation of verbal dark humour elements from the SL (English) into the TL (Italian) through the filter of dubbing. Four translational strategies were identified as far as the rendering of verbal dark humour is concerned: i) complete omission; ii) weakening; iii) close rendering; and iv) increased effect. Complete omission was found to be the most common among these strategies, with 80.9% of dark humour examples being transposed in a way that kept the ST’s function substantially intact. Weakening of darkly humorous lines was applied in 12% of cases, whereas increased effect accounted for 4.6% and complete omission for 2.5%. The fact that for most examples of Nsp VEH (84.9%) and V-AC (V+VE) (91.4%) a close rendering effect was observed and that 12 out of 21 examples of V-AC (PL) (a combined 57%) were either omitted or weakened seemed to confirm, on the one hand, the complexity of the translation process required by cases of V-AC (PL) and V-AC (CS). On the other hand, as suggested in the second part of the research hypothesis, the data might be interpreted as indicating that lesser effort on the translator/adaptor’s part is involved in the adaptation of V-AC (Nsp VEH) and V-V (V+VE). The issue of the possible censorial intervention undergone by examples of verbal dark humour in the sample still remains unclear.

Akt (also called PKB) is a 63 kDa serine/threonine kinase involved in promotion of cell survival, proliferation a nd metabolic responses downstream the phosphoinositide-3-kinase (PI 3-kinase) signaling pathway. In resting cells, Akt is a predominantly cytosolic enzyme; however generation of PI 3-kinase lipid products recruits Akt to the plasma membrane, resulting in a conformational change which confers full enzymatic activity through the phosphorylation of the membrane-bound protein at two residues, Thr308, and Ser473. Activated Akt redistributes to cytoplasm and nucleus, where phosphorylation of specific substrates occurs. Both the presence and the activity of Akt in the nucleus have been described. An interesting mechanism that mediates nuclear translocation of Akt has been described in human mature T-cell leukemia: the product of TCL1 gene, Tcl1, interacts with the PH domain of phosphorylated Akt, thus driving Akt to the nucleus. In this context, Tcl1 may act as a direct transporter of Akt or may contribute to the formation of a complex that promotes the transport of active Akt to the nucleus, where it can phosphorylate nuclear substrates. A well described nuclear substrate if Foxo. IGF-1 triggers phosphorylation of Foxo by Akt inside the nucleus, where phospho-Foxo associates to 14.3.3 proteins that, in turn, promote its export to the cytoplasm where it is sequestered. Remarkably, Foxo phosphorylation by Akt has been shown to be a crucial event in Akt-dependent myogenesis. However, most Akt nuclear substrates have so far remained elusive, as well as nuclear Akt functions. This lack of information prompted us to undertake a search of substrates of Akt in the nucleus, by the combined use of 2D-separation/mass spectrometry and anti-Akt-phosphosubstrate antibody. This study presents evidence of A-type lamins as novel nuclear substrates of Akt. Lamins are type V intermediate filaments proteins found in the nucleus of higher eukaryotes where, together with lamin-binding proteins, they form the lamina at the nuclear envelope, providing mechanical stability for the nuclear membrane. By coimmunoprecipitation, it is demonstrated here that endogenous lamin A and Akt interact, and that A-type lamins are phosphorylated by Akt both in vitro and in vivo. Moreover, by phosphoaminoacid analysis and mutagenesis, it is further demonstrated that Akt phosphorylates lamin A at Ser404, and, more importantly, that while lamin A/C phosphorylation is stable throughout the cell cycle, phosphorylation of the precursor prelamin A becomes detectable as cells enter the G2 phase, picking at G2/M. This study also shows that lamin phosphorylation by Akt creates a binding site for 14.3.3 adaptors which, in turn, promote prelamin A degradation. While this mechanism is in agreement with a general role of Akt in the regulation of a subset of its substrates, opposite to what has been described, degradation is not mediated through a ubiquitination and proteasomal mechanism but through a lysosomal pathway, as indicated by the reverting action of the lysosomal inhibitor cloroquine. Phosphorylation is a key event in the mitotic breakdown of the nuclear lamina. However, the kinases and the precise sites of phosphorylation are scarcely known. Therefore, these results represent an important breakthrough in this very significant but understudied area. The phosphorylation of the precursor protein prelamin A and its subsequent degradation at G2/M, when both the nuclear envelop and the nuclear lamina disassemble, can be view as part of a mechanism to dispose off the precursor that is not needed in this precise context. The recently reported finding that patients affected by Emery-Dreifuss muscular dystrophy carry a mutation at Arg 401, in the Akt phosphorylation motif, open new perspective that warrant further investigation in this very important field.

Despite over 30 years of research, the molecular mechanisms of nonsense-mediated mRNA decay (NMD) are still not well understood. NMD appears to exist in most eukaryotes and is intensively studied in S. cerevisiae, C. elegans, D. melanogaster and in mammalian cells. Current evidence suggests that the core of NMD – involving UPF1, UPF2 and UPF3 – is evolutionarily conserved, but that different species may have evolved slightly different ways to identify target mRNAs for NMD and to degrade them. Our lab has shown that the exon junction complex (EJC) is not absolutely required for NMD in human cells (Bühler et al., NSMB 2006) and that it is neither restricted to CBP80-bound mRNAs as classical models claim (Rufener & Mühlemann, NSMB 2013). Together with the finding that long 3’ UTRs often are an NMD-inducing feature (Eberle et al, PLoS Biol 2008; Yepiskoposyan et al., RNA 2011), our data is consistent with much of the data from other species and hence has led to a “unified” working model for NMD (Stalder & Mühlemann, Trends Cell Biol 2008; Schweingruber et al., Biochim Biophys Acta 2013). Our recent iCLIP experiments with endogenous UPF1 indicate that UPF1 binds mRNAs indiscriminately with respect to being an NMD target or not before they engage with ribosomes (Zünd et al., NSMB 2013). After onset of translation, UPF1 is cleared from the coding region but remains bound to the 3’ UTR of mRNAs. Why this 3’ UTR-associated in some cases induces NMD and in others not is currently being investigated and not yet understood. Following assembly of a phospho-UPF1-containing NMD complex, decay adaptors (SMG5, SMG7, PNRC2) and/or the endonuclease SMG6 are recruited. While the latter cleaves the mRNA in the vicinity of the termination codon, the former proteins induce deadenylation, decapping and exonucleolytic degradation of the mRNA. In my talk, I will give an overview about the latest developments in NMD – with a focus on our own work – and try to integrate the bits and pieces into a somewhat coherent working model.

Las transiciones en guía de onda son las estructuras utilizadas en microondas para transferir y adaptar la señal que viaja en un determinado sistema de transmisión (por ejemplo, un cable coaxial) a otro sistema de transmisión o a un sistema radiante (por ejemplo, una antena de bocina). Los dos sistemas de transmisión entre los que la transición adapta la señal pueden ser distintos o del mismo tipo pero con alguna de sus dimensiones diferente. Existen diferentes transiciones de guía de onda que dependiendo de su utilidad son diseñadas y construidas con diferentes secciones: circular, rectangular, elíptica o incluso combinaciones de éstas. No es necesario que la sección de la guía presente una forma geométrica conocida pero los estándares que se van a seguir hacen referencia en concreto a secciones rectangulares y circulares. En el trabajo que aquí se desarrolla se pretende optimizar por medio de simulaciones paramétricas una transición entre cable coaxial con conector tipo K y una guía de onda de sección circular que sigue el estándar presentado por Flann, Millitech y Antarfs para la banda WR34. La transición que va a ser objeto de este estudio se denomina transición tapered o transición conformada. Este tipo de transiciones se caracterizan por variar una de sus dimensiones progresivamente hasta llegar al tamaño definido en el estándar correspondiente. La manera de realizar la optimización de la transición se basará en el estudio del parámetro S11 que presente la estructura a lo largo de la banda de trabajo. Ya que se sigue el estándar WR34 la banda de trabajo que éste comprende va de 21,7 a 33 GHz. Se pretende conseguir que la respuesta del parámetro S11 se encuentre por debajo de -20 dB en la banda de WR34 como resultado del diseño para poder contar de esta manera con una buena adaptación. Finalmente se propondrá un criterio a seguir para optimizar este tipo de transiciones siguiendo el objetivo de mejor adaptación teniendo en cuenta el impacto de cada tramo sobre el rango de frecuencias en el que influye y se presentarán las características finales que presenta la transición bajo estudio. En este documento se introduce de manera breve la utilidad de los transformadores de impedancias lambda cuartos en líneas de transmisión, el estado del arte de las diferentes técnicas para su diseño, y la propuesta de diseño y caracterización objeto del presente trabajo. Posteriormente, se presenta el caso de estudio para el diseño de la transición para ser integrada a una bocina de choke. Luego, se introduce el marco teórico del trabajo presentando algunos ejemplos ilustrativos de tramos de guía de onda rectangular y guía de onda circular, introduciendo adaptadores de λ/4 en simulaciones. A continuación, se explica la implementación del modelo bajo estudio en CST (Computer Simulation Technology) Studio Suite. Finalmente se presenta la discusión de los resultados obtenidos, las conclusiones y líneas futuras de trabajo. ABSTRACT. Waveguide transitions are structures used in microwave in order to transfer and adapt the signal that travels from a certain transmission system (e.g. coaxial cable) to other transmission system or to a radiant system (e.g. horn antenna). Both transmission systems between which the transition adapts the signal can be different or from the same type, but with differences in some of their dimensions. There are different waveguide transitions that, depending on their utility, are designed and constructed in different sections: circular, rectangular, elliptic or combinations of the former. The section of the guide does not have to have a known geometric shape, although the standards to be followed in this thesis apply to rectangular and circular sections. In the work here presented, we aim to optimize by means of parametric simulations, a transition between a coaxial cable with a K-type connector and a waveguide with circular section following the standard presented by Flann, Millitech y Antarfs for the band WR34. The transition studied is called tapered transition, which is characterized by the progressive variation of its dimensions, until reaching the defined size of the corresponding standard. The way of optimizing the transition will be based in the study of the parameter S11 presented by the structure along the bandwidth. Since the standard used is the WR34, the bandwidth can be defined from 21.7 up to 33 GHz. It is aimed that the response of the S11 parameter be lower equal than -20dB in the frequency band under study according to the design in order to be well matched. Finally, a criterion to follow is proposed in order to optimize these transitions type, following the better-match objective. That will be done taking into account the impact of each section on the frequency range in which influences and the final characteristics of the studied transition will be presented. In this document, it is briefly introduced the utility of quarter-wave impedance transformers in transmission lines, the state-of-art of the different techniques for their design, and the proposal of design and characterization aimed with this work. Afterwards, the study case for the design of the transition to be integrated with a choke horn antenna will be presented. Later, the theoretical frame work is introduced, giving some illustrative examples of rectangular and circular waveguide sections, and also introducing λ/4 adaptors in the simulations. Subsequently, the implementation of the model under study in CST (Computer Simulation Technology) Studio Suite will be explained. Finally, the discussion of the results, conclusion and future lines of the work are presented.

βarrestins mediate the desensitization of the β2-adrenergic receptor (β2AR) and many other G protein-coupled receptors (GPCRs). Additionally, βarrestins initiate the endocytosis of these receptors via clathrin coated-pits and interact directly with clathrin. Consequently, it has been proposed that βarrestins serve as clathrin adaptors for the GPCR family by linking these receptors to clathrin lattices. AP-2, the heterotetrameric clathrin adaptor protein, has been demonstrated to mediate the internalization of many types of plasma membrane proteins other than GPCRs. AP-2 interacts with the clathrin heavy chain and cytoplasmic domains of receptors such as those for epidermal growth factor and transferrin. In the present study we demonstrate the formation of an agonist-induced multimeric complex containing a GPCR, βarrestin 2, and the β2-adaptin subunit of AP-2. β2-Adaptin binds βarrestin 2 in a yeast two-hybrid assay and coimmunoprecipitates with βarrestins and β2AR in an agonist-dependent manner in HEK-293 cells. Moreover, β2-adaptin translocates from the cytosol to the plasma membrane in response to the β2AR agonist isoproterenol and colocalizes with β2AR in clathrin-coated pits. Finally, expression of βarrestin 2 minigene constructs containing the β2-adaptin interacting region inhibits β2AR endocytosis. These findings point to a role for AP-2 in GPCR endocytosis, and they suggest that AP-2 functions as a clathrin adaptor for the endocytosis of diverse classes of membrane receptors.

Analysis of the genetic changes in human tumors is often problematical because of the presence of normal stroma and the limited availability of pure tumor DNA. However, large amounts of highly reproducible “representations” of tumor and normal genomes can be made by PCR from nanogram amounts of restriction endonuclease cleaved DNA that has been ligated to oligonucleotide adaptors. We show here that representations are useful for many types of genetic analyses, including measuring relative gene copy number, loss of heterozygosity, and comparative genomic hybridization. Representations may be prepared even from sorted nuclei from fixed and archived tumor biopsies.

The trans-Golgi network is the major sorting compartment of the secretory pathway for protein, lipid and membrane traffic. There is a constant flow of membrane and cargo to and from this compartment. Evidence is emerging that the trans-Golgi network has multiple biochemically and functionally distinct subdomains, each of which contributes to the combined sorting and transport requirements of this dynamic compartment. The recruitment of distinct arrays of protein complexes to trans-Golgi network membranes is likely to produce the diversity of structure and biochemistry observed amongst subdomains that serve to generate different carriers or maintain resident trans-Golgi network components. This review discusses how these subdomains may be formed and examines the molecular players involved, including G proteins, clathrin adaptors and golgin tethers. Diversity within these protein families is highlighted and shown to be critical for the functionality of the trans-Golgi network, as a mediator of protein sorting and membrane transport, and for the maintenance of Golgi structure.

Nanopore-based sequencer will open the path to the fourth-generation DNA sequencing technology. The main differences between this technique and the previous ones are: DNA molecule that will be sequenced does not need a previous amplification step, is not necessary any type of specific label both molecular adaptors, and it has been abolished enzymatic process in the nucleotide sequence identification event. These differences have as result a more economic method since don’t spend the necessary reagents for the previous techniques, furthermore it lets to sequence samples with a low DNA concentration. This technique is based in the use of a membrane with a biologic nanopore inserted in it whereby the molecule to analyze (analyte) it made to pass, this membrane is placed between two reservoirs containing ions, when an external volatage is applied in both sides this lead to an ion current through the nanopore. When an analyte cross the nanopore the ion current is modified, that modification in the amplitude and duration of ion current determine the physical and chemical properties of that analyte. By means of subsequent statistical analyzes it can be determined to what sequence own this ion current blockade patterns. More used nanopores are the biologic ones, although they are working to develop synthetic nanopores. The main biologic nanopores are: α-Hemolysin from Staphylococcus aureus (α-HL), Mycobacterium smegmatis porin A (MspA) and bacteriophage phi29 pore (phi29). Α-HL and MspA have in their narrowest point a diameter similar to nucleotide size, they are functional at high temperature both wide range of pH (2-12) but MspA is able to read four nucleotide at the same time while α- HL just can read one by one. Finally, phi29 present a bigger diameter what let to get information about DNA spatial conformation and their interaction with proteins (Feng et al., 2015). Nowaday Oxford Nanopore Technologies (ONT) is the only company which has developed Nanopore technology; they have two devices available to sequencing (PromethION and MinION). The MinION is a single-use DNA sequencing device with the size of a USB memory with a total of 3000 nanopores that can sequence until 200kb. The PrometheION is big size sequencer that own 48 different cells, what let to sequence different samples at the same time, with a total of 144.000 nanopores and reading of several megabases (https://www.nanoporetech.com/). The high processivity and low cost become this technique in a great option to massive- sequencing.

Polarized trafficking of adhesion receptors plays a pivotal role in controlling cellular behavior during morphogenesis. Particularly, clathrin-dependent endocytosis of integrins has long been acknowledged as essential for cell migration. However, little is known about the contribution of integrin trafficking to epithelial tissue morphogenesis. Here we show how the transmembrane protein Opo, previously described for its essential role during optic cup folding, plays a fundamental role in this process. Through interaction with the PTB domain of the clathrin adaptors Numb and Numbl via an integrin-like NPxF motif, Opo antagonizes Numb/Numbl function and acts as a negative regulator of integrin endocytosis in vivo. Accordingly, numb/numbl gain-of-function experiments in teleost embryos mimic the retinal malformations observed in opo mutants. We propose that developmental regulator Opo enables polarized integrin localization by modulating Numb/Numbl, thus directing the basal constriction that shapes the vertebrate retina epithelium.
