Stingless bees of the genus Partamona are distributed from southern Mexico to southern Brazil. This genus has been subject to different approaches to solve questions concerning general biology, taxonomy, systematics and biogeography, but population studies applying molecular techniques are inexistent. We analyzed the genetic structure of P. helleri across its geographic distribution along the coastal Atlantic tropical rainforest in Brazil. Ten mtDNA haplotypes were observed in 47 colonies of P. helleri of which some were exclusive and others shared among geographic sub-groups. Statistical analysis showed high genetic differentiation between geographic areas sampled. Fragmentation of the Atlantic forest during Pleistocene glaciations is discussed as a possible cause of the present haplotype distribution and frequency.

The destruction of Brazilian natural habitats has reduced bee populations and negative impacts of native flora pollination have been noticed. This work describes the isolation and characterization of microsatellite loci and evaluates them as molecular markers to study genetic variability of the stingless bee Plebeia remota. A microsatellite enriched genomic library was constructed and 15 primer pairs were designed for this species. The survey was conducted by analyzing 21 unrelated individuals. Genetic diversity indexes were calculated. The mean allelic richness was 6.3, the observed heterozygosity was 0.568, and the percentage of polymorphic loci was 93.33%. Also the primers were tested in cross-species amplification and showed promising results for P. droryana, P. emerina, P. lucii, P. meridionalis, P. pugnax, and P. saiqui. The microsatellite loci described here will be useful to evaluate genetic variability of stingless bees, and certainly will improve our knowledge about population dynamics especially in threatened environments.

An enriched genomic library was constructed from Tetragonisca angustula, a stingless bee species widely distributed in Brazil. The library was screened using two simple-repeat oligonucleotide probes and 21 microsatellite primer pairs were designed flanking a selection of repeat sequences within positive clones. The polymorphism of the microsatellite loci was analyzed by screening a sample of 19 unrelated T. angustula workers. Fifteen out of 21 loci were shown to be polymorphic, with observed heterozygosity estimates ranging from 0.00 to 0.89. The primers were also successfully used to amplify microsatellite loci from other stingless bee species, Tetragonisca fiebrigi, Tetragonisca weyrauchi, Lestrimelitta maracaia and Schwarziana quadripunctata. The results from variability analyses suggest that the microsatellite loci isolated from T. angustula will be useful in further population studies for the species and also for other Meliponini.

We see today many efforts to quantify biodiversity in different biomes. It is very important then to develop and to apply other methodologies that allow us to assess biodiversity. Here we present an example of application of three tools with this goal. We analyzed two populations of Plebeia remota from two distinct biomes that already showed several differences in morphology and behavior. Based on these differences, it has been suggested that the populations of Cunha and Prudentopolis do not represent a single species. In order to verify the existence or absence of gene flow between these two groups, we characterized the patterns of mtDNA through RFLP, the patterns of wing venation through geometric morphometry, and the cuticular hydrocarbons through gas chromatography-mass spectrometry. We used bees collected in these two locations and also from colonies which have being kept for around 9 years at Sao Paulo University. We found six different haplotypes in these specimens, of which three of them occurred exclusively in the population of Cunha and three only in the Prudentopolis population. The fact that the populations do not share haplotypes suggests no maternal gene flow between them. The two populations were differentiated by the pattern of the wing veins. They also had different mixtures of cuticle hydrocarbons. Furthermore it was shown that the colonies kept at the university did not hybridize. These two groups may constitute different species. We also show here the importance of using other methodologies than traditional taxonomy to assess and understand biodiversity, especially in bees.

Five new species of the bee genus Augochlorodes from Brazil are described and the type species, A. turrifaciens Moure, 1958, is redescribed. The new species are: A. clementis sp. nov. (from the states of Parana, Santa Catarina and Rio Grande do Sul), A. incomitatus sp. nov. (Rio Grande do Sul), A. politus sp. nov. (Parana), A. rostratus sp. nov. (Rio Grande do Sul) and A. vachali sp. nov. (Parana, Santa Catarina, Sao Paulo). A discriminant function analysis using head measurements is presented to help distinguish the species. An identification key for the species is provided.

Avaliou-se a infestação, flutuação populacional e horário de visitação de Trigona spinipes (Fabr.) (Hymenoptera: Apidae) em espécies de maracujazeiro. As espécies utilizadas foram Passiflora coccinea, P. setacea, P. alata, P. edulis f. flavicarpa, P. laurifolia e P. nitida, aos três anos de idade. O ensaio foi conduzido em parcelas de 1,5 m de comprimento, avaliando-se os dois lados da espaldeira, considerando-se apenas o 0,5 m superior desta e totalizando, para os dois lados, a área de 1,5 m². Avaliaram-se os seguintes parâmetros: número de irapuás e porcentagem de dano nos botões florais, flores, frutos, ramos, folhas e pedúnculos. Para a determinação do horário de visitação, foi utilizada P. coccinea, sendo as avaliações realizadas às 9:00h, 12:00h e 15:00h. A correlação entre o número médio de T. spinipes presentes nas flores de espécies de maracujazeiro e a porcentagem de danos foi positiva e significativa (r = 0,99). em nenhuma das observações efetuadas, constatou-se a presença de T. spinipes nos botões florais, frutos, ramos, folhas ou pedúnculos das espécies de maracujazeiro, não ocorrendo danos nessas estruturas. Esse resultado sugere que as abelhas são atraídas pelas flores ocasionando danos nessas estruturas, provavelmente, por utilizar o tecido floral ou resinas contidas neste para a construção de ninhos. P. coccinea foi a espécie mais danificada por T. spinipes, apresentando suscetibilidade ao ataque desse inseto. Os horários de maior incidência de T. spinipes foram 9:00h e 12:00h, ocorrendo decréscimo significativo no número de abelhas por flor às 15:00h. Constatou-se maior infestação de T. spinipes nas flores das plantas de maracujá nos meses de outubro a novembro, coincidindo com a primavera, em Jaboticabal, SP.

The present study analyzed, the influence of the treatment with juvenile hormone on the ultrastructure of Apis mellifera L. workers' venom glands. Newly emerged workers received topical application of 1 mu l of juvenile hormone diluted in hexane, in the concentration of 2 mu g/mu l. Two controls were used; one control received no treatment (group C1) and other received topical application of 1 mu l of hexane (group C2). The aspect of the glandular cells, in not treated newly emerged workers, showed that they are not yet secreting actively. Cellular modifications happened according to the worker age and to the glandular area considered. The most active phase of the gland happened from the emergence to the 14th day. At the 25th day the cells had already lost their secretory characteristic, being the distal area the first to suffer degeneration. The treatment with juvenile hormone and hexane altered the temporal sequence of the glandular cycle, forwarding the secretory cycle and degeneration of the venom gland.

The paper presents a study of the pericardial cells of Scaptotrigona postica an eusocial Brazilian stingless bee. Light and electron microscopy was used in a comparative study on workers and queens of different ages, exerting different functions in the colony. The pericardial cells are found only in the peticardial sinus, mainly in groups around the dorsal vessel. Each cell is enclosed by the basal membrane and its peripheral region is characterized by folds of the plasma membrane, which form canals and loops. The points where the plasma membrane folds is frequently closed by diaphragms, that along with the basal lamina form a barrier to substances from hemolymph. Along the membrane limiting the canals and loops, an intense endocytic activity through coated vesicles takes place indicating a selective absorption of hemolymph components. In older individuals, workers or queens, the cells exhibit larger quantities of cytoplasm inclusions, heterogeneous vacuoles containing the final products of intracellular digestion, and autophagic vacuoles with concentric membranous structures. The pericardial cells general morphology is in accordance with the role in processing metabolites captured from hemolymph and storage of indigested residues. (C) 2006 Elsevier Ltd. In S. postica the hypopharyngeal glands are present in workers, males and queens. The glands of newly emerged workers are in a prefunctional phase, in nurse workers they reach their highest development and in forager workers they suffer reabsorption. The newly emerged males and queens, however, have well developed glands that soon start involution. The electrophoretical pattern of worker and male hypopharyngeal glands is compared with the pattern of nurse workers of A. mellifera. The results show simularities, as well as differences among the S. postica males and workers, and among the phases of worker life. The electrophoretic band pattern suggests that the hypopharyngeal glands of nurse workers of S. postica produce substances similar to the ones produced by A. mellifera. The similarity of the extracts of nurse glands of A. mellifera and S. postica, as well as the behavioural similarities of the workers, suggest the same function of this gland in both species.

SDS electrophoresis of midgut proteins from two Trigonini species with different feeding habits shows many similarities and the absence of unique protein in the necrophagous stingless bee Trigona hypogea, suggesting that biochemical adaptation to necrophagy, in this bee, occurs more in the qualitative than in quantitative level. Workers of different ages display some variations in the midgut protein pattern, suggesting that it is important to know the age in studies regarding to bee digestion.

The total number and distribution per antennal flagellomere of sensilla placodea (olfactory disks), sensilla coeloconica, sensilla ampullacea and sensilla campaniformia were determined in workers of Nannotrigona testaceicornis Lepeletier a stingless bee species quite common in Brazil. The distribution of the sensilla was uniform, with the largest number occurring in flagellomere 10 and gradually decreasing in the direction of the basal flagellomeres in a way similar to that observed in Scaptotrigona postica Latreille.Nannotrigona testaceicornis had a larger number of sensilla ampullacea and a smaller number of sensilla coeloconica and sensilla campaniformia than Scaptotrigona postica. Although Nan notrigona testaceicornis does not communicate through the formation of pheromone trails, this species presents a larger quantity of sensilla placodea (relative to the length of the flagellum) than Scaptotrigona postica.

The technique of osmium imidazol for the ultrastructural detection of lipids in the secretory cells of the venom gland of 14-days old worker bees of Apis mellifera L. demonstrated the presence of these components at various sites of the gland. These lipids were found mainly associated to the external region of the basal lamina and the microvilli, in the intercellular spaces, in the cuticle of the collecting canaliculi and in the secretion contained in the glandular lumen. Therefore, in addition to revealing the presence of lipids in the secretion, this technique also allowed us to attribute an exogenous origin to the lipids in the secretion; they are taken up from the haemolymph.

In Apis mellifera the acid or venom gland is composed of secretory cells that surround a channel that opens into a reservoir devoid of musculature. This gland can at times present apical branching. In this study we recorded the frequency of branched venom glands in workers of Africanized bees (Apis mellifera Linnaeus) from six localities in the Pantanal region of Mato Grosso do Sul, and analyzed the relation among the length of the main duct, the length of the duct from the reservoir to the beginning of branching, the length of the branched segment (when present) and the total length of the gland. We sought to determine the probable genotypes of the bees from each population by using the model proposed by Alves-Junior. The frequency of branched glands varied from 50% to 83% in the worker bees coming from those places, indicating that this characteristic is primitive in these bees. The results of the Analysis of Discriminant Functions indicated significant differences in the morphometrical segments of the venom gland (Wilk's Lambda = 0.065; F-(27,F-30) = 4.507; P < 0.001), and permitted a differentiation of the populations studied. The genotypes inferred for the bees of each locality agree with the results obtained in the Analysis of Discriminant Functions and form three distinct groups, with some overlapping areas among them. In all of the populations considered the phenotype largevenom gland was predominant. It is inferred that bees with this phenotype (venom gland larger than S. 15 mm) have Gm(1) Gm(1) genotype, being therefore homozygotes for the major alleles and also for the modifier genes that codify this morphological trait. The high frequency of worker bees with large venom gland in all the places considered makes viable the development of a selection program in order to obtain bees with longer venom glands, aimed at the commercial production of venom by the beekeepers of the Pantanal region of Mato Grosso do Sul.

A series of experiments was performed to assess possible roles of the honey bee Dufour gland secretion. Bioassays with extracts of queen and worker glands from two colonies were made under artificial conditions, in which nestmate and non-nestmate forager workers were tested. The results demonstrate that forager workers display behavioral responses when exposed to Dufour gland extracts of nestmates, but remain indifferent when exposed to non-nestmate extracts. Also, the results demonstrate that forager workers are attracted by virgin queen gland extracts, and repelled by forager worker extracts. The data demonstrate that the Dufour gland secretion is colony- and caste-specific. The attractant remains in the nest. The repellent effect of forager worker extracts is interpreted as an alarm-like pheromone. The attractant effect of virgin queen extracts could be useful in the swarming process to attract scout bees to the eventual founder virgin queen or to attract old foragers to the virgin queen that remains in the nest.
