1-42 beta-Amyloid (A beta(1-42)) peptide is a key molecule involved in the development of Alzheimer's disease. Some of its effects are manifested at the neuronal morphological level. These morphological changes involve loss of neurites due to cytoskeleton alterations. However, the mechanism of A beta(1-42) peptide activation of the neurodegenerative program is still poorly understood. Here, A beta(1-42) peptide-induced transduction of cellular death signals through the phosphatidylinositol 3-kinase (PI3K)/phosphoinositol- dependent kinase (PDK)/novel protein kinase C (nPKC)/Rac 1 axis is described. Furthermore, pharmacological inhibition of PDK1 and nPKC activities blocks Rac 1 activation and neuronal cell death. Our results provide insights into an unsuspected connection between PDK1, nPKCs and Rac 1 in the same signal-transduction pathway and points out nPKCs and Rac 1 as potential therapeutic targets to block the toxic effects of A beta(1-42) peptide in neurons.

Planktobenthos was sampled in 1957-58 in the river Amur. A determination of the kind of organisms drifting in the mass of water of the Amur was carried out. Of special interest for the authors was the activity of drifting of benthic larvae.

Despite the fact that it is a long time sinc e the methods of collection and quantitative estimatio n of protozoa were worked out data on the abundance of these animals in freshwaters are not very numerous. Nevertheless, Infusoria and Rhizopoda at high densities are widespread in the water and sediments of lakes and reservoirs. In order to find ou t the importance of infusoria as a food source for cyclopoids we carried out experimental investigations using C14. For this three species of cyclopoids, widely distributed in open regions of Rybinsk reservoir were selected: Cyclops vicinus, Mesocyclops oithonoides and Kesocyclops leuckarti. The data presented give evidence that the species investigated are able as predators to consume infusorian plankton arid bacteria in the form of flocs or small clumps of detritus.

The output spectrum of Yb-doped double-clad fiber superfluorescent source (SFS) is tailored by placing a broadband dichroic mirror in the pump end of conventional single-pass forward configuration, which constitutes double-pass forward configuration. The 3 dB bandwidth is increased from I I to 42 nm. A maximum output SFS power of 2.12 W and a slope efficiency of 43.2% are obtained. The double-clad fiber is 25 in and the pump power is adequate to saturate the fiber as far as the feedback-induced lasing appears. (c) 2004 Elsevier Ltd. All rights reserved.

A blood coagulation factor IX-binding protein (TSV-FIX-BP) was isolated from the snake venom of Trimeresurus stejnegeri. On SDS-polyacrylamide gel electrophoresis, TSV-FIX-BP showed a single band with an apparent molecular weight of 23,000 under non-reducing conditions. and two distinct bands with apparent molecular weights of 14,800 and 14,000 under reducing conditions. cDNA clones containing the coding sequences of TSV-FIX-BP were isolated and sequenced to determine the structure of the precusors of TSV-FIX-BP subunits. The deduced amino acid sequences of two subunits of TSV-FIX-BP were confirmed by N-terminal protein sequencing and trypsin-digested peptide mass fingerprinting. TSV-FIX-BP was a nonenzymatic C-type lectin-like anti-coagulant. The anti-coagulant activity of TSV-FIX-BP was mainly caused by its dose dependent interaction with blood coagulation factor IX but not with blood coagulation factor X. (C) 2003 Elsevier Science Ltd. The ability to feed on vertebrate blood has evolved many times in various arthropod clades. Consequently, saliva of blood-feeding arthropods has proven to be a rich source of antihemostatic molecules. A variety of platelet aggregation inhibitors antagonize platelet responses to wound-generated signals, including ADP, thrombin, and collagen. Anticoagulants disrupt elements of both the intrinsic and extrinsic pathways. Vasodilators include nitrophorins (nitric oxide storage and transport heme proteins), a variety of peptides that mimic endogenous vasodilatory neuropeptides, and proteins that catabolize or sequester endogenous vasoconstrictors. Multiple salivary proteins may be directed against each component of hemostasis, resulting in both redundancy and in some cases cooperative interactions between antihemostatic proteins. The complexity and redundancy of saliva ensures an efficient blood meal for the arthropod, but it also provides a diverse array of novel antihemostatic molecules for the pharmacologist.
