One approach to understanding common human diseases is to determine the genetic defects responsible for similar diseases in animal models and place those defective genes in their corresponding biochemical pathways. Our laboratory is working with an animal model for human rheumatoid arthritis called collagen-induced arthritis (CIA). We are particularly interested in determining the location of disease-predisposing loci. To that end, we performed experiments to localize susceptibility loci for CIA in an F2 cross between the highly susceptible mouse strain DBA/1j and the highly resistant mouse strain SWR/j. Specifically, a quantitative trait locus analysis was performed to localize regions of the mouse genome responsible for susceptibility/severity to CIA. One susceptibility locus, Cia1 in the major histocompatibility locus, had been identified previously. Two additional loci were detected in our analysis that contribute to CIA severity (Cia2, Cia3) on chromosomes 2 and 6. A third locus was detected that contributes to the age of onset of the disease. This locus (Cia4) was located on chromosome 2 and was linked to the same region as Cia2. Determining the identity of these loci may provide insights into the etiology of human rheumatoid arthritis.

Total organic carbon, total inorganic carbon, biogenic silica content and total organic carbon/total nitrogen ratios of the Laguna Potrok Aike lacustrine sediment record are used to reconstruct the environmental history of south-east Patagonia during the past 51 ka in high resolution. High lake level conditions are assumed to have prevailed during the Last Glacial, as sediments are carbonate-free. Increased runoff linked to permafrost and reduced evaporation due to colder temperatures and reduced influence of Southern Hemispheric Westerlies (SHW) may have caused these high lake levels with lake productivity being low and organic matter mainly of algal or cyanobacterial origin. Aquatic moss growth and diatom blooms occurred synchronously with southern hemispheric glacial warming events such as the Antarctic A-events, the postglacial warming following the LGM and the Younger Dryas chronozone. During these times, a combination of warmer climatic conditions with related thawing permafrost could have increased the allochthonous input of nutrients and in combination with warmer surface waters increased aquatic moss growth and diatom production. The SHW were not observed to affect southern Patagonia during the Last Glacial. The Holocene presents a completely different lacustrine system because (a) permafrost no longer inhibits infiltration nor emits meltwater pulses and (b) the positioning of the SHW over the investigated area gives rise to strong and dry winds. Under these conditions total organic carbon, total organic carbon/total nitrogen ratios and biogenic silica cease to be first order productivity indicators. On the one hand, the biogenic silica is influenced by dissolution of diatoms due to higher salinity and pH of the lake water under evaporative stress characterizing low lake levels. On the other hand, total organic carbon and total organic carbon/total nitrogen profiles are influenced by reworked macrophytes from freshly exposed lake level terraces during lowstands. Total inorganic carbon remains the most reliable proxy for climatic variations during the Holocene as high precipitation of carbonates can be linked to low lake levels and high autochthonous production. The onset of inorganic carbon precipitation has been associated with the southward shift of the SHW over the latitudes of Laguna Potrok Aike. The refined age-depth model of this record suggests that this shift occurred around 9.4 cal. ka BP.

The remarkable astrometric capabilities of the Chandra Observatory offer the possibility to measure proper motions of X-ray sources with an unprecedented accuracy in this wavelength range. We recently completed a proper motion survey of three of the seven thermally emitting radio-quiet isolated neutron stars (INSs) discovered in the ROSAT all-sky survey. These INSs (RXJ0420.0-5022, RXJ0806.4-4123 and RXJ1308.6+2127) either lack an optical counterpart or have one so faint that ground based or space born optical observations push the current possibilities of the instrumentation to the limit. Pairs of ACIS observations were acquired 3 to 5 years apart to measure the displacement of the sources on the X-ray sky using as a reference the background of extragalactic or remote Galactic X-ray sources. We derive 2 sigma upper limits of 123 mas yr(-1) and 86 mas yr(-1) on the proper motion of RXJ0420.0-5022 and RXJ0806.4-4123, respectively. RXJ1308.6+2127 exhibits a very significant displacement (similar to 9 sigma) yielding mu = 220 +/- 25 mas yr(-1), the second fastest measured among all ROSAT-discovered INSs. The source is probably moving away rapidly from the Galactic plane at a speed which precludes any significant accretion of matter from the interstellar medium. Its transverse velocity of similar to 740 (d/700 pc) km s(-1) might be the largest of all ROSAT INSs and its corresponding spatial velocity lies among the fastest recorded for neutron stars. RXJ1308.6+2127 is thus a middle-aged (age similar to 1 My) high velocity cooling neutron star. We investigate its possible origin in nearby OB associations or from a field OB star. In most cases, the flight time from birth place appears significantly shorter than the characteristic age derived from spin down rate. Overall, the distribution in transverse velocity of the ROSAT INSs is not statistically different from that of normal radio pulsars.

We investigated the effect of an extract from a helminth (Ascaris suum) in zymosan-induced arthritis (ZYA) or collagen-induced arthritis (CIA). Rats and mice, respectively, received 1 mg and 0.1 mg zymosan intra-articularly (i.a.). Test groups received an A. suum extract either per os (p.o.) or intraperitoneally (i.p.) 30 min prior to i.a. zymosan. Controls received saline. Hypernociception was measured using the articular incapacitation test. Cell influx, nitrite, and cytokine levels were assessed in joint exudates. The synovia and distal femoral extremities were used for histopathology. Cartilage damage was assessed through determining glycosaminoglycan (GAG) content. DBA/1J mice were subjected to CIA. The test group received A. suum extract i.p. 1 day after CIA became clinically detectable. Clinical severity and hypernociception were assessed daily. Neutrophil influx was determined using myeloperoxidase activity. The A. suum extract, either i.p. or p.o., significantly and dose-dependently inhibited cell influx and hypernociception in ZYA in addition to reducing GAG loss and ameliorating synovitis. The A. suum extract reduced i.a. levels of NO, interleukin-1 beta (IL-1 beta), and IL-10 but not tumor necrosis factor alpha (TNF-alpha) in rats subjected to ZYA while reducing i.a. IL-10, but not IL-1 beta or TNIT-alpha, levels in mice. Clinically, mice subjected to CIA treated with the A. suum extract had less severe arthritis. Hypernociception, myeloperoxidase activity, and synovitis severity were significantly reduced. These data show that a helminth extract given p.o. protects from arthritis severity in two classical arthritis models. This A. suum effect is species independent and functions orally and parenterally. The results show clinical and structural benefits when A. suum extract is given either prophylactically or therapeutically.

In this paper we consider a complex-order forced van der Pol oscillator. The complex derivative Dα1jβ, with α, β ∈ ℝ+, is a generalization of the concept of an integer derivative, where α = 1, β = 0. The Fourier transforms of the periodic solutions of the complex-order forced van der Pol oscillator are computed for various values of parameters such as frequency ω and amplitude b of the external forcing, the damping μ, and parameters α and β. Moreover, we consider two cases: (i) b = 1, μ = {1.0, 5.0, 10.0}, and ω = {0.5, 2.46, 5.0, 20.0}; (ii) ω = 20.0, μ = {1.0, 5.0, 10.0}, and b = {1.0, 5.0, 10.0}. We verified that most of the signal energy is concentrated in the fundamental harmonic ω0. We also observed that the fundamental frequency of the oscillations ω0 varies with α and μ. For the range of tested values, the numerical fitting led to logarithmic approximations for system (7) in the two cases (i) and (ii). In conclusion, we verify that by varying the parameter values α and β of the complex-order derivative in expression (7), we accomplished a very effective way of perturbing the dynamical behavior of the forced van der Pol oscillator, which is no longer limited to parameters b and ω.

Objectives: Children have a greater risk from radiation, per unit dose, due to increased radiosensitivity and longer life expectancies. It is of paramount importance to reduce the radiation dose received by children. This research concerns chest CT examinations on paediatric patients. The purpose of this study was to compare the image quality and the dose received from imaging with images reconstructed with filtered back projection (FBP) and five strengths of Sinogram-Affirmed Iterative Reconstruction (SAFIRE). Methods: Using a multi-slice CT scanner, six series of images were taken of a paediatric phantom. Two kVp values (80 and 110), 3 mAs values (25, 50 and 100) and 2 slice thicknesses (1 mm and 3 mm) were used. All images were reconstructed with FBP and five strengths of SAFIRE. Ten observers evaluated visual image quality. Dose was measured using CT-Expo. Results: FBP required a higher dose than all SAFIRE strengths to obtain the same image quality for sharpness and noise. For sharpness and contrast image quality ratings of 4, FBP required doses of 6.4 and 6.8 mSv respectively. SAFIRE 5 required doses of 3.4 and 4.3 mSv respectively. Clinical acceptance rate was improved by the higher voltage (110 kV) for all images in comparison to 80 kV, which required a higher dose for acceptable image quality. 3 mm images were typically better quality than 1 mm images. Conclusion: SAFIRE 5 was optimal for dose reduction and image quality.

La gestión de los residuos sólidos urbanos presenta recurrentes problemas ambientales y sociosanitarios, con especial crudeza en el ámbito de municipios de pequeña escala. Esta problemática requiere del desarrollo de nuevas tecnologías y rediseño de la gestión de residuos. En la Provincia de Córdoba, la mayoría de las localidades, disponen sus residuos sólidos en sitios abiertos y en forma incontrolada. Estos basurales, son frecuentados por cirujas, trabajo no formal, quienes conjuntamente con otros integrantes de la familia, incluyendo niños, realizan las actividades de recupero de residuos en forma precaria, poniendo en riesgo la salud de dicha población. A su vez, estos lugares son muy propensos a incendios espontáneos ó provocados, lo que conduce a la generación de humos altamente tóxicos que alcanzan a las zonas urbanas. Esta problemática es compartida por todas las regiones con baja densidad poblacional (10 a 40 hab/Km2). A pesar de los incesantes esfuerzos, no se ha logrado aún alcanzar aproximaciones válidas para dar soluciones a esta problemática.La implementación de sistemas de gestión integral con la aportación de tecnologías de bajo costo y provistas por el mercado local, es la única manera que se pueda llegar a buen término con la gestión de residuos y lograr así un desarrollo sustentable de toda la provincia. Los objetivos que se proponen son lograr una solución al grave problema de contaminación para regiones de la provincia de Córdoba debido a la incorrecta disposición y manejo de los residuos sólidos mediante la implementación de un sistema de gestión integral de residuos acorde a las características particulares de cada región, utilizando nuevos indicadores de gestión. Dentro de los objetivos particulares, podemos citar la implementación de sistemas tecnológicos innovadores en el tratamiento de residuos para una correcta disposición final de tecnología sencilla y bajo costo, la implementación de microemprendimientos productivos de clasificación y recupero de materiales, la valoración de los residuos recuperados en función de Bonos de Carbono, disponer de justificativos técnicos y nuevos indicadores económicos para la gestión de subsidios, valorizar el trabajo del ciruja y su familia, capacitación y toma de conciencia a los sectores involucrados. Para el desarrollo del presente trabajo se plantea implementar un sistema de gestión y recupero de residuos regional e integral, que incluya microemprendimientos productivos para lograr la sustentabilidad y a las familias que actualmente trabajan en el recupero de residuos, diseñar y ensayar equipos con características innovadoras para la disposición final de residuos e implementar un proyecto de educación integral que aúne el conocimiento, capacitación y toma de conciencia. Para lograr los objetivos citados se proponen realizar actividades de relevamiento de indicadores facilitadores de vinculación para las localidades de la provincia, generar una logística de funcionamiento para los municipios, relevar las actividades económicas y sanitarias relacionadas a residuos y de las personas involucradas, desarrollar emprendimientos productivos locales, realizar y ensayar tecnologías para el recupero y disposición de residuos, implementar prototipos a escala piloto, implementar talleres de capacitación y material didáctico en las temáticas medio ambiente, desarrollo sustentable, gestión integral de residuos sólidos, reutilización de residuos, salud pública. El presente proyecto estará llevado a cabo por profesionales docentes y alumnos universitarios de distintas especialidades, como así también docentes y técnicos pertenecientes a un establecimiento de educación superior no universitaria, Itec Río Cuarto. Esto, motivado por el convencimiento y necesidad que los problemas ambientales deben ser abordados en forma interdisciplinaria, y las estrategias de intervención que se proponen deben estar integradas con los aportes de cada una de las disciplinas intervinientes.

The aim of the study was to analyse the degree to which gestational age (GA) has been shortened due to prenatal diagnosis of gastrointestinal malformations (GIM). The data source for the study was 14 population-based registries of congenital malformations (EUROCAT). All liveborn infants with GIMs and without chromosomal anomalies, born 1997-2002, were included. The 14 registries identified 1047 liveborn infants with one or more GIMs (oesophageal atresia, duodenal atresia, omphalocele, gastroschisis and diaphragmatic hernia). Median GA at birth was lower in prenatally diagnosed cases for all five malformations, although not statistically significant for gastroschisis. There was little difference in median birthweight by GA for the pre- and postnatally diagnosed infants. The difference in GA at birth between prenatally and postnatally diagnosed infants with GIMs is enough to increase the risk of mortality for the prenatally diagnosed infants. Clinicians need to balance the risk of early delivery against the benefits of clinical convenience when making case management decisions after prenatal diagnosis. Very few studies have been able to show benefits of prenatal diagnosis of congenital malformations for liveborn infants. This may be because the benefits of prenatal diagnosis are outweighed by the problems arising from a lower GA at birth.

We offer new evidence on multi-level determinants of the gender division of housework. Using data from the 2004 European Social Survey (ESS) for 26 European, we study the micro and macro-level factors which increase the likelihood of men doing an equal or greater share of housework than their female partners. A sample of 11,915 young men and women is analysed with a multi-level logistic regression in order to test at individual level the classic relative-income, time-availability and gender-role values, and a new couple conflict hypothesis. At individual level we find significant relationships between relative resources, values, couple's disagreement, and the division of housework which support more economic dependency than "doing gender" perspectives. At the macro-level, we find important composition effects and also support for gender empowerment, family model and social stratification explanations of cross-country differences.

Gastroschisis is an abdominal wall defect more prevalent in offspring of young mothers. It is known to be increasing in prevalence despite the general decrease in the proportion of births to young European women. We investigated whether the increase in prevalence was restricted to the high-risk younger mothers. We analysed 936 cases of gastroschisis from 25 population-based registries in 15 European countries, 1980-2002. We fitted a Bayesian Hierarchical Model which allowed us to estimate trend, to estimate which registries were significantly different from the common distribution, and to adjust simultaneously for maternal age, time (in grouped years) and the random variation between registries. The maternal age-standardised prevalence (standardised to the year 2000 European maternal age structure) increased almost fourfold from 0.54 [95% Credible Interval (CrI) 0.37, 0.75] per 10,000 births in 1980-84 to 2.12 [95% CrI 1.85, 2.40] per 10,000 births in 2000-02. The relative risk of gastroschisis for mothers <20 years of age in 1995-2002 was 7.0 [95% CrI 5.6, 8.7]. There were geographical differences within Europe, with higher rates of gastroschisis in the UK, and lower rates in Italy after adjusting for maternal age. After standardising for regional variation, our results showed that the increase in risk over time was the same for mothers of all ages--the increase for mothers <20 years was 3.96-fold compared with an increase of 3.95-fold for mothers in the other age groups. These findings indicate that the phenomenon of increasing gastroschisis prevalence is not restricted to younger mothers only.

Contexte: Parmi les complications de toute chirurgie, on retrouve l'embolie pulmonaire, dont l'issue est potentiellement fatale. Peu de travaux cependant ont été effectués pour étudier les facteurs de risques associés à une embolie pulmonaire après une chirurgie hépato-biliaire en particulier. Objectif: Le but de notre travail est d'étudier et comparer aux rares résultats existants les facteurs de risque ainsi que les conséquences d'une embolie pulmonaire post- hépatectomie. Les données pourront éventuellement être utilisées pour cibler les patients à risque et améliorer la prévention. Résultats: Le diagnostic d'embolie pulmonaire a été posé chez 24 patients (8%) au CT-scan dans la période post-opératoire. Le délai moyen entre l'opération et le diagnostic était de 5 jours (1-15j). La comparaison des patients du groupe EP (n = 24) au groupe Non-EP (n = 272) montre un BMI médian plus élevé (27 vs 24 kg/m2, p=0.006), un taux plus élevé de résections majeures (71 vs 43%, p=0.01) ainsi qu'une durée opératoire plus longue (310 vs 260 min, p=0.001). La durée totale d'hospitalisation était également supérieure dans le groupe EP (22 vs 11j, p<0.05), de même que la durée de séjour aux soins intensifs (3 vs 1j, p<0.05). La mortalité globale à 90j post-opératoires était de 3.3%, la mortalité du groupe EP étant nulle. Conclusion: Les résultats de cette étude rétrospective ont démontré l'association des embolies pulmonaires post-hépatectomie avec l'obésité, les hépatectomies majeures, ainsi qu'une longue durée d'opération. L'incidence non négligeable des embolies prouve la nécessité d'une prophylaxie thrombo-embolique.
