A patent foramen ovale (PFO), present in ∼40% of the general population, is a potential source of right-to-left shunt that can impair pulmonary gas exchange efficiency [i.e., increase the alveolar-to-arterial Po2 difference (A-aDO2)]. Prior studies investigating human acclimatization to high-altitude with A-aDO2 as a key parameter have not investigated differences between subjects with (PFO+) or without a PFO (PFO-). We hypothesized that in PFO+ subjects A-aDO2 would not improve (i.e., decrease) after acclimatization to high altitude compared with PFO- subjects. Twenty-one (11 PFO+) healthy sea-level residents were studied at rest and during cycle ergometer exercise at the highest iso-workload achieved at sea level (SL), after acute transport to 5,260 m (ALT1), and again at 5,260 m after 16 days of high-altitude acclimatization (ALT16). In contrast to PFO- subjects, PFO+ subjects had 1) no improvement in A-aDO2 at rest and during exercise at ALT16 compared with ALT1, 2) no significant increase in resting alveolar ventilation, or alveolar Po2, at ALT16 compared with ALT1, and consequently had 3) an increased arterial Pco2 and decreased arterial Po2 and arterial O2 saturation at rest at ALT16. Furthermore, PFO+ subjects had an increased incidence of acute mountain sickness (AMS) at ALT1 concomitant with significantly lower peripheral O2 saturation (SpO2). These data suggest that PFO+ subjects have increased susceptibility to AMS when not taking prophylactic treatments, that right-to-left shunt through a PFO impairs pulmonary gas exchange efficiency even after acclimatization to high altitude, and that PFO+ subjects have blunted ventilatory acclimatization after 16 days at altitude compared with PFO- subjects.

Os herpesvrus bovino tipos 1 e 5 (BoHV-1; BoHV-5) so gentica e antigenicamente muito semelhantes e por isso so indistinguveis pela maioria dos testes diagnsticos. Como o BoHV-1 tem sido classicamente associado com doena respiratria e genital, os herpesvrus isolados dessas enfermidades tm sido provisoriamente - e s vezes definitivamente - identificados como BoHV-1. Da mesma forma, os casos de infeco neurolgica por herpesvrus em bovinos tm sido atribudos em sua totalidade ao BoHV-5. Este trabalho relata a identificao de 40 amostras de herpesvrus isoladas de diferentes casos clnicos na regio Centro-Sul do Brasil, Argentina e Uruguai entre 1987 e 2006, pelo uso de um PCR capaz de diferenciar esses vrus. As amostras identificadas como BoHV-1 (n=16) foram isoladas de doena respiratria (n=3), balanopostite e/ou vulvovaginite (n=3), do smen de touros saudveis (n=5) e de casos doena neurolgica (n=5). As amostras virais identificadas como BoHV-5 (n=24) foram em sua maioria isoladas de doena neurolgica (n=21), mas tambm do smen de touros saudveis (n=2) e do bao de um bezerro com doena sistmica (n=1). Esses resultados demonstram que tanto o BoHV-1 como o BoHV-5 no esto estritamente associados s suas respectivas sndromes clnicas e que podem estar freqentemente envolvidos em casos clnicos classicamente atribudos ao outro vrus. Esses achados tambm reforam a necessidade de se identificar corretamente os isolados de herpesvrus para um melhor conhecimento da sua patogenia e epidemiologia.

Larthrose (OA) est une maladie dgnrative et multifactorielle caractrise par une destruction de cartilage, une formation dostophytes et une inflammation au niveau de la membrane synoviale. Le 4-hydroxynonnal (HNE), un produit final de la peroxydation lipidique, a t identifi rcemment comme un facteur catabolique et un mdiateur inflammatoire dans le cartilage arthrosique humain. Notre projet vise tudier leffet du HNE sur la rgulation de la prostaglandine E2 synthase-1 microsomale (mPGES-1) et de la protine activante 5-lipoxygnase (FLAP)/5-lipoxygnase (5-LOX) dans les chondrocytes arthrosiques humains. Lorsque les cellules sont traites une seule fois avec 10 M HNE, les rsultats de Western blot et de PCR en temps rel montrent que lexpression de la cyclooxygnase-2 (COX-2) et de la mPGES-1 augmente de manire significative et atteint respectivement le maximum aprs 8 et 16 heures dincubation puis diminue graduellement. Cependant, lorsque les cellules sont traites plusieurs fois avec 10 M HNE 2 heures dintervalle, lexpression de la COX-2 et de la mPGES-1 augmente en fonction du temps sans subir une baisse aprs 24 heures dincubation. Le HNE induit lactivit du promoteur de la mPGES-1 via lactivation du facteur de transcription Egr-1. Linvestigation de la 2me voie du mtabolisme de lacide arachidonique, savoir 5-LOX/FLAP, montre que le HNE induit lexpression de FLAP aprs 24 heures de stimulation et celle de 5-LOX seulement aprs 48 heures. Ceci semble survenir ltape de transcription au cours de laquelle HNE induit lexpression de lARNm et lactivit du promoteur du gne 5-LOX. Nous avons dmontr aussi que le niveau de leukotrine B4 (LTB4) augmente et suit le mme profil que celui de la 5-LOX. Ltude des mcanismes molculaires susceptibles dtre impliqus dans la rgulation de la 5-LOX/FLAP par le HNE montre que ce dernier stimule leur expression via laction de prostaglandine E2 (PGE2) et du facteur de croissance transformant-beta 1 (TGF-1). En conclusion, notre tude dmontre que le HNE induit court-terme dincubation la voie de COX-2/mPGES-1 puis par la suite stimule celle de FLAP/5-LOX long-terme dincubation dans les chondrocytes arthrosiques humains. Ces rsultats suggrent que la mPGES-1 et 5-LOX/FLAP sont des potentielles cibles thrapeutiques intressantes pour contrler la production de PGE2 et LTB4 dans OA.

Metathesis reactions were used to prepare a range of dicopper(II), monocopper(I), diruthenium(II, III), dimolybdenum(II,II) and dirhodium(II,II) complexes of either racemic or resolved forms of endo- and exo-bicyclo[2.2.1]hept-5-ene-2-carboxylic acid (C7H9CO2H). The X-ray crystal structure of [Cu2{()-endo--O2CC7H9}4(CH3OH)2]2CH3OH shows the two copper(II) ions bridged by two (+)-endo-bicyclo[2.2.1]hept-5-ene-2-carboxylate anions and two ()-endo-bicyclo[2.2.1]hept-5-ene-2-carboxylate anions. Methanol molecules occupy the two trans axial sites, and there are also two methanol molecules hydrogen bonded to opposite carboxyl oxygens.

The presence of paramagnetic species in the aqueous ring opening metathesis polymerizations of the exo,exo-7-oxabicyclo[2.2.1]hept-5-ene-2,3-dicarboxylic acid monomer with RuCl(3) and K(2)[RuCl(5)H(2)O] compounds was studied using ESR techniques. It was observed that the intensities of the Ru(III) signals in the ESR spectra decrease on the time scale of the induction period so that the ROMP can take place. The intensity of the Ru(III) signal almost disappeared 50 min after reacting with K(2)[RuCl(5)H(2)O] and after 100 mm in the case of RuCl(3). Reactions of the cis-[Ru(NH(3))(4)(H(2)O)(2)](tfms)(3) and [Ru(NH(3))(5)H(2)O](tfms)(3) complexes with the monomer and different organic compounds representing the organic functions in the monomer (furan, norbornene, but-2-ene-1,4-diol and formic, acetic, oxalic and maleic acids) were also monitored by ESR and UV/vis spectra. It was deduced that the organic acids provide the disappearance of the Ru(III) signal. The proton NMR relaxation times of the residual water in D(2)O for reactions with oxalic acid suggested that the presence of paramagnetic ions in the solution decreases along with
