Influence of La3+ on the accumulation of trace elements (Se-75, Co-56, Rb-83, V-48, (95)mTc, and Ga-67) in chloroplasts of cucumber seedling leaves was studied by a radioactive multitracer technique. At the same time, chloroplast contents of different concentrations of La3+ treatment were calculated. It was observed that chloroplast contents peaked at 0.02 mM La3+ treatment and that the uptake and distribution of these trace elements in chloroplasts of cucumber seedling leaves are different under different La3+, treatments. With the increase of lanthanum concentrations from 0.002 to 2 mM, the uptake percentages of Se-75, Co-56, and Rb-83 presented an obvious increase and then sharply decreased in contrast to the nonlanthanum treatment, whereas there appeared a sharp decrease and then restored control level in the uptake of V-48. The other two trace elements, namely Tc-95m and Ga-67, were accumulated only in the presence of 0.02 mM La3+. The results indicate that lanthanum treatments to growing the cucumber lead to the change of trace element uptake in the chloroplasts of leaves, which suggest that lanthanum might influence the accumulation of trace elements in chloroplasts of cucumber seedling leaves by regulation of various ion transport mechanisms, thus affecting the photosystem of leaves.

The purpose of this paper is to demonstrate the potential of the EXODUS evacuation model in building environments. The latest PC/workstation version of EXODUS is described and is also applied to a large hypothetical supermarket/restaurant complex measuring 50 m x 40 m. A range of scenarios is presented where population characteristics (such as size, individual travel speeds, and individual response times), and enclosure configuration characteristics (such as number of exits, size of exits, and opening times of exits) are varied. The results demonstrate a wide range of occupant behavior including overtaking, queuing, redirection, and conflict avoidance. Evacuation performance is measured by a number of model predicted parameters including individual exit flow rates, overall evacuation flow rates, total evacuation time, average evacuation time per occupant, average travel distance, and average wait time. The simulations highlight the profound impact that variations in individual travel speeds and occupant response times have in determining the overall evacuation performance. 1. A key aspect of glucose homeostasis is the constant monitoring of blood glucose concentrations by specific glucose sensing units. These sensors, via stimulation of hormone secretion and activation of the autonomic nervous system (ANS), regulate tissue glucose uptake, utilization or production. The best described glucose detection system is that of the pancreatic beta-cells which controls insulin secretion. Secretion of other hormones, in particular glucagon, and activation of the ANS, are regulated by glucose through sensing mechanisms which are much less well characterized. Here I review some of the studies we have performed over the recent years on a mouse model of impaired glucose sensing generated by inactivation of the gene for the glucose transporter GLUT2. This transporter catalyzes glucose uptake by pancreatic beta-cells, the first step in the signaling cascade leading to glucose-stimulated insulin secretion. Inactivation of its gene leads to a loss of glucose sensing and impaired insulin secretion. Transgenic reexpression of the transporter in GLUT2/beta-cells restores their normal secretory function and rescues the mice from early death. As GLUT2 is also expressed in other tissues, these mice were then studied for the presence of other physiological defects due to absence of this transporter. These studies led to the identification of extra-pancreatic, GLUT2-dependent, glucose sensors controlling glucagon secretion and glucose utilization by peripheral tissues, in part through a control of the autonomic nervous system.

Ultrafine ferrite grain sizes were produced in a 0.11C-1.6Mn-0.2Si steel by torsion testing isothermally at 675 °C after air cooling from 1250 °C. The ferrite was observed to form intragranularly beyond a von Mises equivalent tensile strain of approximately 0.7 to 0.8 and the number fraction of intragranular ferrite grains continued to increase as the strain level increased. Ferrite nucleated to form parallel and closely spaced linear arrays or “rafts” of many discrete ultrafine ferrite grains. It is shown that ferrite nucleates during deformation on defects developed within the austenite parallel to the macroscopic shear direction (i.e., dynamic strain-induced transformation). A model austenitic Ni-30Fe alloy was used to study the substructure developed in the austenite under similar test conditions as that used to induce intragranular ferrite in the steel. It is shown that the most prevalent features developed during testing are microbands. It is proposed that high-energy jogged regions surrounding intersecting microbands provide potential sites for ferrite nucleation at lower strains, while at higher strains, the walls of the microbands may also act as nucleation sites.

A retenção de Se pelos colóides do solo constitui importante processo para a manutenção da sanidade ambiental. A informação sobre a adsorção de Se em solos altamente intemperizados é restrita e existem poucos padrões quantitativos disponíveis para a definição de estratégias de remediação de áreas contaminadas. Quantidades crescentes de Se (5, 10, 25, 50, 100 e 250 mg L-1), na forma de Na2SeO3, foram adicionadas a amostras de dez Latossolos brasileiros [três Latossolos Vermelho-Amarelos (LVA-1, LVA-2 e LVA-3), dois Latossolos Vermelhos (LV-1 e LV-2), um Latossolo Vermelho eutroférrico (LVef), um Latossolo Vermelho acriférrico (LVwf), dois Latossolos Amarelos (LA-1 e LA-2) e um Latossolo Amarelo acriférrico (LAwf)]. Isotermas de adsorção foram construídas e foi verificado o ajuste dos resultados experimentais aos modelos de Langmuir e de Freundlich. A equação de Langmuir ajustou melhor os resultados de adsorção de Se do que a isoterma de Freundlich. Todas as isotermas apresentaram o formato tipo-L (exponencial), com exceção daquelas obtidas para o LVA-1 e para o LVA-2, que apresentaram comportamento tipo-C (linear). Valores de adsorção máxima (Ads máx), estimada pelo modelo de Langmuir, variaram de 135 (LVA-3) a 2.245 mg kg-1 (LA-1), enquanto os coeficientes de afinidade (K L) estiveram entre 0,002 (LVA-2) e 0,326 (LVA-3). A constante de afinidade estimada pelo modelo de Freundlich (Kf) variou de 13,7 (LVA-2) a 180,1 (LAwf). A adsorção máxima de Se foi mais elevada no LVef e nos Latossolos ácricos (LAwf e LVwf), enquanto os maiores valores de Kf foram encontrados no LV-2, LVef, LVA-3 e LVwf. Não houve correlação entre os atributos dos solos e as constantes de Langmuir. Valores de Kf correlacionaram-se com os teores de argila (r = 0,42*) e com a capacidade de troca de ânions (r = 0,64*).

OBJETIVO: analisar a influência da via de parto sobre a força muscular do assoalho pélvico (FM-AP). MÉTODOS: estudo clínico de corte transversal, para avaliar a FM-AP pelo teste da avaliação da força do assoalho pélvico (AFA) e uso do perineômetro em primíparas, entre 20-30 anos de idade, 4-6 meses pós-parto. A contração, medida pelos dois testes, foi classificada em: zero - ausência, um - leve, dois - moderada e três - normal, sustentada por 6 segundos. Avaliaram-se 94 mulheres, entre 20 e 30 anos, divididas em três grupos: pós-parto vaginal (n=32); pós-cesárea (n=32) e nulíparas (n=30). A variável independente foi a via de parto e a dependente, a FM-AP. A comparação entre os graus de contração foi realizada pelo teste de Kruskal-Wallis e o teste de Dunn para comparações múltiplas; a influência da via de parto pelo teste chi2, o risco relativo (RR) para alteração da FM-AP e o coeficiente kappa para avaliar equivalência entre os testes. RESULTADOS: a mediana e 1º e 3º quartil da FM-AP foram menores (p=0,01) pós-parto vaginal (2,0;1-2) e intermediários pós-cesárea (2,0; 2-3) em relação às nulíparas (3,0;2-3), tanto analisadas pelo AFA como pelo perineômetro. Aumentou o RR de exame alterado pós-parto vaginal (RR=2,5; IC 95%: 1,3-5,0; p=0,002); (RR=2,3; IC 95%: 1,2-4,3; p=0,005) e pós-cesárea (RR=1,5; IC 95%: 0,94-2,57; p=0,12); (RR=1,3; IC 95%: 0,85-2,23; p=0,29) pelo PFSE e perineômetro, respectivamente. CONCLUSÕES: o parto vaginal diminuiu a força muscular do AP de primíparas quando comparado com os casos submetidos à cesárea e com as nulíparas.

Objective: the urethra is the main port of entry of sexually transmitted pathogens. However, papers on the morphology of the urethra are scarce. The Mongolian gerbil is a rodent native of the Mongolia and China and has been utilized as a laboratory animal since the 1960s. This work describes the ultrastructure of the urethra of the Mongolian gerbil to provide data for future experimental studies. Methods: the urethra of ten adult male gerbils was studied by transmission electron microscopy. Results: the epithelium of the pelvic urethra possesses two cell types: I and II, without the formation of cellular layers, while the penile urethra possesses cellular layers: basal, intermediate and superficial. The urethra presents neurosecretory cells belonging to the amine precursor uptake and decarboxylation system. Conclusions: the urethral epithelium of the gerbil is a neurosecretory epithelium, part of the amine precursor uptake and decarboxylation system.

Two simple methods were developed to determine, 11 pesticides in coconut water, a natural isotonic drink rich in salts, sugars and vitamins consumed by the people and athletes. The first procedure involves solid-phase extraction using Sep-Pak Vac C-18 disposable cartridges with methanol for elution. Isocratic analysis was carried out by means of high-performance liquid chromatography with ultraviolet detection at 254 nm to analyse captan, chlorothalonil, carbendazim, lufenuron and diafenthiuron. The other procedure is based on liquid-liquid extraction with hexane-dichloromethane (1:1, v/v), followed by gas chromatographic analysis with effluent splitting to electron-capture detection for determination of endosulfan, captan, tetradifon and trichlorfon and thermionic specific detection for determination of malathion, parathion-methyl and monocrotophos. The methods were validated with fortified samples at different concentration levels (0.01-12.0 mg/kg). Average recoveries ranged from 75 to 104% with relative standard deviations between 1.4 and 11.5%. Each recovery analysis was repeated at least five times. Limits of detection ranged from 0.002 to 2.0 mg/kg. The analytical procedures were applied to 15 samples and no detectable amounts of the pesticides were found in any samples under the conditions described. (C) 2002 Elsevier B.V. B.V.
