Given the paucity of information on the potential roles of bone morphogenetic proteins (BMPs) in the ruminant ovary we conducted immunolocalization and functional studies on cells isolated from bovine antral follicles. Immunocytochemistry revealed expression of BMP-4 and -7 in isolated theca cells whereas granulosa cells and oocytes selectively expressed RMP-6. All three cell types expressed a range of BMP-responsive type-I (BMPRIB, ActRI) and type-II (BMPRII, ActRII, ActRIIB) receptors supporting autocrine/paracrine roles within the follicle. This was reinforced by functional experiments on granulosa cells which showed that BMP-4, -6 and -7 promoted cellular accumulation of phosphorylated Smad-1 but not Smad-2 and enhanced 'basal' and IGF-stimulated secretion of oestradiol (E2), inhibin-A, activin-A and follistatin (FS). Concomitantly, each BMP suppressed 'basal' and IGF-stimulated progesterone secretion, consistent with an action to prevent or delay atresia and/or luteinization. BMPs also increased viable cell number under 'basal' (BMP-4 and -7) and IGF-stimulated (BMP-4, -6 and -7) conditions. Since FS, a product of bovine granulosa cells, has been shown to bind several BMPs, we used the Biacore technique to compare its binding affinities for activin-A (prototype FS ligand) and BMP-4, -6 and -7. Compared with activin-A (K-d 0.28 +/- 0.02 nM; 100%), the relative affinities of FS for BMP-4, -6 and -7 were 10, 5 and 1% respectively. Moreover, studies on granulosa cells showed that preincubation of ligand with excess FS abolished activin-A-induced phosphorylation of Smad-2 and BMP-4-induced phosphorylation of Smad-1. However, FS only partially reversed BMP-6-induced Smad-1 phosphorylation and had no inhibitory effect on BMP-7-induced Smad-1 phosphorylation. These findings support functional roles for BMP-4, -6 and -7 as paracrine/autocrine modulators of granulosa cell steroidogenesis, peptide secretion and proliferation in bovine antral follicles. The finding that FS can differentially modulate BMP-induced receptor activation and that this correlates with the relative binding affinity of FS for each BMP type implicates FS as a potential modulator of BMP action in the ovary.

The title radical (F4BlmNN) is a stable nitronylnitroxide that forms hydrogen-bonded NH center dot center dot center dot ON chains in the solid state. The chains assemble the F4BlmNN molecules to form stacked contacts between the radical groups, in a geometry that is expected to exhibit ferromagnetic (FM) exchange based on spin polarization (SP) models. The experimental magnetic susceptibility of F4BlmNN confirms the expectation, showing 1-D Heisenberg chain FM exchange behavior over 1.8-300 K with an intrachain exchange constant Of J(chain)/k = +22 K. At lower temperatures, ac magnetic susceptibility and variable field heat capacity measurements show that F4BlmNN acts as a quasi-1-D ferromagnet. The dominant ferromagnetic exchange interaction is attributable to overlap between spin orbitals of molecules within the hydrogen-bonded chains, consistent with the SP model expectations. The chains appear to be antiferromagnetically exchange coupled, giving cusps in the ac susceptibility and zero field heat capacity at lower temperatures. The results indicate that the sample orders magnetically at about 0.7 K. The magnetic heat capacity ordering cusp shifts to lower temperatures as external magnetic field increases, consistent with forming a bulk antiferromagnetic phase below a Neel temperature of T-N(0) = 0.72 K, with a critical field of H-c approximate to 1800 Oe. The interchain exchange is estimated to be zJ/k congruent to (-)0.1 K.

To study the viability of detached leaf culture technique, studies were carried out with detached leaves from cotton apex (true trilobed leaves). The prepared leaves were sprayed with 2,4-D amine and ester, at rates of 10, 30, 70, and 100% of the recommended doses. Detached leaves without herbicide spray were used as controls. Simultaneously, a greenhouse experiment was conducted with the same treatments as used for the detached leaves experiment. Toxicity was measured through a 0-to-5 grading according to the percentage of affected leaf area in the detached leaves experiment or examining the affected rate of whole plant as indicated in the greenhouse. Results showed that the ester form of the herbicide induced earlier and more severe toxicity symptoms in detached leaves and greenhouse grown plants. Positive and significant correlations (p < 0.001) were found between toxicity results obtained at 7 and 14 days after application in detached leaves and greenhouse plants (r = 0.97 and 0.92, respectively). Negative, significant correlations (p < 0.005) were found between the toxicity levels found at 7 and 14 days after application in detached leaves and dry matter of cotton plants grown in the greenhouse (r = -0.92 and -0.92, respectively).

The aim of this study was to evaluate the efficacy of glyphosate and 2,4-D for the Commelina benghalensis, Commelina diffusa and Commelina erecta. Three experiments were conducted in a greenhouse in a completely randomized in factorial scheme 3x7+1 (three herbicides x seven periods - hour after application) for two methods for measuring absorption of herbicides (simulated rain and cut the leaves applied), and a control without application, with four replications. The herbicides used were: glyphosate (1,080 g ha(-1)), 2,4-D amine (720 g ha(-1)) and, mixture glyphosate + 2,4-D (720 + 720 g ha(-1)). It was evaluated seven time intervals for washing (simulating rainfall after application) and cut (simulation abortion as a strategy of defense) the leaves applied: 0, 2, 4, 6, 8, 12 and 24 hours after herbicide application (HHA). The minimum period for absorption of glyphosate alone and in mixture with 2,4 -D presented a satisfactory control (> 90.0%) was around 12 HHA to C. benghalensis, C. diffusa and C. erecta, independently of washing or cut the leaves applied. This observed behavior can influence the time required without rain after herbicide application, besides that, the plants aborted part of the stem with injuries to avoid the herbicide translocation, especially when was used the 2,4-D alone. Concluded that glyphosate alone and in mixture with 2,4-D were efficient and that the differences in the control of species may be due to variation of time to absorb the herbicide.

O objetivo deste estudo foi avaliar a eficiência de controle de duas doses do herbicida 2,4-D pulverizado em plantas de M. aquaticum e submetido à simulação de chuva em diferentes períodos de tempo após sua aplicação. O delineamento experimental utilizado foi o inteiramente casualizado, com quatro repetições, disposto em um esquema fatorial 2 x 8 (duas doses de herbicida e oito intervalos de chuva). Utilizou-se o herbicida 2,4-D, na formulação comercial DMA 806, em duas doses: 670 e 1.340 g e.a. ha-1. A simulação de uma chuva de 15 mm, com duração de cinco minutos, foi realizada em oito intervalos de tempo após a pulverização do herbicida (0,25h, 0,5h, 1h, 2h, 4h, 8h, 12h e sem chuva). Foram realizadas avaliações visuais de controle aos 7, 14, 21 e 28 dias após a aplicação dos tratamentos. As duas doses avaliadas do herbicida 2,4-D foram eficientes no controle das plantas de M. aquaticum, mesmo com ocorrência de chuva 15 minutos após sua aplicação.

O objetivo do presente trabalho foi avaliar a eficácia de glyphosate e 2,4-D, isolados e em mistura, no controle de Commelina villosa. Foram estudadas duas metodologias de avaliação de absorção de herbicidas em oito intervalos de tempo para a lavagem (simulando chuva após a aplicação) e corte (simulando abortamento, como estratégia de defesa) das folhas: 2, 4, 6, 8, 12, 24 e 48 horas após a aplicação dos herbicidas, além de um tratamento sem lavagem ou corte das folhas, em delineamento experimental inteiramente casualizado com quatro repetições, dispostos em um esquema fatorial 3 x 7 + 1 (três herbicidas x sete períodos - horas após a aplicação). Os herbicidas e doses testados foram: glyphosate (1.440 g ha-1), 2,4-D (720 g ha-1) e a mistura glyphosate + 2,4-D (1.080 + 720 g ha-1). A simulação de chuva interferiu de forma negativa no controle das plantas com o herbicida glyphosate. O controle com o herbicida 2,4-D foi influenciado apenas no período de 2 horas. Os períodos de simulação de chuva não influenciaram no controle das plantas com a mistura de glyphosate + 2,4-D. Para o estudo com corte das folhas tratadas, todos os tratamentos independente do período para corte das folhas foram influenciados de forma negativa no controle, sendo que as plantas apresentaram rebrotas quando tratadas com o herbicida 2,4-D isolado.

Reactive species generated by Fe0 oxidation promoted by O2 (catalyzed or not by ligands) are able to degrade contaminant compounds like the herbicide 2,4-dichlorophenoxyacetic acid. The degradation of 2,4-D was influenced by the concentrations of zero valent iron (ZVI) and different ligands, as well as by pH. In the absence of ligands, the highest 2,4-D degradation rate was obtained at pH 3, while the highest percentage degradation (50%) was achieved at pH 5 after 120 min of reaction. Among the ligands studied (DTPA, EDTA, glycine, oxalate, and citrate), only ethylenediaminetetraacetic acid (EDTA) and diethylenetriaminepentaacetic acid (DTPA) significantly enhanced oxidation of 2,4-D. This increase in oxidation was observed at all pH values tested (including neutral to alkaline conditions), indicating the feasibility of the technique for treatment of contaminated water. In the presence of EDTA, the oxidation rate was greater at pH 3 than at pH 5 or 7. Increasing the EDTA concentration increased the rate and percentage of 2,4-D degradation, however increasing the Fe0 concentration resulted in the opposite behavior. It was found that degradation of EDTA and 2,4-D occurred simultaneously, and that the new methodology avoided any 2,4-D removal by adsorption/coprecipitation. © 2013 Elsevier Ltd.

The aim of this study was to evaluate the efficacy of glyphosate and 2,4-D alone and in combination, in the control of Commelina villosa. We studied two methodologies for evaluating herbicide absorption in eight time intervals for washing (simulating rainfall after application) and cutting of leaves (simulating abortion as a defense strategy): 2, 4, 6, 8, 12, 24 and 48 hours after herbicide application, and a treatment without washing or cutting the leaves in a completely randomized design with four replications in a 3 x 7 + 1 factorial design (three herbicides x seven periods – hours after application). Herbicides and doses tested were: glyphosate (1,440 g ha-1), 2,4-D (720 g ha-1) and a mixture of glyphosate + 2,4-D (1,080 + 720 g ha-1). The simulation of rain interfered negatively in the plant control with glyphosate. The control with the herbicide 2,4-D was affected only for the period of 2 hours. Periods of rain simulation did not influence the control of plants with a mixture of glyphosate + 2,4-D. For the study with the cutting of treated leaves, all treatments regardless of the period of cutting the leaves were influenced negatively in terms of plant control, the plants showing regrowth when treated with 2,4-D alone.

A highly concentrated aqueous saline-containing solution of phenol, 2,4-dichlorophenoxyacetic acid (2,4-D) and 2,4-dichlorophenol (2.4-DCP) was treated by the photo-Fenton process in a system composed of an annular reactor with a quartz immersion well and a medium-pressure mercury lamp (450 W). The study was conducted under special conditions to minimize the costs of acidification and neutralization, which are usual steps in this type of process. Photochemical reactions were carried out to investigate the influence of some process variables such as the initial concentration of Fe2+ ([Fe2+](0)) from 1.0 up to 2.5 mM, the rate in mmol of H2O2 fed into the system (F-H2O2,F-in) from 3.67 up to 7.33 mmol of H2O2/min during 120 min of reaction time, and the initial pH (pH(0)) from 3.0 up to 9.0 in the presence and absence of NaCl (60.0 g/L). Although the optimum pH for the photo-Fenton process is about 3.0, this particular system performed well in experimental conditions starting at alkaline and neutral pH. The results obtained here are promising for industrial applications, particularly in view of the high concentration of chloride, a known hydroxyl radical scavenger and the main oxidant present in photo-Fenton processes. ZUSAMMENFASSUNGDer glutamaterge N-Methyl-D-aspartat-Rezeptor (NMDA) ist ein wichtiger ionotroper Rezeptor, der die exzitatorische synaptische Transmission im zentralen Nervensystem von Säugetieren vermittelt. Der NMDA-Rezeptor nimmt unter den Glutamatrezeptoren dabei eine Sonderstellung ein, da er mit einer Reihe von neurodegenerativen Erkrankungen wie dem Morbus Parkinson, dem Morbus Huntington, dem Morbus Alzheimer, der Schizophrenie und der Epilepsie in Zusammenhang gebracht wird. Daher besteht ein großes Interesse an der Entwicklung geeigneter 18F-markierter NMDA-Rezeptorliganden zur nicht-invasiven Visualisierung des NMDA-Rezeptorkomplexes mittels der Positronenemissionstomographie.Die 19F-Analoga ADTC1, tADTC1 und tADTC3 - 5 und das nicht-fluorierte 12C-Analogon tADTC2 wurden synthetisiert und ihre in-vitro Affinität und Lipophilie bestimmt. Mit Ausnahme von ADTC1 und tADTC5 die mikromolare Affinitäten besitzen, haben die Liganden in [H-3]MDL-105,519 Rezeptorbindungsassays niedrige nanomolare Affinitäten für die Glycinbindungsstelle. Die Lipophilie der Verbindungen wurde mit drei verschiedenen Verfahren untersucht und ergab logD7,4-Werte von ungefähr 1 für cADTC1 und tADTC1 – 4, während tADTC5 mit einem logD7,4 von –1,15 eine sehr niedrige Lipophilie aufwies. Die Radiosynthesen der 18F-Liganden wurden hinsichtlich der Umsetzung der Markierungsvorläufer mit 2-[F-18]Fluorethyltosylat oder [F-18]Fluorid untersucht und optimiert. Die höchsten radiochemischen Ausbeuten von ungefähr 90% wurden, unter Verwendung von NaOH als Hilfsbase, bei der 18F-Fluorethylierung von t[F-18]ADTC4 und t[F-18]ADTC5 mit 2-[F-18]Fluorethyltosylat erzielt.
