We have used a transgenic cell line of Catharanthus roseus (L.) G. Don to study the relative importance of the supply of biosynthetic precursors for the synthesis of terpenoid indole alkaloids. Line S10 carries a recombinant, constitutively overexpressed version of the endogenous strictosidine synthase (Str) gene. Various concentrations and combinations of the substrate tryptamine and of loganin, the immediate precursor of secologanin, were added to suspension cultures of S10. Our results indicate that high rates of tryptamine synthesis can take place under conditions of low tryptophan decarboxylase activity, and that high rates of strictosidine synthesis are possible in the presence of a small tryptamine pool. It appears that the utilization of tryptamine for alkaloid biosynthesis enhances metabolic flux through the indole pathway. However, a deficiency in the supply of either the iridoid or the indole precursor can limit flux through the step catalyzed by strictosidine synthase. Precursor utilization for the synthesis of strictosidine depends on the availability of the cosubstrate; the relative abundance of these precursors is a cell-line-specific trait that reflects the metabolic status of the cultures.

The aim of this work was to investigate the alkaloid patterns of Lapiedra martinezii and their relation to biogeography and phenology focused in a phylogenetic comparison. Plants from 14 populations of L. martinezii, covering almost its entire distribution area, were subjected to morphological, ecological, and phytochemical analysis. Experiments for different alkaloid-type content are proposed as a new tool for analysis of plant distribution. Several plants were transplanted for weekly observation of their phenological changes, and alkaloids from different plant organs were extracted, listed, and compared. The alkaloid pattern of L. martinezii comprises 49 compounds of homolycorine, lycorine, tazettine, haemantamine, and narciclasine types. The populations located in the north and south margins of the distribution area displayed alkaloid patterns different from those of the central area. Changes in these patterns during their phenological cycle may be related to a better defence for plant reproduction. L. martinezii is an old relict plant, and it has maintained some of the more primitive morphological features and alkaloid profiles of the Mediterranean Amaryllidaceae. The variations in alkaloid content observed could be interpreted in a phylogenetic sense, and those found in their phenological changes, in an adaptive one.

In an attempt to reveal the relationships between alkaloid biosynthesis and phylogeny, we investigated by GC–MS the alkaloid patterns of 22 species and 3 hybrids (from 45 locations) from seven main sections of the genus Narcissus (Amaryllidaceae). The results indicate that the first alkaloids to evolve in the genus Narcissus were of the lycorine- and homolycorine-type. The alkaloid pattern of the Nevadensis section supports its recent separation from the Pseudonarcissus section. The plants of Narcissus pallidulus (Ganymedes section) show a predominance of Sceletium-type compounds, which are quite rare in the Amaryllidaceae family. Two successful evolutionary strategies involving alkaloid biosynthesis and leading to an expansion in taxa and occupied area were determined. Firstly, a diversification of alkaloid patterns and a high alkaloid concentration in the organs of the large Narcissus species (in the Pseudonarcissus section) resulted in an improved chemical defence in diverse habitats. Secondly, both plant size and alkaloid biosynthesis were reduced (in the Bulbocodium and Apodanthi sections) relegated to dry pastures and rocky places.

Chemical defenses are common among organisms and represent some of the most complex adaptations for avoiding predation, yet our understanding of the ecological nature of these systems remains incomplete. Poison frogs are a group of chemically defended organisms that are dependent entirely on diet for chemical defense. In this study, I identified the dietary arthropods responsible for chemical defense in poison frogs, described spatial and temporal patterns in alkaloid composition of poison frogs, and established links between patterns of variation in alkaloid defense and arthropod diet in poison frogs. Identifying dietary sources and studying patterns of variation in alkaloid composition is fundamental to understanding the ecology and evolution of chemical defense in poison frogs. ^ The dendrobatid poison frog Oophaga pumilio shares many alkaloids in common with other poison frogs and is known to vary in alkaloid composition throughout its geographic range. I designed my dissertation to take advantage of these characteristics and use O. pumilio as a model species for the study of chemical defense in poison frogs. Here, I identified siphonotid millipedes as a source for spiropyrrolizidine alkaloids, formicine ants as a source for pumiliotoxin alkaloids, and oribatid mites as dietary sources for the majority of alkaloids found in poison frogs. I found that alkaloid composition varied spatially and temporally, on both small and large scales, within and among populations of O. pumilio. Alkaloid variation between populations was related to geographic distance, and closer populations tended to have alkaloid compositions more similar to each other than to distant populations. ^ The findings of my study suggest that oribatid mites are the most important dietary source of alkaloids in poison frogs. However, overall alkaloid defense in poison frogs is based on a combination of dietary arthropods, including mites, ants, millipedes, and beetles. Variation in chemical defenses of poison frogs is due to (1) spatial and temporal differences in the presence of alkaloids in certain arthropods and (2) differences in the availability of certain alkaloid-containing arthropods, which are likely the result of differences as well as successional changes in forest structure among locations and through time. ^

A batata está sujeita a stresses bióticos (doenças, pragas, infestantes) e abióticos (condições climáticas, secura, excesso de água, ou fitotoxicidade provocada por herbicidas, entre outros). Os adubos foliares contendo nutrientes (macro e micronutrientes) poderão contribuir para uma melhor nutrição da planta, para além de exercer uma ação benéfica ao nível do potencial hídrico da folha e sua proteção contra os diversos inimigos bióticos. Instalou-se um ensaio em Marinhais na variedade de batata de indústria Lady Rosetta, para avaliar o efeito da adubação foliar de uma gama de adubos foliares “Humigel”, contendo, entre outros nutrientes, o enxofre e o potássio, numa situação de fitotoxidade causada pela aplicação de um herbicida. O ensaio foi instalado com um dispositivo experimental de parcelas totalmente aleatórias, com 4 repetições. Os tratamentos foram a aplicação de “Humigel” e sem aplicação (testemunha). A produção comercial nas parcelas com “Humigel” foi significativamente mais elevada (48 192 kg/ha) do que nas parcelas testemunha (43 458 kg/ha). Não se registaram diferenças significativas ao nível do número de tubérculos (comercial e total) por planta, nem ao nível dos parâmetros de qualidade (peso específico e teste de fritura).

El estudio de microtuberización del cultivar de papa Burren se realizó en el Laboratorio de Cultivo de Tejidos de la Universidad Nacional Agraria entre los meses de junio a diciembre del 2015. Se estudiaron las fases de multiplicación y formación de microtubérculos en Biorreactores Económicos de Inmersión Temporal (BEIT). En la fase de multiplicación se evaluó la formación de plantas completas cuando se sembraron 70 microesquejes en cinco variantes de medios de cultivo. Se empleó un diseño de Bloques Completos al Azar con arreglo unifactorial con réplicas conformadas por 2 BEIT, y para definir las diferencias estadísticas entre las medias (ANDEVA) de los tratamientos se realizó la prueba de rangos múltiples de Duncan-Waller ( = 0.05). En la formación de plantas se evaluaron la longitud de plantas, número de hojas y número de brotes que se les realizó la prueba de medias de diferencia mínima significativa con α = 0.05, se calculó el porcentaje de supervivencia. Como resultado se obtuvo en las variantes de medios de cultivo que contenían 0.20 mg l-1 de GA3 con 0.50 mg l-1 de BAP y 0.10 mg l-1 de GA3 con 1 mg l-1 de BAP, se obtuvieron medias respectivas en número de entrenudos de 6.32-5.7 y en número de hojas por planta de 7.37-6.93. Adiciones de sacarosa de 80 g l-1 y 110 g l-1 favorecieron la formación de microtubérculos de peso fresco con medias entre 0.69-0.75 gramos. La adición de sacarosa entre 80 g l-1 y 120 g l-1 no se registró diferencias estadísticas significativas entre las medias de las variables diámetro y longitud de microtubérculos. Con microtubérculos con diámetro mayor a los 10 mm se obtuvieron medias en longitud de planta, número de hojas y número de brotes de 21, 16.35 y 5.65 cm respectivamente y con diámetros entre 8-10 mm las medias respectivas fueron de 16.37, 11.35 y 5.35 cm.
