The activity of growing living bacteria was investigated using real-time and in situ rheology-in stationary and oscillatory shear. Two different strains of the human pathogen Staphylococcus aureus-strain COL and its isogenic cell wall autolysis mutant, RUSAL9-were considered in this work. For low bacteria density, strain COL forms small clusters, while the mutant, presenting deficient cell separation, forms irregular larger aggregates. In the early stages of growth, when subjected to a stationary shear, the viscosity of the cultures of both strains increases with the population of cells. As the bacteria reach the exponential phase of growth, the viscosity of the cultures of the two strains follows different and rich behaviors, with no counterpart in the optical density or in the population's colony-forming units measurements. While the viscosity of strain COL culture keeps increasing during the exponential phase and returns close to its initial value for the late phase of growth, where the population stabilizes, the viscosity of the mutant strain culture decreases steeply, still in the exponential phase, remains constant for some time, and increases again, reaching a constant plateau at a maximum value for the late phase of growth. These complex viscoelastic behaviors, which were observed to be shear-stress-dependent, are a consequence of two coupled effects: the cell density continuous increase and its changing interacting properties. The viscous and elastic moduli of strain COL culture, obtained with oscillatory shear, exhibit power-law behaviors whose exponents are dependent on the bacteria growth stage. The viscous and elastic moduli of the mutant culture have complex behaviors, emerging from the different relaxation times that are associated with the large molecules of the medium and the self-organized structures of bacteria. Nevertheless, these behaviors reflect the bacteria growth stage.

Esta dissertação enquadra-se no âmbito dos Sistemas de Informação, em concreto, no desenvolvimento de aplicações Web, como é o caso de um website. Com a utilização em larga escala dos meios tecnológicos tem-se verificado um crescimento exponencial dos mesmos, o que se traduz na facilidade com que podem ser encontradas na Internet diversos tipos de plataformas informáticas. Além disso, hoje em dia, uma grande parte das organizações possui o seu próprio sítio na Internet, onde procede à divulgação dos seus serviços e/ou produtos. Pretende-se com esta dissertação explorar estas novas tecnologias, nomeadamente, os diagramas UML - Unified Modeling Language e a concepção de bases de dados, e posteriormente desenvolver um website. Com o desenvolvimento deste website não se propõe a criação de uma nova tecnologia, mas o uso de diversas tecnologias em conjunto com recurso às ferramentas UML. Este encontra-se organizado em três fases principais: análise de requisitos, implementação e desenho das interfaces. Na análise de requisitos efectuou-se o levantamento dos objectivos propostos para o sistema e das necessidades/requisitos necessários à sua implementação, auxiliado essencialmente pelo Diagrama de Use Cases do sistema. Na fase de implementação foram elaborados os arquivos e directórios que formam a arquitectura lógica de acordo com os modelos descritos no Diagrama de Classes e no Diagrama de Entidade-Relação. Os requisitos identificados foram analisados e usados na composição das interfaces e sistema de navegação. Por fim, na fase de desenho das interfaces foram aperfeiçoadas as interfaces desenvolvidas, com base no conceito artístico e criativo do autor. Este aperfeiçoamento vai de encontro ao gosto pessoal e tem como objectivo elaborar uma interface que possa também agradar ao maior número possível de utilizadores. Este pode ser observado na maneira como se encontram distribuídas as ligações (links) entre páginas, nos títulos, nos cabeçalhos, nas cores e animações e no seu design em geral. Para o desenvolvimento do website foram utilizadas diferentes linguagens de programação, nomeadamente a HyperText Markup Language (HTML), a Page Hypertext Preprocessor (PHP) e Javascript. A HTML foi utilizada para a disposição de todo o conteúdo visível das páginas e para definição do layout das mesmas e a PHP para executar pequenos scripts que permitem interagir com as diferentes funcionalidades do site. A linguagem Javascript foi usada para definir o design das páginas e incluir alguns efeitos visuais nas mesmas. Para a construção das páginas que compõem o website foi utilizado o software Macromedia Dreamweaver, o que simplificou a sua implementação pela facilidade com que estas podem ser construídas. Para interacção com o sistema de gestão da base de dados, o MySQL, foi utilizada a aplicação phpMyAdmin, que simplifica o acesso à base de dados, permitindo definir, manipular e consultar os seus dados.

Sera of Chaga's disease patients containing anti-T. cruzi lytic antibodies were submitted to affinity chromatography using Sepharose 4B conjugated with antigen extracted from epimasiigote or trypomasiigote forms of the parasite. Epimastigotes were obtained from culture at the exponential growth phase and the trypomastigotes from blood of infected and immunosuppressed mice. Antigen of both parasite forms was obtained by sonication of the parasites followed by centrifugation. Both antigens were then conjugated to activated Sepharose 4B. Affinity chromatography was performed by passing sera from chagasic patients through an immunoadsorbent column containing either epimasiigote or trypomasiigote antigens. Antibodies bound to the column were eluted with cold 0,2 M glycine buffer pH 2,8. The eluted antibodies were analysed regarding their isotype and lytic activity. The results showed that anti-T. cruzi lytic antibodies present in sera from chagasic patients are mainly located in the IgG isotype and recognize epitopes present in both trypomasiigote and epimastigote forms. Abstract Background: Nanotechnology has the potential to provide agriculture with new tools that may be used in the rapid detection and molecular treatment of diseases and enhancement of plant ability to absorb nutrients, among others. Data on nanoparticle toxicity in plants is largely heterogeneous with a diversity of physicochemical parameters reported, which difficult generalizations. Here a cell biology approach was used to evaluate the impact of Quantum Dots (QDs) nanocrystals on plant cells, including their effect on cell growth, cell viability, oxidative stress and ROS accumulation, besides their cytomobility. Results: A plant cell suspension culture of Medicago sativa was settled for the assessment of the impact of the addition of mercaptopropanoic acid coated CdSe/ZnS QDs. Cell growth was significantly reduced when 100 mM of mercaptopropanoic acid -QDs was added during the exponential growth phase, with less than 50% of the cells viable 72 hours after mercaptopropanoic acid -QDs addition. They were up taken by Medicago sativa cells and accumulated in the cytoplasm and nucleus as revealed by optical thin confocal imaging. As part of the cellular response to internalization, Medicago sativa cells were found to increase the production of Reactive Oxygen Species (ROS) in a dose and time dependent manner. Using the fluorescent dye H2DCFDA it was observable that mercaptopropanoic acid-QDs concentrations between 5-180 nM led to a progressive and linear increase of ROS accumulation. Conclusions: Our results showed that the extent of mercaptopropanoic acid coated CdSe/ZnS QDs cytotoxicity in plant cells is dependent upon a number of factors including QDs properties, dose and the environmental conditions of administration and that, for Medicago sativa cells, a safe range of 1-5 nM should not be exceeded for biological applications.

RESUMO - Assiste-se a um crescimento exponencial das despesas em saúde, quer na Europa como nos Estados Unidos. Em Portugal, os gastos totais com a saúde ascenderam a 10,2% do PIB, em 2006, contra os 8,8% registados no início da década anterior. É importante perceber o que motiva este crescimento quer em termos globais, quer no que diz respeito ao consumo de recursos, bem como até em termos da despesa pública. Este projecto tem dois objectivos fundamentais: em primeiro lugar, contribuir para o estudo dos factores determinantes da procura de cuidados de saúde em Portugal e, consequentemente, determinar as elasticidades procura – preço para diferentes tipos de cuidados de saúde. Metodologia: Estudo observacional baseado na análise empírica de dados administrativos (claims) respeitantes à utilização dos cuidados de saúde por parte de 12.230 indivíduos detentores de um plano de seguro de saúde individual, numa seguradora privada em Portugal. As elasticidades procura – preço para os diferentes tipos de cuidados de saúde obtiveram-se utilizando as variações percentuais das quantidades dos diferentes cuidados de saúde, antes e depois da variação do preço pago pelo indivíduo, para cada tipo de cuidado de saúde. Resultados: De acordo com a teoria económica tradicional o aumento do preço a pagar reduz o consumo de cuidados de saúde, e a procura é elástica, ou seja, os valores da elasticidade procura – preço obtidos são superiores a 1, em valor absoluto, logo o aumento do preço levou a uma redução mais do que proporcional das quantidades procuradas. A procura de cuidados de saúde em ambulatório é mais sensível à variação do preço do que a procura de cuidados de internamento. ------- ABSTRACT - We are witnessing an exponential growth of health care expenditures around the world. In Portugal, the total expenditure on health amounted to 10.2% of GDP in 2006, against 8.8% at the beginning of previous decade. It is important to understand what motivates this growth both in overall terms, with respect to resource consumption, and even in terms of public spending. This study was designed two achieve two objectives: first, to contribute to the study of demand for health care and, more specifically, to analyze the effect of price changes on the utilization of health care services; and secondly, to estimate the demand elasticity for different types of heath care. Methodology: Observational study based on empirical analysis of administrative data (claims) from a private health insurance Company in Portugal. The sample used had information regarding 12.230 individuals. Demand elasticity for the different types of health care services was obtained by the quotient between the percentage changes in the quantity of health care services, before and after the change in the price paid by the corresponding percentage change in the price. Results: This study showed that, for all medical services, price increases were associated with reductions in the quantity of care consumed as predicted by neoclassical demand theory, and we are in the presence of an elastic demand. This means that price elasticity is greater than 1 in absolute value so the increase in the price led to a more than proportional reduction in the quantity demanded. Demand elasticity was more responsive to changes in the price of specialist and emergency care than to changes in the price of inpatient care.

Siderophore production by Bacillus megaterium was detected, in an iron-deficient culture medium, during the exponential growth phase, prior to the sporulation, in the presence of glucose; these results suggested that the onset of siderophore production did not require glucose depletion and was not related with the sporulation. The siderophore production by B. megaterium was affected by the carbon source used. The growth on glycerol promoted the very high siderophore production (1,182 μmol g−1 dry weight biomass); the opposite effect was observed in the presence of mannose (251 μmol g−1 dry weight biomass). The growth in the presence of fructose, galactose, glucose, lactose, maltose or sucrose, originated similar concentrations of siderophore (546–842 μmol g−1 dry weight biomass). Aeration had a positive effect on the production of siderophore. Incubation of B. megaterium under static conditions delayed and reduced the growth and the production of siderophore, compared with the incubation in stirred conditions.

In the present paper we assess the performance of information-theoretic inspired risks functionals in multilayer perceptrons with reference to the two most popular ones, Mean Square Error and Cross-Entropy. The information-theoretic inspired risks, recently proposed, are: HS and HR2 are, respectively, the Shannon and quadratic Rényi entropies of the error; ZED is a risk reflecting the error density at zero errors; EXP is a generalized exponential risk, able to mimic a wide variety of risk functionals, including the information-thoeretic ones. The experiments were carried out with multilayer perceptrons on 35 public real-world datasets. All experiments were performed according to the same protocol. The statistical tests applied to the experimental results showed that the ubiquitous mean square error was the less interesting risk functional to be used by multilayer perceptrons. Namely, mean square error never achieved a significantly better classification performance than competing risks. Cross-entropy and EXP were the risks found by several tests to be significantly better than their competitors. Counts of significantly better and worse risks have also shown the usefulness of HS and HR2 for some datasets.
