Mesenchymal stem cells (MSCs) are a heterogeneous population of cells that proliferate in vitro as plastic-adherent cells, have fibroblast-like morphology and can differentiate into bone, cartilage and fat cells. Therapeutic potential of MSCs have been studied in experimental models, such as rabbit, in Laboratory of Cell Engineering of Botucatu. However, no specific markers have been reported for expanded rabbit MSCs, which hampers the isolation of pure MSC populations by immunophenotypic characterization. Thus, the objective of this study was to produce monoclonal antibodies (mAbs) to rabbit MSCs. MSCs derived from rabbit bone marrow (BM) were isolated, cultured, expanded ex vivo, and immunized into three BALB/c mices, and spleen cells subsequently harvested were used to generate hibridoma cell lines secreting antibodies against MSCs. Hybridoma cells were screened by flow cytometry and antibody-producing cells were subjected to subsequent rounds of retests. MSC1-160 obtained the best positivity for IgG expression and was cloned by limiting dilutions and micromanipulation. Ascitic fluid from ten best clones was purified by affinity chromatography in Protein A-sepharose CL-4B column and purification control was performed by electrophoresis in agarose gels. The purified IgG were tested against rabbit MSCs, obtaining high positivity by flow Cytometry. In conclusion, we developed 10 mAbs, MSC1-160 A20, A30, A41, A47, A55, A60, A63, A69, A81, and A82, that recognize rabbit MSC cell surface antigens showing potential for immunophenotypic characterization of rabbit MSC cell lines

Water is an essential factor in maintaining the vital functions of living beings. We have observed a growing commitment of quality, are due to pollution from many sources and even entire watersheds, whether for industrial waste, sewage, or for substances used in farming such as pesticides, herbicides and fertilizers. Nickel is the 24th most abundant element on earth, is a heavy metal that, in the form of chloride, is a proven genotoxic and mutagenic. Due to its industrial use, there was considerable increase of its concentration in surface sediments. Fish combine characteristics that make them excellent experimental models for aquatic toxicology studies, which are particularly usable as warn about the potential danger of chemicals or the possibility of environmental pollution. Due to impaired water quality and the few published studies relating the nickel with the tissue change, this study aimed at assessing the consequences of the presence of nickel in the aquatic environment. For this analysis, we used individuals of Oreochromis niloticus, exposed for 96 hours at three different concentrations of nickel dissolved in water compared to a control group. After exposure, the gills were removed and these were analyzed by ultramorphological, histological and histochemical analysis. The results indicate that all concentrations used in the experiment altered the histophysiology of exposed individuals. We observed the following changes: rupture of paviment cells, thus resulting in bleeding, loss of microridges surface of these cells and epithelial loss in the gills of all animals in all treatments with nickel chloride, the histochemical analysis showed non-proliferation of chloride cells. However, there was a dose-dependent increase of mucus cells in all animals. Experimental models composed by human and animal cell lines are simplified and informative, allowing them to be widely used for biomedical research. Most laboratories that use in vitro cultivated cells maintain a variation of cell lines stored and cultivated. Therefore, misidentification and cross-contamination events can happen during cell lines handling. This problem can generate a repertoire of dubious results and papers, which may prejudice biomedical research. Recently it was created the International Cell Line Authentication Committee (ICLAC), which aims to spread knowledge about cross-contamination and misidentification of in vitro cell lines. Despite of the efforts spent trying to aware scientific community about the importance of the correct identification of cells, the number of papers based on misidentified cell lines it´s still worrying, compromising the reliability of out coming results and conclusions regarding them. The present study aims to analyze and discuss the main advantages and limitations of eukaryote in vitro cell lines use, characterizing the cell lines authentication problems. Therefore, compilation and critical analyses of literature data was realized, aiming to improve the understanding about this subject. Based on information about 445 cell lines with issues published by ICLAC it´s clear that contamination in human cell lines represented 89,2 % of mentioned problems. HeLa cell line was the responsible for most contamination, especially in 92 normal tissue cell lines, representing 44,6% of the contamination. These results reinforce the importance of periodic maintenance of cell lines cultures by labs and implementation of authentication methods as polymorphic STRs, besides obtaining cell lines from reliable sources and cell banks

Apocynin has been used as an efficient inhibitor of the multi-enzymatic complex NADPH oxidase in many experimental models involving phagocytic and nonphagocytic cells. The mechanism of inhibition has been linked with the previous activation of apocynin through the action of cellular peroxidases leading to the formation of a dimeric oxidation product, diapocynin. In this study we compared apocynin with pure diapocynin regarding their effects as scavenger of hydrogen peroxide and hypochlorous generated by glucose/glucose oxidase and myeloperoxidase respectively, and as inhibitors of the production of hydrogen peroxide and hypochlorous acid by activated neutrophils. The production of hydrogen peroxide was measured by the oxidation of the fluorescent substance Amplex Red and the production of hypochlorous acid by was measured as taurine-chloramine derivative using the chromogenic substrate 3,3’,5,5’- tetramethylbenzidine (TMB). Neutrophils (1 x106 cells/mL) were pre-incubated in PBS buffer supplemented with 1 mM calcium chloride, 0.5 mM magnesium chloride, 1 mg/mL glucose and 5 mM taurine in the presence or absence of inhibitors. The reactions were triggered by adding the soluble stimulus Forbol Miristate Acetate PMA or zymosan and incubated by additional 30 minutes. We found that pure diapocynin was not better than apocynin regarding its scavenger and inhibitory properties. These results suggest that the formation of diapocynin is not essential for the action of apocynin as inhibitor of NADPH oxidase activation

Experimental models are necessary to elucidate pathophysiological mechanisms not yet understood in humans. To evaluate the repercussions of the diabetes, considering two methodologies, on the pregnancy of Wistar rats and on the development of their offspring. In the 1st induction, female offspring were distributed into two experimental groups: Group streptozotocin (STZ, n=67): received the β-cytotoxic agent (100mg STZ/kg body weight - sc) on the 1st day of the life; and Non-diabetic Group (ND, n=14): received the vehicle in a similar time period. In the adult life, the animals were mated. After a positive diagnosis of pregnancy (0), female rats from group STZ presenting with lower glycemia than 120 mg/dL received more 20 mg STZ/kg (ip) at day 7 of pregnancy (2nd induction). The female rats with glycemia higher than 120mg/dL were discarded because they reproduced results already found in the literature. In the mornings of days 0, 7, 14 and 21 of the pregnancy glycemia was determined. At day 21 of pregnancy (at term), the female rats were anesthetized and killed for maternal reproductive performance and fetal development analysis. The data were analyzed using Student-Newman-Keuls, Chi-square and Zero-inflated Poisson (ZIP) Tests (p<0.05). STZ rats presented with increased rates of pre (STZ=22.0%; ND=5.1%) and post-implantation losses (STZ=26.1%; ND=5.7%), reduced rates of fetuses with appropriate weight for gestational age (STZ=66%; ND=93%) and reduced degree of development (ossification sites). Conclusion: Mild diabetes led a negative impact on maternal reproductive performance and caused intrauterine growth restriction and impaired fetal development

Even though the presence of the female prostate has been reported in many species, including humans, bats and several rodents, it has many anatomical and histological variations. There is still plentiful discussion on the biological function of this organ. Many authors state that paraurethral ducts and glands are functional and homologous to the male prostate. The use of experimental models and a better knowledge of the female prostate gland in other species, can be useful to veterinary medicine as well as human medicine. Therefore the aim of this study is to check for the presence of this gland in female dogs of various breeds and age. For that purpose 25 urethras, from the bladder to the vulva, were collected, fixed in 10% neutral buffered formalin, routinely processed and sectioned into 4 slides of 4 µm, each with 40 µm gap between each set of 4 slides, using an automatic microtome and stained with Hematoxylin and Eosine (HE). The HE sections were evaluated for the presence of prostatic gland in the sample. Unstained tissue sections cut from paraffin blocks were marked with a polyclonal anti-PSA primary antibody. The prevalence of the gland was 32% (8/25). The structure of the paraurethral PSA-positive gland was acinar, organized in buds, with secretory epithelium varying from cubic to columnar; eccentric nuclei, with lose chromatin and a layer of basal cells, very similar to the male prostate were observed. In view of these characteristics, for the first time in the literature, was demonstrate that those glands. may be considered as female prostate in dogs, as they are in other vertebrates

The purpose of this literature review is to present the state-of-the-art on the application of tissue engineering techniques in sinus lifts procedures, reporting the influence of these techniques in increasing bone height and volume in the maxillary sinus, and the osseointegration of the implants installed in the grafted areas. PubMed and Scopus databases were searched using the keywords “scaffold” OR “engineered tissue” AND “sinus augmention” OR “sinus floor elevation”. In the initial search 463 articles were selected, of which 19 were selected abstracts reading, with nine papers selected by the end, for evaluation of the application of tissue engineering techniques in sinus lift procedures. Despite the positive results of tissue engineering procedures in sinus floor elevation reported in the selected articles, further studies are still needed, for a better standardization of experimental models and materials used, leading to definitive conclusions about the effects of the application of tissue engineering procedures on bone formation in maxillary sinus lifting procedures.

AIM: The aim of this study was to evaluate, through a systematic review of the literature, the effects of different implant surface modifications on osseointegration in diabetic subjects. METHODS: A search was performed of the PubMed database, using a combination of the following keywords: “Implant surface” OR “Dental implants” AND “Diabetes” OR “Hyperglycemia”. Papers published in English between January 1960 and November 2013 were selected. All experimental models were considered in this search, but case reports and in vitro studies were excluded from this review. RESULTS: The initial search identified 182 articles. After reading the titles and abstracts, 39 articles were selected for full reading. Finally, 4 papers were selected after evaluation of all the papers, and these papers are discussed in this review. Due to the methodological heterogeneity of the selected studies, it was not possible to perform a meta-analysis of the data. CONCLUSION: It can be concluded that although the benefits of surface modifications present in individuals with diabetes have biological plausibility, there is little evidence of the benefits of these modifications.

Results of the analysis of dynamic behavior of flashover phenomenon on the high voltage-polluted insulators are presented. These results were taken from a mathematical and an experimental model that introduce the variable thickness influence of the layer pollution deposited on the high-voltage insulator surface. Analysis of the flashover was done by way of introducing a variation in the thickness of the channel of Obenaus' model, simulating a layer pollution of variable thickness. The objective was to obtain a better reproduction of the real layer pollution deposited on the insulator that works in the polluted regions. Two types of thickness variations were used: a sudden variation, using a step; and a soft variation, using a ramp; that were put along the way of the discharge. Comparison between the mathematical and experimental models showed that introduction of a ramp makes Obenaus' model more efficient in analyzing behavior of flashover phenomenon.
