A kinetic study of the hydrolytic stabilities of mono-, di-, and 2-chloro-2-deoxy septanosides, under acid-catalysis, is reported herein. A comparison of mono-and diseptanosides, shows that the glycosidic bond in the disaccharide is more stable than the monosaccharide. Further the glycosidic bond at the reducing end hydrolyzes almost twice as faster than that of the non-reducing end of the disaccharide. 2-Chloro-2-deoxy septanoside is found to be the most stable and its glycosidic bond hydrolysis occurs at elevated temperatures only. The orientation of the exo-cyclic hydroxymethyl group and the inductive effect are suggested to play a role in the rates of hydrolysis. (C) 2014 Elsevier Ltd. All rights reserved.

Bacteria have evolved to survive the ever-changing environment using intriguing mechanisms of quorum sensing (QS). Very often, QS facilitates formation of biofilm to help bacteria to persist longer and the formation of such biofilms is regulated by c-di-GMP. It is a well-known second messenger also found in mycobacteria. Several methods have been developed to study c-di-GMP signaling pathways in a variety of bacteria. In this review, we have attempted to highlight a connection between c-di-GMP and biofilm formation and QS in mycobacteria and several methods that have helped in better understanding of c-di-GMP signaling. The bacterial second messengers (p)ppGpp and bis-(3'-5')-cyclic dimeric GMP (c-di-GMP) regulate important functions, such as transcription, virulence, biofilm formation, and quorum sensing. In mycobacteria, they regulate long-term survival during starvation, pathogenicity, and dormancy. Recently, a Pseudomonas aeruginosa strain lacking (p) ppGpp was shown to be sensitive to multiple classes of antibiotics and defective in biofilm formation. We were interested to find out whether Mycobacterium smegmatis strains lacking the gene for either (p)ppGpp synthesis (Delta rel(Msm)) or c-di-GMP synthesis (Delta dcpA) would display similar phenotypes. We used phenotype microarray technology to compare the growth of the wild-type and the knockout strains in the presence of several antibiotics. Surprisingly, the Delta rel(Msm) and Delta dcpA strains showed enhanced survival in the presence of many antibiotics, but they were defective in biofilm formation. These strains also displayed altered surface properties, like impaired sliding motility, rough colony morphology, and increased aggregation in liquid cultures. Biofilm formation and surface properties are associated with the presence of glycopeptidolipids (GPLs) in the cell walls of M. smegmatis. Thin-layer chromatography analysis of various cell wall fractions revealed that the levels of GPLs and polar lipids were reduced in the knockout strains. As a result, the cell walls of the knockout strains were significantly more hydrophobic than those of the wild type and the complemented strains. We hypothesize that reduced levels of GPLs and polar lipids may contribute to the antibiotic resistance shown by the knockout strains. Altogether, our data suggest that (p)ppGpp and c-di-GMP may be involved in the metabolism of glycopeptidolipids and polar lipids in M. smegmatis.

A series of 2,5-di(4-aryloylaryloxymethyl)-1,3,4-oxadiazoles 9a-j were obtained via multistep synthesis from hydroxybenzophenones 4a-e. The cytotoxicity of compounds 9a-j was evaluated against human leukemia cell lilies (K562 and CEM). The compounds exhibited moderate to good anti-cancer activity with compounds 9b and 9i having a chloro group exhibiting the best activity (IC50 = 10 mu M). Compound 9i exhibited activity against both the cell lines and 9b only exhibited activity against CEM. Further, a lactate dehydrogenase (LDH) assay and DNA fragmentation studies of the compounds 9a-j were also performed. (C) 2013 Elsevier Masson SAS. All rights reserved.

The relative energies of triangular face sharing condensed macro polyhedral carboranes: CB20H18 and C2B19H18+ derived from mono- and di-substitution of carbons in (4) B21H18- is calculated at B3LYP/6-31G* level. The relative energies, H center dot center dot center dot H non-bonding distances, NICS values, topological charge analysis and orbital overlap compatibility connotes the face sharing condensed macro polyhedral mono-carboranes, 8 (4-CB20H18) to be the lowest energy isomer. The di-carba- derivative, (36) 4,4'a-C2B19H18+ with carbons substituted in a different B-12 cage in (4) B21H18- in anti-fashion is the most stable isomer among 28 possibilities. This structure has less non-bonding H center dot center dot center dot H interaction and is in agreement with orbital-overlap compatibility, and these two have the pivotal role in deciding the stability of these clusters. An estimate of the inherent stability of these carboranes is made using near-isodesmic equations which show that CB20H18 (8) is in the realm of the possible. (C) 2015 Elsevier B.V. Sepsis is a life threatening condition resulting from a high burden of infection. It is a major health care problem and associated with inflammation, organ dysfunction and significant mortality. However, proper understanding and delineating the changes that occur during this complex condition remains a challenge. A comparative study involving intra-peritoneal injection of BALB/c mice with Salmonella Typhimurium (infection), lipopolysaccharide (endotoxic shock) or thioglycollate (sterile peritonitis) was performed. The changes in organs and sera were profiled using immunological assays and Fourier Transform Infrared (FTIR) micro-spectroscopy. There is a rapid rise in inflammatory cytokines accompanied with lowering of temperature, respiratory rate and glucose amounts in mice injected with S. Typhimurium or lipopolysaccharide. FTIR identifies distinct changes in liver and sera: decrease in glycogen and protein/lipid ratio and increase in DNA and cholesteryl esters. These changes were distinct from the pattern observed in mice treated with thioglycollate and the differences in the data obtained between the three models are discussed. The combination of FTIR spectroscopy and other biomarkers will be valuable in monitoring molecular changes during sepsis. It was demonstrated in earlier work that, by approximating its range kernel using shiftable functions, the nonlinear bilateral filter can be computed using a series of fast convolutions. Previous approaches based on shiftable approximation have, however, been restricted to Gaussian range kernels. In this work, we propose a novel approximation that can be applied to any range kernel, provided it has a pointwise-convergent Fourier series. More specifically, we propose to approximate the Gaussian range kernel of the bilateral filter using a Fourier basis, where the coefficients of the basis are obtained by solving a series of least-squares problems. The coefficients can be efficiently computed using a recursive form of the QR decomposition. By controlling the cardinality of the Fourier basis, we can obtain a good tradeoff between the run-time and the filtering accuracy. In particular, we are able to guarantee subpixel accuracy for the overall filtering, which is not provided by the most existing methods for fast bilateral filtering. We present simulation results to demonstrate the speed and accuracy of the proposed algorithm.

The alarmone (p)ppGpp regulates transcription, translation, replication, virulence, lipid synthesis, antibiotic sensitivity, biofilm formation, and other functions in bacteria. Signaling nucleotide cyclic di-GMP (c-di-GMP) regulates biofilm formation, motility, virulence, the cell cycle, and other functions. In Mycobacterium smegmatis, both (p) ppGpp and c-di-GMP are synthesized and degraded by bifunctional proteins Rel(Msm) and DcpA, encoded by rel(Msm) and dcpA genes, respectively. We have previously shown that the Delta rel(Msm) and Delta dcpA knockout strains are antibiotic resistant and defective in biofilm formation, show altered cell surface properties, and have reduced levels of glycopeptidolipids and polar lipids in their cell wall (K. R. Gupta, S. Kasetty, and D. Chatterji, Appl Environ Microbiol 81:2571-2578, 2015, http://dx.doi.org/10.1128/AEM.03999-14). In this work, we have explored the phenotypes that are affected by both (p) ppGpp and c-di-GMP in mycobacteria. We have shown that both (p) ppGpp and c-di-GMP are needed to maintain the proper growth rate under stress conditions such as carbon deprivation and cold shock. Scanning electron microscopy showed that low levels of these second messengers result in elongated cells, while high levels reduce the cell length and embed the cells in a biofilm-like matrix. Fluorescence microscopy revealed that the elongated Delta rel(Msm) and Delta dcpA cells are multinucleate, while transmission electron microscopy showed that the elongated cells are multiseptate. Gene expression analysis also showed that genes belonging to functional categories such as virulence, detoxification, lipid metabolism, and cell-wall-related processes were differentially expressed. Our results suggests that both (p) ppGpp and c-di-GMP affect some common phenotypes in M. smegmatis, thus raising a possibility of cross talk between these two second messengers in mycobacteria. IMPORTANCE Our work has expanded the horizon of (p) ppGpp and c-di-GMP signaling in Gram-positive bacteria. We have come across a novel observation that M. smegmatis needs (p) ppGpp and c-di-GMP for cold tolerance. We had previously shown that the Delta rel(Msm) and Delta dcpA strains are defective in biofilm formation. In this work, the overproduction of (p) ppGpp and c-di-GMP encased M. smegmatis in a biofilm-like matrix, which shows that both (p) ppGpp and c-di-GMP are needed for biofilm formation. The regulation of cell length and cell division by (p) ppGpp was known in mycobacteria, but our work shows that c-di-GMP also affects the cell size and cell division in mycobacteria. This is perhaps the first report of c-di-GMP regulating cell division in mycobacteria.

采用一种非接触的光学方法傅立叶变换莫尔法(Fourier transform method),结合数字图像处理技术,对微幅振荡的水表面波的振幅进行测量.它是对全场中每一个像素点进行测量,比接触测量法具有更高的灵敏度.它为微幅水表面波振幅的测量提供了一种手段.通过将计算机生成的周期性光栅图像经投影机直接投影到被测物体的参考平面,经CCD摄像头、图像板捕捉存储形成数字化的光栅图像,利用傅立叶变换莫尔法处理光栅图像,从而获得包含有水表面波的振幅的相位信息,再经适当的几何变换获得振幅信息.我们在垂直振荡装置上进行了不同激励频率和不同振幅的表面波的振幅测量.

Resumen: La vera intenzione teologica di Anselmo d’Aosta nello scrivere il Proslogion e il vero significato del suo celebre unum argumentum vanno visti nella funzione che la ragione svolge necessariamente all’interno della vita di fede del cristiano. Anselmo, come Tommaso d’Aquino, non sostiene che l’esistenza di Dio sia un “articulus fidei” ma piuttosto uno dei “praeambula fidei”. La ragione naturale ha la certezza che Dio esiste, ancora prima della dimostrazione metafisica, e questo non fa che confermare l’assurdità di pensare che non esista il fondamento reale di tutte le cose esistenti.

采用逐次平均方法,构造一种快速逼近Fourier级数和实用算子(逐次平均算子),用这种算子来取代Fourier级数无穷项求和,具有误差小,收敛速度快的功能。志Fejer算子比,它的逼近性要好得多,与Korbin算子比,它更易于实用,理论证明其一般性,计算著名的Saffman-Taylor粘性指进问题时应用逐次平均算子,既不产生大的截断误差,又能避免高频振荡而使计算过程稳定,得到很好的结果。

Abstract: The late antique destruction of two bronze statues of Pausanias - the Spartan general responsible for the Greek victory at the Battle of Plataea (479 BC) - housed in the temple of Athena Chalkioikos in Sparta (Lib. Ep. 1518), has been interpreted as one of the few cases of a violent conflict between pagan and Christian population in Greece. Nevertheless the sources suggest that late antique Sparta was a bastion of Hellenic paganism and give a picture of a small and quiet town ruled by a pagan educated élite, where pagans like Libanius wanted to live. Since there is no evidence of a violent conflict between pagans and Christians in Sparta, and Libanius confirms that in 365 AD all the temples and cult statues were still in place, this paper addresses the issue from a different point of view and offers a new contribution to the history of Sparta in Late Antiquity. By using literary, archaeological and epigraphic evidence the paper explores: 1) the relationship between Roman administration and Spartan élite in the IVth century AD; 2) the historical memory of Pausanias in Late Antiquity. It will be emphasized that the obscure burning of the two statues helped to remove from Sparta the memory of Pausanias - a controversial figure, misrepresented in Late Antiquity and connected to the ancient staseis in Laconia - in order to promote a positive image of Sparta as a city without conflicts and ruled by the political system of Lycurgus (eunomia). As documented by local inscriptions in praise of late Roman governors, the mythical lawgiver Lycurgus was the paradigm of the imperial governors who rebuilded the town in the IVth cent. AD. It can be assumed that while Rome, Constantinople, Antioch and Athens were troubled by political and religious violence or by seditions between different factions, Sparta aimed to revive its traditional model of civic order in the new historical context of Late Antiquity.

La técnica de producción en Floating system constituye una alternativa económica y veloz de producción en hidroponia. Este sistema permite obtener hojas de vegetales de pequeño tamaño “Baby leaf” listas para consumir (IV Gama), cuyo consumo a nivel mundial muestra una tendencia creciente. Dicha técnica no se encuentra aún muy difundida para la producción de radicchio rosso. El objetivo de este trabajo fue evaluar el efecto del tamaño de celda, del substrato y de la técnica de floating system sobre la producción de radicchio. Los ensayos se realizaron en cámara a 25 ºC con un fotoperíodo de 16 hs de luz y 8 de oscuridad y con una intensidad de luz de 10 000 Lux. Se utilizó un diseño experimental factorial totalmente aleatorizado, con 5 repeticiones y 3 factores, cuyos niveles fueron: 1) Tamaño de celda, (T), con 3 niveles: grande, mediano y pequeño, 2) Técnica de producción, (F), con 2 niveles: floating system y sistema convencional, 3) Sustrato, (S), con 2 niveles: vermiculita y mezcla. Se analizaron las siguientes variables: 1) peso fresco, 2) peso seco, 3) largo de planta, 4) número de hojas y 5) área foliar. Los resultados obtenidos resultan alentadores respecto del empleo de la técnica de floating system. Para la obtención de plántulas con calidad de IV Gama resulta más conveniente el empleo de celdas de 80 cm3 y sustrato mezcla.
