Covalently attached benzimidazole molecules on silica gel surface, ≡SiL (where L = N-propyl-benzimidazole), adsorbs Co(ClO4)2 from non-aqueous solvent by forming a surface complex according to the reaction: m ≡SiL + Co(ClO4)2 → (≡SiL)mCo(ClO4)2. The equilibrium constant and the adsorption capacity, determined by applying the Langmuir equation were b = 3.0 × 103 L mol-1 and Ns= 0.098 × 10-3 mol g-1, respectively. The metal is bonded through the nitrogen atom and the perchlorate ion is not coordinated. The ESR study indicated that the complex has essentially an octahedral geometry with tetragonal distortion, with the electrons of the four nitrogen atoms interacting with the cobalt central metal ion in the equatorial plane. Only one complex species was detected on the surface.

The objective of this experiment was to determine the protein requirements for hen pullets from 1 to 18 weeks of age, by factorial method, using the nitrogen balance and the comparative slaughtering techniques. Protein requirements for maintenance, was obtained by the nitrogen balance technique using four diets with different protein levels (18,9, 4 and 2% of CP) aiming to obtain positive balance, next to zero and negative nitrogen balance. The endogenous nitrogen losses (0.2575 g of N/kg·75/day) was obtained by regression of nitrogen balance (NB) on ingestion nitrogen (IN). The requirements of nitrogen for maintenance was estimated by the intercept of axis X (0.3831 g of N/kg·75/day). The slope of the straight line still supplied the efficiency of N of the diet (67.21%). The regression coefficients of the equation represented the requirements of net nitrogen for weight gain. Considering the conversion efficiencies of nitrogen of the diet into nitrogen for weight gain, the requirements of nitrogen were determined for weight gain of .065, .087g and .090 g of N per gram of weight gain, for the phases 1 to 6, 7 to 12, and 13 to 18 weeks of age, respectively. Based on the results, three equations of prediction of the daily nitrogen requirements were fitted in function of live weight (LW in kg) and daily weight gain (G in g): 1 to 6 weeks N=.3831 x BW·75 + G .065, 7 to 12 weeks N=.3831 × BW·75 + G .087, and 13 to 18 weeks N=.3831 × BW·75 + G .090.

We consider some existing relativistic models for the nucleon structure functions, relying on statistical approaches instead of perturbative ones. These models are based on the Fermi-Dirac distribution for the confined quarks, where a density of energy levels is obtained from an effective confining potential. In this context, it is presented some results obtained with a recent statistical quark model for the sea-quark asymmetry in the nucleon. It is shown, within this model, that experimental available observables, such as the ratio and difference between proton and neutron structure functions, are quite well reproduced with just three parameters: two chemical potentials used to reproduce the valence up and down quark numbers in the nucleon, and a temperature that is being used to reproduce the Gottfried sum rule violation. © 2010 American Institute of Physics.

Cochlear root neurons (CRNs) are the first brainstem neurons which initiate and participate in the full expression of the acoustic startle reflex. Although it has been suggested that a cholinergic pathway from the ventral nucleus of the trapezoid body (VNTB) conveys auditory prepulses to the CRNs, the neuronal origin of the VNTB-CRNs projection and the role it may play in the cochlear root nucleus remain uncertain. To determine the VNTB neuronal type which projects to CRNs, we performed tract-tracing experiments combined with mechanical lesions, and morphometric analyses. Our results indicate that a subpopulation of non-olivocochlear neurons projects directly and bilaterally to CRNs via the trapezoid body. We also performed a gene expression analysis of muscarinic and nicotinic receptors which indicates that CRNs contain a cholinergic receptor profile sufficient to mediate the modulation of CRN responses. Consequently, we investigated the effects of auditory prepulses on the neuronal activity of CRNs using extracellular recordings in vivo. Our results show that CRN responses are strongly inhibited by auditory prepulses. Unlike other neurons of the cochlear nucleus, the CRNs exhibited inhibition that depended on parameters of the auditory prepulse such as intensity and interstimulus interval, showing their strongest inhibition at short interstimulus intervals. In sum, our study supports the idea that CRNs are involved in the auditory prepulse inhibition of the acoustic startle reflex, and confirms the existence of multiple cholinergic pathways that modulate the primary acoustic startle circuit. © 2013 Springer-Verlag Berlin Heidelberg.

A number of studies have demonstrated that simple elastic network models can reproduce experimental B-factors, providing insights into the structure-function properties of proteins. Here, we report a study on how to improve an elastic network model and explore its performance by predicting the experimental B-factors. Elastic network models are built on the experimental C coordinates, and they only take the pairs of C atoms within a given cutoff distance r(c) into account. These models describe the interactions by elastic springs with the same force constant. We have developed a method based on numerical simulations with a simple coarse-grained force field, to attribute weights to these spring constants. This method considers the time that two C atoms remain connected in the network during partial unfolding, establishing a means of measuring the strength of each link. Silica gel having a particle size between 0.2 and 0.05 mm and a specific surface area, S BET = 473 m 2 g -1, was chemically modified with benzimidazole. Adsorption isotherms of CuX 2 (X = Cl, Br or ClO 4) from ethanol and acetone solutions were studied at 298 K. The metal is bonded to the surface through the free nitrogen atom of the attached benzimidazole. The average number of ligands co-ordinated to the central metal ion was shown to depend on the solid surface loading by the solute. At low loading the electronic and ESR spectral parameters indicated that the copper ion is in a distorted-tetragonal symmetry field.

This study reports the isolation and biochemical characterization of two different serine proteases from Bothrops pirajai snake venom, thus providing a comparative analysis of the enzymes. The isolation process consisted of three consecutive chromatographic steps (Sephacryl S-200, Benzamidine Sepharose and C2/C18), resulting in two serine proteases, named BpirSP27 and BpirSP41 after their molecular masses by mass spectrometry (27,121 and 40,639 Da, respectively). Estimation by SDS-PAGE under denaturing conditions showed that, when deglycosylated with PNGase F, BpirSP27 and BpirSP41 had their molecular masses reduced by approximately 15 and 42%, respectively. Both are acidic enzymes, with pI of approximately 4.7 for BpirSP27 and 3.7 for BpirSP41, and their N-terminal amino acid sequences showed 57% identity to each other, with high similarity to the sequences of other snake venom serine proteases (SVSPs). The enzymes showed different actions on bovine fibrinogen, with BpirSP27 acting preferentially on the B beta chain and BpirSP41 on both A alpha and B beta chains. The two serine proteases were also able to degrade fibrin and blood clots in vitro depending on the doses and incubation periods, with higher results for BpirSP41. Both enzymes coagulated the human plasma in a dose-dependent manner, and BpirSP41 showed a higher coagulant potential, with minimum coagulant dose (MCD) of similar to 3.5 mu g versus 20 mu g for BpirSP27. The enzymes were capable of hydrolyzing different chromogenic substrates, including S-2238 for thrombin-like enzymes, but only BpirSP27 acted on the substrate S-2251 for plasmin. They also showed high stability against variations of temperature and pH, but their activities were significantly reduced after preincubation with Cu2+ ion and specific serine protease inhibitors. In addition. BpirSP27 induced aggregation of washed platelets to a greater extent than BpirSP41. The results showed significant structural and functional differences between B. pirajai serine proteases, providing interesting insights into the structure-function relationship of SVSPs. The mycotoxin aflatoxin B1 (AFB1) is a carcinogenic food contaminant which is metabolically activated by epoxydation. The metabolism of mycotoxins via the mercapturate metabolic pathway was shown, in general, to lead to their detoxication. Mercapturic acids thus formed (S-substitued-N-acetyl-L-cysteines) may be accumulated in the kidney and either excreted in the urine or desacetylated by Acylase 1 (ACY1) to yield cysteine S-conjugates. To be toxic, the N-acetyl-L-cysteine-S-conjugates first have to undergo deacetylation by ACY 1. The specificity and rate of mercapturic acid deacetylation may determine the toxicity, however the exact deacetylation processes involved are not well known. The aim of this study was to investigate the role of ACY1 in the toxicity of some bioactive epoxides from Aflatoxin B1. We characterized the kinetic parameters of porcine kidney and human recombinant aminoacylase-1 towards some aromatic and aliphatic-derived mercapturates analogue of mycotoxin mercapturic acids and 3,4-epoxyprecocene, a bioactive epoxide derivated from aflatoxin. The deacetylation of mercapturated substrates was followed both by reverse phase HPLC and by TNBS method. Catalytic activity was discussed in a structure function relationship. Ours results indicate for the first time that aminoacylase-1 could play an important role in deacetylating mercapturate metabolites of aflatoxin analogues and this process may be in relation with their cyto- and nephrotoxicity in human. Uca populations have an important functional and structural role in many estuarine ecosystems. These crabs exhibit distinct physiological tolerance to salinity gradients, which may partially explain their heterogeneous distribution. In order to investigate the population structure and distribution of Uca spp. in a tropical estuary, we sampled Uca crabs in replicated 0.75 m2 quadrats at six muddy plain areas during monthly intervals between July and November 2012 in spring tidal conditions. Environmental factors including water temperature, salinity, sediment total organic matter, chlorophyll-a, and granulometry were analyzed. We sampled a total of 2919 individuals distributed in three Uca species (U. uruguayensis, U. thayeri and U. maracoani), from which U. uruguayensis was dominant. The density and biomass of individuals were spatially and temporally heterogeneous. During October and November we found higher Uca spp. densities (71.3 ± 47.3 to 77.6 ± 44,5 ind. 0.75 m-²) and biomass (1.8 ± 1.1 to 2.1 ± 1.0 g 0.75 m-2 AFDW) if compared to the previous months, density (July 55,5± 44,1 August 52,5± 34,9 and September 47,7 ± 25,6 ind. 0,75m-²) and biomass in others months (July 1,0± 0,94 August 1,1 ± 0,72 and September 1,3±0,93 g 0.75 m-2 AFDW ). The same pattern was found for other variables, such as salinity (32 and 34), organic matter (30 and 67%) and chlorophyll-a (89 and 46 μg g-1). In two study areas we found this pattern which suggests that higher Uca productivity and food availability are related. A principal component analysis (PCA) suggests that salinity and granulometry (silt) can influence (60% correspondence) the distribution of U. maracoani. For U. uruguayensis and U. thayeri the PCA suggests chlorophyll-a was important, which is a good indicator for labile organic matter. Our study suggests that the population structure and distribution of Uca species may be regulated by food availability, supporting their utility as biological models for ecosystem monitoring.
