We report a fast (less than 3 h) and cost-effective melting temperature assay method for the detection of single-nucleotide polymorphisms in the MBL2 gene. The protocol, which is based on the Corbett Rotor Gene real time PCR platform and SYBR Green I chemistry, yielded, in the cohorts studied, sensitive (100%) and specific (100%) PCR amplification without the use of costly fluorophore-labeled probes or post-PCR manipulation. At the end of the PCR, the dissociation protocol included a slow heating from 60º to 95ºC in 0.2ºC steps, with an 8-s interval between steps. Melting curve profiles were obtained using the dissociation software of the Rotor Gene-3000 apparatus. Samples were analyzed in duplicate and in different PCR runs to test the reproducibility of this technique. No supplementary data handling is required to determine the MBL2 genotype. MBL2 genotyping performed on a cohort of 164 HIV-1-positive Brazilian children and 150 healthy controls, matched for age and sex and ethnic origin, yielded reproducible results confirmed by direct sequencing of the amplicon performed in blind. The three MBL2 variants (Arg52Cys, Gly54Asp, Gly57Glu) were grouped together and called allele 0, while the combination of three wild-type alleles was called allele A. The frequency of the A/A homozygotes was significantly higher among healthy controls (0.68) than in HIV-infected children (0.55; P = 0.0234) and the frequency of MBL2 0/0 homozygotes was higher among HIV-1-infected children than healthy controls (P = 0.0296). The 0 allele was significantly more frequent among the 164 HIV-1-infected children (0.29) than among the 150 healthy controls (0.18; P = 0.0032). Our data confirm the association between the presence of the mutated MBL2 allele (allele 0) and HIV-1 infection in perinatally exposed children. Our results are in agreement with the literature data which indicate that the presence of the allele 0 confers a relative risk of 1.37 for HIV-1 infection through vertical transmission.

Maalit ovat hyvin komplekseja seoksia, jotka sisältävät erilaisia pinta-aktiivisia aineita. Erilaisia pinta-aktiivisia apuaineita käytetään varsinkin vesiohenteisissa maaleissa veden korkean pintajännityksen takia. Nämä maalin komponentit saat-tavat kuitenkin aiheuttaa maaliin vaahtoamisongelmia. Maalien vaahtoaminen aiheuttaa esimerkiksi erilaisia pintavikoja maalikalvossa ja epäotpimaalista tuo-tantosäiliöiden ja pakkausten täyttöä. Joissain tapauksissa myös maalin funktio-naalisuus saattaa häiriintyä. Maalien vaahtoamista pyritään vähentämään pääasiassa vaahdonestäjiksi kutsu-tuilla apuaineilla. Tärkeimmät vesipohjaisissa maaleissa käytettävät vaahdonestäjät ovat mineraaliöljyjä sekä silikonipohjaisia vaahdonestäjiä. Tässä työssä tutkitaan kolmen eri vesiohenteisen sisäseinämaalin vaahtoamista ja mahdollisia keinoja vähentää sitä. Laboratoriossa kokeillaan nykyisten vaahdonestäjien tilalle mahdollisia korvaavia apuaineita. Lisäksi kullekin maalille luodaan koesuunni-telma, jonka mukaan toteutetaan tehtaalla sarja koeajoja, joilla pyritään löytämään sopiva vaahdonestäjän annostus sekä lisäystapa kullekin maalille, siten että maalien vaahtoaminen vähenee. Laboratoriokokeiden perusteella korvaavaa vaahdonestäjää nykyisten tilalle ei löydetty. Valmistusprosessin yhteydessä sen sijaan havaittiin, että vaahdonestäjän lisäystapaa muuttamalla voisi olla mahdollista vähentää maalien vaahtoamista merkittävästi lisäämättä käytettävää vaahdonestäjän annostusta.

Työn tavoitteena oli kehittää menetelmä, jolla voitaisiin arvioida veto-ominaisuuksien avulla konvertointikoneen prässäysnopeuden ja muottien lämpötilan vaikutuksia kartongin muovautuvuuteen. Kirjallisuusosassa käsiteltiin kartongin muovautuvuuteen vaikuttavia ominaisuuksia sekä vetolujuuteen vaikuttavia tekijöitä. Lisäksi käsiteltiin kosteuden vaikutusta muovautuvuuteen sekä esiteltiin vuokakonvertointiprosessin periaate ja muovin sekä kartongin käyttäytymisen erot konvertointiprosessissa. Kokeellinen osa jakaantui kolmeen osaan. Esikokeissa tutkittiin laboratoriomittauksilla vetonopeuden ja kosteuden muutosten vaikutusta erilaisten kaupallisten kartonkimateriaalien vetolujuuteen ja murtovenymään. Tulosten perusteella valittiin prässäysnopeudet ja kosteustaso 2D- testauslaitteella suoritettuihin esi-pilot-mittakaavan kokeisiin sekä varsinaiseen pilot-koeajoon. Pilot-koeajossa pyrittiin arvioimaan prässäysnopeuden ja muottien lämpötilan vaikutuksia muovautuvuuteen vetolujuusarvojen avulla. Prässäysnopeudella ja vetolujuuslaitteen nopeudella ei havaittu yhteneväisyyksiä, jotta voitaisiin arvioida kartongin muovautuvuutta. Muovautuvuutta voidaan kuitenkin arvioida epäonnistuneisuusluvulla, jonka laskentapa kehitettiin työn aikana. Epäonnistuneisuusluku kuvaa hyvin kartongin muovautuvuutta ja on menetelmänä helppo, mutta sen heikkoutena on repeämien mittaamisen aiheuttama epätarkkuus. Esikokeissa havaittiin myös muovipäällysteiden veto-ominaisuuksien vastaavan kartongin ominaisuuksia 65 %:n suhteellisessa kosteudessa, jonka valittiin siten olevan optimaalinen olosuhde pilot-mittakaavan koeajoihin.

Functional MRI (fMRI) resting-state experiments are aimed at identifying brain networks that support basal brain function. Although most investigators consider a ‘resting-state’ fMRI experiment with no specific external stimulation, subjects are unavoidably under heavy acoustic noise produced by the equipment. In the present study, we evaluated the influence of auditory input on the resting-state networks (RSNs). Twenty-two healthy subjects were scanned using two similar echo-planar imaging sequences in the same 3T MRI scanner: a default pulse sequence and a reduced “silent” pulse sequence. Experimental sessions consisted of two consecutive 7-min runs with noise conditions (default or silent) counterbalanced across subjects. A self-organizing group independent component analysis was applied to fMRI data in order to recognize the RSNs. The insula, left middle frontal gyrus and right precentral and left inferior parietal lobules showed significant differences in the voxel-wise comparison between RSNs depending on noise condition. In the presence of low-level noise, these areas Granger-cause oscillations in RSNs with cognitive implications (dorsal attention and entorhinal), while during high noise acquisition, these connectivities are reduced or inverted. Applying low noise MR acquisitions in research may allow the detection of subtle differences of the RSNs, with implications in experimental planning for resting-state studies, data analysis, and ergonomic factors.

The purpose of this study is to investigate whether commercial Kraft lignin can be treated with pulsed corona discharge apparatus so that it becomes active. Active lignin refers to the kind of lignin that can be precipitated on the surface of a fiber by lowering the pH. A secondary agenda here is to remove the pungent smell of kraft lignin, which is caused by organically bound sulfur. It is expected that the study will identify mild processing conditions and parameters for achievement of the desired outcome. In the literature review, the properties of lignin are explained, as is their impact on any further processing. In addition, a number of processes are described for the oxidation of lignin in a variety of applications. In the experimental part of the study, test runs were conducted to determine the effects of oxygen supply and pulse frequency on oxidation results, where the purpose is to produce reactive lignin and to find a process that is feasible at an industrial scale. Based on the reported experiments, lignin could not be made active or precipitated to the surface of the fiber. Actual changes in the structure of lignin were not observed, but the pungent smell of lignin was removed. The exact reason for this change could not be established because sulfur NMR analysis did not work for the lignin samples.

Solvent extraction of calcium and magnesium impurities from a lithium-rich brine (Ca ~ 2,000 ppm, Mg ~ 50 ppm, Li ~ 30,000 ppm) was investigated using a continuous counter-current solvent extraction mixer-settler set-up. The literature review includes a general review about resources, demands and production methods of Li followed by basics of solvent extraction. Experimental section includes batch experiments for investigation of pH isotherms of three extractants; D2EHPA, Versatic 10 and LIX 984 with concentrations of 0.52, 0.53 and 0.50 M in kerosene respectively. Based on pH isotherms LIX 984 showed no affinity for solvent extraction of Mg and Ca at pH ≤ 8 while D2EHPA and Versatic 10 were effective in extraction of Ca and Mg. Based on constructed pH isotherms, loading isotherms of D2EHPA (at pH 3.5 and 3.9) and Versatic 10 (at pH 7 and 8) were further investigated. Furthermore based on McCabe-Thiele method, two extraction stages and one stripping stage (using HCl acid with concentration of 2 M for Versatic 10 and 3 M for D2EHPA) was practiced in continuous runs. Merits of Versatic 10 in comparison to D2EHPA are higher selectivity for Ca and Mg, faster phase disengagement, no detrimental change in viscosity due to shear amount of metal extraction and lower acidity in stripping. On the other hand D2EHPA has less aqueous solubility and is capable of removing Mg and Ca simultaneously even at higher Ca loading (A/O in continuous runs > 1). In general, shorter residence time (~ 2 min), lower temperature (~23 °C), lower pH values (6.5-7.0 for Versatic 10 and 3.5-3.7 for D2EHPA) and a moderately low A/O value (< 1:1) would cause removal of 100% of Ca and nearly 100% of Mg while keeping Li loss less than 4%, much lower than the conventional precipitation in which 20% of Li is lost.

Tämä raportti käsittelee ”Torrefioidun biohiilipelletin laatu ja varastoitavuus” hankkeen tuloksia. Hankkeen tavoitteena oli tutkia torrefioidun biohiilipelletin prosessiteknologiaa, markkinoita ja tuotantokustannuksia kirjallisuustutkimusosiossa. Hankkeen päätutkimus keskittyi koeajoihin pilottilaitoksella, jossa valmistettiin biohiilipellettiä erilaisista puuraaka-aineista. Pilottilaitos oli perustettu Torrec Oy:n toimesta Etelä-Savon Energian Pursialan voimalaitoksen yhteyteen Mikkelissä ja sen tuotanto oli käynnistynyt kesällä 2014. Kaikki koe-erät valmistettiin vain käyttämällä sidonta-aineena lauhdevettä, jota oli tiivistynyt säiliön pohjalle torrefiointiprosessin aikana. Näin ollen erillistä lisäsidonta-aineita ei tarvittu, jolloin voidaan säästää tuotantokustannuksissa jatkossakin. Euroopan Unioni on asettanut 20 % tavoitteen uusiutuvien energioiden käytölle vuoteen 2020, josta biomassalla voidaan kattaa kaksi kolmannesta. Tutkimushankkeen tavoitteena oli metsään perustuvan bioenergiatuotannon lisääminen ja tuontienergian korvaaminen kotimaisella polttoaineella. Hankkeen tarkoituksena oli tutkimusanalyysien kautta kehittää uutta kilpailukykyistä teknologiavaihtoehtoa puupolttoaineiden hyödyntämiseksi. Torrefiointiteknologiaa ollaan kaupallistamassa ympäri Eurooppaa parasta aikaa ja uusia biohiilen tuotantolaitoksia on kehitteillä ja rakenteilla. Tutkimuksen tulokset osoittavat, että biohiilipelletillä on mahdollisuudet suurimittakaavaiseen energiantuotantoon laadun suhteen, kunhan sen käyttäminen tulee edullisemmaksi laitoksissa. Toisaalta, tämä kehitys vaatii tukimekanismeja valtion puolelta, jotta pelletit lähtisivät todella liikkeelle markkinoilla.

SUMMARY Organizational creativity – hegemonic and alternative discourses Over the course of recent developments in the societal and business environment, the concept of creativity has been brought into new arenas. The rise of ‘creative industries’ and the idea of creativity as a form of capital have attracted the interests of business and management professionals – as well as academics. As the notion of creativity has been adopted in the organization studies literature, the concept of organizational creativity has been introduced to refer to creativity that takes place in an organizational context. This doctoral thesis focuses on organizational creativity, and its purpose is to explore and problematize the hegemonic organizational creativity discourse and to provide alternative viewpoints for theorizing about creativity in organizations. Taking a discourse theory approach, this thesis, first, provides an outline of the currently predominant, i.e. hegemonic, discourse on organizational creativity, which is explored regarding themes, perspectives, methods and paradigms. Second, this thesis consists of five studies that act as illustrations of certain alternative viewpoints. Through these exemplary studies, this thesis sheds light on the limitations and taken-for-granted aspects of the hegemonic discourse and discusses what these alternative viewpoints could offer for the understanding of and theorizing for organizational creativity. This study leans on an assumption that the development of organizational creativity knowledge and the related discourse is not inevitable or progressive but rather contingent. The organizational creativity discourse has developed in a certain direction, meaning that some themes, perspectives, and methods, as well as assumptions, values, and objectives, have gained a hegemonic position over others, and are therefore often taken for granted and considered valid and relevant. The hegemonization of certain aspects, however, contributes to the marginalization of others. The thesis concludes that the hegemonic discourse on organizational creativity is based on an extensive coverage of certain themes and perspectives, such as those focusing on individual cognitive processes, motivation, or organizational climate and their relation to creativity, to name a few. The limited focus on some themes and the confinement to certain prevalent perspectives, however, results in the marginalization of other themes and perspectives. The negative, often unintended, consequences, implications, and side effects of creativity, the factors that might hinder or prevent creativity, and a deeper inquiry into the ontology and epistemology of creativity have attracted relatively marginal interest. The material embeddedness of organizational creativity, in other words, the physical organizational environment as well as the human body and its non-cognitive resources, has largely been overlooked in the hegemonic discourse, although thereare studies in this area that give reason to believe that they might prove relevant for the understanding of creativity. The hegemonic discourse is based on an individual-centered understanding of creativity which overattributes creativity to an individual and his/her cognitive capabilities, while simultaneously neglecting how, for instance, the physical environment, artifacts, social dynamics and interactions condition organizational creativity. Due to historical reasons, quantitative as well as qualitative yet functionally- oriented studies have predominated the organizational creativity discourse, although studies falling into the interpretationist paradigm have gradually become more popular. The two radical paradigms, as well as methodological and analytical approaches typical of radical research, can be considered to hold a marginal position in the field of organizational creativity. The hegemonic organizational creativity discourse has provided extensive findings related to many aspects of organizational creativity, although the con- ceptualizations and understandings of organizational creativity in the hegemonic discourse are also in many respects limited and one-sided. The hegemonic discourse is based on an assumption that creativity is desirable, good, necessary, or even obligatory, and should be encouraged and nourished. The conceptualiza- tions of creativity favor the kind of creativity which is useful, valuable and can be harnessed for productivity. The current conceptualization is limited to the type of creativity that is acceptable and fits the managerial ideology, and washes out any risky, seemingly useless, or negative aspects of creativity. It also limits the possible meanings and representations that ‘creativity’ has in the respective discourse, excluding many meanings of creativity encountered in other discourses. The excessive focus on creativity that is good, positive, productive and fits the managerial agenda while ignoring other forms and aspects of creativity, however, contributes to the dilution of the notion. Practices aimed at encouraging the kind of creativity may actually entail a risk of fostering moderate alterations rather than more radical novelty, as well as management and organizational practices which limit creative endeavors, rather than increase their likelihood. The thesis concludes that although not often given the space and attention they deserve, there are alternative conceptualizations and understandings of organizational creativity which embrace a broader notion of creativity. The inability to accommodate the ‘other’ understandings and viewpoints within the organizational creativity discourse runs a risk of misrepresenting the complex and many-sided phenomenon of creativity in organizational context. Weber and Schumpeter: the Economic action of the entrepreneur. Is there any specificity to be raised in the relations established between entrepreneurs and institutions? Recently, the term entrepreneurship is being widely employed. Enterprising is not anymore a restricted activity to the private sector, but also the Third Sector and the Public Administration. It does not only circumscribe the space of innovation, but also of the adapting changes. In this sense, such elastic concept runs the risk to lose consistency. It seems to be appropriate, therefore, to rescue the meaning that many authors, considered classic in Social Sciences, had attributed to the subject, to show that, although deep socioeconomic transformations occurred since Schumpeter wrote the Theory of the Economic Development, is still necessary to emphasize a basic dimension of the enterprising action: resistance and institutional conflict.

Verkkopalveluiden ylläpitovaiheessa halutaan varmistua, etteivät palveluun tehdyt muutokset aiheuta verkkopalvelussa virhetilanteita ja palvelu toimii moitteetta. Muutoksen hyväksyntätestaus voidaan tehdä regressiotestauksena vertaamalla palvelun tilaa ennen ja jälkeen muutoksen. Sisältöpainotteisessa verkkopalvelussa testaaminen keskittyy loppukäyttäjälle esitetyn sivun semanttiseen sekä visuaaliseen oikeellisuuteen sekä erilaisiin toiminnallisiin testeihin. Työssä tarkastellaan etenkin suositulla WordPress-julkaisujärjestelmällä toteutettujen verkkopalveluiden ylläpitoa. Keskeisenä osana julkaisujärjestelmillä toteutettujen verkkopalveluiden ylläpitoa on julkaisujärjestelmän ja sitä täydentävien lisäosien päivittämistä ajantasaisiin versioihin. Nämä päivitykset paitsi tuovat uusia ominaisuuksia verkkopalvelun kehittäjille, myös paikkaavat järjestelmän tietoturvahaavoittuvuuksia sekä korjaavat aiemmissa versioissa esiintyneitä virheitä. Tässä työssä kehitettiin kohdeyrityksen aiempia verkkopalveluiden ylläpitoprosesseja niissä tunnistettujen kehityskohteiden perusteella. Uudistettu kokonaisuus jakautuu kahteen kokonaisuuteen: päivitystarpeen seurantaan sekä päivitysten tekemiseen. Päivitystarpeen seurantaa varten kehitettiin uusi työkalu helpottamaan kokonaiskuvan hahmottamista. Päivitysten tekemisen osalta työssä keskityttiin automatisoidun regressiotestauksen kehittämiseen, missä tärkeimpänä testauskeinona käytetään verkkopalvelusta tallennettujen kuvankaappausten vertailuun perustuvaa visuaalista testausta. Uusien ylläpitoprosesseille määriteltiin myös seurannan kohteet uudistuksen onnistumisen ja jatkokehityksen arviointia varten.

The growth of the food packaging industry has raised more interest in bio-based fibre packing. The use of petroleum based packages is unfriendly to the environment while bio-based is a sustainable option for food packing. In this Master Thesis the aim was to discover how the press forming machineries runnability is affected by parameters of the press and how it also affects formability of paperboard trays. Familiarisation of the working operation parameters was done with the KAMA ST 75 flat-bed die cutting machine and the VP3-70 mould press. Some small test runs of moulding trays where done to get acquainted to the adjustment parameters of the machines. Literature study was done on how paperboards physical properties react to the forces applied during press forming. The study of what kind of defects to the paperboard tray might occur during forming process and the causes for these defects. Also how the parameters of the press forming machine affects formability of the tray. Maintenance procedures was done to the press forming machine to enhance the reliably of production process. Tool alignment measurement was done to determine proper alignment. Laboratory test of the physical properties of the test material was done to find any connection to how the test material performs in press forming. An evaluation criterion was made to evaluate the dimensions and defects of the tray. From the test result a conclusion can be drawn on how the parameters of the press forming process affect the paperboard material. Based on the results the adjustment the parameters of moulding machines to the mechanical properties of paperboard it is possible to produce high quality fibre passed trays for the food packaging industry.

Nietzsche employed metaphors frequently throughout his works. This is especially true in Thus Spoke Zarathustra. Although this is often aesthetically pleasing, it can be very difficult for the reader to understand the nuances and interconnections with the various metaphors. This is generally considered one of the main drawbacks of Thus Spoke Zarathustra. While it is beautifully written in a style that is incomparable today, much of what it is attempting to communicate is lost on the reader. This thesis explores the connection between the metamorphoses of the spirit and the seasons in Thus'Spoke Zarathustra, with the camel spirit corresponding to autumn, the lion spirit with winter, the child spirit with spring, and finally the Overman with summer. Although the Overman is not included among the three metamorphoses of the spirit, it will be argued that the Overman is a separate metamorphosis and must not be conflated with the child spirit despite their similarities. While Thus Spoke Zarathustra will be the primary text used, Nietzsche's other works will be employed to demonstrate that this connection between the metamorphoses of the spirit and the seasons runs through much of his thought. By demonstrating how the seasons are used in Thus Spoke Zarathustra, a deeper understanding of the work will be revealed. Further, this thesis will demonstrate that it is an intentional connection, and not merely coincidental or something that has been constructed and imposed upon the work. Bringing this correspondence between the metamorphoses of the spirit and the seasons to light will result in the need to rethink particular notions of Nietzsche's philosophy. The most apparent involves the Overman and the process of overcoming. Although the Overman has often been viewed as "the end" in the cycle of metamorphoses, it will be argued that this is not the case. The typical interpretation of the metamorphoses of the spirit regard it as a linear progression; however, it will be shown that the metamorphoses of the spirit is cyclical with the camel, lion, and child spirits endlessly repeating, much like the seasons.
