A 2-year study was carried out on established trees at two sites in southeastern Queensland, Australia, to identify environmental factors that influenced rooting of Backhousia citriodora from cuttings. Complex interactions of rainfall events above 20 mm from the preceding month and mean maximum temperature on stock plants resulted in a correlation with rooting success of r = 0.81 and 0.74 for sites at The University Of Queensland, Gatton Campus, and Cedar Glen, respectively. A more detailed investigation under controlled environmental conditions showed that maintaining stock plants at temperatures between 10 and 30degreesC had no direct effect on rooting capacity. However, temperature was correlated with growth, which may have an indirect effect on root formation. In spring floral initiation was found to only delay rooting and had no effect on the final rooting percentage. A series of seasonal experiments demonstrated that application of the auxins indole-3-acetic acid, indole-3-butyric acid and napthaleneacetic acid over a range of concentrations (1000-8000 mug/ml) did not significantly increase rooting compared to the control and there is no practical advantage in applying auxins. Seasonal results and the temperature experiment also suggest that under a glasshouse environment with higher temperatures in winter and an adequate supply of water, B. citriodora cuttings can be successfully rooted over the whole year. The resource potential of shallow water tables for cropping systems has been investigated using the Australian sugar industry as a case study. Literature concerning shallow water table contributions to sugarcane crops has been summarised, and an assessment of required irrigation for water tables to depths of 2 m investigated using the SWIMv2.1 soil water balance model for three different soils. The study was undertaken because water availability is a major limitation for sugarcane and other crop production systems in Australia and knowledge on how best to incorporate upflow from water tables in irrigation scheduling is limited. Our results showed that for the three soils studied (representing a range of permeabilities as defined by near-saturated hydraulic conductivities), no irrigation would be required for static water tables within 1 m of the soil surface. Irrigation requirements when static water tables exceeded 1 m depth were dependent on the soil type and rooting characteristics (root depth and density). Our results also show that the near-saturated hydraulic conductivities are a better indicator of the ability of water tables below 1 m to supply sufficient upflow as opposed to soil textural classifications. We conclude that there is potential for reductions in irrigation and hence improvements in irrigation water use efficiency in areas where shallow water tables are a low salinity risk: either fresh, or the local hydrology results in net recharge. Grevillea (Proteaceae) is a native Australian plant genus with high commercial value as landscape ornamentals. There has been limited research on the culture and propagation of Australian native species. The effect of indole-3-butyric acid (IBA) on the rooting of G. 'Royal Mantle' and G. 'Coastal Dawn' in winter, spring and summer was evaluated at University of Queensland Gatton, Southern Queensland in order to determine the rooting ability of this species in different seasons. Both Grevillea cultivars showed seasonal rooting. The more difficult-to-root G. 'Coastal Dawn' had a reduced response to IBA application than G. 'Royal Mantle'. Stem and leaf indole-3-acetic acid (IAA) levels were not different between cultivars, therefore rooting ability between the two cultivars does not appear to be due to the differences in endogenous IAA levels. The purpose of this study was to determine the seasonal water use patterns of dominant macrophytes coexisting in the coastal Everglades ecotone. We measured the stable isotope signatures in plant xylem water of Rhizophora mangle, Cladium jamaicense, and Sesuvium portulacastrum during the dry (DS) and wet (WS) seasons in the estuarine ecotone along Taylor River in Everglades National Park, FL, USA. Shallow soilwater and deeper groundwater salinity was also measured to extrapolate the salinity encountered by plants at their rooting zone. Average soil water oxygen isotope ratios (δ 18O) was enriched (4.8 ± 0.2‰) in the DS relative to the WS (0.0 ± 0.1‰), but groundwater δ 18O remained constant between seasons (DS: 2.2 ± 0.4‰; WS: 2.1 ± 0.1‰). There was an inversion in interstitial salinity patterns across the soil profile between seasons. In the DS, shallow water was euhaline [i.e., 43 practical salinity units (PSU)] while groundwater was less saline (18 PSU). In the WS, however, shallow water was fresh (i.e., 0 PSU) but groundwater remained brackish (14 PSU). All plants utilized 100% (shallow) freshwater during the WS, but in the DS R. mangle switched to a soil–groundwater mix (δ 55% groundwater) while C. jamaicense and S. portulacastrum continued to use euhaline shallow water. In the DS, based on δ 18O data, the roots of R. mangle roots were exposed to salinities of 25.4 ± 1.4 PSU, less saline than either C. jamaicense(39.1 ± 2.2 PSU) or S. portulacastrum (38.6 ± 2.5 PSU). Although the salinity tolerance of C. jamaicense is not known, it is unlikely that long-term exposure to high salinity is conducive to the persistence of this freshwater marsh sedge. This study increases our ecological understanding of how water uptake patterns of individual plants can contribute to ecosystem levels changes, not only in the southeast saline Everglades, but also in estuaries in general in response to global sea level rise and human-induced changes in freshwater flows.

The landscape structure of emergent wetlands in undeveloped portions of the southeastern coastal Everglades is comprised of two distinct components: scattered forest fragments, or tree islands, surrounded by a low matrix of marsh or shrub-dominated vegetation. Changes in the matrix, including the inland transgression of salt-tolerant mangroves and the recession of sawgrass marshes, have been attributed to the combination of sea level rise and reductions in fresh water supply. In this study we examined concurrent changes in the composition of the region’s tree islands over a period of almost three decades. No trend in species composition toward more salt-tolerant trees was observed anywhere, but species characteristic of freshwater swamps increased in forests in which fresh water supply was augmented. Tree islands in the coastal Everglades appear to be buffered from some of the short term effects of salt water intrusion, due to their ability to build soils above the surface of the surrounding wetlands, thus maintaining mesophytic conditions. However, the apparent resistance of tree islands to changes associated with sea level rise is likely to be a temporary stage, as continued salt water intrusion will eventually overwhelm the forests’ capacity to maintain fresh water in the rooting zone.

Savannah is the second biome in biodiversity in Brazil, presenting great vegetation endemism. Dipteryx alata Vog. (Fabaceae), native from this biome, is an economically important species, with an incipient market due to the lack of commercial plantations. This highlights the need to develop and provide the basis for the domestication of this species. Thus, this study determined the best conditions for in vitro establishment, multiplication, elongation and rooting of stem tips of D. alata plantlets grown vitro. Two culture media (MS and WPM) were evaluated in different salt concentrations (25, 50, 75 and 100%) for plantlet establishment. Four concentrations of 6– Benzylaminopurine (BAP) (0, 1, 2, 3 and 4 mg L-1) amended with 0.25 mg L-1 naphthalene-acetic acid (NAA) were studied for multiplication. Simultaneous elongation and rooting were studied with four concentrations of NAA (0, 1, 2, 3 and 4 mg L-1) together with 0.5 mg L-1 IBA. The variables analyzed were: shoot length (CPA), root length (CP), fresh matter (MF), dry matter (MSC), stem diameter (DC) and number of leaves (NF), 120 days after inoculation, with the exception of number of shoots, which was evaluated in the multiplication stage only. The medium MS at the original salt concentration (100%) was effective for the in vitro establishment of E. alata, resulting in greater root length (27.65 cm) and number of leaves per plantlet (26.0). The concentration of 4 mg L-1 BAP was the best one for multiplication; however, greater concentrations can boost multiplication. The effect of NAA and IBA were noticeable on in vitro elongation and rooting, with best CPA (3.14 cm) and CR (15.84 cm). Therefore, it is possible to state that the medium MS increases the success probability of in vitro establishment of stem tips of Dipteryx alata. NAA concentrations below 3 mg L-1 were favorable for in vitro development of the species, with essential characteristics for acclimatization success|.

Qatar's rulers established Al-Jazeera Children's Channel in 2005 to nurture Arab and Islamic traditions and values; the channel sought to differentiate itself by ensuring that material would conform to what its management considered to be culturally appropriate guidelines for content. However, having been set up as a child-centred, non-commercial, fully-funded enterprise that aspired to source most of its content within the Arabic-speaking region, its priorities shifted in 2011-13 towards maximizing commercial revenues through foreign imports. In light of the shift, this chapter explores the complex interweaving of commercial and political considerations behind production and commissioning processes. The channel's branding and re-branding shows how a children’s television project can be adopted to reinforce a country’s claim to regional cultural leadership, while being packaged in such a way as to depoliticize that country’s institutions and composition by rooting national identity in a combination of commercial interests and notions of traditional culture.

Although it is well known that nutrient imbalance in shoot tissues may impair plant performance, the interactive effect between foliar phosphorus (P) application and varying P availability in the rooting medium on the nutritional status of sugarcane has not been well studied. To fill this research gap, four sugarcane varieties (IAC91-1099, IACSP94-2101, IACSP94-2094 and IACSP95-5000) were evaluated using a combination of two concentrations of P in nutrient solution (P-deficient, PD = 0.02 mmol L^(−1) and P-sufficient, PS = 0.5 mmol L^(−1)) and foliar P application (none and 0.16 mol L^(−1)). The spray was applied until drip point three times during the experiment with 15 days intervals, after which the plants were harvested to quantify growth and shoot concentration of nitrogen (N), P, magnesium (Mg), sulphur (S) and manganese (Mn). The responses of sugarcane plants to foliar P spray at different levels of P supply in the rooting medium was not genotype-dependent. It was demonstrated for the averaged values across varieties, that foliar P application enhanced sugarcane performance under low P, as revealed by improvements of leaf area and dry matter production of shoot and root of PD plants. Under P limitation we also observed diminished shoot concentration of N, P, Mg, S and increased concentration of Mn. However, foliar P spray increased the concentrations of N, P, S and reduced shoot Mn. Furthermore, shoot P:N, P:Mg, P:S, P:Mn and Mg:Mn concentration ratios exhibited a positive relationship with shoot dry matter production. In conclusion, low P supply in the rooting medium impairs nutrient balance in shoot tissues of sugarcane at early growth; however, this effect was ameliorated by foliar P application which merits further study under field conditions.

The most native fruit trees are belonging to Myrtaceae family, which it have as main marketing potential their fruit. Despite the wide acceptance of the fruits of these native fruit cultura, the establishment of commercial orchards is still necessary, because if it prevails extraction in the forest. To start the cultivo in the orchard, the first point is on the mother plant choice, which should provide superior characteristics when compared to other genotypes. Then, it is necessary to choose the method to can produce satisfactory amount of seedlings and preferably without it to lose the mother plant characteristics. For this, it adopts the asexual thechniques, with option for grafting, cuttings and air layering. These techniques when tested with native fruits tree, it proved limiting in theses results, with this, it should to test other it to recommend its use, especially, those fruit native of higher potential as jabuticaba tree, pitanga tree, sete capote tree and araça amarelo tree. The aim of this study was to test the use of asexual propagation through mini-cuttings in these native fruit trees, according to the time of collection, the mini-cutting length and concentration of IBA, as well as, it to relate the results of rooting with tryptophan extracted at certain times. The work was carried out at Universidade Tecnológica Federal do Paraná – Câmpus Dois Vizinhos, Brazil. The samples were collected each two months. The mini-cutting were prepared with 6 or 8 cm, with a pair of leaves reduced to 25% of the original size. The mini-cuttings had their base immersed in liquid solution of indole-butyric acid (IBA) in the concentrations of 0, 3000 and 6000 mg L-1 and then were placed in tubes containing commercial substrate. The experimental design was completely randomized with factorial 2 x 3 x 6 (mini-cutting length x IBA concentration x time of collection), with four replications, it being each plot varied according to the amount of shoots obtained by period time. After 120 days, the rooting and callus formation (%), average number of roots per mini-cutting and the average length of the roots were evaluated. After 60 days of these evaluations, the survival of mini-cuttings rooted after transplant was evaluated. It was evaluated also the production of mini-cuttings of each size in each period time. At the end of the experiment it was evaluated the percentage of survival of mother plantlets. For analysis of tryptophan was used materials branches, leaves and twigs with leaves, taken from the materials used for the production of mini-cutting. It was recommended for hybrid jabuticaba tree the use mini-cutting with eight cm, treated with 6000 mg L-1 of IBA and collected in June. For jabuticaba tree of cabinho and araça amarelo tree the period for propagation by mini-cuttings should be in August, regardless of IBA concentration and length of the mini-cutting. In the jabuticaba tree sabara and sete capote tree is important to obtain more satisfactory results realized the collect in October or December, with the same independence of other levels tested in other factors. However, for sete capote tree should test other techniques to increase the efficiency of propagation. And with pitanga tree recommended to the collection in June, but with 6cm the application of 3000 mg L-1 of IBA and 8 cm with 6000 mg L-1 of IBA.

Many rainfed wheat production systems are reliant on stored soil water for some or all of their water inputs. Selection and breeding for root traits could result in a yield benefit; however, breeding for root traits has traditionally been avoided due to the difficulty of phenotyping mature root systems, limited understanding of root system development and function, and the strong influence of environmental conditions on the phenotype of the mature root system. This paper outlines an international field selection program for beneficial root traits at maturity using soil coring in India and Australia. In the rainfed areas of India, wheat is sown at the end of the monsoon into hot soils with a quickly receding soil water profile; in season water inputs are minimal. We hypothesised that wheat selected and bred for high yield under these conditions would have deep, vigorous root systems, allowing them to access and utilise the stored soil water at depth around anthesis and grain-filling when surface layers were dry. The Indian trials resulted in 49 lines being sent to Australia for phenotyping. These lines were ranked against 41 high yielding Australian lines. Variation was observed for deep root traits e.g. in eastern Australia in 2012, maximum depth ranged from 118.8 to 146.3 cm. There was significant variation for root traits between sites and years, however, several Indian genotypes were identified that consistently ranked highly across sites and years for deep rooting traits.

Olive (Olea europaea L.), one of the main crops in the Mediterranean basin, is mainly propagated by cuttings, a classical propagation method that relies on the ability of the cuttings to form adventitious roots. While some cultivars are easily propagated by this technique, some of the most interesting olive cultivars are considered difficult-to-root which poses a challenge for their preservation and commercialization. Therefore, increasing the current knowledge on adventitious root formation is extremely important for species like olive. This research focuses on evaluating the role of free auxins and oxidative enzymes on adventitious root formation of two olive cultivars with different rooting ability - ‘Galega vulgar’ (difficult-to-root) and ‘Cobrançosa’ (easy-to-root). In this context, free auxin levels and enzyme activities were determined in in vitro-cultured ‘Galega vulgar’ microshoots and in semi-hardwood cuttings of cvs. ‘Galega vulgar’ and ‘Cobrançosa’. To attain this goal, an analytical method for the quantification of free indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) was developed, which is based on dispersive liquid-liquid microextraction followed by microwave derivatization (DLLME-MAD) and gas chromatography-mass spectrometry (GC/MS) analysis. The developed method was validated in terms of linearity, recovery, limit of detection (LOD) and limit of quantification (LOQ) and proved to be useful in the analysis of two very different types of plant tissues. The results from auxin quantification in olive samples point at a relationship between free auxin levels and rooting ability of both microshoots and semihardwood cuttings. A defective IBA-IAA conversion, resulting in a peak of free IAA during initiation phase, seems to be associated with low rooting ability. Likewise, differences in the activity of oxidative enzymes also appear to be related with rooting ability. Higher polyphenol oxidases (PPO) activity is likely related with an easyto- root behavior, while the opposite is true for peroxidases (POX) (including IAA oxidase (IAAox)) activity. A possible hypothesis for adventitious root formation in olive microcuttings is presented herein for the first time. Free auxins, oxidative enzymes, alternative oxidase (AOX) and reactive oxygen species (ROS) are some of the factors that may be involved in this highly complex physiological process. Interestingly, while temporal changes in auxin levels were similar between microshoots and semihardwood cuttings, the conclusions obtained from enzyme activity results in microshoots didn’t translate to semi-hardwood tissues, showing the emerging need for adaptation of classical agronomical research studies to modern techniques; Resumo: Procurando compreender o papel das auxinas e enzimas oxidativas na formação de raízes adventícias em cultivares de oliveira (Olea europaea L.) A oliveira (Olea europaea L.) é uma das principais culturas da bacia Mediterrânica e é propagada maioritariamente por estacaria, um processo altamente dependente da capacidade das estacas para formar raízes adventícias. Enquanto algumas cultivares são fáceis de propagar desta forma, algumas das cultivares de oliveira mais interessantes são consideradas difíceis de enraizar, o que dificulta a sua preservação e comercialização e torna extremamente importante aprofundar o conhecimento sobre o enraizamento adventício desta espécie. Este trabalho foca-se na avaliação do papel das auxinas livres e das enzimas oxidativas na formação de raízes adventícias em duas cultivares de oliveira com diferente capacidade de enraizamento - ‘Galega vulgar’ (difícil de enraizar) e ‘Cobrançosa’ (fácil de enraizar). Neste contexto, determinaram-se os níveis de auxinas livres e as actividades de enzimas oxidativas em microestacas de ‘Galega vulgar’ cultivadas in vitro bem como em estacas semi-lenhosas das cvs. ‘Galega vulgar’ e ‘Cobrançosa’. Para tal foi necessário desenvolver uma metodologia analítica para a quantificação de ácido indol-3-acético (IAA) e ácido indol-3-butírico (IBA), baseada em microextracção dispersiva líquido-líquido (DLLME) seguida de derivatização em microondas (MAD) e análise por cromatografia gasosa acoplada a espectrometria de massa (GC/MS). O método desenvolvido foi validado em termos de linearidade, recuperação, limite de detecção (LOD) e limite de quantificação (LOQ), e mostrou-se eficaz na análise de dois tipos de tecidos vegetais bastante diferentes. Os resultados da análise de auxinas em amostras de oliveira apontam para uma possível relação entre os níveis de auxinas livres e a capacidade de enraizamento, tanto em microestacas como em estacas semi-lenhosas. Uma conversão IBA-IAA deficiente, que resulta num pico de IAA durante a fase de iniciação, parece estar associada à baixa capacidade de enraizamento. Por outro lado, a capacidade de enraizamento também parece estar relacionada com diferenças na actividade de enzimas oxidativas. Comportamentos fáceis de enraizar estão associados a actividade mais elevada das polifenoloxidases (PPO), enquanto o oposto é verdade para a actividade das peroxidases (POX) (incluindo a IAA oxidase (IAAox)). Neste trabalho propõe-se pela primeira vez uma possível explicação para o enraizamento adventício em microestacas de oliveira. Auxinas livres, enzimas oxidativas, oxidase alternativa (AOX) e espécies reactivas de oxigénio (ROS) são alguns dos factores envolvidos neste processo fisiológico altamente complexo. Curiosamente, enquanto as alterações temporais nos níveis de auxinas foram semelhantes entre microestacas e estacas semi-lenhosas, o mesmo não se observou relativamente à actividade enzimática, o que mostra a necessidade de adaptação dos estudos agronómicos tradicionais às técnicas correntes.

Os híbridos de Eucalyptus globulus representam uma excelente alternativa para o setor de celulose e papel, em razão dos ganhos em qualidade da madeira para a fabricação de celulose. Entretanto, estes híbridos têm apresentado recalcitrância ao enraizamento adventício. Assim, a micropropagação é apontada como a técnica para o rejuvenescimento desses híbridos adultos, viabilizando a propagação clonal dos mesmos. O presente trabalho avaliou o cultivo in vitro de três clones de Eucalyptus grandis x Eucalyptus globulus e de três clones de Eucalyptus urophylla x Eucalyptus globulus, em relação à multiplicação in vitro, no meio MS suplementado com 0,5 mg L-1 de BAP e 0,01 mg L-1 de ANA, bem como o efeito das concentrações de 0,25; 0,50; 0,75 e 1,0 mg L-1 de AIB e dos meios de cultura MS e JADS no alongamento in vitro das brotações. Os clones diferiram quanto à multiplicação in vitro das brotações e apresentaram uma taxa de multiplicação média dos clones de 3,0 tufos de brotações em cada subcultivo, ao longo dos 25 subcultivos realizados. No alongamento in vitro, os clones diferiram quanto às concentrações de AIB adequadas para provocar o alongamento, bem como em relação aos meios de cultura MS e JADS. O intervalo médio entre 0,40 e 0,80 mg L-1 de AIB proporcionou o maior número e comprimento das brotações alongadas in vitro e com maior vigor.

O objetivo deste trabalho foi avaliar o efeito do tipo de miniestacas e a necessidade de aplicação de ácido indolbutírico sobre o enraizamento e qualidade das mudas formadas de Handroanthus heptaphyllus . As miniestacas apicais e intermediárias foram obtidas em minijardim multiclonal formado a partir de sementes. As miniestacas foram preparadas com 5 cm de comprimento, um par de folhas reduzidas a 50% da área foliar e estaqueadas sem e com AIB na concentração de 8000 mg L-1. As avaliações foram realizadas aos 30 dias, na expedição do setor de enraizamento e aos 120 dias, quando a muda se encontrava formada. O experimento foi conduzido em delineamento inteiramente casualizado, em esquema fatorial 3 x 2 x 2 (três épocas de coleta, duas concentrações de AIB e duas posições do propágulo), com quatro repetições, sendo 12 miniestacas por repetição. De acordo com os resultados, o AIB não foi necessário para o enraizamento das miniestacas, entretanto, sua utilização promove incremento do número e comprimento de raízes. As miniestacas intermediárias proporcionaram maior massa seca de raízes aos 30 dias após o estaqueamento, e aos 120 dias, maior número de folhas e de raízes. A época de coleta influenciou a qualidade final das mudas. Aquelas produzidas na última coleta (oitavo) apresentaram valores médios inferiores nas características biométricas, exceto para o comprimento e número de raízes de primeira ordem.

Pigeon pea ( Cajanus cajan (L.) Millsp.) is a drought tolerant pulse legume, mainly grown for grain in the semi-arid tropics, particularly in Africa. Pigeon pea production in countries like Kenya is faced with a number of challenges, particularly lack of high quality seeds. The objective of this study was to develop an in vitro regeneration system for pigeon pea varieties grown in Kenya, that is amenable to genetic transformation. In vitro regeneration of pigeon pea varieties, KAT 60/8 and ICEAP 00557, commonly grown in Kenya was achieved using leaf explants from in vitro grown seedlings, through callus initiation, followed by shoot and root induction. For callus initiation, MS media supplemented with 0.5-4 mg l-1 2, 4-D and TDZ separately were tested, and IBA at 0.1, 0.5 and 1 mg l-1 was tested for rooting of shoots. Embryogenic calli was obtained on MS containing 2, 4- D; whereas TDZ induced non-embryogenic callus alone or with shoots directly on explants. Indirect shoot regeneration frequency of 6.7 % was achieved using 1 mg l-1 2, 4-D-induced embryogenic callus obtained using KAT 60/8 explants. Whereas direct shoot regeneration frequencies of 20 and 16.7% were achieved using ICEAP 00557 and KAT 60/8 explants, using 0.5 mg l-1 and 2 mg l-1 TDZ, respectively. Optimum rooting was achieved using 0.5 mg l-1 IBA; and up to 92% rooted shoots were successfully established in soil after acclimatisation. Genotype and hormone concentrations had a significant (P<0.05) influence on callus, shoot and root induction. The protocol developed can be optimised for mass production and genetic transformation of KAT 60/8 variety.
