Cats, as definitive host, play an important role in the transmission of Toxoplasma gondii. This study aimed to establish the seroprevalence of anti-T. gondii immunoglobulins G and M, and determine the frequency of oocysts in the feces of the domestic cat population in Rio de Janeiro, Brazil. We also aimed to study the association between T. gondii infection and age, sex, breed, lifestyle, diet and retroviral infection. A total of 108 cats were included in the study and fecal samples of 54 of those cats were obtained. Only 5.6% of the cats were seropositive for anti-T. gondii immunoglobulins using the indirect hemagglutination test. None of the 54 cats presented oocysts in their fecal samples. Although not statistically significant, males, mixed-breed, free-roaming and cats aged two years and older were found to be more exposed. Age, lifestyle and the use of litter boxes were found to play an important role as risk factors. Anemia and retroviral infections were independent of T. gondii infection. No antibodies were detected in the majority of cats (94.4%), indicating that those cats had never been exposed to the parasite and, therefore, once infected, they could present the risk of shedding large numbers of oocysts into the environment.

Several researchers have stated that parasites can alter the behavior of their hosts, in order to increase the transmission rate, principally when prey-predator relationships are a reliable way of infection transmission. The aim of this study was to verify the occurrence of changes in anxiety and short-term memory patterns in experimentally infected Mus musculus by Toxocara canis and/or Toxoplasma gondii. Forty male Mus musculus (Balb/c) eight-week-old were divided into four groups of 10 mice each. One group was infected with 300 eggs of Toxocara canis; a second group was submitted to infection with 10 cysts of Toxoplasma gondii; a third group was concomitantly infected with both parasites with the same inoculums and the last group was maintained without infection. The anxiety levels were evaluated using an elevated plus maze and an actometer; the short-term memory was determined by a two-way active avoidance equipment. The determination of anxiety levels were conducted 40 and 70 days after infection and the short-term memory was evaluated 140 days after infection. Mice chronically infected by Toxoplasma gondii showed impaired learning and short-term memory, but no significant differences were found in mice infected by Toxocara canis or concomitantly infected by Toxocara canis and Toxoplasma gondii when compared to non infected mice.

Context and objective:The molecular characterization of local isolates of Toxoplasma gondii is considered significant so as to assess the homologous variations between the different loci of various strains of parasites.Design and setting:The present communication deals with the molecular cloning and sequence analysis of the 1158 bp entire open reading frame (ORF) of surface antigen 3 (SAG3) of two Indian T. gondii isolates (Chennai and Izatnagar) being maintained as cryostock at the IVRI.Method:The surface antigen 3 (SAG3) of two local Indian isolates were cloned and sequenced before being compared with the available published sequences.Results:The sequence comparison analysis revealed 99.9% homology with the standard published RH strain sequence of T. gondii. The strains were also compared with other established published sequences and found to be most related to the P-Br strain and CEP strain (both 99.3%), and least with PRU strain (98.4%). However, the two Indian isolates had 100% homology between them.Conclusion:Finally, it was concluded that the Indian isolates were closer to the RH strain than to the P-Br strain (Brazilian strain), the CEP strain and the PRU strains (USA), with respect to nucleotide homology. The two Indian isolates used in the present study are known to vary between themselves, as far as homologies related to other genes are concerned, but they were found to be 100% homologous as far as SAG3 locus is concerned. This could be attributed to the fact that this SAG3 might be a conserved locus and thereby, further detailed studies are thereby warranted to exploit the use of this particular molecule in diagnostics and immunoprophylactics. The findings are important from the point of view of molecular phylogeny.

Toxoplasmosis is frequently acquired through the oral route by the ingestion of cysts or oocysts of Toxoplasma gondii. Once ingested, the parasites penetrate the intestinal epithelial cells and rapidly disseminate to all organs in the host. During T. gondii infection, the intestinal microbiota plays an important role in stimulating a protective immune response against the parasite. In this sense the use of probiotics is worthy of note since they are live microorganisms that have beneficial effects on the host through stimulation of the immune response that can be important in the control of T. gondii proliferation and dissemination in the host. In the present study, the action of the probiotic Bifidobacterium animalis subsp. lactis was investigated in C57BL/6 mice infected with oocysts of ME49 strain of T. gondii. The probiotic had an immunomodulatory action, inducing CD19 lymphocyte proliferation and consequently increasing anti-T. gondii antibody level.Bifidobacterium animalis subsp. lactisprovided protection in supplemented mice, compared to the control group. In addition, supplemented animals had milder inflammatory process in the small intestine, indicating that the probiotic protects the intestinal mucosa during infection with T. gondii. It was concluded that the probioticB. animalis subsp. lactis induces humoral immune response capable of providing protection against T. gondii infection.

The role of rodents in the epidemiology of toxoplasmosis was investigated inLondrina, Paraná State, Brazil. One hundred and eighty-one Rattus rattus and one Mus musculus were caught in 37 places. Blood and tissues were collected and submitted to the indirect fluorescence antibody test (IFAT) and the bioassay. Serum samples from 61 contacting dogs were also collected. Sixteen rats (8.8%) were positive for Toxoplasma gondii, but just two of them were positive by serology and bioassay test. Antibodies were found in nine (4.9%) rats. Tissues of nine rats bioassayed were positive and four isolates were obtained. Restriction fragment length polymorphism (RFLP) analysis was performed using 12 markers (SAG1, SAG2, SAG2-alt, C22-8, C29-2, L358, PK1, BTUB, GRA6, SAG3, Apico, CS3). Genotyping revealed that the four strains isolated from this study have been isolated before in cats and chickens from Brazil. None of the isolates was identified like clonal archetypal T-types I, II, and III. The rats presented lower serologic Toxoplasma gondii prevalence (8.8%) compared to contacting dogs (70.5%).

Através de inoculação no peritônio de camundongos, demonstraram, os autores a presença do Toxoplasma gondii na saliva de 12 (60%) indivíduos com toxoplasmose, em estudo no qual 20 doentes foram considerados. Esta verificação é comunicada como complementação a outra, anterior e preliminar, relativa ao mesmo assunto. A importância da presença do parasito na saliva mereceu destaque, em virtude do significado que ela pode ter em relação à transmissão da toxoplasmose.

Em virtude da conveniência de tentar aprimorar o tratamento da toxoplasmose, de semelhanças taxonômicas e de informes contidos na literatura científica, foi considerado oportuno verificar se a artemisinina, medicamento eficiente no sentido de poder debelar a malária, é ativa na infecção experimental de camundongos pelo Toxoplasma gondii. Houve, então, administração de diferentes quantidades desse antiparasitário, inclusive após prévio contato in vitro com toxoplasmas e estimulação da produção de macrófagos, para observações diversificadas a propósito da intenção referida. Pelo menos de acordo com a metodologia usada, a artemisinina não se mostrou eficaz e sugestões para prosseguimento de avaliações acerca do assunto ficaram consignadas.

A encefalite por Toxoplasma gondii (ET) é a principal causa de massa no sistema nervoso central (SNC) em pacientes com a síndrome da imunodeficiência adquirida (SIDA). Com o objetivo de determinar a prevalência dessa afecção e da presença de anticorpos específicos no soro e no líquor, bem como a sensibilidade (S) e a especificidade (E) da tomografia computadorizada (TC), dos achados clínicos e dos testes específicos foram revisados todos os prontuários de 516 pacientes com SIDA, internados no HCPA entre maio/85 e dezembro/91. A prevalência através de TC foi de 13% (diagnóstico presuntivo). A pesquisa de anticorpos específicos para toxoplasmose por imunofluorescência indireta no sangue (SS) foi positiva em 65% e no líquor (SL) em 49% dos pacientes em que foi realizada. Necrópsias de 125 pacientes foram revisadas encontrando-se uma prevalência de ET em 27 (22%) casos, em que o diagnóstico foi considerado definitivo. A sensibilidade da TC foi de 65% e a especificidade de 82%. A SS apresentou S de 95% e E de 30%, enquanto a SL apresentou uma S de 77% e E de 56%. Os seguintes achados clínicos foram pesquisados: febre (S=92%; E=56%); sinais neurológicos focais (S=59%; E=82%) e cefaléia (S=41 %; E=69%). Concluímos que é alta a prevalência da ET na SIDA e que a TC e a pesquisa de anticorpos específicos no soro e no líquor, devido à alta especificidade da primeira e a alta sensibilidade da segunda, constituem-se em métodos adequados para o diagnóstico da ET, sendo discutível a necessidade de realizar biópsia cerebral nesses casos.

Foi realizado um estudo retrospectivo ou caso controle em uma amostra de 500 pessoas selecionadas aleatoriamente da população de Ribeirão das Neves, MG, Brasil, durante o período de junho de 1983 a janeiro de 1984, para verificar se variáveis ligadas ao contato com animais no domicílio estariam associadas a ocorrência da infecção humana por Toxoplasma gondii. Foram obtidas associações estatisticamente significativas com relação ao contato com gatos, galinhas e porcos no domicílio e nenhuma associação quanto ao consumo de carne, leite e ovos.

Foi comprovada a eficácia da azitromicina e da pirimetamina no tratamento da infecção experimental de camundongos pelo Toxoplasma gondii. Houve administração cotidiana, pela via oral de 200mg/kg e 12,5mg/kg, respectivamente durante dez dias. O uso dos medicamentos ocorreu mediante associação ou não e o emprego concomitante proporcionou o melhor resultado e convirá efetuar investigação semelhante com cepa cistogênica do parasita.

Este estudo determinou a ocorrência de anticorpos anti-Toxoplasma gondii em população indígena do Mato Grosso, os Enawenê-Nawê. Estes habitam uma vasta região selvagem, com raros contatos com não-índios. Não apresentam animais domésticos, inclusive gatos. A dieta é baseada em insetos, mandioca, milho, mel e fungos e não se alimentam de carne, exceto de peixe. Com base no exposto, desenvolveu-se análise sorológica, por meio de ELISA - IgG e IFI - IgG e IgM. De 148 soros, 80,4% foram ELISA ou IFI- IgG positivos. Não foram detectados casos de IgM reagentes. Nesse grupo as taxas de soropositividade aumentaram significativamente com a idade, de 50% a 95%. Analisando-se os hábitos e costumes, aliados à alta soropositividade encontrada, sugere-se que a presença de felinos silvestres nas imediações da aldeia e coleções de água poderia ter papel importante como fonte de infecção, contaminando o solo e, conseqüentemente, os insetos e fungos consumidos pelos índios.

Foi colhido um total de 442 soros em rebanhos caprinos de sete regiões do Estado de São Paulo e testados para anticorpos contra Toxoplasma gondii pela reação de imunofluorescência indireta. Em todos os rebanhos, foram encontrados caprinos reagentes, totalizando 64 (14,5%) animais com sorologia positiva em diferentes capris.
