A doença de Aujeszky ou pseudoraiva (DA), causada pelo vírus da pseudoraiva (PRV) é a maior preocupação na produção de suínos. No estado do Rio Grande do Sul, Brasil, a DA foi somente detectada em 1954, em bovino. Em 2003, ocorreram dois surtos de encefalite em granjas na região norte do estado, fronteira com o estado de Santa Catarina. O vírus da doença de Aujeszky (VDA) foi isolado a partir de animais coletados em oito granjas distintas da região e submetido a análises antigênicas e moleculares. As amostras de VDA isoladas foram comparadas com as amostras padrão NIA-3 e NP. A caracterização antigênica dos mesmos foi realizada com testes de imunoperoxidase frente a um painel de anticorpos mono-clonais (Mabs) preparado contra epitopos de glicoproteinas virais (gB, gC, gD e gE). A caracterização genômica foi realizada através da análise restrição enzimática (REA) sobre o genoma total das amostras, com a enzima de restrição (REA) Bam HI. O perfil antigênico das oito amostras isoladas no Rio Grande do Sul, bem como os apresentados pelas amostras padrão NIA-3 e NP, foram similares. A REA revelou que todos as oito amostras do Rio Grande do Sul apresentaram um arranjo genômico do tipo II, genótipo frequentemente encontrado em surtos prévios de DA em outros estados do Brasil. Os resultados aqui obtidos indicam que as oito amostras isoladas no Rio Grande do Sul são similares.

Foi realizado um estudo retrospectivo de 80 casos de cirrose em cães no Laboratório de Patologia Veterinária da Universidade Federal de Santa Maria, Rio Grande do Sul. Dos cães em que o sexo e a idade constavam dos protocolos de necropsia, 53,8% eram machos e 46,2% eram fêmeas; 50,0% foram incluídos como idosos, 48,6% como adultos e 1,4% como filhote. Os principais sinais clínicos observados incluíram: ascite (39/80 [48,8%]), icterícia (19/80 [23,8%]), anorexia (13/80 [16,2%]), distúrbios neurológicos (12/80 [15,0%]), dispnéia (12/80 [15,0%]) e edema subcutâneo (10/80 [12,5%]). Dos 63 cães em que a descrição macroscópica constava dos protocolos de necropsia, 76,2% tinham cirrose macronodular e 23,8% tinham cirrose micronodular. Nos 14 cães em que as lesões histológicas foram revistas pode-se observar diferentes graus de fibrose (leve [57,2%], moderada [21,4%] ou acentuada [21,4%]) e nenhuma relação entre as intensidades da fibrose e das outras lesões histológicas observadas (degeneração gordurosa, proliferação de ductos biliares, inflamação, bilestase, hemossiderose e necrose aleatória de hepatócitos). As alterações macro e microscópicas extra-hepáticas incluíram: ascite (39/63 [61,9%]), icterícia (19/63 [30,2%]), status spongiosus (15/63 [23,8%]), hidrotórax (12/63 [19,0%]), edema subcutâneo (10/63 [15,9%]), derivações portossistêmicas (11/63 [17,5%]), úlceras gástricas ou duodenais (11/63 [17,5%]) e nefrose colêmica (4/63 [6,3%]).

Dentre os 253 atendimentos realizados em aves selvagens entre agosto de 2003 a agosto de 2006 no Ambulatório de Animais Selvagens do Hospital Veterinário da Universidade Federal do Paraná, 45 casos (17,8%) referiram-se a consultas à espécie Serinus canarius (canário-belga). Dentre as aves atendidas e suas respectivas ordens obteve-se uma maior ocorrência da ordem Psittaciforme. As enfermidades mais freqüentemente visualizadas foram as afecções traumáticas com 56 casos (22,13%). Destas, 17 animais (30,91%) possuíam algum tipo de fratura, sendo a fratura rádio-ulnar a mais comum, com 17,65% de ocorrência. As outras moléstias mais relatadas foram a presença de ectoparasitos (12,50%) e endoparasitos (10,68%), doenças respiratórias (10,42%), procedimentos preventivos (7,55%), afecções dermatológicas (6,51%), neoplasias (4,95%), afecções oftálmicas (4,43%), afecções gastrintestinais (3,91%), caquexia (3,39%), afecções neurológicas (2,86%), automutilação (2,86%), obesidade (2,34%), agressão por outros animais (1,56 %), doenças nutricionais (1,30%), retenção de ovo (1,04%), bouba aviária (0,78%) e gota úrica (0,52%). Tendo em vista a alta prevalência de traumatismos e presença de ecto e endo parasitas que poderiam ser evitados se estivesse ocorrendo um manejo adequado com a ave, sugere-se a necessidade que o Médico Veterinário assuma um papel mais efetivo na Medicina Veterinária Preventiva buscando informar e debater questões referentes ao modo correto de alimentação, criação e manejo das aves, assim como também o esclarecimento acerca das questões referentes às zoonoses quando da consulta veterinária.

Mutations of the tumor suppressor gene p53 have been considered to be important determinants in several kinds of human cancer. Accumulation of p53 protein has been reported to correlate with more aggressive clinical behavior in some neoplasms. The role of p53 expression in adrenal cortical tumors (ACT) has not been elucidated but some studies have suggested its correlation with malignant behavior. Our objective was to determine if there is a correlation between the expression of immunoreactive p53 and the biological behavior of ACT. Fifty-seven ACT (21 from children and 36 from adults) were evaluated for p53 expression by immunohistochemistry in formalin-fixed paraffin-embedded tissue and analyzed in terms of outcome. The p53 parameter was utilized semiquantitatively. Tumors were classified as p53 negative when no positivity was observed, or when only few cells showed weak positivity (0/1+) and scored as p53 positive when there was a diffuse and strong nuclear positivity (2+/3+). In children, p53 positivity was associated with clinically malignant ACT and p53 negativity was associated with clinically benign ACT (P = 0.026). In adults' ACT, p53 positivity had an effect on disease-free survival (P<0.001) and also correlated with Weiss score, with a cutoff = 4 (P = 0.04). p53 expression was related to the clinical behavior of ACT in both children and adults and these findings seem to support a role for p53 in ACT progression.

Patients with sickle-cell anemia submitted to frequent blood transfusions are at risk of contamination with hepatitis C virus (HCV). Determination of HCV RNA and genotype characterization are parameters that are relevant for the treatment of the viral infection. The objective of the present study was to determine the frequency of HCV infection and the positivity for HCV RNA and to identify the HCV genotype in patients with sickle-cell anemia with a history of blood transfusion who had been treated at the Hospital of the HEMOPE Foundation. Sera from 291 patients were tested for anti-HCV antibodies by ELISA 3.0 and RIBA 3.0 Chiron and for the presence of HCV RNA by RT-PCR. HCV genotyping was performed in 19 serum samples. Forty-one of 291 patients (14.1%) were anti-HCV positive by ELISA and RIBA. Both univariate and multivariate analysis showed a greater risk of anti-HCV positivity in those who had started a transfusion regime before 1992 and received more than 10 units of blood. Thirty-four of the anti-HCV-positive patients (34/41, 82.9%) were also HCV RNA positive. Univariate analysis, used to compare HCV RNA-negative and -positive patients, did not indicate a higher risk of HCV RNA positivity for any of the variables evaluated. The genotypes identified were 1b (63%), 1a (21%) and 3a (16%). A high prevalence of HCV infection was observed in our patients with sickle-cell anemia (14.1%) compared to the population in general (3%). In the literature, the frequency of HCV infection in sickle-cell anemia ranges from 2 to 30%. The serological screening for anti-HCV at blood banks after 1992 has contributed to a better control of the dissemination of HCV infection. Because of the predominance of genotype 1, these patients belong to a group requiring special treatment, with a probable indication of new therapeutic options against HCV.

The objectives of the present study were to assess the in vitro-induced anti-hepatitis C virus (HCV) antibody production (IVIAP) in relation to the clinical, biochemical, virologic and histologic variables of patients with HCV infection. The study included 57 patients (60% males) with HCV infection (anti-HCV and HCV-RNA positive). Alanine aminotransferase (ALT) was elevated in 89% of the patients. Mean viral load was 542,241 copies/ml and histology of the liver showed chronic hepatitis in 27/52 (52%) and cirrhosis in 11/52 (21%) patients. IVIAP levels were determined by immunoenzymatic assay at median absorbance of 0.781 at 450 nm. IVIAP was negative in 14% of the patients. When groups with IVIAP levels above and below the median were compared, high IVIAP levels were associated with the male sex, elevated ALT levels and more advanced disease stage. After logistic regression analysis, advanced histologic damage to the liver remained as the only independent variable associated with elevated IVIAP levels. Using a receiver operator characteristic curve, the best cut-off level for IVIAP was established (= 1.540), with 71% sensitivity and 94% specificity for the detection of more advanced disease stages (grades 3 and 4). These findings are consistent with the participation of immunological mechanisms in the genesis of the hepatic lesions induced by HCV and indicate that the IVIAP test may be useful as a noninvasive marker of liver damage either alone or in combination with other markers.

In laboratory animals, acupuncture needs to be performed on either anesthetized or, if unanesthetized, restrained subjects. Both procedures up-regulate c-Fos expression in several areas of the central nervous system, representing therefore a major pitfall for the assessment of c-Fos expression induced by electroacupuncture. Thus, in order to reduce the effect of acute restraint we used a protocol of repeated restraint for the assessment of the brain areas activated by electroacupuncture in adult male Wistar rats weighing 180-230 g. Repeated immobilization protocols (6 days, 1 h/day and 13 days, 2 h/day) were used to reduce the effect of acute immobilization stress on the c-Fos expression induced by electroacupuncture at the Zusanli point (EA36S). Animals submitted to immobilization alone or to electroacupuncture (100 Hz, 2-4 V, faradic wave) in a non-point region were compared to animals submitted to electroacupuncture at EA36S (4 animals/subgroup). c-Fos expression was measured in 41 brain areas by simple counting of cells and the results are reported as number of c-Fos-immunoreactive cells/10,000 µm². The protocols of repeated immobilization significantly reduced the immobilization-induced c-Fos expression in most of the brain areas analyzed (P < 0.05). Animals of the EA36S groups had significantly higher levels of c-Fos expression in the dorsal raphe nucleus, locus coeruleus, posterior hypothalamus and central medial nucleus of the thalamus. Furthermore, the repeated immobilization protocols intensified the differences between the effects of 36S and non-point stimulation in the dorsal raphe nucleus (P < 0.05). These data suggest that high levels of stress can interact with and mask the evaluation of specific effects of acupuncture in unanesthetized animals.

Both genetic background and diet have profound effects on plasma lipid profiles. We hypothesized that a high-carbohydrate (high-CHO) diet may affect the ratios of serum lipids and apolipoproteins (apo) differently in subjects with different genotypes of the SstI polymorphism in the apoCIII gene (APOC3). Fifty-six healthy university students (27 males and 29 females, 22.89 ± 1.80 years) were given a washout diet of 54% carbohydrate for 7 days, followed by a high-CHO diet of 70% carbohydrate for 6 days without total energy restriction. Serum triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), apoB100, apoAI, and the APOC3 SstI polymorphism were analyzed. The ratios of serum lipids and apoB100/apoAI were calculated. At baseline, the TG/HDL-C ratio was significantly higher in females, but not in males, with the S2 allele. The differences in the TG/HDL-C ratio between genotypes remained the same after the washout and the high-CHO diet in females. When compared with those before the high-CHO diet, the TC/HDL-C (male S2 carriers: 3.13 ± 1.00 vs 2.36 ± 0.65, P = 0.000; male subjects with the S1S1 genotype: 2.97 ± 0.74 vs 2.09 ± 0.55, P = 0.000; female S2 carriers: 2.68 ± 0.36 vs 2.24 ± 0.37, P = 0.004; female subjects with the S1S1 genotype: 2.69 ± 0.41 vs 2.09 ± 0.31, P = 0.000) and LDL-C/HDL-C (male S2 carriers: 1.44 ± 0.71 vs 1.06 ± 0.26, P = 0.012; male subjects with the S1S1 genotype: 1.35 ± 0.61 vs 1.01 ± 0.29, P = 0.005; female S2 carriers: 1.18 ± 0.33 vs 1.00 ± 0.18, P = 0.049; female subjects with the S1S1 genotype: 1.18 ± 0.35 vs 1.04 ± 0.19, P = 0.026) ratios were significantly decreased after the high-CHO diet regardless of gender and of genotype of the APOC3 SstI polymorphism. However, in female S2 carriers, the TG/HDL-C (1.38 ± 0.46 vs 1.63 ± 0.70, P = 0.039) ratio was significantly increased after the high-CHO diet. In conclusion, the high-CHO diet has favorable effects on the TC/HDL-C and LDL-C/HDL-C ratios regardless of gender and of genotype of the APOC3 SstI polymorphism. Somehow, it enhanced the adverse effect of the S2 allele on the TG/HDL-C ratio only in females.

A acerola é um fruto altamente perecível e que contém altos teores de vitamina C, sendo este o seu principal atrativo em termos nutricionais. A atual legislação brasileira prevê uma variação de, no máximo, 20% do teor dos nutrientes especificados no rótulo. Devido a essa exigência seria importante que os fabricantes considerassem tanto o teor inicial de vitamina C quanto a perda ao longo da armazenagem dos produtos de acerola. Neste trabalho, foi feito o acompanhamento da estabilidade da vitamina C em polpa pasteurizada e acerola in natura congeladas, ambas armazenadas a -12ºC e -18ºC, e em suco de acerola pasteurizado engarrafado, mantido a temperatura ambiente, ao longo de 4 meses de armazenagem. As polpas congeladas não apresentaram degradação significativa durante este período, já as in natura apresentaram cinética de degradação de 1ª ordem e o suco de ordem zero. Após 4 meses de armazenagem as acerolas armazenadas a -12ºC e -18ºC apresentaram teores de 869±12 e 1.223±148 mg vit.C/100g, representando uma perda de 43% e 19%, respectivamente, em relação ao teor inicial. Polpas a -12ºC e -18ºC apresentaram teores de 1.314±6 e 1.322±2 mg vit.C/100g, respectivamente, representando uma perda de, aproximadamente, 3% e o suco apresentou uma perda de 32%, correspondendo a um teor final de 673±17mg vit.C/100g.

A ampla variabilidade genética nos pomares de aceroleiras brasileiros tem gerado frutos de coloração amarela a vermelha púrpura dificultando a obtenção de produtos de coloração avermelhada, cor esperada pelos consumidores. As antocianinas são pigmentos instáveis, responsáveis pela cor vermelha deste fruto maduro. Com o objetivo de determinar o teor destes pigmentos em polpa de acerola submetida ao congelamento foi instalado um experimento inteiramente casualisado. As polpas obtidas de frutos de 12 acessos (plantas) do Banco Ativo de Germoplasma da Universidade Federal Rural de Pernambuco (UFRPE) foram armazenadas a -18ºC, por um período de 06 meses. Unidades amostrais de 30g foram coletadas ao acaso, no período inicial e final do experimento, e submetidas a determinação quantitativa de antocianinas totais utilizando um método espectrofotométrico. Grande variação nos teores de antocianinas totais foi observada nos acessos estudados. No tempo zero de armazenamento o valor mínimo e máximo deste fitoquímico foi de 59,74mg e 3,79mg.100g -1 de polpa, respectivamente. Após seis meses de congelamento foi constatada uma redução de 3,4% a 23,6% no teor desse pigmento nas amostras avaliadas. Evidenciou-se, portanto, que o armazenamento a -18ºC por seis meses reduziu o teor de antocianinas e que os pigmentos antociânicos dos acessos 08 e 13 apresentaram-se mais estáveis ao congelamento.

A falta de classificação da acerola por classes varietais e a variabilidade de plantas, quanto ao porte, arquitetura da copa, coloração e forma dos frutos despertou o interesse quanto a sua caracterização química. Neste trabalho foram estudados frutos de três matrizes com características físicas distintas, nos estádios de maturação "de vez" e "maduro" e em distintas safras. Observaram-se diferenças significativas através da análise ANOVA e teste de Tukey (p<0,05), entre alguns parâmetros físicos (tamanho, peso), físico-químicos (pH e sólidos solúveis), químicos (açúcares redutores e totais, ácido ascórbico e pectina).

Os objetivos deste trabalho foram descrever e ilustrar os aspectos morfológicos da semente e do desenvolvimento da planta jovem de Amburana cearensis. Os frutos foram coletados de cinco matrizes distantes no mínimo 100m, levando-se em consideração o aspecto fitossanitário e a intensidade de frutificação. O endocarpo permanece aderido à semente formando uma ala, mesmo após a deiscência; o hilo apresenta fenda longitudinal típica da sub-família; a germinação é do tipo semi-hipógea fanerocotiledonar e, com o desenvolvimento observou-se a presença de uma tuberosidade na raiz primária. Os caracteres descritos e ilustrados mostraram-se bastante homogêneos entre as cinco matrizes, podendo ser empregados para identificação da referida espécie, tanto em trabalhos de laboratório e de produção de mudas quanto em estudos ecológicos.

Mild hypothermia has a protective effect on brain edema and encephalopathy in both experimental and human acute liver failure. The goals of the present study were to examine the effects of mild hypothermia (35°C) on brain metabolic pathways using combined 1H and 13C-Nuclear Magnetic Resonance (NMR) spectroscopy, a technique which allows the study not only of metabolite concentrations but also their de novo synthesis via cell-specific pathways in the brain. :1H and 13C NMR spectroscopy using [1-13C] glucose was performed on extracts of frontal cortex obtained from groups of rats with acute liver failure induced by hepatic devascularization whose body temperature was maintained either at 37°C (normothermic) or 35°C (hypothermic), and appropriate sham-operated controls. At coma stages of encephalopathy in the normothermic acute liver failure animals, glutamine concentrations in frontal cortex increased 3.5-fold compared to sham-operated controls (P < 0.001). Comparable increases of brain glutamine were observed in hypothermic animals despite the absence of severe encephalopathy (coma). Brain glutamate and aspartate concentrations were respectively decreased to 60.9% ± 7.7% and 42.2% ± 5.9% (P < 0.01) in normothermic animals with acute liver failure compared to control and were restored to normal values by mild hypothermia. Concentrations of lactate and alanine in frontal cortex were increased to 169.2% ± 15.6% and 267.3% ± 34.0% (P < 0.01) respectively in normothermic rats compared to controls. Furthermore, de novo synthesis of lactate and alanine increased to 446.5% ± 48.7% and 707.9% ± 65.7% (P < 0.001), of control respectively, resulting in increased fractional 13C-enrichments in these cytosolic metabolites. Again, these changes of lactate and alanine concentrations were prevented by mild hypothermia. Mild hypothermia (35°C) prevents the encephalopathy and brain edema resulting from hepatic devascularization, selectively normalizes lactate and alanine synthesis from glucose, and prevents the impairment of oxidative metabolism associated with this model of ALF, but has no significant effect on brain glutamine. These findings suggest that a deficit in brain glucose metabolism rather than glutamine accumulation is the major cause of the cerebral complications of acute liver failure.

Our knowledge of the agricultural sustainability of the millennia-old mountain oases in northern Oman is restricted in particular with respect to C and N turnover. A laboratory study was conducted (1) to analyse the effects of rewetting and drying on soil microorganisms after adding different manures, (2) to investigate the effects of mulching or incorporating of these manures, and (3) to evaluate the relationships between C and N mineralisation rates and manure quality indices. During the first 9-day rewetting and drying cycle, i.e. the “mulch” period, the content of extractable organic C decreased by approximately 40% in all four treatments. During the second 9-day rewetting and drying cycle, i.e. the “incorporation” period, this fraction decreased insignificantly in almost all treatments. The control and mature manure treatments form the first pair with a low percentage of total organic C evolved as CO2 (0.3% in 18 days) and a considerable percentage of total N mineralised as NH4 and NO3 (1% in 18 days), the fresh and immature manure treatments form the second pair with a higher amount of total organic C evolved as CO2 (0.5% in 18 days) and no net N mineralisation. During the first 9-day rewetting and drying cycle, the contents of microbial biomass C and biomass N increased by approximately 150% in all four treatments. During the second 9-day rewetting and drying cycle, no further increase was observed in the control and immature manure treatments and a roughly 30% increase in the other two treatments.
