The microbiotic crust study is among new focuses in investigating on the desertification control. Based on determination of algal crusts with different successive ages (4-, 8-, 17-, 34-, 42-year-old) and unconsolidated sand in the desert area, species composition and clustering analyses were carried out in this study. Results on successional orientation revealed that (1) the abundance of Cyanophyta, specially of Scytonema javanicum gradually decreased; (2) the abundance of Chlorophyta, Bacillariophyta and a species of Cyanophyta, Phormidium tenue increased; (3) the biodiversity increased gradually with the community succession; and (4) biomass of microalgae increased at the early stage, but decreased at the later stage due to the abundance of lichens and mosses. But, the speed of natural succession was so slow that the community-building species was still the first dominant species after 42 years, except that its dominant degree decreased just slightly. However, successive speed and trend were affected by water, vegetation coverage, terrain, time and soil physico-chemical properties as well, especially Mn content in the soil appeared to have a threshold effect.

Four filamentous cyanobacteria, Microcoleus vaginatus, Phormidium tenue, Scytonema javanicum (Kutz.) and Nostoc sp., and a single-celled green alga, Desmococcus olivaceus, all isolated from Shapotou (Ningxia Hui Autonomous Region of China), were batch cultured and inoculated onto unconsolidated sand in greenhouse and field experiments. Their ability to reduce wind erosion in sands was quantified by using a wind tunnel laboratory. The major factors related to cohesion of algal crusts, such as biomass, species, species combinations, bioactivity, niche, growth phase of algae, moisture, thickness of the crusts, dust accretion (including dust content and manner of dust added) and other cryptogams (lichens, fungi and mosses) were studied. The best of the five species were M. vaginatus and P. tenue, while the best mix was a blend of 80% M. vaginatus and 5% each of P. tenue, S. javanicum, Nostoc sp. and D. olivaceus. The threshold friction velocity was significantly increased by the presence of all of the cyanobacterial species, while the threshold impact velocity was notably increased only by the filamentous species. Thick crusts were less easily eroded than thin crusts, while biomass was more effective than thickness. Dust was incorporated best into Microcoleus crust when added in small amounts over time, and appeared to increase growth of the cyanobacterium as well as strengthen the cohesion of the crust. Microbial crust cohesion was mainly attributed to algal aggregation, while lichens, fungi and mosses affected more the soil structure and physico-chemical properties.

采用时空互代法,以典型侵蚀环境纸坊沟流域生态恢复过程中不同年限的人工刺槐林为研究对象,选取坡耕地和天然侧柏林为参照,分析了植被恢复过程中土壤水稳性团聚体变化规律及其与土壤养分状况之间的相互关系。结果表明,侵蚀环境下的坡耕地由于人为耕作干扰,土壤水稳性团聚体含量低下,抗蚀性能较差。营造刺槐林前5 a土壤水稳性团聚体含量较坡耕地显著快速增加,随后增幅变缓,成对数增长,恢复25 a后土壤水稳性团聚体含量已经达到天然侧柏林水平。在植被恢复过程中土壤小粒径的水稳性团聚体逐渐聚集转变形成大粒径的团聚体。相关性分析和回归分析表明,植被恢复过程中>0.25 mm土壤水稳性团聚体与有机碳、全氮、碱解氮、速效钾、容重等相关性达到显著(p<0.05)或极显著水平(p<0.01),与全磷和速效磷相关性较弱。坡耕地退耕营造刺槐林后可以减少人为干扰,增加碳素和氮素供给,提高水稳性团聚体含量,使土壤抗蚀性能提高。

采用时空互代法,以典型侵蚀环境纸坊沟流域不同封禁年限的狼牙刺群落和杂灌群落为研究对象,选取放牧地和天然次生林为参照,分析了生态恢复过程中土壤微生物生物量、呼吸强度、代谢商及理化性质的演变特征。结果表明,封禁后土壤理化性质明显改善;微生物生物量随封禁年限的延长变化显著,阳坡随封禁年限增加土壤微生物生物量逐渐增加,25 a后微生物生物量碳、氮、磷较封禁前分别增加252%、161%和174%,但显著低于天然侧柏林,仅为其39.0%、41.8%和53.7%;阴坡封禁前10年微生物生物量迅速增加,随后增加幅度减缓,呈波动式缓慢上升趋势;封禁25 a后微生物生物量碳、氮、磷分别增加108%、93%和102%,但明显低于天然杂灌丛群落和辽东栎林,仅为辽东栎林的54.4%、49.1%和40.1%。土壤呼吸强度在封禁5 a后增大明显,且随着年限增加逐渐上升,阳坡25 a时达到最大值,而阴坡15 a时达到最大值,随后开始有所下降,25 a后降至最低点,但仍显著高于放牧地,相同封禁年限的土壤呼吸强度阴坡明显高于阳坡。qCO2随着封禁进程逐渐降低,25 a后达到最低值。相关性分析显示微生物生物量碳、氮、磷、呼吸强度、qCO2与土壤养...

本文对东北地区松辽平原不同纬度农田土壤碳氮磷剖面分布特征进行比较研究,从北到南依次采集了黑土区的海伦、哈尔滨、德惠、公主岭和棕壤区的昌图、沈阳、大石桥玉米地土壤样品。所得主要结论如下: 各样点土壤有机碳含量随土层深度的增加而下降。海伦、哈尔滨和公主岭样点40~60cm土层土壤有机碳含量及其储量显著低于0~40cm土层;海伦、哈尔滨、德惠、公主岭和昌图样点土壤水溶性有机碳表现出随深度增加先升高后降低,在沈阳和大石桥样点土壤水溶性有机碳表现出随深度而下降的趋势;各样点0~20cm土层土壤微生物量碳含量高于20~40cm土层。典型黑土区海伦点0~100cm的SOC储量为213.4t•hm-2, 棕壤区昌图、沈阳、大石桥样点分别为69.9、87.9和73.4t•hm-2,海伦点SOC储量是棕壤区三样点的3倍左右。 土壤全氮、碱解氮、硝态氮及氮储量随剖面深度增加而下降。德惠点在20~40cm土层、沈阳点在40~60cm土层、昌图点在60~80cm土层的全磷含量最低;其他样点土壤全磷、有机磷含量和磷储量总体上呈现随土层深度增加而下降的趋势。黑土区样点土壤有机磷含量在40cm以下各土层迅速下降,而棕壤区各样点20cm以下各土层差异不显著。除公主岭和大石桥点外, 其他各样点土壤Olsen-P含量在0~20cm 土层显著高于20~40cm土层。 土壤有机碳、全氮、碱解氮、全磷和有机磷含量随纬度增加而增加。营养元素在纬度上的分异主要受成土母质、气候条件等自然因素影响,施肥、耕作等人为活动对表层土壤营养元素分布的影响较大。除土壤水溶性有机碳外,土壤碳、氮和磷之间及其与其他基本理化性质间均存在显著的相关关系。

This review, covering reports published from 2001 to December 2008, shows how ionic liquids (ILs) have made significant contributions in the improvement of capillary and microchip electrophoresis (CE and mu CE) for the separation and detection of analytes such as phenols and aromatic acids, metal ions, medicines, enantiomers. biological materials, etc. Furthermore, CE methods applied in the sensitive and accurate determination of physico-chemical properties of ILs have been summarized. Accordingly, research vacancies and future development trends in these areas are discussed.

The mixed oxides LaNiO3, La0.1Sr0.9NiO3, La2NiO4 and LaSrNiO4 were prepared and used as catalysts for the direct decomposition of NO. The catalysts were characterized by means of XRD, XPS, O-2-TPD, NO-TPD and chemical analysis. By comparing the physico-chemical properties and catalytic activity for NO decomposition, a conclusion could be drawn as follows. The direct decomposition of NO over perovskite and related mixed oxide catalysts follows a redox mechanism. The lower valent metal ions Ni2+ and disordered oxygen vacancies seem to be the active sites in the redox process. The oxygen vacancy plays an important role favorable for the adsorption and activation of NO molecules on one hand and on the other hand for increasing the mobility of lattice oxygen which is beneficial to the reproduction of active sites. The presence of oxygen vacancies is one of the indispensable factors to give the mixed oxides a steady activity for NO decomposition.

Geological fluids are important components in the earth system. To study thephysical chemistry properties and the evolution of fluid system turns out to be one of the most challenging issues in geosciences. Besides the conventional experimental approaches and theoretical or semi-theoretical modeling, molecular level computer simulation(MLCS) emerges as an alternative tool to quantificationally study the physico-chemical properties of fluid under extreme conditions in order to find out the characteristics and interaction of geological fluids in and around earth. Based on our previous study of the intermolecular potential for pure H2O and thestrict evaluation of the competitive potential models for pure CH4 and the ab initio fitting potential surface across H2O-CH4 molecules in this study, we carried out more than two thousand molecular dynamics simulations for the PVTx properties of pure CH4 and the H2O-CH4 mixtures. Comparison of 1941 simulations with experimental PVT data for pure CH4 shows an average deviation of 0.96% and a maximum deviation of 2.82%. The comparison of the results of 519 simulations of the mixtures with the experimental measurements reveals that the PVTx properties of the H2O-CH4 mixtures generally agree with the extensive experimental data with an average deviation of 0.83% and 4% in maximum, which is equivalent to the experimental uncertainty. Moreover, the maximum deviation between the experimental data and the simulation results decreases to about 2% as temperature and pressure increase,indicating that the high accuracy of the simulation is well retained in the high temperature and pressure region. After the validation of the simulation method and the intermolecular potential models, we systematically simulated the PVTx properties of this binary system from 673 K and 0.05 GPa to 2573 K and 10 GPa. In order to integrate all the simulation results and the experimental data for the calculation of thermodynamic properties, an equation of state (EOS) is developed for the H2O-CH4 system covering 673 to 2573 K and 0.01 to 10 GPa. Isochores for compositions < 4 mol% CH4 up to 773 K and 600 MPa are also determined in this thesis.

The composition of equine milk differs considerably from that of the milk of the principal dairying species, i.e., the cow, buffalo, goat and sheep. Because equine milk resembles human milk in many respects and is claimed to have special therapeutic properties, it is becoming increasingly popular in Western Europe, where it is produced on large farms in several countries. Equine milk is considered to be highly digestible, rich in essential nutrients and to possess an optimum whey protein:casein ratio, making it very suitable as a substitute for bovine milk in paediatric dietetics. There is some scientific basis for the special nutritional and health-giving properties of equine milk but this study provides a comprehensive analysis of the composition and physico-chemical properties of equine milk which is required to fully exploit its potential in human nutrition. Quantification and distribution of the nitrogenous components and principal salts of equine milk are reported. The effects of the high concentration of ionic calcium, large casein micelles (~ 260 nm), low protein, lack of a sulphydryl group in equine β-lactoglobulin and a very low level of κ-casein on the physico-chemical properties of equine milk are reported. This thesis provides an insight into the stability of equine casein micelles to heat, ethanol, high pressure, rennet or acid. Differences in rennet- and acid-induced coagulation between equine and bovine milk are attributed not only to the low casein content of equine milk but also to differences in the mechanism by which the respective micelles are stabilized. It has been reported that β-casein plays a role in the stabilization of equine casein micelles and proteomic techniques support this view. In this study, equine κ-casein appeared to be resistant to hydrolysis by calf chymosin but equine β-casein was readily hydrolysed. Resolution of equine milk proteins by urea-PAGE showed the multi-phosphorylated isoforms of equine αs- and β-caseins and capillary zone electrophoresis showed 3 to 7 phosphorylated residues in equine β-casein. In vitro digestion of equine β-casein by pepsin and Corolase PP™ did not produce casomorphins BCM-5 or BCM-7, believed to be harmful to human health. Electron microscopy provided very clear, detailed images of equine casein micelles in their native state and when renneted or acidified. Equine milk formed flocs rather then a gel when renneted or acidified which is supported by dynamic oscillatory analysis. The results presented in this thesis will assist in the development of new products from equine milk for human consumption which will retain some of its unique compositional and health-giving properties.

In recent years, extensive research has been carried out on the health benefits of milk proteins and peptides. Biologically active peptides are defined as specific protein fragments which have a positive impact on the physiological functions of the body; such peptides are produced naturally in vivo, but can also be generated by physical and/or chemical processes, enzymatic hydrolysis and/or microbial fermentation. The aims of this thesis were to investigate not only the traditional methods used for the generation of bioactive peptides, but also novel processes such as heat treatment, and the role of indigenous milk proteases, e.g., in mastitic milk, in the production of such peptides. In addition, colostrum was characterised as a source of bioactive proteins and peptides. Firstly, a comprehensive study was carried out on the composition and physical properties of colostrum throughout the early-lactation period. Marked differences in the physico-chemical properties of colostrum compared with milk were observed. Various fractions of colostrum were also tested for their effect on the secretion of pro- and anti-inflammatory cytokines from a macrophage cell line and bone marrow dendritic cells, as well as insulin secretion from a pancreatic beta cell line. A significant reduction in the secretion of the pro-inflammatory cytokines, TNF-α, IL-6, IL-1β and IL-12, a significant increase in the secretion of the anti-inflammatory cytokine, IL-10, as well as a significant increase in insulin secretion were observed for various colostrum fractions. Another study examined the early proteomic changes in the milk of 8 cows in response to infusion with the endotoxin lipopolysaccharide (LPS) at quarter level in a model mastitic system; marked differences in the protein and peptide profile of milk from LPS challenged cows were observed, and a pH 4.6-soluble fraction of this milk was found to cause a substantial induction in the secretion of IL-10 from a murine macrophage cell line. Heat-induced hydrolysis of sodium caseinate was investigated from the dual viewpoints of protein breakdown and peptide formation, and, a peptide fraction produced in this manner was found to cause a significant increase in the secretion of the anti-inflammatory cytokine, IL-10, from a murine macrophage cell line. The effects of sodium caseinate hydrolysed by chymosin on the gut-derived satiety hormone glucagon-like peptide-1 (GLP-1) were investigated; the resulting casein-derived peptides displayed good in vitro and in vivo secretion of GLP-1. Overall, the studies described in this thesis expand on current knowledge and provide good evidence for the use of novel methods for the isolation, generation and characterisation of bioactive proteins and/or peptides.

The ammonia oxidation reaction on supported polycrystalline platinum catalyst was investigated in an aluminum-based microreactor. An extensive set of reactions was included in the chemical reactor modeling to facilitate the construction of a kinetic model capable of satisfactory predictions for a wide range of conditions (NH3 partial pressure, 0.01-0.12 atm; O-2 partial pressure, 0.10-0.88 atm; temperature, 523-673 K; contact time, 0.3-0.7 ms). The elementary surface reactions used in developing the mechanism were chosen based on the literature data concerning ammonia oxidation on a Pt catalyst. Parameter estimates for the kinetic model were obtained using multi-response least squares regression analysis using the isothermal plug-flow reactor approximation. To evaluate the model, the behavior of a microstructured reactor was simulated by means of a complete Navier-Stokes model accounting for the reactions on the catalyst surface and the effect of temperature on the physico-chemical properties of the reacting mixture. In this way, the effect of the catalytic wall temperature non-uniformity and the effect of a boundary layer on the ammonia conversion and selectivity were examined. After further optimization of appropriate kinetic parameters, the calculated selectivities and product yields agree very well with the values actually measured in the microreactor. Literature data on the toxicity of chlorophenols for three luminescent bacteria (Vibrio fischeri, and the lux-marked Pseudomonas fluorescens 10586s pUCD607 and Burkholderia spp. RASC c2 (Tn4431)) have been analyzed in relation to a set of computed molecular physico-chemical properties. The quantitative structure-toxicity relationships of the compounds in each species showed marked differences when based upon semi-empirical molecular-orbital molecular and atom based properties. For mono-, di- and tri-chlorophenols multiple linear regression analysis of V. fischeri toxicity showed a good correlation with the solvent accessible surface area and the charge on the oxygen atom. This correlation successfully predicted the toxicity of the heavily chlorinated phenols, suggesting in V. fischeri only one overall mechanism is present for all chlorophenols. Good correlations were also found for RASC c2 with molecular properties, such as the surface area and the nucleophilic super-delocalizability of the oxygen. In contrast the best QSTR for P. fluorescens contained the 2nd order connectivity index and ELUMO suggesting a different, more reactive mechanism. Cross-species correlations were examined, and between V. fischeri and RASC c2 the inclusion of the minimum value of the nucleophilic susceptibility on the ring carbons produced good results. Poorer correlations were found with P. fluorescens highlighting the relative similarity of V. fischeri and RASC c2, in contrast to that of P. fluorescens.

Autoclaved soil is commonly used for the study of xenobiotic sorption and as an abiotic control in biodegradation experiments. Autoclaving has been reported to alter soil physico-chemical and xenobiotic sorption characteristics such that comparison of autoclaved with non-autoclaved treatments in soil aging and bioavailability studies may yield misleading results. Experiments could be improved by using autoclaved soil re-inoculated with indigenous microorganisms as an additional or alternative non-sterile treatment for comparison with the sterile, autoclaved control. We examined the effect of autoclaving (3 x 1 h, 121°C, 103.5 KPa) on the physico-chemical properties of a silt loam soil (pH 7.2, 2.3% organic carbon) and the establishment of indigenous microorganisms reintroduced after autoclaving. Sterilisation by autoclaving significantly (p ≤ 0.05) decreased pH (0.6 of a unit) and increased concentrations of water-soluble organic carbon (WSOC; nontreated = 75 mg kg-1; autoclaved = 1526 mg kg-1). The initial first-order rate of 14C-2,4-dichloro-UL-phenol (2,4-DCP) adsorption to non-treated, autoclaved and re-inoculated soil was rapid (K1 = 16.8-24.4 h-1) followed by a slower linear phase (K2). In comparison with autoclaved soil (0.038% day-1), K2 values were higher for re-inoculated (0.095% day-1) and nontreated (0.181% day-1) soil. This was attributed to a biological process. The Freundlich adsorption coefficient (K(f)) for autoclaved soil was significantly (p ≤ 0.05) higher than for re-inoculated or non-treated soil. Increased adsorption was attributed to autoclaving-induced changes to soil pH and solution composition. Glucose-induced respiration of autoclaved soil after re-inoculation was initially twice that in the non-treated control, but it decreased to control levels by day 4. This reduction corresponded to a depletion of WSOC. 2,4-DCP mineralisation experiments revealed that the inoculum of nonsterile soil (0.5 g) contained 2,4-DCP-degrading microorganisms capable of survival in autoclaved soil. The lag phase before detection of significant 2,4-DCP mineralisation was reduced (from 7 days to ≤3 days) by pre-incubation of re-inoculated soils for 7 and 14 days before 2,4-DCP addition. This was attributed to the preferential utilisation of WSOC prior to the onset of 2,4-DCP mineralisation. Cumulative 14CO2 evolved after 21 days was significantly lower (p ≤ 0.05) from non-treated soil (25.3%) than re-inoculated soils (ca 45%). Experiments investigating sorption-biodegradation interactions of xenobiotics in soil require the physico-chemical properties of sterile and non-sterile treatments to be as comparable as possible. For fundamental studies, we suggest using re-inoculated autoclaved soil as an additional or alternative non-sterile treatment.
