Mixture modeling methodology was used to investigate interactions of sugar, oligofructose and inulin in papaya nectars as related to sensory liking and chemical characteristics. Mixing sugar and inulin and increasing the sugar proportion raised the liking of flavor and sweetness and the overall acceptability of papaya nectars. Addition of the three components, along with raising the sugar proportion, increased the ash and soluble solids content in papaya nectars. The internal preference mappings showed that all nectars with oligofructose and inulin were as well liked as nectar containing sugar alone, except for some formulations with lower quantities of sugar. Formulations with 6 g/100 g sugar and 6 g/100 g inulin, or with 8 g/100 g sugar, 2 g/100 g inulin and 2 g/100 g oligofructose, can be considered to be the best formulations to produce, with regard to sensory liking and adequacy of chemical parameters, besides all papaya nectars with addition of oligofructose and inulin can potentially be claimed as prebiotic. Petroleum contamination impact on macrobenthic communities in the northeast portion of Todos os Santos Bay was assessed combining in multivariate analyses, chemical parameters such as aliphatic and polycyclic aromatic hydrocarbon indices and concentration ratios with benthic ecological parameters. Sediment samples were taken in August 2000 with a 0.05 m(2) van Veen grab at 28 sampling locations. The predominance of n-alkanes with more than 24 carbons, together with CPI values close to one, and the fact that most of the stations showed UCM/resolved aliphatic hydrocarbons ratios (UCM:R) higher than two, indicated a high degree of anthropogenic contribution, the presence of terrestrial plant detritus, petroleum products and evidence of chronic oil pollution. The indices used to determine the origin of PAH indicated the occurrence of a petrogenic contribution. A pyrolytic contribution constituted mainly by fossil fuel combustion derived PAH was also observed. The results of the stepwise multiple regression analysis performed with chemical data and benthic ecological descriptors demonstrated that not only total PAH concentrations but also specific concentration ratios or indices such as >= C24:< C24, An/178 and Fl/Fl + Py, are determining the structure of benthic communities within the study area. According to the BIO-ENV results petroleum related variables seemed to have a main influence on macrofauna community structure. The PCA ordination performed with the chemical data resulted in the formation of three groups of stations. The decrease in macrofauna density, number of species and diversity from groups III to I seemed to be related to the occurrence of high aliphatic hydrocarbon and PAH concentrations associated with fine sediments. Our results showed that macrobenthic communities in the northeast portion of Todos os Santos Bay are subjected to the impact of chronic oil pollution as was reflected by the reduction in the number of species and diversity. These results emphasise the importance to combine in multivariate approaches not only total hydrocarbon concentrations but also indices, isomer pair ratios and specific compound concentrations with biological data to improve the assessment of anthropogenic impact on marine ecosystems. Previous studies have shown that heparin induces vascular relaxation via integrin-dependent nitric oxide (NO)-mediated activation of the muscarinic receptor. The aim of this study was to identify the structural features of heparin that are necessary for the induction of vasodilatation. To address this issue, we tested heparin from various sources for their vasodilatation activities in the rat aorta ring. Structural and chemical characteristics of heparin, such as its molecular weight and substitution pattern, did not show a direct correlation with the vasodilation activity. Principal component analysis (PCA) of circular dichroism (CD), 1H-nuclear magnetic resonance (NMR) and vasodilation activity measurements confirmed that there is no direct relationship between the physico-chemical nature and vasodilation activity of the tested heparin samples. To further understand these observations, unfractionated heparin (UFH) from bovine intestinal mucosa, which showed the highest relaxation effect, was chemically modified. Interestingly, non-specific O- and N-desulfation of heparin reduced its anticoagulant, antithrombotic, and antihemostatic activities, but had no effect on its ability to induce vasodilation. On the other hand, chemical reduction of the carboxyl groups abolished heparin-induced vasodilation and reduced the affinity of heparin toward the extracellular matrix (ECM). In addition, dextran and dextran sulfate (linear non-sulfated and highly sulfated polysaccharides, respectively) did not induce significant relaxation, showing that the vasodilation activity of polysaccharides is neither charge-dependent nor backbone unspecific. In this thesis we focussed on the characterization of the reaction center (RC) protein purified from the photosynthetic bacterium Rhodobacter sphaeroides. In particular, we discussed the effects of native and artificial environment on the light-induced electron transfer processes. The native environment consist of the inner antenna LH1 complex that copurifies with the RC forming the so called core complex, and the lipid phase tightly associated with it. In parallel, we analyzed the role of saccharidic glassy matrices on the interplay between electron transfer processes and internal protein dynamics. As a different artificial matrix, we incorporated the RC protein in a layer-by-layer structure with a twofold aim: to check the behaviour of the protein in such an unusual environment and to test the response of the system to herbicides. By examining the RC in its native environment, we found that the light-induced charge separated state P+QB - is markedly stabilized (by about 40 meV) in the core complex as compared to the RC-only system over a physiological pH range. We also verified that, as compared to the average composition of the membrane, the core complex copurifies with a tightly bound lipid complement of about 90 phospholipid molecules per RC, which is strongly enriched in cardiolipin. In parallel, a large ubiquinone pool was found in association with the core complex, giving rise to a quinone concentration about ten times larger than the average one in the membrane. Moreover, this quinone pool is fully functional, i.e. it is promptly available at the QB site during multiple turnover excitation of the RC. The latter two observations suggest important heterogeneities and anisotropies in the native membranes which can in principle account for the stabilization of the charge separated state in the core complex. The thermodynamic and kinetic parameters obtained in the RC-LH1 complex are very close to those measured in intact membranes, indicating that the electron transfer properties of the RC in vivo are essentially determined by its local environment. The studies performed by incorporating the RC into saccharidic matrices evidenced the relevance of solvent-protein interactions and dynamical coupling in determining the kinetics of electron transfer processes. The usual approach when studying the interplay between internal motions and protein function consists in freezing the degrees of freedom of the protein at cryogenic temperature. We proved that the “trehalose approach” offers distinct advantages with respect to this traditional methodology. We showed, in fact, that the RC conformational dynamics, coupled to specific electron transfer processes, can be modulated by varying the hydration level of the trehalose matrix at room temperature, thus allowing to disentangle solvent from temperature effects. The comparison between different saccharidic matrices has revealed that the structural and dynamical protein-matrix coupling depends strongly upon the sugar. The analyses performed in RCs embedded in polyelectrolyte multilayers (PEM) structures have shown that the electron transfer from QA - to QB, a conformationally gated process extremely sensitive to the RC environment, can be strongly modulated by the hydration level of the matrix, confirming analogous results obtained for this electron transfer reaction in sugar matrices. We found that PEM-RCs are a very stable system, particularly suitable to study the thermodynamics and kinetics of herbicide binding to the QB site. These features make PEM-RC structures quite promising in the development of herbicide biosensors. The studies discussed in the present thesis have shown that, although the effects on electron transfer induced by the native and artificial environments tested are markedly different, they can be described on the basis of a common kinetic model which takes into account the static conformational heterogeneity of the RC and the interconversion between conformational substates. Interestingly, the same distribution of rate constants (i.e. a Gamma distribution function) can describe charge recombination processes in solutions of purified RC, in RC-LH1 complexes, in wet and dry RC-PEM structures and in glassy saccharidic matrices over a wide range of hydration levels. In conclusion, the results obtained for RCs in different physico-chemical environments emphasize the relevance of the structure/dynamics solvent/protein coupling in determining the energetics and the kinetics of electron transfer processes in a membrane protein complex.

Zusammenfassung Die vorliegende Arbeit hat zum Ziel, pharmazeutisch-technologische Möglichkeiten der Retardierung bei ausgewählten Antiasthmatika zur pulmonalen Applikation anzuwenden. Dafür sollten Mikropartikel hergestellt und pharmazeutisch sowie biopharmazeutisch charakterisiert werden. Als Modellsubstanzen werden das Glukokortikoid Budesonid und das β2-Sympathomimetikum Salbutamol in Form seiner Base und seines Salzes verwendet. Die Auswahl erfolgt nach physikochemischen (Lipophilie, Molekulargewicht) und therapeutischen (Halbwertszeit der Wirkung, Applikationsfrequenz) Gesichtspunkten. Mikropartikel auf Polymerbasis ermöglichen eine kontrollierte Freigabe der Arzneistoffe über einen vorausbestimmten Zeitraum. Es erfolgt die Auswahl physiologisch unbedenklicher Hilfsstoffe (Polylaktide R 202H/ Poly(laktid-co-glykolide) RG 502H, RG 752-S) mit unterschiedlichen Anteilen an Coglykolid sowie unterschiedlichen Molekulargewichten, die sich prinzipiell zur Verzögerung der Freisetzung eignen und sich bei der parenteralen Applikation bereits bewährt haben. Die Sprühtrocknung wird als geeignetes pharmazeutisch-technologisches Verfahren zur Präparation von Mikropartikeln im Teilchengrößenbereich von 1- 10 Mikrometern beschrieben, welche den Wirkstoff mit möglichst hoher Beladung verkapselt. Die sprühgetrockneten Pulver sollen pharmazeutisch physikochemisch mittels Rasterelektronenmikroskopie (Morphologie), Laserdiffraktometrie (Teilchengrößenverteilung), DSC und Röntgenpulverdiffraktometrie (thermisches Verhalten) und mittels Stickstoff-Tief-Temperatur Adsorptionsverfahren (spezifische Oberfläche) charakterisiert werden. Zusätzlich wird die Wirkstoffbeladung der sprühgetrockneten Polymer-Mikropartikel mittels HPLC ermittelt. Die biopharmazeutische Charakterisierung der sprühgetrockneten Pulver erfolgt über die in-vitro Freigabekinetik und die Stabilität der Mikropartikel. Zusätzlich werden Versuche an Zellkulturen und in-vivo Versuche an Mäusen durchgeführt, um die Effekte der sprühgetrockneten Mikropartikel und des Hilfsstoffs hinsichtlich der Freisetzungsretardierung zu testen. Bei den in-vivo Versuchen werden der Atemwegswiderstand und die Verlängerung der exspiratorischen Phase (penh) als Parameter für einen antiasthmatischen Effekt gewählt. Die Lungenlavage Flüssigkeit wird zusätzlich überprüft. Die Ergebnisse zeigen, dass es mit Hilfe der Sprühtrocknung möglich ist, Polymer-Mikropartikel herzustellen, die aufgrund ihrer Partikelgröße von d50 ≤ 5,8 µm fähig sind, die unteren Abschnitte der Lunge zu erreichen. Die Morphologie der Mikropartikel ist abhängig vom zu versprühenden Produkt. Thermodynamisch und röntgenpulverdiffraktometrisch betrachtet handelt es sich um amorphe Produkte, die aber über lange Zeit in diesem Zustand stabil sind. Die Wiederfindung der eingesetzten Arzneistoffmenge in den sprühgetrockneten Polymer-Mikropartikeln und die Freigabeversuche zur Charakterisierung der Retardierungseigenschaften der verwendeten Polymere ergeben, dass es mit Hilfe der Sprühtrocknung von Budesonid und Salbutamol mit den Polymeren möglich ist, retardierende Mikropartikel herzustellen. Die Wiederfindung von Budesonid und Salbutamol in den sprühgetrockneten Polymer-Mikropartikeln entspricht nahezu der eingesetzten Menge. Bei Salbutamolsulfat ist dies nicht der Fall. In Zellkulturversuchen der murinen Zellinie RAW 264.7 ergaben sich Hinweise darauf, dass bei Konzentrationen von 10-6 M und 10-8 M, die Downregulation der IL-6 Konzentration durch die Sprüheinbettung von 9,1 % Budesonid mit PLGA in stärkerem Ausmaß erfolgte, als bei unverkapseltem Budesonid. Zusätzlich wurden in-vivo Versuche mit intranasaler und intraperitonealer Gabe durchgeführt. Die Budesonid-Polymer Sprüheinbettung wurde mit unverkapseltem Budesonid vergleichen. Nach intraperitonealer Gabe hatte die Sprüheinbettung mit Budesonid die besten Effekte hinsichtlich der Unterdrückung des penh und des Atemwegswiderstands auch bei steigenden Metacholinkonzentrationen. Die Auswertung der Lungenlavage Flüssigkeit zeigt sehr deutlich die Downregulation der IL-6 Konzentration in der Lunge durch die Sprüheinbettung mit Budesonid. Zur Zeit werden Vorbereitungen getroffen, ein Gerät zu testen, das in der Lage ist, ein Mikrospray zu generieren, so dass eine intratracheale Verabreichung möglich wäre.

Soil is a critically important component of the earth’s biosphere. Developing agricultural production systems able to conserve soil quality is essential to guarantee the current and future capacity of soil to provide goods and services. This study investigates the potential of microbial and biochemical parameters to be used as early and sensitive soil quality indicators. Their ability to differentiate plots under contrasting fertilization regimes is evaluated based also on their sensitivity to seasonal fluctuations of environmental conditions and on their relationship with soil chemical parameters. Further, the study addresses some of the critical methodological aspects of microplate-based fluorimetric enzyme assays, in order to optimize assay conditions and evaluate their suitability to be used as a toll to asses soil quality. The study was based on a long-term field experiment established in 1966 in the Po valley (Italy). The soil was cropped with maize (Z. mays L.) and winter wheat (T. aestivum L.) and received no organic fertilization, crop residue or manure, in combination with increasing levels of mineral N fertilizer. The soil microbiota responded to manure amendment increasing it biomass and activity and changing its community composition. Crop residue effect was much more limited. Mineral N fertilization stimulated crop residue mineralization, shifted microbial community composition and influenced N and P cycling enzyme activities. Seasonal fluctuations of environmental factors affected the soil microbiota. However microbial and biochemical parameters seasonality did not hamper the identification of fertilization-induced effects. Soil microbial community abundance, function and composition appeared to be strongly related to soil organic matter content and composition, confirming the close link existing between these soil quality indicators. Microplate-based fluorimetric enzyme assays showed potential to be used as fast and throughput toll to asses soil quality, but required proper optimization of the assay conditions for a precise estimation of enzymes maximum potential activity.

With the increasing importance that nanotechnologies have in everyday life, it is not difficult to realize that also a single molecule, if properly designed, can be a device able to perform useful functions: such a chemical species is called chemosensor, that is a molecule of abiotic origin that signals the presence of matter or energy. Signal transduction is the mechanism by which an interaction of a sensor with an analyte yields a measurable form of energy. When dealing with the design of a chemosensor, we need to take into account a “communication requirement” between its three component: the receptor unit, responsible for the selective analyte binding, the spacer, which controls the geometry of the system and modulates the electronic interaction between the receptor and the signalling unit, whose physico-chemical properties change upon complexation. A luminescent chemosensor communicates a variation of the physico-chemical properties of the receptor unit with a luminescence output signal. This thesis work consists in the characterization of new molecular and nanoparticle-based system which can be used as sensitive materials for the construction of new optical transduction devices able to provide information about the concentration of analytes in solution. In particular two direction were taken. The first is to continue in the development of new chemosensors, that is the first step for the construction of reliable and efficient devices, and in particular the work will be focused on chemosensors for metal ions for biomedical and environmental applications. The second is to study more efficient and complex organized systems, such as derivatized silica nanoparticles. These system can potentially have higher sensitivity than molecular systems, and present many advantages, like the possibility to be ratiometric, higher Stokes shifts and lower signal-to-noise ratio.

Ziel dieser Dissertation ist die experimentelle Charakterisierung und quantitative Beschreibung der Hybridisierung von komplementären Nukleinsäuresträngen mit oberflächengebundenen Fängermolekülen für die Entwicklung von integrierten Biosensoren. Im Gegensatz zu lösungsbasierten Verfahren ist mit Microarray Substraten die Untersuchung vieler Nukleinsäurekombinationen parallel möglich. Als biologisch relevantes Evaluierungssystem wurde das in Eukaryoten universell exprimierte Actin Gen aus unterschiedlichen Pflanzenspezies verwendet. Dieses Testsystem ermöglicht es, nahe verwandte Pflanzenarten auf Grund von geringen Unterschieden in der Gen-Sequenz (SNPs) zu charakterisieren. Aufbauend auf dieses gut studierte Modell eines House-Keeping Genes wurde ein umfassendes Microarray System, bestehend aus kurzen und langen Oligonukleotiden (mit eingebauten LNA-Molekülen), cDNAs sowie DNA und RNA Targets realisiert. Damit konnte ein für online Messung optimiertes Testsystem mit hohen Signalstärken entwickelt werden. Basierend auf den Ergebnissen wurde der gesamte Signalpfad von Nukleinsärekonzentration bis zum digitalen Wert modelliert. Die aus der Entwicklung und den Experimenten gewonnen Erkenntnisse über die Kinetik und Thermodynamik von Hybridisierung sind in drei Publikationen zusammengefasst die das Rückgrat dieser Dissertation bilden. Die erste Publikation beschreibt die Verbesserung der Reproduzierbarkeit und Spezifizität von Microarray Ergebnissen durch online Messung von Kinetik und Thermodynamik gegenüber endpunktbasierten Messungen mit Standard Microarrays. Für die Auswertung der riesigen Datenmengen wurden zwei Algorithmen entwickelt, eine reaktionskinetische Modellierung der Isothermen und ein auf der Fermi-Dirac Statistik beruhende Beschreibung des Schmelzüberganges. Diese Algorithmen werden in der zweiten Publikation beschrieben. Durch die Realisierung von gleichen Sequenzen in den chemisch unterschiedlichen Nukleinsäuren (DNA, RNA und LNA) ist es möglich, definierte Unterschiede in der Konformation des Riboserings und der C5-Methylgruppe der Pyrimidine zu untersuchen. Die kompetitive Wechselwirkung dieser unterschiedlichen Nukleinsäuren gleicher Sequenz und die Auswirkungen auf Kinetik und Thermodynamik ist das Thema der dritten Publikation. Neben der molekularbiologischen und technologischen Entwicklung im Bereich der Sensorik von Hybridisierungsreaktionen oberflächengebundener Nukleinsäuremolekülen, der automatisierten Auswertung und Modellierung der anfallenden Datenmengen und der damit verbundenen besseren quantitativen Beschreibung von Kinetik und Thermodynamik dieser Reaktionen tragen die Ergebnisse zum besseren Verständnis der physikalisch-chemischen Struktur des elementarsten biologischen Moleküls und seiner nach wie vor nicht vollständig verstandenen Spezifizität bei.

Die Ladungsdichte eines Polyelektrolyten bestimmt im Wesentlichen das spezielle physikochemische Verhalten in Lösung und stellt einen der wichtigsten Parameter bei deren Anwendung dar. Folglich ist ein quantitatives Verständis über die effektive Ladung unabdingbar und stellt nach wie vor einen Schwerpunkt aktueller Polymerforschungen dar. Allgemein anerkannt ist die Tatsache, dass die effektive Ladungsdichte in allen Fällen niedriger ist, als es die chemische Struktur vorgibt. Die vorliegende Arbeit befasst sich mit der quantitativen Bestimmung der effektiven Ladung eines linearen, anionischen Polyelektrolyts in einem organischen Lösungsmittel. Zu diesem Zweck wurde ein Polymer mit geschützten Carboxylatgruppen synthetisiert, das eine nachträgliche quantitative Anpassung der Ladungsdichte durch Verseifung eines Esters erlaubt. Der auf diesemWeg synthetisierte Polyelektrolyt besitzt eine theoretische Ladungsdichte von 20% und wurde mit Lichtstreutechniken, Fluoreszenzkorrelationsspektroskopie und elektrophoretischen Methoden untersucht um anschließend aus den erhaltenen Daten, unter Verwendung verschiedener Theorien, die effektive Ladung in Abhängigkeit der Polymer- und Salzkonzentration zu bestimmen. Die Ergebnisse liefern, in Abhängigkeit der verwendeten Theorie, eine konstante oder von den äußeren Parametern abhängige Ladungsdichte, die immer deutlich unter der theoretisch möglichen liegt.

Particulate matter (PM) pollution is a leading cause of premature death, particularly in those with pre-existing lung disease. A causative link between particle properties and adverse health effects remains unestablished mainly due to complex and variable physico-chemical PM parameters. Controlled laboratory experiments are required. Generating atmospherically realistic Aerosols and performing cell-exposure studies at relevant particle-doses are challenging. Here we examine gasoline-exhaust particle toxicity from a Euro-5 passenger car in a uniquely realistic exposure scenario, combining a smog chamber simulating atmospheric ageing, an aerosol enrichment System varying particle number concentration independent of particle chemistry, and an aerosol Deposition chamber physiologically delivering particles on air-liquid interface (ALI) cultures reproducing normal and susceptible health status. Gasoline-exhaust is an important PM source with largely unknown health effects. We investigated acute responses of fully-differentiated normal, distressed (antibiotics treated) normal, and cystic fibrosis human bronchial epithelia (HBE), and a proliferating, single-cell type bronchial epithelial cell-line (BEAS-2B). We show that a single, short-term exposure to realistic doses of atmospherically-aged gasoline-exhaust particles impairs epithelial key-defence mechanisms, rendering it more vulnerable to subsequent hazards. We establish dose-response curves at realistic particle-concentration levels. Significant differences between cell models suggest the use of fully differentiated HBE is most appropriate in future toxicity studies.

n the framework of the FRUELA project, two oceanographic surveys were conducted by R/V Hespérides in the eastern Bellingshausen Sea, western basin of the Bransfield Strait and Gerlache Strait area during December 1995 and January 1996. The main hydrographic structures of the study domain were the Southern Boundary of the ACC and the Bransfield Front. The characteristics and zonation of local water masses are discussed in terms of temperature, salinity, dissolved oxygen, nutrient and inorganic carbon concentrations. Concentration intervals for water mass labelling, on the basis of chemical parameters in addition to the common theta/S-based classification, are defined. Silicate seems to be a very good discriminator for local water masses.
