The potential reproductive value of arbuscular mycorrhizal fungi (Gloinus intraradices and Glomus invermaium), root pathogenic fungi (Rhizoctonia solani and Fusarium culmorum) and saprotrophic fungi (Penicillium hordei and Trichoderma harzianum) were examined for the collembolans Folsomia candida Willem and Folsomia fimetaria L. Dried baker's yeast (Saccharomyces cerevisiae) was used as a reference standard food in laboratory cultures. Collembolan performance was determined as final size, fecundity and population growth rate after when fed the fungal food sources for 31 days. The mycorrhizal fungi gave the least growth and fecundity compared with the other fungi, but G. intraradices gave good fecundity for F. candida. In terms of growth, Baker's yeast was a high-quality food for both adults and juveniles of both species, but it was a poorer food in terms of fecundity of F. candida. Preference of the fungi in all possible pairwise combinations showed that although F. fimetaria did not perform well on Glomus spp. and F. candida did not grow well on Glomus spp. their preference for these fungi did not reflect this. The highest fecundity was seen with the root pathogen F. culmorum. Different quality indicators such as the C:N ratio of the fungal food sources as well as other biological parameters are discussed in relation to their reproductive value and Collembola preferential feeding. Truly continuous solid-state fermentations with operating times of 2-3 weeks were conducted in a prototype bioreactor for the production of fungal (Penicillium glabrum) tannase from a tannin-containing model substrate. Substantial quantities of the enzyme were synthesized throughout the operating periods and (imperfect) steady-state conditions seemed to be achieved soon after start-up of the fermentations. This demonstrated for the first time the possibility of conducting solid-state fermentations in the continuous mode and with a constant noninoculated feed. The operating variables and fermentation conditions in the bioreactor were sufficiently well predicted for the basic reinoculation concept to succeed. However, an incomplete understanding of the microbial mechanisms, the experimental system, and their interaction indicated the need for more research in this novel area of solid-state fermentation. Possible evidence is presented for Maillard glycation of enzymes during oligosaccharide synthesis by reverse hydrolysis. In 70% (w/v) mannose solutions, 1,2-alpha-mannosidase from Penicillium citrinum lost 40% and alpha-mannosidase from almonds lost 60% activity at 55 degreesC over 2 weeks. Oligosaccharide yields were 15 and 45% respectively. Higher molecular weight glycation adducts were formed in a time-dependent manner as seen by MALDI-TOF. Inhibitors of the Maillard. reaction were able to partially alleviate these effects resulting in reduced loss of enzyme activity and oligosaccharide yield increases of 27-53% relative to the control. The aim of this study was to estimate the indoor and outdoor concentrations of fungal spores in the Metropolitan Area of Sao Paulo (MASP), collected at different sites in winter/spring and summer seasons. The techniques adopted included cultivation (samples collected with impactors) and microscopic enumeration (samples collected with impingers). The overall results showed total concentrations of fungal spores as high as 36,000 per cubic meter, with a large proportion of non culturable spores (around 91% of the total). Penicillium sp. and Aspergillus sp. were the dominant species both indoors and outdoors, in all seasons tested, occurring in more than 30% of homes at very high concentrations of culturable airborne fungi [colony forming units(CFU) m(-3)]. There was no significant difference between indoor and outdoor concentrations. The total fungal spore concentration found in winter was 19% higher than that in summer. Heat and humidity were the main factors affecting fungal growth; however, a non-linear response to these factors was found. Thus, temperatures below 16A degrees C and above 25A degrees C caused a reduction in the concentration (CFU m(-3)) of airborne fungi, which fits with MASP climatalogy. The same pattern was observed for humidity, although not as clearly as with temperature given the usual high relative humidity (above 70%) in the study area. These results are relevant for public health interventions that aim to reduce respiratory morbidity among susceptible populations.

Filamentous fungi and yeasts associated with the marine algae Adenocystis utricularis, Desmarestia anceps, and Palmaria decipiens from Antarctica were studied. A total of 75 fungal isolates, represented by 27 filamentous fungi and 48 yeasts, were isolated from the three algal species and identified by morphological, physiological, and sequence analyses of the internal transcribed spacer region and D1/D2 variable domains of the large-subunit rRNA gene. The filamentous fungi and yeasts obtained were identified as belonging to the genera Geomyces, Antarctomyces, Oidiodendron, Penicillium, Phaeosphaeria, Aureobasidium, Cryptococcus, Leucosporidium, Metschnikowia, and Rhodotorula. The prevalent species were the filamentous fungus Geomyces pannorum and the yeast Metschnikowia australis. Two fungal species isolated in our study, Antarctomyces psychrotrophicus and M. australis, are endemic to Antarctica. This work is the first study of fungi associated with Antarctic marine macroalgae, and contributes to the taxonomy and ecology of the marine fungi living in polar environments. These fungal species may have an important role in the ecosystem and in organic matter recycling.

The aim of the present study was to analyze the mycobiota, occurrence of mycotoxins (aflatoxins and cyclopiazonic acid), and production of phytoalexin (trans-resveratrol) in two peanut varieties (Runner IAC 886 and Caiapo) during plant growth in the field. Climatic factors (rainfall, relative humidity and temperature) and water activity were also evaluated. The results showed a predominance of Fusarium spp. in kernels and pods, followed by Penicillium spp. and Aspergillus flavus. Aflatoxins were detected in 20% and 10% of samples of the IAC 886 and Caiapo varieties, respectively. Analysis showed that 65% of kernel samples of the IAC 886 variety and 25% of the Caiapo variety were contaminated with cyclopiazonic acid. trans-Resveratrol was detected in 6.7% of kernel samples of the IAC 886 variety and in 20% of the Caiapo variety. However, trans-resveratrol was found in 73.3% of leaf samples in the two varieties studied. (C) 2011 Published by Elsevier Ltd.

The aim of this study was to investigate the effects of nutrients (nitrogen, zinc and boron) on fungal growth and fumonisins production in corn samples obtained at the beginning of grain formation and at harvest. Three nitrogen doses were applied to the corn plants through soil in combination with three zinc doses and two boron doses during sowing. Mycological analysis of grains, using Dichloran Rose-Bengal Chloramphenicol Agar, collected at the beginning of formation demonstrated a fungal population predominantly of yeasts. Analysis of freshly harvested corn revealed a higher frequency of Penicillium spp. (72%) and F verticillioides (27%). High Performance Liquid Chromatography analysis revealed that 100% of grains were contaminated with fumonisins B, at levels ranging from 0.3 to 24.3 mg/kg and 93% contaminated with fumonisin B(2) at levels ranging from 0.05 to 5.42 mg/kg. Nitrogen (50 kg/ha) in combination with boron (0.5 kg/ha) resulted in an increased fumonisin B2 production. (c) 2007 Elsevier Ltd. All rights reserved.

Although the production of patulin in apple fruits is mainly by Penicillium expansum, there is no information on the ability of heat resistant moulds that may survive pasteurization to produce this mycotoxin in juice packages during storage and distribution. In this study, the production of patulin by Byssochlamys spp (Byssochlamys nivea FRR 4421, B. nivea ATCC 24008 and Byssochlamys fulva IOC 4518) in cloudy and clarified apple juices packaged in laminated paperboard packages or in polyethylene terephthalate bottles (PET) and stored at both 21 degrees C and 30 degrees C, was investigated. The three Byssochlamys strains were able to produce patulin in both cloudy and clarified apple juices. Overall, the lower the storage temperature, the lower the patulin levels and mycelium dry weight in the apple juices (p<0.05). The greatest variations in pH and degrees Brix were observed in the juices from which the greatest mycelium dry weights were recovered. The maximum levels of patulin recovered from the juices were ca. 150 mu g/kg at 21 degrees C and 220 mu g/kg at 30 degrees C. HPLC-UV, HPCL-DAD and mass spectrometry analyses confirmed the ability of B. fulva IOC 4518 to produce patulin. Due to the heat resistance of B. nivea and B. fulva and their ability to produce patulin either in PET bottles or in laminated paperboard packages, the control of contamination and the incidence of these fungi should be a matter of concern for food safety. Control measures taken by juice industries must also focus on controlling the ascospores of heat resistant moulds. (C) 2010 Elsevier B.V. All rights reserved.

The fungi Aspergillus sydowii Ce15, Aspergillus sydowii Ce19, Aspergillus sydowii Gc12, Bionectria sp. Ce5, Penicillium miczynskii Gc5, Penicillium raistrickii Ce16 and Trichoderma sp. Gc1, isolated from marine sponges Geodia corticostylifera and Chelonaplysylla erecta, were evaluated for their ability to grow in the presence of DDD pesticide. Increasing concentrations of DDD pesticide, i.e., 5.0 mg (1.56 x 10(-12) mmol), 10.0 mg (3.12 x 10(-2) mmol) and 15.0 mg (4.68 x 10(-2) mmol) in solid and liquid culture media were tested. The fungi Trichoderma sp. Gc1 and Penicillium miczynskii Gc5 were able to grow in the presence of up to 15.0 mg of DDD, suggesting their potential for biodegradation. A 100% degradation of DDD was attained in liquid culture medium when Trichoderma sp. Gc1 was previously cultivated for 5 days and supplemented with 5.0 mg of DDD in the presence of hydrogen peroxide. However, the quantitative analysis showed that DDD was accumulated on mycelium and biodegradation level reached a maximum value of 58% after 14 days.

O conhecimento dos fungos anemófilos em determinada cidade ou região é importante para o diagnóstico etiológico e o tratamento específico das manifestações alérgicas respiratórias. Várias técnicas quantitativas e qualitativas são preconizadas para coleta e identificação desses fungos na dependência do local estudado. Nesta pesquisa, sobre esporos de fungos do ar, foi utilizado o equipamento Rotorod Sampler®, que retira a amostra do ar através de um bastão de plástico preso a um motor elétrico que o faz girar rapidamente, sendo as partículas suspensas no ar recolhidas pelo bastão. As amostras foram coletadas uma vez por semana, durante 24 horas, correspondendo a um ciclo de coleta. Foram realizadas 52 coletas entre abril de 2000 e março de 2001. Os resultados mostraram prevalência de ascosporos (50,49%), Cladosporium (17,86%), Aspergillus/Penicillium (15,03%), basidiosporos (3,84%), rusts (3,82%), Helminthosporium (2,49%), Botrytis (1,22%), Alternaria (1,19%), smuts (0,90%), Curvularia (0,87%), Nigrospora (0,61%) e Fusarium (0,08%). Não foi possível identificar 1,59% dos esporos de fungos coletados no período. O maior número de esporos foi observado nos meses de verão e o menor, durante o outono. Utilizando provas in vivo e in vitro, avaliou-se a hipersensibilidade a fungos entre 39 pacientes atópicos sofrendo de rinite e ou asma brônquica. Os testes cutâneos identificaram sensibilização em 17,94% dos pacientes, enquanto as provas sorológicas caracterizaram presença de IgE específica em 12,82% dos casos avaliados. A detecção de significativo número de esporos de fungos no ar de Porto Alegre, com muitas espécies comprovadamente alergênicas, e os índices de sensibilização observados em indivíduos atópicos confirmam a importância do estudo dos fungos anemófilos nessa cidade, com vistas a aprimorar o diagnóstico e o manejo de pacientes com manifestações alérgicas respiratórias.

O papel do milho na alimentação humana e animal é importante por causa da composição química e do valor nutritivo; por isso, é um dos cereais mais cultivados e consumidos no mundo. Uma das principais causas de danos à qualidade dos grãos é a infecção fúngica e a conseqüente contaminação por micotoxinas. Neste trabalho, foi identificado os efeitos do retardamento da colheita, do grau de umidade e do período de armazenamento dos grãos na ocorrência de fungos patogênicos e de grãos ardidos e a possível contaminação por micotoxinas na lavoura e durante o armazenamento, com o objetivo de caracterizar a influência dos fungos de pré e pós colheita na qualidade dos grãos de milho nas safras 98/99 e 99/00. A incidência de fungos patogênicos foi quantificada em 400 grãos e de grãos ardidos (GA), a percentagem de grãos com injúria mecânica visível (IMV) e de grãos normais (GN) em quatro repetições de 250 g de grãos. A contaminação por micotoxinas foi analisada por cromatografia de camada delgada e as características nutricionais foram analisadas por espectrometria de reflectância no infravermelho proximal. O retardamento da colheita gerou condições favoráveis para a elevação da incidência dos fungos das espécies Aspergillus spp., Cephalosporium spp., Fusarium graminearum e Penicillium spp. A incidência de GA se elevou enquanto os grãos apresentavam umidade acima de 20%. A incidência de Fusarium moniliforme foi reduzindo à medida que os grãos perdiam água. O percentual de grãos com IMV somados a incidência GA foi de 15,90% no silo de Lodi e de 15,78% na Cotrel. O retardamento de colheita influenciou na ocorrência de fungos, de GA e de micotoxinas na lavoura. As práticas de manejo das lavouras e a operação de colheita afetaram qualidade comercial dos grãos de milho e o armazenamento conservou as características nutricionais.

Cerca de 15% da safra de arroz é perdida anualmente, principalmente em práticas inadequadas de pós-colheita. Nesse contexto, a avaliação de técnicas de secagem e armazenamento é necessária em função das perdas quali e quantitativas de grãos, resultantes do desenvolvimento de fungos. O objetivo desse trabalho foi analisar a contaminação fúngica e por micotoxinas do arroz submetido aos sistemas de secagem intermitente e estacionário, com armazenamento em sacaria e em silo-secador, avaliando o comportamento das principais variáveis de influência no processo. O trabalho foi realizado entre os meses de maio de 2003 a maio de 2004. As amostras de arroz com casca foram coletadas a cada dois meses, em duplicatas. Foram, ainda, analisadas amostras de arroz branco polido e parboilizado de ambos sistemas. O número de colônias de bolores e leveduras foi determinado e a capacidade produtora de micotoxinas dos isolados do gênero Aspergillus foi investigada. A detecção de aflatoxinas, ocratoxina A, zearalenona e citrinina foi realizada por cromatografia em camada delgada. Os grãos secados no sistema estacionário apresentaram maior contagem de colônias fúngicas. Enquanto no sistema intermitente, a contaminação foi mais uniforme ao longo do armazenamento. Predominaram representantes do gênero Penicillium nos dois sistemas, principalmente P. commune e P. islandicum. Entre os isolados do gênero Aspergillus, destacou-se A. flavus. Foram encontrados 7 (13, 20%) isolados de A.flavus produtores de aflatoxina B1, mas não foram detectadas micotoxinas nas amostras. A umidade dos grãos afetou significativamente a contaminação fúngica no manejo intermitente e no terço superior (altura 2) do silo-secador. A temperatura no interior do silo influenciou a contaminação no terço inferior (altura 1).

Presentemente as pessoas passam cerca de 90% da sua vida no interior de edifícios, onde a poluição atinge níveis superiores aos registados no exterior. Os infantários são espaços onde a qualidade do ar interior (QAI) deverá ser alvo de maior preocupação, visto ser o local onde as crianças passam uma fase importante do seu desenvolvimento físico-motor. São vários os poluentes que podem comprometer a saúde humana, de entre os quais se destacam os poluentes biológicos, os químicos, e os físico-químicos. O presente trabalho teve como objectivo analisar a QAI em onze infantários da Ilha da Madeira durante as quatro estações do ano. Para o estudo foi levado em consideração a dispersão geográfica e a envolvente rural e urbana. Complementarmente, foi efectuado um estudo aprofundado dos compostos orgânicos voláteis (COVs) em dois dos infantários, com o emprego de amostragem activa e passiva. Nos infantários em análise, verificou-se a existência de um sério problema de renovação do ar no interior das salas, reflectido nas elevadas concentrações de CO2 e bactérias encontradas durante a monitorização dos espaços. As concentrações de fungos raramente excederam o valor máximo de referência (VMR), sendo o Penicillium spp. e o Cladosporium sp., as espécies mais comuns encontradas. Contudo foram detectadas algumas espécies consideradas perigosas para a saúde humana, como o Aspergillus niger e o A. versicolor. Relativamente aos restantes parâmetros (CO, O3, HCHO, NO2, NO, SO2, H2S, PM10, T e HR), na sua generalidade apresentaram concentrações inferiores aos respectivos VMR: excepção para o HCHO que em 40% das análises revelou valores acima do seu VMR, bem como a T e HR, que apresentaram valores fora das respectivas escalas de referência. Não foram encontradas diferenças significativas na QAI entre os onze infantários no que diz respeito à sua localização geográfica e envolvente. No entanto, em alguns parâmetros foram observadas variações sazonais. O estudo dos perfis de COVs revelou que o tipo de amostragem a empregar deve ter em consideração o objectivo da recolha: se for necessária a obtenção de dados como um flash instantâneo, a utilização da amostragem activa é aconselhada, se forem requeridos dados médios de um determinado período de tempo, o emprego da amostragem passiva será a melhor escolha.

he present model of agriculture is based on intensive use of industrial inputs, due to its rapid response, but it brings harmful consequences to the environment, and it is necessary the use of modern inputs. And an alternative is the use of rock biofertilizers in agriculture, a product easy to use, with higher residual effect and does not harm the environment. The objective of study was to evaluate the inoculation and co-inoculation of different microorganisms in the solubilization of rock phosphate and potash ground microbial evaluating the best performance in the production of biofertilizers comparing with rocks pure in soil chemical properties and, verify effect of inoculation of the bacterium Paenibacillus polymyxa in the absorption of minerals dissolved in the development of cowpea (Vigna unguiculata [L.] Walp.). The first bioassay was conducted in Laboratory (UFRN) for 72 days in Petri dishes, where the rock powder was increased by 10% and sulfur co-inoculated and inoculated with bacterial suspension of Paenibacillus polymyxa grown in medium tryptone soy broth, Ralstonia solanacearum in medium Kelman, Cromobacterium violaceum in medium Luria-Bertani and Acidithiobacillus thiooxidans in medium Tuovinen and Kelly,and fungi Trichoderma humatum and Penicillium fellutanum in malt extract. Every 12 days, samples were removed in order to build up the release curve of minerals. The second bioassay was conducted in a greenhouse of the Agricultural Research Corporation of Rio Grande do Norte in experimental delineation in randomized block designs, was used 10 kg of an Yellow Argissolo Dystrophic per pot with the addition of treatments super phosphate simple (SS), potassium chloride (KCl), pure rock, biofertilizers in doses 40, 70, 100 and 200% of the recommendation for SS and KCl, and a control, or not inoculated with bacteria P. polymyxa. Were used seeds of cowpea BRS Potiguar and co-inoculated with the bacterial suspension of Bradyrhizobium japonicum and P. polymyxa. The first crop was harvested 45 days after planting, were evaluated in the dry matter (ADM), macronutrients (N, P, K, Ca, Mg) and micronutrients (Zn, Fe, Mn) in ADM. And the second at 75 days assessing levels of macro end micronutrients in plants and soil, and the maximum adsorption capacity of P in soil. The results showed synergism in co-inoculations with P. polymyxa+R. solanacearum and, P. polymyxa+C. violaceum solubilizations providing higher P and K, respectively, and better solubilization time at 36 days. The pH was lower in biofertilizers higher doses, but there was better with their addition to P at the highest dose. Significant reduction of maximum adsorption capacity of phosphorus with increasing dose of biofertilizer. For K and Ca was better with SS+KCl, and Mg to pure rock. There was an effect of fertilization on the absorption, with better results for P, K and ADM with SS+KCL, and N, Ca and Mg for biofertilizers. Generally, the P. polymyxa not influence the absorption of the elements in the plant. In treatments with the uninoculated P. polymyxa chemical fertilizer had an average significantly higher for weight and number of grains. And in the presence of the bacteria, biofertilizers and chemical fertilizers had positive values in relation to rock and control. The data show that the rocks and biofertilizers could meet the need of nutrients the plants revealed as potential for sustainable agriculture

Este trabalho teve por objetivo determinar a ocorrência e a freqüência de fungos em banana 'Prata anã' e elucidar o agente causal das podridões em pós-colheita de frutos provenientes do norte de Minas Gerais. Dois métodos de isolamento foram adotados: diluição em placas, a partir da lavagem de frutos verdes, e direto de frutos maduros. Os fungos Colletotrichum musae, Trichoderma harzianum, Fusarium equisetii, Penicillium sp. Aspergillus parasiticus, Trichothecium roseum, Colletotrichum acutatum, Alternaria sp., Cladosporium musae e Curvularia lunata foram os mais freqüentemente associados aos frutos. A patogenicidade desses fungos foi testada pela substituição de discos da casca de frutos verdes por discos de micélio. Colletotrichum musae apresentou área média lesionada em torno do ponto de inoculação igual a 5,8 cm², enquanto para os demais fungos testados não passou de 1,50 cm². Os resultados mostraram que C. musae é o agente primário das podridões dos frutos examinados com 100 % de incidência e os demais fungos limitaram-se a necrosar os ferimentos em torno do ponto de inoculação. O modo de infecção latente, causada por C. musae, parece favorecer, primeiramente, a colonização interna dos tecidos e, posteriormente, a ação dos fungos oportunistas, que aceleram as podridões nos frutos e na coroa.
