The petrochemical industry represents a sector of intense activity, in constant expansion and great economic importance for Brazil. In order to be used in various human activities petroleum needs to suffer a refinement process that, besides requiring large amounts of water, produces large amounts of wastes, which are discharged in hydric resources. Petroleum is a complex mixture mainly comprised by hydrocarbons, many of them are recognized as toxic chemicals, which are able to induce a considerable environmental pollution. Since water is an important resource for the maintenance of ecosystems and is also the final receiver of effluent discharges of the petroleum industry, there is an imminent need to monitor, constantly, the hydric resources that are influenced by this industrial activity, so that it can be ensured the environmental health. This study aimed, mainly, to analyze the quality of water samples derived from refinement process performed of the largest petroleum refinery in Brazil (REPLAN), in distinct steps of the industry treatment, and waters of rivers associated to its activity. Micronuclei and Nuclear Abnormalities tests were performed in erythrocytes of the fish species Oreochromis niloticus exposed to water samples derived from refinery use and to rivers under the influence of their effluents, in order to evaluate its possible toxic, genotoxic and mutagenic effects. The results obtained show that the treatment carried out by the refinery, during the evaluated periods, was effective, since the substances present in the water samples did not induce significant genotoxic and/or mutagenic alterations in the genetic material of the test organisms. Junto ao Laboratório de Fracionamento de Proteínas, desenvolveu atividades aplicando técnicas eletroforéticas (gel de agarose, eletroforese nativa em gel de poliacrilamida) em amostras de músculos de aves de interesse comercial (frangos, perus e avestruzes), tendo como co-orientador o Prof. Paulo Tadeu Figueira da Faculdade de Medicina Veterinária, PUC/PR - Toledo, onde desenvolveu atividades de análise de géis de eletroforese. Aplicou as técnicas de eletroforeses citadas em amostras de músculos e soro sanguíneo (em andamento) de tilápias (Oreochromis niloticus), bem como desenvolveu atividades de análises computacionais (VDS – PHARMACIA) e estatísticas (ASSISTAT) nos dados obtidos, resultando na participação em 7 congressos com apresentação de trabalhos. Cursou a disciplina de Genética, como aluno especial, junto à turma de Licenciatura em Ciências Biológicas, num total de 8 créditos, sendo aprovado com média 7,2

Contamination of rivers and lakes, waste disposed by infiltration into the soil as fertilizer, for example, has been highlighted around the world, since such compounds can contain toxic elements, causing changes in aquatic and/or terrestrial ecosystem. Due to the possible presence of these elements in these wastes, it is possible assess damage to organisms exposed through breaks in genetic material and mutations. In this sense, this work aimed at exploring the cytotoxic, mutagenic and genotoxic vinasse pontential, residue used as fertilizer, from the processing of sugar cane into alcohol, using fish (Oreochromis niloticus - Cichlidae) as a test organism. The individuals were exposed to different dilutions of vinasse in the water and later carried out analysis of their erythrocytes using the micronucleus test and nuclear abnormalities associated and the comet assay. In the first bioassay, tilapia exhibited 5% and 10% vinasse concentrations died after 48h. The number of micronucleated erythrocytes was statistically significant for the 1% vinasse concentration and also for nuclear abnormalities such as broken-egg and nuclear bud, when compared with negative and positive controls; were also significant lobed type erythrocytes, when compared to the negative control. The test results of the comet assay were not significant for 1% vinasse when compared to the negative control, due to the high rate of comets with damage control, which is unusual. The same was observed for changes as notched and blebbed type. In the second chemical analyzes conducted, the detection of heavy metals in the vinasse sample, charged only the presence of copper and chromium, the latter with a concentration 89 times higher than in the first test. Um dos caminhos para identificar os contaminantes e seus efeitos em ecossistemas aquáticos é examinar a resposta de alguns organismos considerados bioindicadores. O presente projeto teve por objetivo avaliar a bioconcentração de cádmio e cobre pelo peixe de água doce Oreochromis niloticus (tilapia), visando, principalmente, estabelecer seu potencial bioindicador de contaminação destes metais em ambientes fluviais.Dentre os vários metais pesados passíveis de contaminação ambiental, o cobre um elemento essencial e o cádmio um elemento não essencial, são altamente tóxicos, mesmo em níveis de concentrações muito baixos e podem se elevar quanto maior for o nível trófico do organismo, podendo chegar ao homem em teores magnificados. Em condições laboratoriais, foram determinadas as toxicidades aguda de Cu, de Cd, e de Cu+Cd em tilápias em períodos de 24 á 96 horas de exposição. A partir dos resultados obtidos nos testes de toxicidade aguda, foi estabelecida a dose letal que levou a óbito metade da população (CL50). Os valores encontrados para o presente estudo foram: CL50-96h para cobre – 3,53 mg/L;CL50-96h pra cádmio – 20,13 mg/L;CL50-96h para cobre combinado ao cádmio – 1,36 mg/L. Foram avaliadas as concentrações dos referidos metais nos fígados e nas brânquias. O presente estudo mostra que em linhas gerais o fígado acumulou mais metal do que as brânquias, e que o processo de absorção do cobre é mais rápido do que o do cádmio para a espécie de peixe testada. Os resultados obtidos mostraram que o cobre foi mais tóxico do que o que o cádmio, entretanto, a combinação de ambos metais apresentou maior toxicidade

Water is an essential factor in maintaining the vital functions of living beings. We have observed a growing commitment of quality, are due to pollution from many sources and even entire watersheds, whether for industrial waste, sewage, or for substances used in farming such as pesticides, herbicides and fertilizers. Nickel is the 24th most abundant element on earth, is a heavy metal that, in the form of chloride, is a proven genotoxic and mutagenic. Due to its industrial use, there was considerable increase of its concentration in surface sediments. Fish combine characteristics that make them excellent experimental models for aquatic toxicology studies, which are particularly usable as warn about the potential danger of chemicals or the possibility of environmental pollution. Due to impaired water quality and the few published studies relating the nickel with the tissue change, this study aimed at assessing the consequences of the presence of nickel in the aquatic environment. For this analysis, we used individuals of Oreochromis niloticus, exposed for 96 hours at three different concentrations of nickel dissolved in water compared to a control group. After exposure, the gills were removed and these were analyzed by ultramorphological, histological and histochemical analysis. The results indicate that all concentrations used in the experiment altered the histophysiology of exposed individuals. We observed the following changes: rupture of paviment cells, thus resulting in bleeding, loss of microridges surface of these cells and epithelial loss in the gills of all animals in all treatments with nickel chloride, the histochemical analysis showed non-proliferation of chloride cells. However, there was a dose-dependent increase of mucus cells in all animals.
