This issue of the FAL Bulletin shows why urban logistics is a relevant topic in the discussion of urbanization and provides arguments why the last-mile should be included in policies on sustainable mobility in the region. It also delivers insights and arguments how a modern perception of the cargo bike and its inclusion in sustainable logistics policy can deliver towards more sustainable urban logistics development.

En el presente Boletín Fal se expone la relevancia de la logística urbana en el marco del debate sobre la urbanización y se argumenta la necesidad de tener en cuenta el último eslabón de la cadena de abastecimiento en las políticas de movilidad sostenible de la región. Además, este número aporta información sobre el nuevo concepto de la bicicleta de carga y cómo podría contribuir a lograr un desarrollo más sostenible de la logística urbana de incluirse en las políticas de logística.

Photodynamic therapy (PDT) is a promising method for localized and specific inactivation of fungi and bacteria. A nontoxic light-sensitive compound is taken up by cells, which are then exposed selectively to light, which activates toxicity of the compound. We investigated the potential of sublethal PDT using light-sensitive curcumin (CUR) in combination with blue (455 nm) light to promote reactive oxygen species (ROS) formation in the form of singlet oxygen and DNA damage of Candida albicans. Surprisingly, CUR-mediated PDT but also light alone caused significantly longer comet tails, an indication of DNA damage of C. albicans when compared with the negative control. The intracellular ROS production was also significantly higher for the group treated only with light. However, PDT compared to blue light alone significantly slowed DNA repair. Comet tails decreased during 30 min visualized as a 90% reduction in length in the absence of light for cells treated with light alone, while comet tails of cells treated with PDT only diminished in size about 45%. These results indicate that complex mechanisms may result in PDT in a way that should be considered when choosing the photosensitive compound and other aspects of the treatment design.
Myoglobin from the burrowing reptile Amphisbaena alba: concentrations and functional characteristics

1. 1. Myoglobin from the subterranean reptile Amphisbaena alba was isolated for measurement of concentrations and physico-chemical properties. 2. 2. The concentrations (averaging 12.1 mg.g-1 wet weight in the temporal muscles and 5.8-6.0 in the muscles that motivate the wedge-shaped head which forms the burrowing tool) far exceed those earlier reported for reptiles and other terrestrial vertebrates. 3. 3. The myoglobin has a low O2 affinity compared to mammals (P50 = 2mmHg at 25°C). In the presence of the same myoglobin O2 tension as in mammals this appears to favour similar in vivo O2 saturations at the lower reptilian body temperature. 4. 4. The temperature sensitivity of P50 reflect a heat of oxygenation, ΔH near -13 kcal· mol-1. The myoglobin is monomeric and thus lacks cooperativity in O2 binding and there is no Bohr effect. 5. 5. The pattern of microheterogeneity is similar to that of myoglobin of terrestrial vertebrates but different to aquatic mammals and reptiles. The major and two minor components exhibit very similar O2 affinities. 6. 6. The concentrations and oxygen-binding characteristics of Amphisbaena myoglobin are discussed with regard to the flow of O2 to the mitochondria during digging activity in hypoxic burrow environments. © 1981.

Phase diagrams of poly(ethylene glycol)/polyacrylate/Na2SO4 systems have been investigated with respect to polymer size and pH. Plasmid DNA from Escherichia coil can depending on pH and polymer molecular weight be directed to a poly(ethylene glycol) or to a polyacrylate-rich phase in an aqueous two-phase system formed by these polymers. Bovine serum albumin (BSA) and E. coil homogenate proteins can be directed opposite to the plasmid partitioning in these systems. Two bioseparation processes have been developed where in the final step the pDNA is partitioned to a salt-rich phase giving a total process yield of 60-70%. In one of them the pDNA is partitioned between the polyacrylate and PEG-phases in order to remove proteins. In a more simplified process the plasmid is partitioned to a PEG-phase and back-extracted into a Na2SO4-rich phase. The novel polyacrylate/PEG system allows a strong change of the partitioning between the phases with relatively small changes in composition or pH. (C) 2012 Elsevier B.V. All rights reserved.

The partitioning of Clavulanic Acid (CA) in a novel inexpensive and stable aqueous two-phase system (ATPS) composed by poly(ethylene glycol) (PEG) and sodium polyacrylate (NaPA) has been studied. The aqueous two-phase systems are formed by mixing both polymers with a salt (NaCl or Na2SO4) and an aqueous solution of CA. The stability of CA on the presence of both polymers was investigated and it was observed that these polymers do not degrade the biomolecule. The effect of PEG-molecular size, polymer concentrations on the commercial CA partitioning has been studied, at 25 degrees C. The data showed that commercial CA was preferentially partitioned for the PEG-rich phase with a partition coefficient (K-CA) between 1 and 12 in the PEG/NaPA aqueous two phase systems supplemented with NaCl and Na2SO4. The partition to the PEG phase was increased in the systems with high polymer concentrations. Furthermore, Na2SO4 caused higher CA preference for the PEG-phase than NaCl. The systems having a composition with 10 wt.% of PEG4000, 20 wt.% of NaPA8000 and 6 wt.% of Na2SO4 were selected as the optimal ones in terms of recovery of CA from fermented broth of Streptomyces clavuligerus. The partitioning results (K-CA = 9.15 +/- 1.06) are competitive with commercial extraction methods of CA (K-CA = 11.91 +/- 2.08) which emphasizes that the system PEG/NaPA/Na2SO4 can be used as a new process to CA purification/concentration from fermented broth. Background: Genome-wide association studies (GWAS) require large sample sizes to obtain adequate statistical power, but it may be possible to increase the power by incorporating complementary data. In this study we investigated the feasibility of automatically retrieving information from the medical literature and leveraging this information in GWAS. Methods: We developed a method that searches through PubMed abstracts for pre-assigned keywords and key concepts, and uses this information to assign prior probabilities of association for each single nucleotide polymorphism (SNP) with the phenotype of interest - the Adjusting Association Priors with Text (AdAPT) method. Association results from a GWAS can subsequently be ranked in the context of these priors using the Bayes False Discovery Probability (BFDP) framework. We initially tested AdAPT by comparing rankings of known susceptibility alleles in a previous lung cancer GWAS, and subsequently applied it in a two-phase GWAS of oral cancer. Results: Known lung cancer susceptibility SNPs were consistently ranked higher by AdAPT BFDPs than by p-values. In the oral cancer GWAS, we sought to replicate the top five SNPs as ranked by AdAPT BFDPs, of which rs991316, located in the ADH gene region of 4q23, displayed a statistically significant association with oral cancer risk in the replication phase (per-rare-allele log additive p-value [p(trend)] = 2.5 x 10(-3)). The combined OR for having one additional rare allele was 0.83 (95% CI: 0.76-0.90), and this association was independent of previously identified susceptibility SNPs that are associated with overall UADT cancer in this gene region. We also investigated if rs991316 was associated with other cancers of the upper aerodigestive tract (UADT), but no additional association signal was found. Conclusion: This study highlights the potential utility of systematically incorporating prior knowledge from the medical literature in genome-wide analyses using the AdAPT methodology. Abstract Background The etiology of Bell's palsy can vary but anterograde axonal degeneration may delay spontaneous functional recovery leading the necessity of therapeutic interventions. Corticotherapy and/or complementary rehabilitation interventions have been employed. Thus the natural history of the disease reports to a neurotrophic resistance of adult facial motoneurons leading a favorable evolution however the related molecular mechanisms that might be therapeutically addressed in the resistant cases are not known. Fibroblast growth factor-2 (FGF-2) pathway signaling is a potential candidate for therapeutic development because its role on wound repair and autocrine/paracrine trophic mechanisms in the lesioned nervous system. Methods Adult rats received unilateral facial nerve crush, transection with amputation of nerve branches, or sham operation. Other group of unlesioned rats received a daily functional electrical stimulation in the levator labii superioris muscle (1 mA, 30 Hz, square wave) or systemic corticosterone (10 mgkg-1). Animals were sacrificed seven days later. Results Crush and transection lesions promoted no changes in the number of neurons but increased the neurofilament in the neuronal neuropil of axotomized facial nuclei. Axotomy also elevated the number of GFAP astrocytes (143% after crush; 277% after transection) and nuclear FGF-2 (57% after transection) in astrocytes (confirmed by two-color immunoperoxidase) in the ipsilateral facial nucleus. Image analysis reveled that a seven days functional electrical stimulation or corticosterone led to elevations of FGF-2 in the cytoplasm of neurons and in the nucleus of reactive astrocytes, respectively, without astrocytic reaction. Conclusion FGF-2 may exert paracrine/autocrine trophic actions in the facial nucleus and may be relevant as a therapeutic target to Bell's palsy.

Syftet med denna uppsats var att utforska om och hur den nationella handlingsplanen mot kvinnlig könsstympning efterlevs inom socialtjänsten i Nyköpings kommun. Kommunen som studien avgränsats till valdes för att den hade en ökad invandring under 1990-talet från länder där ingreppet är omfattande. Studien har avgränsats till att undersöka en familjecentral, vuxen- och ungdomsenheten och barn och familj. Studien omfattar en fallstudie för att få ett kvalitativt resultat. Det som framkommit i studien är paradoxen att Nyköpings kommun genomför ett förebyggande arbete mot kvinnlig könsstympning men kommunen har föga kunskap om handlingsplanen. På grund av att en implementering inte har skett är de anställda omedvetna om arbetets relevans till den nationella handlingsplanen. Behovet av ett förebyggande arbete beträffande kvinnlig könsstympning har påträffats då individer i bostadsområdet Brandkärr bett om hjälp. Arbetet som utförs i kommunen mot kvinnlig könsstympning har sitt ankare i en person, därmed gör sig eldsjälsproblematiken gällande.

Missbruksproblematiken bland kvinnor har ökat och anses vara ett samhällsproblem. Inom såväl den ämnesspecifika forskningen som i praktisk verksamhet har missbrukande kvinnor ofta betraktats som utsatta och traumatiserade. Studiens syfte var att belysa socialsekreterares syn på kvinnor och kvinnligheter i samband med missbruksproblematik. Detta studerades utifrån frågeställningar om deras arbetssätt, attityder, värderingar och syn på kvinnligheter. För att få förståelse av fenomenets komplexitet användes kvalitativ metod med halvstrukturerade intervjuer samt de teoretiska perspektiven; socialkonstruktivism och symbolisk interaktionism. Analysen skedde med hjälp av meningskoncentrering och utifrån tidigare forskning. Resultatet visade att socialsekreterarnas syn huvudsakligen var samstämmig, kvinnorna beskrevs generellt som utsatta och traumatiserade. Vidare framträdde ett behov av ytterligare reflektion gällande könsperspektivet. Avslutningsvis diskuterades behovet av forskning kring män som kön.

Abstrakt Bakgrund. Anknytning är ett viktigt band som skapas mellan mor och barn, detta är viktigt för att barnet ska utvecklas till en trygg person. För att anknytning ska uppstå krävs mycket närhet och värme. Vid brist på anknytning blir barnet passivt och apatiskt. Syfte. Syftet med denna studie var att belysa hur anknytningsprocessen mellan mor och barn kan påverkas vid olika förlossningssätt. Metod. En litteratursökning av artiklar publicerade mellan 1989-2007 gjordes, sökorden var attachment, cesarean section, delivery, mother-child relations och nurs*. 28 artiklar granskades noga och av dessa stämde 14 helt överens med inklusionskriterierna och vårt syfte. Dessa bearbetades och analyserades i litteraturstudien. Resultat. Analysen resulterade i tre kategorier. Den första var faktorer som kan påverka anknytningsprocessen, den andra var en komplicerad förlossnings inverkan på anknytningsprocessen och den tredje var omvårdnadshandlingar som kan underlätta anknytningsprocessen och sjukhusvistelsen för modern. Diskussion. Komplicerade förlossningar bidrog ofta till att kvinnan inte fick träffa sitt barn direkt efter förlossningen, något som var psykiskt påfrestande för kvinnan och negativt för anknytningsprocessen. Kvinnor som genomgick komplicerade förlossningar var i extra stort behov av stöd och information från personalen för att känna närhet till sitt barn. Konklusion. När en kvinna får känna sitt barns närhet direkt efter förlossningen så startar anknytningsprocessen fortare än om det första mötet blir uppskjutet. Om omvårdnadspersonalen använder sig av den kunskap som finns angående interaktionen mellan mor och barn så kan det underlätta för kvinnan i samspelet med sitt barn.

Bakgrund: Intensivvård består definitionsmässigt av monitorering, diagnos och vård av patienter med tänkbar reversibel svikt i ett eller flera organsystem. Vårdpersonal bör arbeta med en holistisk syn på patienten, ta vara på de känslomässiga, fysiska, andliga och sociala behoven. För att alla delar skall kunna tillfredställas bör vårdpersonalen även uppmärksamma de anhöriga och göra dessa delaktiga i vården. En nödvändighet för att klara detta är att sjuksköterskan är insatt i vad patientens familj går igenom och vilka behov de har. Syfte: Syftet med litteraturstudien var att belysa de upplevelser familjemedlemmar hade när deras anhörige vårdas på en intensivvårdsavdelning. Metod: Litteraturstudien byggde på femton vetenskapliga artiklar varav sju var kvalitativa och åtta var kvantitativa studier. Dessa artiklar fann författarna via de internationella databaserna PubMed och Cinahl. Artiklarna granskades sedan med en granskningsmall och de studier som matchade litteraturstudiens syfte gick till vidare granskning för klassificering, värdering och kategoriindelning. Resultat: Närhet ansågs som ett av de viktigaste behoven bland anhöriga till patienter på en intensivvårdsavdelning. Majoriteten av de anhöriga uppvisade en höggradig stress som var jämförbar med posttraumatiskt stresssyndrom och de uppvisade symtom såsom hjärtklappning, sömnsvårigheter och ångestkänslor. Behovet av god information betydde oerhört mycket för alla anhöriga, den information som de anhöriga uppskattade mest var sjuksköterskans förklaring av diagnos, prognos och olika behandlingar som patienten genomgick. Diskussion: Sjukvårdspersonalen kan gärna uppmuntra anhöriga till att hjälpa till med omvårdnaden om den sjuke men det är viktigt att förklara att detta ej är ett tvång och att inget är mer rätt än det andra. Kommunikation är nyckeln till att minska stress och osäkerhet hos de anhöriga.

Conservatism is a central theme of organismic evolution. Related species share characteristics due to their common ancestry. Some concern have been raised among evolutionary biologists, whether such conservatism is an expression of natural selection or of a constrained ability to adapt. This thesis explores adaptations and constraints within the plant reproductive phase, particularly in relation to the evolution of fleshy fruit types (berries, drupes, etc.) and the seasonal timing of flowering and fruiting. The different studies were arranged along a hierarchy of scale, with general data sets sampled among seed plants at the global scale, through more specific analyses of character evolution within the genus Rhamnus s.l. L. (Rhamnaceae), to descriptive and experimental field studies in a local population of Frangula alnus (Rhamnaceae). Apart from the field study, this thesis is mainly based on comparative methods explicitly incorporating phylogenetic relationships. The comparative study of Rhamnus s.l. species included the reconstruction of phylogenetic hypotheses based on DNA sequences. Among geographically overlapping sister clades, biotic pollination was not correlated with higher species richness when compared to wind pollinated plants. Among woody plants, clades characterized by fleshy fruit types were more species rich than their dry-fruited sister clades, suggesting that the fleshy fruit is a key innovation in woody habitats. Moreover, evolution of fleshy fruits was correlated with a change to more closed (darker) habitats. An independent contrast study within Rhamnus s.l. documented allometric relations between plant and fruit size. As a phylogenetic constraint, allometric effects must be considered weak or non-existent, though, as they did not prevail among different subclades within Rhamnus s.l. Fruit size was correlated with seed size and seed number in F. alnus. This thesis suggests that frugivore selection on fleshy fruit may be important by constraining the upper limits of fruit size, when a plant lineage is colonizing (darker) habitats where larger seed size is adaptive. Phenological correlations with fruit set, dispersal, and seed size in F. alnus, suggested that the evolution of reproductive phenology is constrained by trade-offs and partial interdependences between flowering, fruiting, dispersal, and recruitment phases. Phylogenetic constraints on the evolution of phenology were indicated by a lack of correlation between flowering time and seasonal length within Rhamnus cathartica and F. alnus, respectively. On the other hand, flowering time was correlated with seasonal length among Rhamnus s.l. species. Phenological differences between biotically and wind pollinated angiosperms also suggested adaptive change in reproductive phenology.

In this thesis, mechanistic and synthetic studies on transformations of H-phosphonates into DNA analogues containing P-S or P-C bonds are described. Configurational stability of dinucleoside H-phosphonates and the stereochemical course of their sulfurisation in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) were investigated. In light of these studies, the reported stereoselective sulfurisation of dinucleoside H-phosphonates and benzoylphosphonates in the presence of DBU was proved to be incorrect. Efficient protocols for the synthesis of new nucleotide analogues with non-ionic C-phosphonate internucleotide linkages were developed. The synthesis of dinucleoside 2-pyridylphosphonates was successfully performed by a DBU-promoted reaction of H-phosphonate diesters with N-methoxypyridinium salts. The thio analogues, 2-pyridyl- and 4-pyridyl phosphonothioate diesters, could be obtained by modifying the reactions developed for their oxo counterparts. Dinucleoside 3-pyridylphosphonates were prepared via a palladium(0)-catalysed cross coupling strategy that could be extended also to the synthesis of nucleotide analogues with metal-complexing properties, i.e. terpyridyl- and bipyridylphosphonate derivatives. Oligonucleotides modified with pyridylphosphonate internucleotide linkages have been prepared and preliminary studies on their hybridisation properties and resistance towards enzymatic degradation were performed. Finally, nucleotidic units for the incorporation of pyridylphosphonate groups at the 5’-terminus of oligonucleotides were designed. Condensations of such units with a suitably protected nucleoside afforded after oxidation the expected dinucleoside (3’-5’)-phosphates with pyridylphosphonate monoester functions at the 5’-ends.

A Swiss-specific FRAX model was developed. Patient profiles at increased probability of fracture beyond currently accepted reimbursement thresholds for bone mineral density (BMD) measurement by dual X-ray absorptiometry (DXA), and osteoporosis treatment were identified.
