In the present study, the GPD2 gene from Saccharomyces cerevisiae, which codifies for the enzyme glycerol-3-phosphate dehydrogenase (GPDH), was cloned from the pPICZ-alpha expression vector and used with the purpose of inducing the extracellular expression of the glycerol-3-phosphate dehydrogenase under the control of the methanol-regulated AOX promoter. The presence of the GPD2 insert was confirmed by PCR analysis. Pichia pastoris X-33 (Mut(+)) was transformed with linearized plasmids by electroporation and transformants were selected on YPDS plates containing 100 mu g/mL of zeocin. Several clones were selected and the functionality of this enzyme obtained in a culture medium was assayed. Among the mutants tested, one exhibited 3.1 x 10(-2) U/mg of maximal activity. Maximal enzyme activity was achieved at 6 days of growth. Medium composition and pre-induction osmotic stress influenced protein production. Pre-induction osmotic stress (culturing cells in medium with either 0.35 M sodium chloride or 1.0 M sorbitol for 4h prior to induction) led to an increase in cell growth with sorbitol and resulted in a significant increase in GPDH productivity with sodium chloride in 24h of induction approximately fivefold greater than under standard conditions (without pre-induction). OBJETIVO: Descrever resultados da aplicação de um sistema de monitoramento de fatores de risco para doenças crônicas não transmissíveis (DCNT) por meio de entrevistas telefônicas (SIMTEL) no município de Botucatu/SP. MÉTODOS: Entrevistou-se amostra probabilística (n = 1.410) da população de indivíduos com 18 ou mais anos de idade residente em domicílios do município de Botucatu/SP, conectados à rede de telefonia fixa. A amostragem foi realizada em três etapas: sorteio de linhas do cadastro da companhia telefônica; seleção de linhas residenciais ativas; sorteio para entrevista de um morador com 18 ou mais anos de idade por linha elegível. A taxa de sucesso (entrevistas realizadas: linhas elegíveis sorteadas) foi de 86,9%, sendo de 5,8% a proporção de recusas. Foi aplicado um questionário com 74 questões sobre consumo alimentar, atividade física, tabagismo, consumo de bebidas alcoólicas, peso e estatura recordados e auto-referência a diagnósticos médicos de hipertensão arterial e diabetes. Apresentam-se estimativas brutas da prevalência de fatores de risco/proteção para DCNT e estimativas ajustadas que levam em conta a distribuição segundo idade, sexo e escolaridade da população adulta total do município no Censo Demográfico de 2000. RESULTADOS: Foram observadas altas prevalências de excesso de peso (46.7%) e sedentarismo (57.9%). Houve desvantagem para os homens quanto ao consumo excessivo de bebidas alcoólicas e vantagem no que se refere à prática de atividade física em 1 ou mais dias da semana. Nas mulheres, observou-se associação inversa entre escolaridade e os seguintes fatores de risco: obesidade, excesso de peso, sedentarismo, consumo de carnes com gordura e hábito de fumar. Resultado semelhante foi observado para homens, exceto com relação a obesidade e excesso de peso. CONCLUSÕES: A segunda experiência de aplicação do SIMTEL confirmou o desempenho satisfatório e a utilidade do sistema em nosso meio.

The behavior of microorganisms towards the antibiotic action of propolis has been widely investigated. Since reports dealing with seasonal effect on propolis activity are not available, this assay was carried out aiming to observe the in vitro antimicrobial activity of propolis, collected during the four seasons, on bacterial strains isolated from human infections. Dilution of ethanolic extract of propolis (EEP) in agar was the method performed, with serial concentrations ranging from 0.4 to 14.0% (% v/v). The behavior of some bacteria was analysed according to the incubation period in medium plus propolis, and the survival curve was plotted. It was verified that the growth of Gram-positive bacteria is inhibited by low propolis concentrations (0.4%) whereas Gram-negative bacteria were less susceptible to this substance, the minimal inhibitory concentration ranging from 4.5 to 8.0%. There was no significant difference with regards to the seasonal effect on the survival curve of Staphylococcus aureus and Escherichia coli; after incubation with propolis, there was an efficient antimicrobial action, mainly towards Gram-positive bacteria. The establishment of plant species depends crucially on where the seeds are deposited. However, since most studies have been conducted in continuous forests, not much is known about the effects of forest fragmentation on the maintenance of abiotic and biotic characteristics in microhabitats and their effects on seed survival. in this study, we evaluated the effects of forest fragmentation on the predation upon the seeds of the palm Syagrus romanzoffiana in three microhabitats (interior forest, forest edge and gaps) in eight fragments of semi-deciduous Atlantic forest ranging in size from 9.5 ha to 33,845 ha in southeastern Brazil. Specifically, we examined the influence of the microhabitat structure, fauna and fragment size on the pattern of seed predation. Fragments < 100 ha showed similar abiotic and biotic characteristics to those of the forest edge, with no seed predation in these areas. Forest fragments 230-380 ha in size did not present safe sites for S. romanzoffiana seed survival and showed high seed predation intensity in all microhabitats evaluated. In fragments larger than 1000 ha, the seed predation was lower, with abiotic and biotic differences among gaps, interior forests and forest edges. In these fragments, the survival of S. romanzoffiana seeds was related to squirrel abundance and interior forest maintenance. Based on these results, we concluded that there are no safe sites for S. romanzoffiana seed establishment in medium- and small-sized fragments as result of the biotic and abiotic pressure, respectively We suggest that on these forest fragments, management plans are needed for the establishment of S. romanzoffiana, such as interior forest improvement and development in small-sized sites in order to minimize the edge effects, and on medium-sized fragments, we suggest post-dispersal seed protection in order to avoid seed predation by vertebrates. our findings also stress the importance of assessing the influence of forest fragmentation on angiosperm reproductive biology as part of the effective planning for the management of fragmented areas. Nas últimas duas décadas, o crescimento do interesse pela metodologia Seis Sigma intensificou a aplicação da abordagem estatística e de outras abordagens quantitativas com o intuito de melhorar não apenas a qualidade de produtos, serviços e processos, como também aumentar o desempenho organizacional e o processo de tomada de decisão. Este artigo trata da aplicação da abordagem estatística no contexto da gestão da qualidade em indústrias de alimentos de médio e grande porte do Estado de São Paulo com o propósito de: identificar quais ferramentas e técnicas estatísticas são mais amplamente empregadas por indústrias do setor para garantir e controlar a qualidade; avaliar a interdependência entre o sucesso da implementação de programas de qualidade e segurança alimentar como Boas Práticas de Fabricação (BPF) e sistema de Análise de Perigos e Pontos Críticos de Controle (APPCC) e o uso de estatística; e analisar estimativas do grau de relevância do pensamento estatístico e de seus benefícios como ferramenta de melhoria da qualidade. Um survey exploratório-descritivo foi realizado e os resultados revelaram que a abordagem estatística começa a ser mais valorizada nas indústrias de alimentos pela relevância de seus benefícios assim como já ocorre em outros setores. Há evidências de que a implantação bem sucedida dos programas de segurança alimentar seja uma condição primordial para o uso efetivo de estatística e de outras abordagens quantitativas.

Alkalophilic Bacillus licheniformis 77-2 produced an extracellular alkali-tolerant xylanase with negligible cellulase activity in medium containing corn straw. The effectiveness of crude xylanase on treatment of eucalyptus Kraft pulp was evaluated. A biobleaching experiment was carried out to compare the chlorine saving with pulp treated and untreated by the enzyme. Two-stage bleaching was employed, using a ClO2 chlorination and NaOH extraction (DE sequence). With the enzymatic treatment, in order to obtain the same value of Kappa number and brightness, respectively 28.5 and 30% less ClO2 was required in comparison to the enzymatically untreated samples.

Back-to-back correlations of particle-antiparticle pairs are related to the in-medium mass-modification and squeezing of the quanta involved. They are predicted to appear when hot and dense hadronic matter is formed in high energy nucleus-nucleus collisions. The survival and magnitude of the back-to-back correlations (BBC) of boson-antiboson pairs generated by in-medium mass modifications are studied here in the case of a thermalized, finite-sized, spherically symmetric expanding medium. We show that the BBC signal indeed survives the finite-time emission, as well as the expansion and flow effects, with sufficient intensity to be observed at BNL Relativistic Heavy Ion Collider (RHIC).

The back-to-back correlations (BBC) of particle-antiparticle pairs, signalling in-medium mass modification, are studied in a finite size thermalized medium. The width of BBC function is explicitly evaluated in the case of a nonrelativistic spherically symmetric expanding fireball. The effect of the flow is to reduce the BBC signal as compared to the case of non flow. Nevertheless, a significant signal survives finite-time emission plus expansion effects.

Back-to-back correlations of asymptotic fermion-anti-fermion pairs appear if in-medium interactions lead to mass modifications of fermion states in a thermalized medium. The back-to-back correlations of protons and anti-protons will be experimentally observable in ultrarelativistic heavy ion collisions. The strength of back-to-back correlations of fermions can be unlimitedly large, diverging as the momentum of the pair increases and the net baryon density decreases.

Squeezed correlations of hadron-antihadron pairs are predicted to appear if their masses are modified in the hot and dense medium formed in high-energy heavy ion collisions. If discovered experimentally, they would be an unequivocal evidence of in-medium mass shift found by means of hadronic probes. We discuss a method proposed to search for this novel type of correlation, illustrating it by means of D(s)-mesons with in-medium shifted masses. These particles are expected to be more easily detected and identified in future upgrades at RHIC.

A novel type of correlation involving particle-antiparticle pairs was found out in the 1990's. Currently known as squeezed or Back-to-Back Correlations (BBC), they should be present if the hadronic masses are modified in the hot and dense medium formed in high energy heavy ion collisions. Although well-established theoretically, such hadronic correlations have not yet been observed experimentally. In this phenomenological study we suggest a promising way to search for the BBC signal, by looking into the squeezed correlation function of phi phi and K(+)K(-) pairs at RHIC energies, as function of the pair average momentum, K(12) = (k(1) + k(2))/2. The effects of in-medium mass-shift on the identical particle correlations (Hanbury-Brown and Twiss effect) are also discussed.
