Hereditary or acquired cerebellar hypoplasia (CH) is commonly diagnosed in Holstein, Guernsey, Shorthorn and Jersey cattle. Bovine viral diarrhea (BVD) has been associated to acquired CH due to viral infection during the second trimester of pregnancy. Stricken calf usually shows ataxia, hypermetria, opisthotonus, intentional tremor and wide-based stance when in standing position. Three newborn calves were referred to the FCAV/Unesp Veterinary Teaching Hospital because of neurological distress. The clinical presentation, similar in all cases, indicated CH. Two weeks later, clinical signs did not improve and euthanasia was performed. Macroscopic examination revealed a gelatinous serosanguineous fluid over the brain surface and within the cervical spinal canal. Histologically the cerebellum had disorganization of the internal granular layer and moderate disappearance of Purkinje cells. The observed clinical signs and nervous tissue lesions were consistent with congenital cerebellar syndrome, possibly associated to viral infection during fetal development. Despite CH has been assumed to be related to BVD, blue tongue and Akabane viruses, only the BVD etiology has been already identified in Brazil.

A pilomyxoid astrocytoma is a recently described tumor that occurs predominantly in the hypothalamic-chiasmatic region and is rarely found elsewhere. It has similar features as pilocytic astrocytomas, but has distinct histological characteristics and a poorer prognosis. A pilomyxoid astrocytoma is an aggressive tumor, and increased awareness is necessary with a suspect case. We present the first case of a pilomyxoid astrocytoma of the brainstem described after the newest World Health Organization classification of central nervous system tumors. © F.O. Chronic hepatitis C virus (HCV) infection is an important cause of morbidity and mortality globally, and often leads to end-stage liver disease. The DNA damage checkpoint pathway induces cell cycle arrest for repairing DNA in response to DNA damage. HCV infection has been involved in this pathway. In this study, we assess the effects of HCV NS2 on DNA damage checkpoint pathway. We have observed that HCV NS2 induces ataxia-telangiectasia mutated checkpoint pathway by inducing Chk2, however, fails to activate the subsequent downstream pathway. Further study suggested that p53 is retained in the cytoplasm of HCV NS2 expressing cells, and p21 expression is not enhanced. We further observed that HCV NS2 expressing cells induce cyclin E expression and promote cell growth. Together these results suggested that HCV NS2 inhibits DNA damage response by altering the localization of p53, and may play a role in the pathogenesis of HCV infection. © 2013 Bitter et al.

To determine the behavioral and antinociceptive effects of narcotic and non-narcotic analgesics administered by intravenous injection in horses, 10 thoroughbred mares weighing between 450 and 550 kg and ranging in age from 8 to 13 years old were analyzed. The effects of alfentanil, butorphanol, flunixin, and saline solution on the general activity of the horses were investigated by measuring spontaneous locomotor activity (SLA) and head height (HH) in two behavior stalls. The antinociceptive effects of alfentanil (0.02 mg kg-1), butorphanol (0.1 mg kg-1), flunixin meglumine (0.5 mg kg-1), and saline were determined by measuring skin twitch reflex latency (STRL) after thermal cutaneous nociceptive stimulation. A paired Student t-test was used to compare SLA and HH between the groups of horses receiving different doses of the same drug at various time points. The Tukey test was used to compare the antinociceptive effect of the treatments. Differences were considered significant when P value was <.05. Horses treated with opioid analgesics demonstrated excitation, as shown by a significant increase in SLA at all doses tested and by neighing and demonstrating attentive attitudes with movement of the ears, stereotypical walking, and ataxia in most of the animals. HH was elevated only in animals treated with alfentanil. Antinociception was observed at 5 and 30 minutes after administration of alfentanil and butorphanol, respectively. Increased SLA was observed at 30 and 90 minutes after administration of alfentanil and butorphanol, respectively. We observed no effect on antinociception in horses given flunixin. In conclusion, this study suggests that alfentanil has a faster onset and a shorter duration than butorphanol; however, both drugs are able to stimulate the central nervous system. © 2013 Elsevier Inc. The antinociceptive and behavioral effects of methadone (MET) alone or combined with detomidine (DET) were studied in horses. Intravenous treatments were randomly administered in a two-phase crossover study. In phase 1, six horses were treated with saline (control) or 0.2 or 0.5 mg/kg methadone (MET0.2; MET0.5, respectively). In phase 2, six horses were treated with 0.01 mg/kg DET alone or with DET combined with 0.2 mg/kg MET (DET/MET0.2). Thermal nociceptive threshold (TNT) and electrical nociceptive thresholds (ENT) were recorded by using a heat projection lamp and electrodes placed in the coronary band of the thoracic limbs, respectively. Spontaneous locomotor activity (SLA) was studied by movement sensors in the stall (phase 1). Chin-to-floor distance was assessed in phase 2. In phase 1, the TNT increased significantly for 30 minute after MET0.5 but not after saline or MET0.2. Hyperesthesia and ataxia were observed in 2 of 6 and 6 of 6 horses after MET0.2 and MET0.5, respectively. SLA increased significantly for 120 minutes after MET in a dose-dependent way, but not after placebo. In phase 2, DET and DET/MET0.2 significantly increased the TNT and ENT above baseline for 15 and 30 minutes, respectively; thresholds were significantly higher with DET/MET0.2 than with DET at the same times. Chin-to-floor distance decreased significantly from baseline for 30 minutes, and no excitatory behavior was observed in both treatments. Although the higher dose of MET induced short-acting antinociception, the associated adverse effects may contraindicate its clinical use. The lower dose of MET potentiated DET-induced antinociception without adverse effects, which might be useful under clinical circumstances. © 2013 Elsevier Inc. All rights reserved.

O mercúrio é um perigoso metal e uma importante fonte de contaminação ambiental no Brasil e sobretudo na Amazônia. O principal órgão-alvo deste metal é o SNC onde causa danos que podem levar aos sintomas clássicos: ataxia, parestesia, disartria e alterações no desenvolvimento do sistema nervoso de crianças. A contaminação mercurial nos rios amazônicos aumenta a quantidade encontrada nos peixes, principalmente os que estão no topo da cadeia alimentar, expondo dessa forma a populações ribeiras à intoxicação mercurial, uma vez que o peixe é um elemento central na dieta dessas populações. Por isso, é fundamental o monitoramento periódico dos níveis de mercúrio nas espécies de peixes consumidas nessa região. Nosso trabalho se propôs a identificar os níveis de metilmercúrio e mercúrio inorgânico nas espécies de peixes mais consumidas pelas populações ribeirinhas da região do Tapajós e comparálos com os níveis encontrados em peixes da mesma espécie obtidos na região de Belém. Além disso, realizar uma comparação com os resultados obtidos por Dos Santos et al. (2000) e analisando os nível atuais e os antigos. Os peixes foram coletados no mercado municipal de Itaituba, no Tapajós, e no mercado do Ver-o-peso, em Belém. Amostras de músculo de cada peixe foram liofilizadas e analisadas pela Universidad de Castilla-La Mancha (Espanha) para quantificação do metilmercúrio e do mercúrio inorgânico. Os resultados obtidos no presente trabalho mostraram que somente os peixes piscívoros da região do Tapajós apresentam níveis de metilmercúrio acima do limite preconizado pela OMS (0,5 μg/g). Em todos todos os grupos do estudo, os níveis de mercúrio inorgânico estão bem abaixo deste limite. A espécie mais contaminada foi a Brachyplatystoma flavicans (dourada) chegando a ultrapassar até cinco vezes o limite de tolerância da OMS. Com nossos dados, pode-se dizer que os peixes da região do Tapajós continuam atualmente expostos a altas concentrações de mercúrio. As espécies não-piscívoras tiveram baixas concentrações de mercúrio orgânico, sendo aptas para consumo humano. O presente estudo apoia a importância do monitoramento continuado dos ambientes considerados expostos e não expostos na Amazônia. O conhecimento originado por este monitoramento fomentará definitivamente o desenvolvimento de estratégias de prevenção e de ações governamentais perante o problema da contaminação mercurial na Amazônia.

A Malária Cerebral (MC) apresenta-se como uma severa complicação resultante da infecção por Plasmodium falciparum. Esta condição encontra-se comumente associada a disfunções cognitivas, comportamentais e motoras, sendo a retinopatia uma das mais graves conseqüências da doença. Diversos modelos experimentais já foram descritos no intuito de elucidar os mecanismos fisiopatológicos relacionados a esta síndrome, no entanto, estes ainda permanecem pouco compreendidos. Dentro deste contexto, o presente trabalho procurou investigar as alterações neuroquímicas envolvidas na patologia da MC. Os camundongos C57Bl/6 (fêmeas e machos) inoculados com ≈106 eritrócitos parasitados (PbA) apresentaram baixa parasitemia (15-20%) com sinais clínicos evidentes como: deficiência respiratória, ataxia, hemiplegia e coma seguido de morte, condizentes com o quadro de MC. A análise no tecido retiniano demonstrou uma diminuição nos níveis de GSH com 2 dias após a inoculação. Entretanto, essa diminuição não foi tão evidente com o decorrer da infecção (4º e 6º dias após infecção). Concomitante a este aumento durante o processo infeccioso, observamos um progressivo aumento na captação de 3H-glutamato (4º e 6º dia após infecção) por um sistema independente de Na+, sugerindo que o quadro de MC é responsável por um aumento na atividade de uma proteína transportadora. Dados obtidos com a imunofluorescência demonstram que além de aumentar a atividade do sistema de transporte, o quadro de MC também estimula o aumento na expressão do sistema xCG - no tecido retiniano. O presente trabalho demonstra ainda que estes eventos neuroquímicos no tecido retiniano são independentes de ativação inflamatória, visto que os níveis de TNF-α e expressão de NOS-2, apresentam-se alterados somente no tecido retiniano.

Ipomoea carnea subsp. fistulosa é uma planta que contém swainsonina causando doença de depósito lisossomal em ruminantes, principalmente em caprinos na região Nordeste do Brasil. Para o estudo das plantas tóxicas da Ilha de Marajó, foram visitadas sete propriedades rurais na Ilha de Marajó, seis localizadas no município de Cachoeira do Arari e uma no município de Soure. Em todas as propriedades visitadas as pastagens eram constituídas de campo nativo, tinham pouca disponibilidade de forragem e I. carnea subsp. fistulosa encontrava-se em grande quantidade. Nas três propriedades onde eram criados caprinos foram observados animais com sinais nervosos, incluindo tremores de intenção, aumento da base de sustentação quando em estação, ataxia, hipermetria, nistagmo, paresia espástica ou debilidade, alterações posturais, perda de equilíbrio e quedas. Em duas fazendas a prevalência foi de 32% (23/71) e 100% (32/32) e em outra havia um animal com sinais acentuados e o resto do rebanho, de 19 caprinos, não foi examinado clinicamente. Bovinos, ovinos e bubalinos não foram afetados. Foram eutanasiados e necropsiados seis caprinos que apresentavam sinais clínicos acentuados. Macroscopicamente não foram observadas alterações. Na histologia observou-se vacuolização do pericário de neurônios e do citoplasma de células epiteliais da tireóide, rim, fígado, pâncreas e macrófagos de diversos órgãos. No sistema nervoso central a vacuolização era mais grave nos neurônios de Purkinje do cerebelo e nos neurônios dos núcleos cerebelares e do tronco encefálico. Observaram-se também degeneração walleriana dos axônios e gliose. A alta freqüência da intoxicação nas três fazendas que criavam caprinos sugere que a intoxicação por I. carnea subsp. fistulosa é muito importante para caprinos na Ilha de Marajó, onde há abundante quantidade da planta, que permanece verde durante todo o período seco.

Este trabalho é um estudo prospectivo e descritivo dos aspectos epidemiológicos e clínicos de 72 envenenamentos por escorpiões admitidos no Hospital Municipal de Santarém, Estado do Pará, Brasil, entre fevereiro de 2000 a fevereiro de 2001. Trouxeram o animal 8,3% das vítimas, os quais foram identificados como T. cambridgei. O sexo masculino foi acometido em 83,3%. A idade das vítimas e o tempo para o socorro médico foram respectivamente de 33,6±18,3 anos e 4,6±3,2 horas em média. Os membros superiores foram acometidos em 51,5% dos casos. As manifestações locais estiveram presentes em 91,7% e as sistêmicas em 98,6% dos envenenamentos. Entre os sintomas locais encontramos: parestesia em 79,2%, dor em 52,8%, e edema em 26,4% dos casos. Nas manifestações sistêmicas predominou as queixas neurológicas em 97,2% das vítimas, sendo o sintoma de sensação de "choque elétrico" pelo corpo (88,9%) o mais freqüente. No exame neurológico os sinais mais encontrados foram: mioclonias (93,0%), dismetria (86,1%), disartria (80,6%) e ataxia de marcha (70,8%). Classificou-se como moderados 76,4% dos envenenamentos, sem nenhum caso grave. Deixaram de realizar a soroterapia 32,7% dos casos moderados, por ausência de soro específico no momento do atendimento. O escorpionismo da região de Santarém mostra um comportamento clínico regional diferente daqueles descritos no Brasil e de outros locais da Amazônia e, apresenta uma clínica predominantemente neurológica, ainda não descrita na literatura brasileira.

Background: Fumonisins produced by Fusarium verticillioides are among the most important medical mycotoxins known. The intake of concentrate based on corn and corn by-products contaminated with fumonisins can cause severe poisoning in horses. The injuries are observed mainly in the white matter of the brain, and the disease is known as Equine Leukoencephalomalacia (ELEM). This study aims to describe and discuss the epidemiological, clinical and diagnostic aspects of an outbreak of ELEM occurred in three farms in the municipalities of Canarana and Agua Boa, in the eastern region of Mato Grosso, Brazil.Materials, Methods & Results: The outbreak occurred between May and August 2010. The disease affected six horses and four mules of different ages and sex. Clinical examination was only possible in animals with chronic evolution of the disease. All the affected animals showed neurological clinical signs such as ataxia and recumbency, which progressed to death or sudden death. Histopathological analysis showed foci of necrosis that predominantly affected the white matter, and the presence of gitter cells. Degenerative lesions were observed in the liver of the animals. Mortality rate ranged from 12.5 to 71%, and lethality reached 100%. The cases were preceded by sudden drops in the weather temperature. Fumonisins levels of 6.6 ppm were detected in the feed of the animals.Discussion: The presumptive diagnosis of leukoencephalomalacia was consistent and based on clinical and epidemiological studies. However, the definitive diagnosis was based upon the histological features of the brain including the presence of extensive areas of malacia. Moreover, the animals were being fed with corn or corn by-products contaminated with fumonisins levels considered to be toxic to equids. The mortality and lethality rates are in agreement with outbreaks described in previous studies. The animals showed neurological signs as the predominant clinical manifestation, with gait ataxia followed by recumbency, prostration and death between 24 h and 29 days. Similarly to other reports, the disease was more frequent in adult animals, which succumbed in 24-48 h. Conversely, the evolution of the disease in young animals was of 10 to 29 days. Sudden death was more prevalent in the mules. Previous studies have shown a predominance of cerebral and brainstem lesions in horses, whereas in mules the clinical signs are related to brainstem lesions. Corn and corn by-products are commonly used as energy supplementation to horses in the southern of Mato Grosso state, but outbreaks of the disease are uncommon. This may be influenced by the prevalence of hot climate conditions, which does not favor the production of toxin by the fungus. The atypical low weather temperatures (9-11 degrees C) observed prior to the outbreak could have contributed to the mycotoxin production by F. verticillioides, which requires temperatures between 8-12 degrees C to produce toxins. However, the disease in the region may be underestimated, considering that the practice of necropsies is not common among field technicians, mainly in the occurrence of sporadic deaths among horses intended for work. Preventive measures include avoiding the use of corn and corn by-products for horses after periods of sudden drops in temperature in the region. Furthermore, clinical and epidemiological surveys and post-mortem and histopathological analyses are undoubtedly important for appropriate differential diagnosis, especially in equids with neurological signs.

Background: Scorpion envenomations are a major public health problem in Brazil, whose most dangerous cases are attributable to the genus Tityus. This study was designed to compare the clinical and demographic features of envenomations by Tityus obscurus in two areas of the state of Para located in the Amazon basin. Were compared demographic findings, local and systemic signs and symptoms of human envenomations caused by T. obscurus that occurred in western and eastern areas of the state.Results: Forty-eight patients with confirmed envenomation by T. obscurus were evaluated from January 2008 to July 2011. Most of them came from the eastern region, where male and female patients were present in similar numbers, while males predominated in the west. Median age groups were also similar in both areas. Most scorpion stings took place during the day and occurred significantly more frequently on the upper limbs. The time between the sting and admission to the health center was less than three hours in both areas. Most eastern patients had local manifestations while in the west, systemic manifestations predominated. Local symptoms were similar in both areas, but systemic signs and symptoms were more common in the west. Symptoms frequently observed at the sting site were local and radiating pain, paresthesia, edema, erythema, sweating, piloerection and burning. The systemic manifestations were significantly higher in patients from the west. Futhermore, neurological symptoms such as general paresthesia, ataxia, dysarthria, myoclonus, dysmetria, and electric shock-like sensations throughout the body were reported only by patients from the west.Conclusion: The present study shows that two regions of Para state differ in the clinical manifestations and severity of confirmed envenomation by T. obscurus which suggests a toxicity variation resulting from the diversity of T. obscurus venom in different areas of the Brazilian Amazon basin, and that T. serrulatus antivenom can be successfully used against T. obscurus.
