     This study was undertaken to further understanding of the mechanisms which regulate mucus secretion by rat stomach cells. Particular objectives were: (i) to develop and use a radiochemical assay to estimate the secretion of mucin by a suspension of gastric mucosal cells in vitro, (ii) to develop and use a solid-phase enzyme immunoassay (EIA) to study the regulation of the release of bulk gastric mucin from the isolated cells and (iii) to compare the results obtained with the two procedures.      Cells were isolated by exposure of gastric mucosa to pronase and EDTA. Cell suspensions were preincubated with D-[6-3H]glucosamine. [3H]-labelled material of high molecular mass released into the incubation medium, was purified by Fast Protein Liquid Chromatography, and appeared to be gastric mucin. Some unidentified [3H]-labelled material of lower molecular mass was also found in the medium. Release of [3H]-labelled high molecular mass material was essentially linearly related to time. Secretin, isoprenaline and carbachol stimulated release of [3H]-labelled high molecular mass material. The half-maximally effective concentrations of secretin and isoprenaline were 2.3nM and 34nM respectively. Histamine, gastrin and epidermal growth factor were without effect.      A rabbit polyclonal antibody was raised by using purified 'native' rat gastric mucin as immunogen. The antibody preparation appeared specific for rat gastric mucin and was used to establish a quantitative solid-phase EIA. Release of bulk mucin was essentially linearly related to time. Phorbol-12-myristate-13-acetate (PMA), forskolin and A23187 dose-dependently stimulated bulk mucin release. Synergistic interactions were observed between PMA and forskolin, and PMA and A23187. Secretin and isoprenaline were confirmed as mucin secretogogues.      In conclusion gastric mucin release was investigated for the first time by using a suspension of gastric mucosal cells. Two different assay procedures were developed. Some pathways and agents responsible for controlling mucin secretion were identified.

The promoters of the large groundwater developments implemented in the 1970's paid little attention to the effects of pumping on soil moisture. A field study, conducted in 1979 in the Tern Area of the Shropshire Groundwater Scheme, revealed that significant quantities of the available moisture could be removed from the root zone of vegetation when drawdown of shallow watertables occurred. Arguments to this effect, supported by the field study evidence, were successfully presented at the Shropshire Groundwater Scheme public inquiry. The aim of this study has been to expand the work which was undertaken in connection with the Shropshire Groundwater Scheme, and to develop a method whereby the effects of groundwater pumping on vegetation can be assessed, and hence the impacts minimised. Two concepts, the critical height and the soil sensitivity depth, formulated during the initial work are at the core of the Environmental Impact Assessment method whose development is described. A programme of laboratory experiments on soil columns is described, as is the derivation of relationships for determining critical heights and field capacity moisture profiles. These relationships are subsequently employed in evaluating the effects of groundwater drawdown. In employing the environmental assessment technique, digitised maps of relevant features of the Tern Area are combined to produce composite maps delineating the extent of the areas which are potentially sensitive to groundwater drawdown. A series of crop yield/moisture loss functions are then employed to estimate the impact of simulated pumping events on the agricultural community of the Tern Area. Finally, guidelines, based on experience gained through evaluation of the Tern Area case study, are presented for use in the design of soil moisture monitoring systems and in the siting of boreholes. In addition recommendations are made for development of the EIA technique, and further research needs are identified.

The enteroinsular axis (EIA) constitutes a physiological signalling system whereby intestinal endocrine cells secrete incretin hormones following feeding that potentiate insulin secretion and contribute to the regulation of blood glucose homeostasis. The two key hormones responsible are named glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). Recent years have witnessed sustained development of antidiabetic therapies that exploit the EIA. Current clinical compounds divide neatly into two classes. One concerns analogues or mimetics of GLP-1, such as exenatide (Byetta) or liraglutide (NN2211). The other group comprises the gliptins (e.g. sitagliptin and vildagliptin) which boost endogenous incretin activity by inhibiting the enzyme dipeptidyl peptidase 4 (DPP 4) that degrades both GLP-1 and GIP. Ongoing research indicates that further incretin and gliptin compounds will become available for clinical use in the near future, offering comparable or improved efficacy. For incretin analogues there is the prospect of prolonged duration of action and alternative routes of administration. This review focuses on recent advances in pre-clinical research and their translation into clinical studies to provide future therapies for type 2 diabetes targeting the EIA. © 2009 Bentham Science Publishers Ltd.

The cross-country petroleum pipelines are environmentally sensitive because they traverse through varied terrain covering crop fields, forests, rivers, populated areas, desert, hills and offshore. Any malfunction of these pipelines may cause devastating effect on the environment. Hence, the pipeline operators plan and design pipelines projects with sufficient consideration of environment and social aspects along with the technological alternatives. Traditionally, in project appraisal, optimum technical alternative is selected using financial analysis. Impact assessments (IA) are then carried out to justify the selection and subsequent statutory approval. However, the IAs often suggest alternative sites and/or alternate technology and implementation methodology, resulting in revision of entire technical and financial analysis. This study addresses the above issues by developing an integrated framework for project feasibility analysis with the application of analytic hierarchy process (AHP), a multiple attribute decision-making technique. The model considers technical analysis (TA), socioeconomic IA (SEIA) and environmental IA (EIA) in an integrated framework to select the best project from a few alternative feasible projects. Subsequent financial analysis then justifies the selection. The entire methodology has been explained here through a case application on cross-country petroleum pipeline project in India.

One of the most widespread renewable energy sources in Brazil is ethanol, from sugarcane, therefore, the sugar and alcohol sector is expanding, with positive impacts for the economy of the country. Sugar cane was introduced in Brazil as a crop during its colonization, for the production of sugar, and put the country in the global scenario. The expansion of this crop occurred in the seventies, to reduce the reliance in fossil energy sources and to stimulate the development of the agricultural activity. Thus, the federal government has promoted the sugar cane crop and the production of ethanol as a fuel. However, it is important to minimize possible impacts that the crop may cause to the environment. Sugar cane has expanded in the frontiers of the mesoregion of Triângulo Mineiro and Alto Paranaíba-MG, and, in this perspective, the agroindustrial complex known as Companhia Energética Vale do São Simão Ltda., with the Mill located in the county of Santa Vitória, Minas Gerais, was adopted to evaluate the environmental impacts caused by the sugarcane in the area of influence of the mill. The mill has a polygonal area corresponding to 53,525.20 hectares, and for its establishment a Study and Report of Environmental Impacts (EIA/RIMA) was presented, as required as an environment protection instrument by the Environment National Policy (Law nº 6.938/81), and detailed by the Resolution CONAMA nº 01/1986. These studies pointed that native vegetation fragments in the Area of Influence of the Mill, before its implantation, corresponded to approximately 20.7% of the area. Therefore, this study evaluated the impacts of the installation of Usina Vale do São Simão, between 2007 and 2012, determining its reflex on the environmental regularization of the farms, and the vegetation fragments existing in the area, in the recovery and recomposition of areas defined as Legal Reserve and Permanent Preservation. Previous studies of the area were analyzed, soil use and occupation was mapped for the years 2007 and 2012, and the areas of permanent preservation and native vegetation fragments were marked. In general, there was a decline in native vegetation coverage in the period, although it cannot be stated that such reduction was a direct effect of the milling activity. Therefore, the legal requirement of preserving such areas was not capable of bringing the positive effects of protection and recovery as demanded by the Law, highlighting that the current legislation was not enough to protect such areas.

In this paper we analyze the set of Bronze Age bone tools recovered at the archaeological site of El Portalón of Cueva Mayor in the Sierra de Atapuerca (Burgos). The Bronze Age cultural period is the best represented in the cavity and its study has forced us to unify the different excavation and stratigraphical criteria undertaken from the earliest archaeological excavations developed by J.M. Apellániz during the 70s until the excavations of the current research team (EIA) since 2000. We propose here for the first time a relationship between the initial system of “beds” used by Apellániz and our recent sedimentary sequence that recognizes eleven stratigraphic levels radiometrically dated from the late Upper Pleistocene to the Middle Age. Within the bone industry assemblage we recognize a large variety of utensils and ornamental elements, with native and allochthonous features, that make evident a regional as well as long distance relationships of these populations of the interior of the Iberian Peninsula during the recent Prehistory.

Tsar Peter the Great ruled Russia between 1689 and 1725. Its domains, stretching from the Baltic Sea in the west to the Pacific Ocean in the east. From north to south, its empire stretching from the Arctic Ocean to the borders with China and India. Tsar Peter I tried to extend the geographical knowledge of his government and the rest of the world. He was also interested in the expansion of trade in Russia and in the control of trade routes. Feodor Luzhin and Ivan Yeverinov explored the eastern border of the Russian Empire, the trip between 1719 and 1721 and reported to the Tsar. They had crossed the peninsula of Kamchatka, from west to east and had traveled from the west coast of Kamchatka to the Kuril Islands. The information collected led to the first map of Kamchatka and the Kuril Islands. Tsar Peter ordered Bering surf the Russian Pacific coast, build ships and sail the seas north along the coast to regions of America. The second expedition found equal to those of the previous explorers difficulties. Two ships were eventually thrown away in Okhotsk in 1740. The explorers spent the winter of 1740-1741 stockpiling supplies and then navigate to Petropavlovsk. The two ships sailed eastward and did together until June 20, then separated by fog. After searching Chirikov and his boat for several days, Bering ordered the San Pedro continue to the northeast. There the Russian sailors first sighted Alaska. According to the log, "At 12:30 (pm July 17) in sight of snow-capped mountains and between them a high volcano." This finding came the day of St. Elijah and so named the mountain.

Exponemos los resultados de una observación exploratoria en la que se analizan los impactos funcionales en el sistema administrativo como consecuencia de la construcción de la Central Hidroeléctrica de Belo Monte (CHB) en el municipio de Altamira (Amazonia oriental brasileña). En la perspectiva teórico-conceptual de la teoría de sistemas sociales de Niklas Luhmann, realizamos una observación del sistema administrativo local sobre sus operaciones y funciones, en base a cuatro códigos preestablecidos: toma de decisiones, contingencia, significación simbólica del proyecto y planificación. Las conclusiones permiten entrever en un aplano conceptual la utilidad de los conceptos sistémicos de Luhmann, y en el empírico, una planificación y administración pública municipal reactivas.

The physiological mechanisms of parental and alloparental care in cooperatively breeding nonhuman primate species such as the common marmoset (Callithrix jacchus) are poorly known. In this study, we examined prolactin and cortisol plasma levels of fathers and older offspring of both sexes, with and without previous experience in infant carrying, around parturition and during infant carrying. Blood samples were collected from fathers and older offspring and prolactin and cortisol were measured by RIA and EIA, respectively. Prolactin levels of both caretakers were not influenced by infant’s birth, previous experience or proximity to parturition. However, prolactin levels increased in both caretakers while in physical contact with infants and also with the number of infants being carried in older offspring. These findings suggest that increased prolactin seems to be mainly due to physical effort rather than a physiological trigger of paternal and alloparental care in common marmosets. Cortisol levels were higher for experienced fathers shortly before parturition which could act to reinforce affiliative bonds between breeding males and females at this time or in the ability of males to detect the proximity of the parturition or both
