In order to assess the potential risk of anti-HBc-positive blood donors for post-transfusional hepatitis and to investigate whether other HBV serological markers are capable of identifying the presence of the virus, 1000 first-time blood donors were enrolled between June and July 1997. These donors were screened using routine Brazilian blood center tests (HIV 1 and 2, HTLV 1 and 2, Chagas disease, Syphilis, HCV, HBsAg, anti-HBc and ALT ). The 120 (12%) found to be anti-HBc-positive underwent further tests: HBe, anti-HBe, anti-HBs and HBV-DNA by PCR. Ten cases were HBsAg positive and all were HBV-DNA positive by PCR. Three HBsAg-negative donors were HBV-DNA-positive. Two HBV-DNA-positive donors were also anti-HBs-positive. All the HBV-positive donors had at least one HBV marker other than anti-HBc. Anti-HBc is an important cause of blood rejection. Testing for HBsAg alone is not fully protective and anti-HBc remains necessary as a screening test. The presence of anti-HBs is not always indicative of absence of the virus. The addition of other HBV serological markers could represent an alternative in predicting the presence of the virus when compared with PCR. It is recommended that other studies should be carried out to confirm this finding.

The wide use of antibiotics in aquaculture has led to the emergence of resistant microbial species. It should be avoided/minimized by controlling the amount of drug employed in fish farming. For this purpose, the present work proposes test-strip papers aiming at the detection/semi-quantitative determination of organic drugs by visual comparison of color changes, in a similar analytical procedure to that of pH monitoring by universal pH paper. This is done by establishing suitable chemical changes upon cellulose, attributing the paper the ability to react with the organic drug and to produce a color change. Quantitative data is also enabled by taking a picture and applying a suitable mathematical treatment to the color coordinates given by the HSL system used by windows. As proof of concept, this approach was applied to oxytetracycline (OXY), one of the antibiotics frequently used in aquaculture. A bottom-up modification of paper was established, starting by the reaction of the glucose moieties on the paper with 3-triethoxysilylpropylamine (APTES). The so-formed amine layer allowed binding to a metal ion by coordination chemistry, while the metal ion reacted after with the drug to produce a colored compound. The most suitable metals to carry out such modification were selected by bulk studies, and the several stages of the paper modification were optimized to produce an intense color change against the concentration of the drug. The paper strips were applied to the analysis of spiked environmental water, allowing a quantitative determination for OXY concentrations as low as 30 ng/mL. In general, this work provided a simple, method to screen and discriminate tetracycline drugs, in aquaculture, being a promising tool for local, quick and cheap monitoring of drugs.

Studies on microbial characterization of cold-smoked salmon and salmon trout during cold storage were performed on samples available in the Portuguese market. Samples were also classified microbiologically according to guidelines for ready-to-eat (RTE) products. Further investigations on sample variability and microbial abilities to produce tyramine and histamine were also performed. The coefficient of variation for viable counts of different groups of microorganisms of samples collected at retail market point was high in the first 2 wk of storage, mainly in the Enterobacteriaceae group and aerobic plate count (APC), suggesting that microbiological characteristics of samples were different in numbers, even within the same batch from the same producer. This variation seemed to be decreased when storage and temperature were controlled under lab conditions. The numbers of Enterobacteriaceae were influenced by storage temperature, as indicated by low microbial numbers in samples from controlled refrigeration. Lactic acid bacteria (LAB) and Enterobacteriaceae were predominant in commercial products, a significant percentage of which were tyramine and less histamine producers. These results might be influenced by (1) the technological processes in the early stages of production, (2) contamination during the smoking process, and (3) conditions and temperature fluctuations during cold storage at retail market point of sale.

Introdução: A síndrome dolorosa miofascial é um dos principais problemas encontrados na prática clínica, tendo como principais características os pontos gatilho (PG), ativos ou latentes. Os PG latentes têm uma elevada prevalência na musculatura da cintura escapular, nomeadamente ao nível do trapézio superior (TS), influenciando o controlo motor do ombro. A compressão isquémica aplicada no PG do TS poderá influenciar o comportamento muscular dos sinergistas durante o movimento de abdução no plano da omoplata. Objetivos: Este estudo visa estudar a influência da compressão isquémica aplicada no PG do TS na magnitude de ativação dos músculos TS, trapézio inferior (TI), grande dentado (GD), infra-espinhoso (IE) e deltóide médio (DM), assim como a relação muscular através da razão entre o TS e os restantes músculos em análise, durante o movimento de abdução no plano da omoplata. Métodos: O presente estudo é experimental, aleatório e controlado. Foram criados dois grupos, grupo controlo (GC) (n=14) e grupo experimental (GE) (n=15), a partir de uma amostra de 67 indivíduos. No GC foi aplicado um procedimento placebo e no GE foi aplicada no PG latente do TS a técnica de compressão isquémica. Antes e após a intervenção foi recolhida a atividade eletromiográfica dos músculos em análise, assim como os dados cinemáticos, durante o movimento de elevação do ombro no plano da omoplata. Para a análise registou-se a amplitude máxima de abdução e foi analisada a atividade muscular individual dos músculos em estudo assim como a razão entre o TS e os restantes músculos. Esta análise foi realizada em intervalos de 30° até ao final do movimento. Resultados: Não se verificaram diferenças na amplitude máxima de abdução, nem entre grupos (1º momento p=0,608, t=0,816; 2º momento p=0,119; t=1,252) nem entre os dois momentos em cada grupo (GC, t=-1,119; p=0,256; GE, t=-1,604, p=0,135). Na magnitude de ativação individual de cada músculo também não se verificaram diferenças significativas com a aplicação da técnica, tendo-se verificado no DM uma tendência para o aumento da ativação aquando da intervenção, ao longo de todo o arco de movimento, em comparação com a pré-intervenção, já no GC de controlo não se verificaram alterações entre momentos. Também na análise da razão entre músculos não se verificaram diferenças entre grupos. Conclusão: A intervenção realizada não influenciou nenhum dos parâmetros em análise do complexo articular do ombro nem a amplitude de movimento de abdução.

While testing 414 sera for the diagnosis of Chagas' disease, the conventional reactions of indirect hemagglutination, indirect immunofluorescence and the immunosorbent assay showed a sensitivity of 95.7%, 100% and 98.2% and a specificity of 98%, 98% and 96.4%, respectively, and an excellent association using Fisher's exact test. Chemiluminescence presented 100% sensitivity and 89.6% specificity, while PCR showed 100% specificity and 1.2% sensitivity. It is believed that the three conventional serological reactions are still adequate for diagnosing Chagas' disease.

Clostridium difficile is a gram positive, spore former, anaerobic bacterium that is able to cause infection and disease, with symptoms ranging from mild diarrhea to pseudomembranous colitis, toxic megacolon, sepsis and death. In the last decade new strains have emerged that caused outbreaks of increased disease severity and higher recurrence, morbidity and mortality rates, and C. difficile is now considered both a main nosocomial pathogen associated with antibiotic therapy as well as a major concern in the community.(...)

The aim of the present study was to outline the serological profile of hepatitis C among blood donors seen at the Uberaba Regional Blood Center, Hemominas Foundation, over the last 14 years. The frequency of hepatitis C was compared between first-time and repeat donors and the epidemiological characteristics of those with positive and indeterminate ELISA anti-HCV (third and fourth generation) were analyzed based on the donor histories kept in the archives of the Uberaba Regional Blood Center. The serological ineligibility rate was 0.3%, with higher prevalence in the group of first-time donors. We did not find any significant differences regarding age, skin color, marital status or place of residence between eligible and ineligible donors; however, the frequency of positive serology was higher among men. The lower (0.3%) rate of ineligibility due to hepatitis C that was observed at the Uberaba Regional Blood Center, in relation to most Brazilian blood centers, is probably due to the large number of repeat donors (83.3%). This reinforces the importance of achieving donor commitment for increasing transfusion safety.

A Fundação Robinson, em Portalegre, adquiriu em 2006 uma colecção de arte sacra com um historial muito peculiar, e desde então tem vindo a trabalhar neste espólio, com o objetivo de o preservar, promovendo a sua conservação, estudo e divulgação. Apenas uma pequena percentagem de peças será restaurada e exposta, permanecendo a maioria em reserva. Durante o ultimo ano procedeu-se à avaliação das condições da sala de reserva, cujos resultados se apresentam neste trabalho. Foi caracterizada a coleção, o edifício onde se encontra a sala de reserva e as condições desta mesma sala. Para avaliar os riscos a que a coleção está sujeita foram identificando os agentes de deterioração - forças físicas, fogo, água, ação criminosa, pragas, contaminantes, luz visível e radiação ultravioleta, temperatura incorreta e humidade relativa incorreta e dissociação por negligência, e a ação específica de cada um destes agentes na coleção. Com o objetivo de elaborar um plano de mitigação para cada um dos riscos específicos encontrados realizou-se o cálculo da sua magnitude, recorrendo à metodologia desenvolvida por Robert Waller, o que permitiu estabelecer uma escala de prioridades relativamente à urgência de intervenção. Os resultados desta avaliação de risco permitiram traçar um plano de atuação, baseado em decisões bem informadas de forma a rentabilizar os recursos existentes, apontando métodos de mitigação para reduzir as perdas, de forma a melhorar as condições de conservação deste espólio.
Analysis of metabolic flux distributions in relation to the extracellular environment in Avian cells

Continuous cell lines that proliferate in chemically defined and simple media have been highly regarded as suitable alternatives for vaccine production. One such cell line is the AG1.CR.pIX avian cell line developed by PROBIOGEN. This cell line can be cultivated in a fully scalable suspension culture and adapted to grow in chemically defined, calf serum free, medium [1]–[5]. The medium composition and cultivation strategy are important factors for reaching high virus titers. In this project, a series of computational methods was used to simulate the cell’s response to different environments. The study is based on the metabolic model of the central metabolism proposed in [1]. In a first step, Metabolic Flux Analysis (MFA) was used along with measured uptake and secretion fluxes to estimate intracellular flux values. The network and data were found to be consistent. In a second step, Flux Balance Analysis (FBA) was performed to access the cell’s biological objective. The objective that resulted in the best predicted results fit to the experimental data was the minimization of oxidative phosphorylation. Employing this objective, in the next step Flux Variability Analysis (FVA) was used to characterize the flux solution space. Furthermore, various scenarios, where a reaction deletion (elimination of the compound from the media) was simulated, were performed and the flux solution space for each scenario was calculated. Growth restrictions caused by essential and non-essential amino acids were accurately predicted. Fluxes related to the essential amino acids uptake and catabolism, the lipid synthesis and ATP production via TCA were found to be essential to exponential growth. Finally, the data gathered during the previous steps were analyzed using principal component analysis (PCA), in order to assess potential changes in the physiological state of the cell. Three metabolic states were found, which correspond to zero, partial and maximum biomass growth rate. Elimination of non-essential amino acids or pyruvate from the media showed no impact on the cell’s assumed normal metabolic state.

The objective of this study was to differentiate benign ovarian tumors from malignant ones before surgery using color and pulsed Doppler sonography, and to compare results obtained before and after use of contrast medium, thereby verifying whether contrast results in an improvement in the diagnostic sensitivity. METHODS: Sixty two women (mean age 49.9 years) with ovarian tumors were studied, 45 with benign and 17 with malignant tumors. All women underwent a transvaginal color Doppler ultrasonographic exam. A study of the arterial vascular flow was made in all tumor areas, as well as an impedance evaluation of arterial vascular flow using the resistance index. RESULT: Localization of the vessels in the tumor revealed a greater proportion of malignant tumors with detectable internal vascular flows (64%) than benign tumors with such flows (22%). There was a considerable overlap of these findings. The use of contrast identified a greater number of vessels with confirmation in the totality of tumors, but did not improve the Doppler capacity in tumoral differentiation. Malignant tumors presented lower values of resistance index than the benign ones, whether or not contrast was used. The cutoff value for resistance index that better maximized the Doppler sensitivity and specificity was 0.55. Through this value, an increase of the sensitivity after contrast use was obtained, varying from 47% to 82%, while specificity remained statistically unchanged. CONCLUSION: Although the injection of a microbubble agent improved the sensitivity of the method detecting vascularization of tumors, a positive finding for vascularization by this method was not clinically useful in the differentiation of benign and malignant ovarian tumors.
