Differential protein expression analysis based on modification of selected amino acids with labelling reagents has become the major method of choice for quantitative proteomics. One such methodology, two-dimensional difference gel electrophoresis (2-D DIGE), uses a matched set of fluorescent N-hydroxysuccinimidyl (NHS) ester cyanine dyes to label lysine residues in different samples which can be run simultaneously on the same gels. Here we report the use of iodoacetylated cyanine (ICy) dyes (for labelling of cysteine thiols, for 2-D DIGE-based redox proteomics. Characterisation of ICy dye labelling in relation to its stoichiometry, sensitivity and specificity is described, as well as comparison of ICy dye with NHS-Cy dye labelling and several protein staining methods. We have optimised conditions for labelling of nonreduced, denatured samples and report increased sensitivity for a subset of thiol-containing proteins, allowing accurate monitoring of redox-dependent thiol modifications and expression changes. Cysteine labelling was then combined with lysine labelling in a multiplex 2-D DIGE proteomic study of redox-dependent and ErbB2-dependent changes in epithelial cells exposed to oxidative stress. This study identifies differentially modified proteins involved in cellular redox regulation, protein folding, proliferative suppression, glycolysis and cytoskeletal organisation, revealing the complexity of the response to oxidative stress and the impact that overexpression of ErbB2 has on this response.

Self-report measures of obsessive-compulsive disorder (OCD) in children and adolescents are needed for practical evaluation of severity and treatment response. We compared the self- and parent-report Obsessional Compulsive Inventory Revised (CHOCI-R) to the interview-based Child Yale-Brown Obsessive-Compulsive Scale (CY-BOCS) in a clinical sample of 285 children and adolescents with OCD. Classical test theory and item-response theory were applied to compare the instruments. The self- and parent-report CHOCI-R had good internal consistency and were strongly related to each other. The self- and parent-report CHOCI-R severity scores correlated with the CY-BOCS (Pearson's r 0.55 and 0.45 respectively). The CY-BOCS discriminated better at the severe end of the spectrum. The CHOCI-R provided better discrimination in the mild to moderate range. The time-efficient self- and parent-report alternatives will enable routine measurement of OCD severity in clinical practice. The clusters [Fe3(CO)11(RCN)] (1: R = Me, C3H5, C6H5, or C6H4-2-Me) have been prepared at low temperature from [Fe3(CO)12] and RCN in the presence of Me3NO. Compounds 1 react essentially quantitatively with a wide range of two-electron donors, L, (viz.: CO, PPh3, P(OMe)3, PPh2H, PPh2Me, PF3, CyNC (Cy = cyclohexyl), P(OEt)3, SbPh3, PBu3, AsPh3, or SnR2 (R = CH(SiMe3)2)) to give [Fe3(CO)11L] (2). In some cases (2), on treatment with Me3NO and then L′ (L′ = a second two-electron donor) yields [Fe3(CO)10LL′] in high yield. The crystal and molecular structures of 1 (L = NCC6H4Me-2) have been determined by a full single crystal structure analysis, and shown to have an axial nitrile coordinated at the unique iron atom, with two CO groups bridging the other two metal atoms.

The response of North Atlantic and European extratropical cyclones to climate change is investigated in the climate models participating in phase 5 of the Coupled Model Intercomparison Project (CMIP5). In contrast to previous multimodel studies, a feature-tracking algorithm is here applied to separately quantify the re- sponses in the number, the wind intensity, and the precipitation intensity of extratropical cyclones. Moreover, a statistical framework is employed to formally assess the uncertainties in the multimodel projections. Under the midrange representative concentration pathway (RCP4.5) emission scenario, the December–February (DJF) response is characterized by a tripolar pattern over Europe, with an increase in the number of cyclones in central Europe and a decreased number in the Norwegian and Mediterranean Seas. The June–August (JJA) response is characterized by a reduction in the number of North Atlantic cyclones along the southern flank of the storm track. The total number of cyclones decreases in both DJF (24%) and JJA (22%). Classifying cyclones according to their intensity indicates a slight basinwide reduction in the number of cy- clones associated with strong winds, but an increase in those associated with strong precipitation. However, in DJF, a slight increase in the number and intensity of cyclones associated with strong wind speeds is found over the United Kingdom and central Europe. The results are confirmed under the high-emission RCP8.5 scenario, where the signals tend to be larger. The sources of uncertainty in these projections are discussed.

First published February 17, 2010; doi: 10.1152/ajpcell.00417.2009.-Cell mechanical properties on a whole cell basis have been widely studied, whereas local intracellular variations have been less well characterized and are poorly understood. To fill this gap, here we provide detailed intracellular maps of regional cytoskeleton (CSK) stiffness, loss tangent, and rate of structural rearrangements, as well as their relationships to the underlying regional F-actin density and the local cytoskeletal prestress. In the human airway smooth muscle cell, we used micropatterning to minimize geometric variation. We measured the local cell stiffness and loss tangent with optical magnetic twisting cytometry and the local rate of CSK remodeling with spontaneous displacements of a CSK-bound bead. We also measured traction distributions with traction microscopy and cell geometry with atomic force microscopy. On the basis of these experimental observations, we used finite element methods to map for the first time the regional distribution of intracellular prestress. Compared with the cell center or edges, cell corners were systematically stiffer and more fluidlike and supported higher traction forces, and at the same time had slower remodeling dynamics. Local remodeling dynamics had a close inverse relationship with local cell stiffness. The principal finding, however, is that systematic regional variations of CSK stiffness correlated only poorly with regional F-actin density but strongly and linearly with the regional prestress. Taken together, these findings in the intact cell comprise the most comprehensive characterization to date of regional variations of cytoskeletal mechanical properties and their determinants.

Tidigare studier har visat att hästnäringen inklusive spridningseffekter årligen omsätter ca 46 miljarder kronor och bidrar till sysselsättning för ca 30 000 helårsverken. Motsvarande beräkningar för travsportens del av hästnäringen eller för en mindre region finns däremot inte. Denna rapport syftar till att uppskatta regionalekonomisk betydelse av travsport i Sverige med utgångspunkt i en mindre travbanas verksamhet. Dalatravet Rättvik (DT Rättvik) används som en fallstudie. Resultaten baseras dels på registerdata över faktiskt registrerade hästar med koppling till DT Rättvik dels på enkätmaterial insamlat från ett urval av besökare vid tävlingar under sommaren 2014. Totalt används svar från 444 besökare i analysen. Resultaten från beräkningarna visar att den direkta ekonomiska effekten av DT Rättviks 333 travhästar i amatör- eller proffsträning genererar 20 miljoner i travhästtrelaterad konsumtion. DT Rättviks travsportevenemang uppskattas generera ca 29 miljoner varav 11,1 miljoner ärett tillskott till den regionala ekonomin i form av turistekonomisk omsättning. Travsportens sammanlagda regionalekonomiska effekt beräknas således uppgå till minst 31,1 miljonerkronor. Inkluderas även utbildning, avel och uppfödning samt sysselsättning vid ATG-ombudså uppskattas travsporten generera ca 40 helårsverken inom DT Rättviksregionen.

Esta dissertação teve como objetivo o estudo e desenvolvimento da técnica de síntese por combustão em solução para obtenção de nanocristais de alumina, e a caracterização desse material por diferentes técnicas. O estudo da obtenção incluiu o modelamento termodinâmico das reações investigadas e a influência da razão combustíveloxidante no tamanho de partícula. A síntese da alumina por combustão foi realizada a partir do par precursor químico glicina-nitrato de alumínio em água, para diferentes razões combustível-oxidante, temperaturas de chama e número total de moles gasosos, relacionando esses parâmetros com tamanho de cristalito no pó, deformação de rede, área superficial e porosidade total. As reações químicas de combustão em solução resultaram na formação in situ de fases metaestáveis de Al2O3 para o pó como-sintetizado. A fase Al2O3-α, cuja formação não ocorreu nem na presença de um extra-oxidante, somente foi obtida após um tratamento térmico a 1100°C do pó como-sintetizado. O modelamento termodinâmico da reação de combustão em solução mostrou que quando a razão combustível-oxidante aumenta, obtém-se uma elevação da temperatura de chama adiabática e da quantidade de gás produzida, definindo características do particulado como morfologia, tamanho de cristalito, área superficial e nível de aglomeração. A caracterização do pó utilizou técnicas como análise termodiferencial (ATD) e termogravimétrica (ATG), granulometria por dispersão de laser (GDL), BET para análise de área superficial e porosidade total, difração de raios X (DRX), microscopia eletrônica de varredura (MEV), e microscopia eletrônica de transmissão (MET). Os pós de alumina alfa obtidos apresentaram-se como constituídos de cristalitos nanométricos, dispostos na forma de agregados de tamanho micrométrico. O tamanho de cristalito médio, calculado pelo método de Williamson-Hall, a partir de dados de análises por DRX, foi de 97 nm, e deformação média calculada de 5x10-3 % A análise granulométrica dos pós como-sintetizados verificou um tamanho de aglomerado de 40 µm, com distribuição monomodal. As análises por MEV e MET confirmaram a aglomeração das partículas do pó como-sintetizado, a partir de partículas com morfologia irregular.

o presente estudo analisa os custos da escola rização de uma amostra de escolas do município do Rio de Janeiro e de outra do Distrito Federal, com o objetivo de determinar sua escala ótima de operação e analisar sua eficiência na utilização dos seus recursos físicos e humanos. A escala ótima, como o trabalho demonstra, e determinadÇl. pe;Lo pontQ de minimo . .das curvas de -custo, estimando o tamanho ideal para o funcionamento das escolas, de sorte a obter o máximo de eficiência e produtividade. A eficiência é analisada em termos da intensidade de uso da capacidade física instalada e da alocação adequada.dos recursos humanos, em função da natureza dos serviços escolares e do número de alunos nela matriculados. Espera-se demonstrar que o planejamento edu cacional pode aumentar a eficiência do sistema tirando proveito do que propoe a teoria microeconômica ao abordar as questões relati vas à função de produção e aos seus custos. E que, ao final, a cor reta utilização desses conceitos pode ser muito útil para ajudar a superar a desnecessária, mas muitas vezes real - dicotomia entre quantidade e qualidade. Planejamento economicamente correto pode permitir oferecer e~sino de qualidade a uma grande quantidade de crianças, adolescentes e jovens. A quantidade, antes de ser obstá culo, pode até favorecer a oferta de um ensino de boa qualidade. Evidentemente, é necessár~o competência técnica e científica para fazê-lo, secundando uma firme vontade política de fazê-lo.

Vetores de expressão são ferramentas moleculares úteis para investigar a função de genes tanto em sistemas procarióticos quanto eucarióticos. O fungo Metarhizium anisopliae, utilizado no controle biológico de artrópodes, é bem caracterizado em nível molecular. Genes candidatos a participar do processo de infecção de hospedeiros tem sido isolados utilizando estratégias em que o gene candidato é predefinido (enzimas hidrolíticas, por exemplo) ou estratégias mais globais como projetos de ESTs e a análise de bibliotecas de subtração. A superexpressão tem sido o método adotado para verificar a participação de genes isolados no processo de infecção. Esta estratégia tem sido baseada no promoter heterólogo PgpdA de Aspergillus nidulans. Neste trabalho, o gene que codifica o fator de alongamento de tradução tef-1 α de Metarhizium anisopliae foi clonado e a sua região promotora foi localizada e utilizada na construção de um vetor de expressão. Somente uma cópia do gene tef-1α está presente no genoma de M. anisopliae e o seu perfil de expressão foi analisado. Uma árvore filogenética foi construída baseada nos ortógos de tef-1 α e mostrou uma alta correlação com o fungo Cordyceps taii. A região de 639 pb à montante do codon de iniciação (ATG) foi utilizada com sucesso para a expressão do gene repórter sGFP e do gene bar, que confere resistência ao glifosinato de amônio, em M. anisopliae. Os transformantes construídos não apresentaram alteração na sua virulência em bioensaios com carrapatos, em relação a linhagem receptora. Além disso, demonstramos que o nível de expressão permite a detecção óptica da fluorescência de sGFP durante a infecção dos carrapatos. Desta forma, o vetor desenvolvido será uma ferramenta útil para a superexpressão em Metarhizium.

Continuous Synthesis by Solution Combustion was employed in this work aiming to obtain tin dioxide nanostructured. Basically, a precursor solution is prepared and then be atomized and sprayed into the flame, where its combustion occurs, leading to the formation of particles. This is a recent technique that shows an enormous potential in oxides deposition, mainly by the low cost of equipment and precursors employed. The tin dioxide (SnO2) nanostructured has been widely used in various applications, especially as gas sensors and varistors. In the case of sensors based on semiconducting ceramics, where surface reactions are responsible for the detection of gases, the importance of surface area and particle size is even greater. The preference for a nanostructured material is based on its significant increase in surface area compared to conventional microcrystalline powders and small particle size, which may benefit certain properties such as high electrical conductivity, high thermal stability, mechanical and chemical. In this work, were employed as precursor solution tin chloride dehydrate diluted in anhydrous ethyl alcohol. Were utilized molar ratio chloride/solvent of 0,75 with the purpose of investigate its influence in the microstructure of produced powder. The solution precursor flux was 3 mL/min. Analysis with X-ray diffraction appointed that a solution precursor with molar ratio chloride/solvent of 0,75 leads to crystalline powder with single phase and all peaks are attributed to phase SnO2. Parameters as distance from the flame with atomizer distance from the capture system with the pilot, molar ratio and solution flux doesn t affect the presence of tin dioxide in the produced powder. In the characterization of the obtained powder techniques were used as thermogravimetric (TGA) and thermodiferential analysis (DTA), particle size by laser diffraction (GDL), crystallographic analysis by X-ray diffraction (XRD), morphology by scanning electron microscopy (SEM), transmission electron microscopy (TEM), specific surface area (BET) and electrical conductivity analysis. The field of "Materials Chemistry" has been developing in recent years and there has been a great increase of interest in the synthesis and chemical and physical properties of new inorganic solids. New routes of synthesis and synthesis modified has been developed with the aim not only to optimize the processes in laboratory scale, but also on an industrial scale, and make them acceptable by current environmental legislation. The phenomenology of current solid state chemistry properties coupled with the high temperature superconductivity, ferromagnetism, porosity molecular and colors are evidence affected by the synthesis method, which in turn can influence the technological application of these materials. From this understanding, mixed oxides of nickel and zinc nanoparticulate were synthesized by microwave-assisted combustion route using three specific types of organic fuels employing the weight ratios 1:1/2 and 1:1 of cation metallic/fuel, in order to investigate the influence of such proportions to obtain the solids. The new fuels were chosen to replace, for example, urea or glycine that are the fuels most commonly preferred in this kind of synthesis. The powders without heat treatment were studied by Thermogravimetric analysis (TGA), X-Ray Diffraction (XRD) and then calcined at 900°C. After heat treatment, the samples were characterized by analysis of X Ray Diffraction (XRD) and Scanning Electron Microscopy (SEM). The modified synthesis route porposed was effective for obtaining powders. Both the alternative fuels chosen as the different weight ratios employed, influenced in the morphology and obtaining oxides
