A Leishmaniose é uma doença infecciosa causada por várias espécies de parasitas do gênero Leishmania. A quimioterapia é o único tratamento efetivo para a doença, mas essas drogas são, em geral, tóxicas e requer um longo período de tratamento. Produtos naturais provenientes de plantas oferecem novas perspectivas e representam uma importante fonte de novos agentes leishmanicidas. Assim, é de grande importância avaliar os efeitos do extrato aquoso da raiz de Physalis angulata, planta amplamente utilizada pela medicina popular, em formas promastigotas e amastigotas de Leishmania (Leishmania) amazonensis e sua ação sobre a célula hospedeira. As fisalinas D, E, F e G foram demonstradas pela primeira vez na raiz de P. angulata pela análise cromatográfica. Uma atividade antiproliferativa e uma inibição dose dependente de promastigotas 74,1% e 99,8 % (IC50 35,5 μg/mL) e amastigotas 70,6% e 70,9% (IC50 32.0 μg/mL) foram observadas quando os parasitas foram tratados com 50 e 100 μg/mL do extrato, respectivamente. A análise da atividade microbicida da célula hospedeira infectada com L. amazonensis mostrou que extrato foi capaz de reverter o efeito causado pelo parasito de inibir a produção de espécies reativas de oxigênio. O tratamento com o extrato também induziu alterações morfológicas importantes em formas promastigotas avaliadas por microscopia óptica, microscopia eletrônica de transmissão e varredura. Foram observadas alterações na morfologia, na divisão celular, principalmente na fase de citocinese, na membrana flagelar, na bolsa flagelar e alterações em organelas importantes, como o cinetoplasto, onde ocorreu duplicação irregular e alteração do seu tamanho. Já por citometria de fluxo foi possível confirmar que o tratamento induziu uma exposição de fosfatidilserina e diminuição no volume celular de promastigotas tratadas. Com relação à célula hospedeira, o extrato promoveu alterações no citoesqueleto, o aumento número de projeções citoplasmáticas, do volume celular e de vacúolos e da habilidade de espraiamento sem causar efeito citotóxico ou alteração ultraestrutural em macrófagos tratados com o extrato. Assim, estes resultados demonstram que o extrato aquoso da raiz de P. angulata foi eficaz na ativação da célula hospedeira e na inibição do crescimento do protozoário, o que representa uma fonte alternativa e promissora de agente leishmanicida.

In special education, high skills designate people who demonstrate the capability, potential or performance in human activities, well above average. Our objective, using neuroscience to understand the early, prodigies, geniuses and high abilities as a result of a unique process of memory formation. It is a literature search, in which the works were identified, located, compiled, analyzed and blacklisted. The material was located from keywords in databases. For the compilation were used as inclusion criteria, adopted the intersection of the keywords as well as the work of researchers from education, psychology and neuroscience, published between 1966 and 2009; qualitatively analyzed 40 references, 18 articles and 22 other texts. The trajectory analysis was developed with the question: no need to explain differently the precocious prodigies, geniuses, and AH? We reached three logical propositions. We demonstrate through this exercise of course, that there would be no need to explain differently the precocious prodigies, geniuses, and AH, but conclude that there is need for such differentiation, even within the field of neuroscience, however, the statement above points to Renzulli’s theory as valid for geniuses and AH, with exceptions for early and wonders that deserve a closer look.

In this work we address contributions from scalars to (g-2)μ. In order to explain the recently measured deviation by the BNL experiment on (g-2)μ, it is necessary that these scalars are either light or couple strongly with muons. Here we explore this last possibility. We show that a scalar with mass of the order of 102 GeV provides significant contribution to (g-2)μ if the Yukawa coupling is about 10-1. We suggest scenarios where this comes about naturally. ©2001 The American Physical Society.

Atmospheric particulate matter (PM) is genotoxic and recently was classified as carcinogenic to humans by the International Agency for Research on Cancer. PM chemical composition varies depending on source and atmospheric conditions. The Salmonella/microsome assay is the most used mutagenicity test and can identify the major chemical classes responsible for observed mutagenicity. The objective of this work was to characterize the mutagenicity of PM samples from a countryside city, Limeira, Brazil, which is influenced by heavy traffic and sugar cane biomass burning. Six samples of total PM were collected. Air mass backward trajectories were calculated. Organic extracts were assayed using the Salmonella/microsome microsuspension mutagenicity assay using TA98, YG1041, and TA1538, with and without metabolic activation (S9). YG1041 was the most sensitive strain and mutagenicity reached 9,700 revertants per m(3) without metabolic activation. Potency for TA1538 was higher than TA98, indicating that this strain should be considered in air mutagenicity studies. The increased response to YG1041 relative to TA98, and the decreased response with S9, suggests that nitroaromatics are the major contributors. Limeira is among the most mutagenic cities in the world. High mutagenicity in Limeira seems to occur when the air mass from the area of sugarcane production is mixed with air from the region impacted by anthropogenic activities such as traffic. An increase in the formation of nitro-polycyclic aromatic hydrocarbons may result from longer contact time between the aromatic compounds and the atmosphere with high NOx and ozone concentration, although more studies are required to confirm this hypothesis. This communication describes the general biochemical properties and some immunological characteristics of the venom from the Peruvian scorpion Hadruroides lunatus, which is the most medically relevant species in Peru. The soluble venom of this scorpion is toxic to mice, the LD50 determined was 0.1 mg/kg and 21.55 mg/kg when the venom was injected intracranial or intraperitoneally, respectively. The soluble venom displayed proteolytic, hyaluronidasic, phospholipasic and cardiotoxic activities. High performance liquid chromatography of the soluble venom resulted in the separation of 20 fractions. Two peptides with phospholipasic activity were isolated to homogeneity and their molecular masses determined by mass spectrometry (MALDI TOF). Anti-H. lunatus venom sera were produced in rabbits. Western blotting analysis showed that most of the protein content of this venom is immunogenic. H. lunatus anti-venom displayed consistent cross-reactivity with venom antigens from the new World-scorpions Tityus serrulatus and Centruroides sculpturatus venoms; however, a weaker reactivity was observed against the venom antigens from the old World-scorpion Androctonus australis Hector. The structures and functional activities of metalloproteinases from snake venoms have been widely studied because of the importance of these molecules in envenomation. Batroxase, which is a metalloproteinase isolated from Bothrops atrox (Para) snake venom, was obtained by gel filtration and anion exchange chromatography. The enzyme is a single protein chain composed of 202 amino acid residues with a molecular mass of 22.9 kDa, as determined by mass spectrometry analysis, showing an isoelectric point of 7.5. The primary sequence analysis indicates that the proteinase contains a zinc ligand motif (HELGHNLGISH) and a sequence C164I165M166 motif that is associated with a "Met-turn" structure. The protein lacks N-glycosylation sites and contains seven half cystine residues, six of which are conserved as pairs to form disulfide bridges. The three-dimensional structure of Batroxase was modeled based on the crystal structure of BmooMP alpha-I from Bothrops moojeni. The model revealed that the zinc binding site has a high structural similarity to the binding site of other metalloproteinases. Batroxase presented weak hemorrhagic activity, with a MHD of 10 mu g, and was able to hydrolyze extracellular matrix components, such as type IV collagen and fibronectin. The toxin cleaves both a and beta-chains of the fibrinogen molecule, and it can be inhibited by EDTA. EGTA and beta-mercaptoethanol. Batroxase was able to dissolve fibrin clots independently of plasminogen activation. These results demonstrate that Batroxase is a zinc-dependent hemorrhagic metalloproteinase with fibrin(ogen)olytic and thrombolytic activity. Published by Elsevier Ltd.

Large conurbations are a significant source of the anthropogenic pollution and demographic differences between cities that result in a different pollution burden. The metropolitan area of Sao Paulo (MASP, population 20 million) accounts for one fifth of the Brazilian vehicular fleet. A feature of MASP is the amount of ethanol used by the vehicular fleet, known to exacerbate air quality. The study describes the diurnal behaviour of the submicron aerosol and relies on total particle number concentration, particle number size distribution, light scattering and light absorption measurements. Modelled planetary boundary layer (PBL) depth and air mass movement data were used to aid the interpretation. During morning rush-hour, stagnant air and a shallow PBL height favour the accumulation of aerosol pollution. During clear-sky conditions, there was a wind shift towards the edge of the city indicating a heat island effect with implications on particulate pollution levels at the site. The median total particle number concentration for the submicron aerosol typically varied in the range 1.6 x 10(4)-3.2 x 10(4) cm(-3) frequently exceeding 4 x 10(4) cm-3 during the day. During weekdays, nucleation-mode particles are responsible for most of the particles by numbers. The highest concentrations of total particle number concentrations and black carbon (BC) were observed on Fridays. Median diurnal values for light absorption and light scattering (at 637 nm wavelength) varied in the range 12-33 Mm(-1) and 21-64 Mm(-1), respectively. The former one is equal to 1.8-5.0 mu g m(-3) of BC. The growth of the PBL, from the morning rush-hour until noon, is consistent with the diurnal cycle of BC mass concentrations. Weekday hourly median single-scattering albedo (omega(0)) varied in the range 0.59-0.76. Overall, this suggests a top of atmosphere (TOA) warming effect. However, considering the low surface reflectance of urban areas, for the given range of omega(0), the TOA radiative forcing can be either positive or negative for the sources within the MASP. On the average, weekend omega(0) values were 0.074 higher than during weekdays. During 11% of the days, new particle formation (NPF) events occurred. The analysed events growth rates ranged between 9 and 25 nm h(-1). Sulphuric acid proxy concentrations calculated for the site were less than 5% of the concentration needed to explain the observed growth. Thus, other vapours are likely contributors to the observed growth.
