Hintergrund: Miniaturisierung ist ein häufig beobachtetes Phänomen bei Pflanzen in arktisch-alpinen Lebensräumen und wird als Anpassung an niedrige Jahresmitteltemperaturen und eine kurze Vegetationsperiode interpretiert. Ziele: In der vorliegenden Arbeit wird im Petasites-Clade (Petasites Mill., Endocellion Turcz. ex Herder, Homogyne Cass., Tussilago L.; Asteraceae) und in Soldanella (Primulaceae) die Evolution der Miniaturisierung arktisch-alpiner Arten untersucht. Zudem wird innerhalb von Homogyne untersucht, ob unterschiedliche edaphische Präferenz von H. alpina (variabel) und H. discolor (kalkliebend) genetisch fixiert ist. rnMethoden: Molekulare Phylogenien des Petasites-Clades und von Soldanella wurden mit nukleären und plastidären Markern erstellt, und mit den in den Alpen vorkommenden Soldanella-Arten wurde zudem eine Fingerprint-Studie (AFLPs) gemacht. Zur Datierung der Diversifizierungsereignisse im Petasites-Clade diente eine molekulare Uhr, und die Evolution von Miniaturisierung wurde rekonstruiert. Mit H. alpina und H. discolor wurde ein vergleichendes Kulturexperiment durchgeführt.rnErgebnisse: Miniaturisierung entstand mehrere Male unabhängig voneinander in den arktisch-alpinen Vertretern des Petasites-Clade, aber nicht alle arktisch-alpinen Arten sind klein. Das Alter der arktisch-alpinen Arten deutet darauf hin, dass diese Taxa ihren Ursprung in der arkto-tertiären Flora haben. In Soldanella sind reduzierte Blütenmorphologie sowie Kleinwüchsigkeit der beiden alpinen Arten zweimal parallel entstanden. Homogyne alpina und H. discolor zeigen keine edaphischen Unterschiede hinsichtlich des Keimverhaltens, aber in Kultur zeigt sich, dass die Präferenz von H. discolor für Kalk wahrscheinlich genetisch fixiert ist.rnSchlussfolgerungen: Miniaturisierung von Pflanzen in größerer Höhe und höherer geographischer Breite kann in der Regel beobachtet werden. Allerdings kann die Evolution arktisch-alpiner Arten auch durch Faktoren wie Nährstoffverfügbarkeit, Konkurrenz und Störung beeinflusst werden, die dem Effekt der Temperatur entgegenwirken, so dass nicht alle Pflanzen in arktisch-alpinen Habitaten klein sind. Blütenmorphologische Reduktion in Soldanella kann als Anpassung an einen höheren Grad an Selbstbestäubung interpretiert werden, um eine geringere Bestäuberaktivität im alpinen Lebensraum zu kompensieren.

Im Rahmen dieser Arbeit wurde ein biologisches Verfahren zur Reduzierung des Methanschlupfes in Gasaufbereitungsanlagen entwickelt. Der Methanschlupf entsteht, wenn das in Biogasanlagen produzierte Biogas auf normierte Erdgasqualität aufgereinigt wird, welches notwendig ist, um es in das bestehende Erdgasnetz einleiten zu können. Bei dieser Aufreinigung wird aus dem Biogas auch ein Teil des Methans mit ausgewaschen und gelangt mit dem Abgas der Gasaufbereitungsanlage in die Umwelt. Bisher wird dieses methanhaltige Abgas verbrannt, da eine Freisetzung des starken Treibhausgases Methan durch das Erneuerbare-Energien-Gesetz untersagt ist. Dies reduziert die ökologische Bilanz und setzt die Wirtschaftlichkeit der gesamten Biogasanlage herab. rnUm das Methan mit Hilfe eines biologischen Verfahrens zu entfernen, wurden zunächst methanoxidierende Bakterien (MOB) aus verschiedenen Habitaten isoliert, darunter auch erstmalig aus Termiten. Der Nachweis erfolgte durch (quantitative) Polymerase-Kettenreaktion und Fluoreszenz-in-situ-Hybridisierung anhand spezifischer Primer bzw. Sonden für das Gen der partikulären Methanmonoxygenase, ein MOB kennzeichnendes Enzym. Ihr Titer wurde durch qPCR auf 10^2 - 10^3 MOB pro Termitendarm durch qPCR bestimmt. Mit Hilfe einer 16S rDNA Sequenzierung, der (n)SAPD-PCR, der Bestimmung der zellulären Fettsäurezusammensetzung sowie MALDI-TOF-MS-Analysen konnten die Termitenisolate der Gattung Methylocystis zugeordnet werden. Die fehlende Artzuweisung spricht jedoch für die Isolierung einer neuen Art. rnFür den Einsatz der Isolate in Gasaufbereitungsanlagen wurde in Zusammenarbeit mit dem Prüf- und Forschungsinstitut in Pirmasens ein Reaktor im Technikumsmaßstab entwickelt und konstruiert. Der Reaktor wurde mit synthetischen Aufwuchskörper befüllt, diese mit einem neu gewonnenen potenten Termitenisolat besiedelt und der methanhaltige Abgasstrom der Gasaufbereitungsanlage darüber geleitet. Es wurde eine Reduktion des Methans um 68 % innerhalb von 30 Stunden erzielt. Medienoptimierungen wiesen das Potential auf, diesen Verbrauch um das bis zu 4-fache weiter zu steigern. Da durch die Oxidation des Methans im Abgasstrom der Gasaufbereitungsanlage Zellmasse und Polyhydroxybuttersäure (PHB) aufgebaut wurde, können diese als Substrat zurück in die Biogasanlagen geleitet werden und die Wirtschaftlichkeit weiter verbessern. Die Wirksamkeit des in diesem Projekt entwickelten Verfahrens wurde somit eindeutig demonstriert.

The purpose of our study was to assess whether prairie voles find alcohol rewarding. Prairie voles have recently become a species of interest for alcohol studies, which have traditionally used other rodent model species including several different strains of mice and rats. The prairie vole is one of only two known rodent species that readily administers high levels of unsweetened alcohol, implicating it as a potentially effective animal model for studying alcohol abuse. However, voluntary consumption does not necessarily imply that prairie voles find it rewarding. Therefore the purpose of our study was to investigate if alcohol has rewarding properties for prairie voles using three different approaches: place conditioning, flavor conditioning, and immunohistochemistry. Furthermore, we sought to characterize their reward profile and compare it to other commonly used rodent models ¿ C57BL/6 mice, DBA/2J mice, and Sprague-Dawley rats. Place and flavor conditioning are behavioral methods that rely on the learned association between a stimulus and the effects of a drug; the drug of interest in these studies is alcohol. To assess whether prairie voles will demonstrate a conditioned preference for alcohol-paired stimuli, seven place conditioning studies were run that investigated a range of different doses, individual conditioning session durations, and trial durations. Video analysis revealed no difference in the amount of time spent on the alcohol-paired floor, suggesting no conditioned place preference for alcohol. Two flavor conditioning tests were conducted to assess whether voles would demonstrate a preference for an alcohol-paired flavored saccharin solution. Voles demonstrated reduced consumption of the alcohol-paired flavored saccharin solution, regardless of dose or flavor, when alcohol administration occurred after conditioning sessions (p=<0.001). When alcohol was administered before conditioning sessions, no difference in consumption of the alcohol-paired and saline-paired flavored saccharin solutions was seen (p=0.545). Previous studies that have documented similar behavior have hypothesized that this is an example of an anticipatory contrast effect. This theory proposes that prairie voles reduce their intake of a hedonic solution (flavored saccharin solution) in anticipation of later drug administration (alcohol). However, conditioning-based behavioral methods of studying alcohol reward are highly sensitive to the parameters of the conditioned stimulus, thus it is possible that voles will not show preference for alcohol-related stimuli, even if they do find alcohol rewarding. Immunohistochemical analysis supplemented this behavioral data by allowing us to identify specific neural regions that were directly activated in response to the acute administration of alcohol. No difference in the number of activated c-Fos neurons in the Nucleus Accumbens (NAc) core or shell was seen (p=0.3364; p=0.6698) in animals that received an acute injection of alcohol or saline. There was a significant increase in the number of activated c-Fos neurons in the Paraventricular Nucleus of the Hypothalamus (PVN) in alcohol-treated animals compared to saline-treated animals (p=0.0034). There was no difference in the pixel count of activated c-Fos neurons or in the % area activated in the Arcuate Nucleus between alcohol and saline-treated animals (p=0.4523; p=0.3304). In conclusion, the place conditioning studies that were conducted in this thesis suggest that prairie voles do not demonstrate preference or aversion towards alcohol-paired stimuli. The flavor conditioning studies suggest that prairie voles do not demonstrate aversion but rather avoidance of the alcohol-paired flavor in anticipation of future alcohol administration. The preliminary immunohistochemical data collected is inconclusive but cannot rule out the possibility of neuronal activation patterns indicative of reward. Vaccination in HIV-infected children is often less effective than in healthy children. The goal of this study was to assess vaccine responses to hepatitis A virus (HAV) in HIV-infected children. Children of the Swiss Mother and Child HIV Cohort Study (MoCHiV) were enrolled prospectively. Recommendations for initial, catch-up, and additional HAV immunizations were based upon baseline antibody concentrations and vaccine history. HAV IgG was assessed by enzyme-linked immunosorbent assay (ELISA) with a protective cutoff value defined as ≥10 mIU/ml. Eighty-seven patients were included (median age, 11 years; range, 3.4 to 21.2 years). Forty-two patients were seropositive (48.3%) for HAV. Among 45 (51.7%) seronegative patients, 36 had not received any HAV vaccine dose and were considered naïve. Vaccine responses were assessed after the first dose in 29/35 naïve patients and after the second dose in 33/39 children (25 initially naïve patients, 4 seronegative patients, and 4 seropositive patients that had already received 1 dose of vaccine). Seroconversion was 86% after 1 dose and 97% after 2 doses, with a geometric mean concentration of 962 mIU/ml after the second dose. A baseline CD4(+) T cell count below 750 cells/μl significantly reduced the post-2nd-dose response (P = 0.005). Despite a high rate of seroconversion, patients with CD4(+) T cell counts of <750/μl had lower anti-HAV antibody concentrations. This may translate into a shorter protection time. Hence, monitoring humoral immunity may be necessary to provide supplementary doses as needed.

Hepatitis A virus (HAV) exposure in unprotected adults may cause severe and serious symptoms, with risk of both morbidity and mortality increasing with age. As seroprevalence of HAV is low in industrialised countries, and an increasing number of people, with an increasing median age, travel from areas of low HAV endemicity to high endemicity, pre-travel vaccination is warranted. Vaccination of the elderly against HAV, however, may be associated with reduced seroprotection, since the immune response decreases with age. Studies with monovalent hepatitis A vaccine or combined hepatitis A and B vaccine show good efficacy in adults in general. Few studies have assessed the immune response in older adults. The only prospective study with monovalent hepatitis A vaccine in the elderly showed a reduced seroprotection of approximately 65% after a single primary dose in subjects over the age of 50 years, while seroprotection was 98% in this age group after receiving a booster dose. The only prospective study with combined hepatitis A and B vaccine in younger subjects or older than 40 years showed similar seroprotection (99-100%) against HAV compared to a monovalent vaccine after receiving three doses. As data on seroprotection for HAV in the elderly are limited, further studies are needed to elucidate how optimal protection in the elderly can be achieved. In the mean time, based on the available data, the suggestion is made to screen elderly travellers to areas endemic for HAV for the presence of naturally acquired immunity, and, if found susceptible, be immunised well in advance of their trip, to allow time for post-vaccination antibody testing and/or administration of a second dose of the vaccine.

BACKGROUND Timing is critical for efficient hepatitis A vaccination in high endemic areas as high levels of maternal IgG antibodies against the hepatitis A virus (HAV) present in the first year of life may impede the vaccine response. OBJECTIVES To describe the kinetics of the decline of anti-HAV maternal antibodies, and to estimate the time of complete loss of maternal antibodies in infants in León, Nicaragua, a region in which almost all mothers are anti-HAV seropositive. METHODS We collected cord blood samples from 99 healthy newborns together with 49 corresponding maternal blood samples, as well as further blood samples at 2 and 7 months of age. Anti-HAV IgG antibody levels were measured by enzyme immunoassay (EIA). We predicted the time when antibodies would fall below 10 mIU/ml, the presumed lowest level of seroprotection. RESULTS Seroprevalence was 100% at birth (GMC 8392 mIU/ml); maternal and cord blood antibody concentrations were similar. The maternal antibody levels of the infants decreased exponentially with age and the half-life of the maternal antibody was estimated to be 40 days. The relationship between the antibody concentration at birth and time until full waning was described as: critical age (months)=3.355+1.969 × log(10)(Ab-level at birth). The survival model estimated that loss of passive immunity will have occurred in 95% of infants by the age of 13.2 months. CONCLUSIONS Complete waning of maternal anti-HAV antibodies may take until early in the second year of life. The here-derived formula relating maternal or cord blood antibody concentrations to the age at which passive immunity is lost may be used to determine the optimal age of childhood HAV vaccination.

BACKGROUND AND OBJECTIVES Nicaragua is highly endemic for hepatitis A. We aimed to provide an estimate of the change in the age-specific risk of hepatitis A virus (HAV) infection based on serological data from cross-sectional and longitudinal samples collected in León, Nicaragua, in 1995/96 (n = 979) and 2003 (n = 494). METHODS The observed age-specific prevalence of anti-HAV antibodies was correlated to the age-specific risk of infection by calculating the probability of freedom from infection at a specific age. RESULTS The proportion of seropositive children aged 1.5 to 6 years was 42% in 2003 compared to 67% in 1995/96. Estimated annual risk of infection for a 3-year old child was 30% (95% CI: 27.0%, 33.1%) in 1995 and 15.5% (95% CI: 12.4%, 19.0%) in 2003. There was good agreement between estimates based on cross-sectional and longitudinal data. The age-specific geometric mean of the quantified anti-HAV antibody levels assessed in 2003 was highest at age 4 and decreased steadily up to age 40. CONCLUSIONS The substantially lower risk of HAV infection in 2003 than in 1995 for young children indicates a beginning transition from high to intermediate endemicity in León, Nicaragua. Consecutive age-stratified serosurveys are useful to assess changes in risk of infection following public health interventions. The decreasing age-specific GMC of anti-HAV antibodies during adulthood in a country with endemic HAV indirectly suggests that ongoing HAV exposure in the community has marginal boosting effect on antibody levels once protective immunity has been established by natural infection.

Several important fundamental and applied problems require a quantification of slow rates of groundwater flow. To resolve these problems helium appears to be a promising tracer. In this contribution we discuss a new approach, which gives the helium inventory in a rock – pore water system by using the relevant mineral record, i.e., without extraction and investigation of the porewater samples. Some U- and Th-poor minerals such as quartz (quartz separates from Permo-Carboniferous Formation, sandstone–shale interlayering, Molasses Basin, Northern Switzerland, hereafter PCF, are used in this study) contain excessive helium having migrated into their internal helium-accessible volume (HAV) from the surrounding porewater [I.N. Tolstikhin, B.E. Lehmann, H.H. Loosli, A. Gautschi, Helium and argon isotopes in rocks, minerals and related groundwaters: a case study in Northern Switzerland, Geochim. Cosmochim. Acta 60 (1996) 1497–1514]. These volumes are estimated by using helium as a nano-size penetrating tool, i.e., by saturation of the minerals with helium under controlled pressure–temperature conditions and subsequent measurements of the helium-saturated concentrations. In the quartz separates HAV/total volume ratios vary from 0.017% to 0.16%; along with the measured initial (unsaturated) He concentration the HAV gives the internal helium pressure, the mean value obtained for 7 samples (25 sample aliquots) is P=0.45F0.15 atm (1 r). The product of helium pressure and solubility (7.35_10_3 cc STP He/cc H2O for the temperature and salinity of PCF aquifers reported in [F.J. Pearson, W. Balderer, H.H. Loosli, B.E. Lehmann, A. Matter, T. Peters, H. Schmassmann, A. Gautschi, Applied Isotope Hydrogeology–A Case Study in Northern Switzerland, Elsevier Amsterdam, 1991, 439 pp.]) is the mineral-derived He concentration in the respective porewater, CPW=0.0035F0.0017 cc He/cc H2O. This value is in full accord with measured He concentrations in PCF aquifers, CPCF, varying from 0.0045 to 0.0016 cc He/cc H2O. This agreement validates the proposed approach and also shows that the mineral–porewater helium–concentration equilibrium has been established. Indeed, estimates of the He-migration rates through our quartz samples show that in ~6000 years the internal pressure should equilibrate with He-concentration in related porewater of PCF, and this time interval is short compared to independent estimates [I.N. Tolstikhin, B.E. Lehmann, H.H. Loosli, A. Gautschi, Helium and argon isotopes in rocks, minerals and related groundwaters: a case study in Northern Switzerland, Geochim. Cosmochim. Acta 60 (1996) 1497–1514]. The helium inventory in the rock–porewater assemblage shows that helium abundance in pore waters is indeed important. In shale samples (with ~15% porosity) porewaters contain more helium than the host minerals altogether. Porewater heliumconcentration profiles, available from the mineral record, along with helium production rates are input parameters allowing model(s) of helium migration through a hydrological structure to be developed. Quite high helium concentrations in PCF porewaters imply slow removal mechanisms, which will be discussed elsewhere.

Hepatitis B virus (HBV) is a significant cause of liver diseases and related complications worldwide. Both injecting and non-injecting drug users are at increased risk of contracting HBV infection. Scientific evidence suggests that drug users have subnormal response to HBV vaccination and the seroprotection rates are lower than that in the general population; potentially due to vaccine factors, host factors, or both. The purpose of this systematic review is to examine the rates of seroprotection following HBV vaccination in drug using populations and to conduct a meta-analysis to identify the factors associated with varying seroprotection rates. Seroprotection is defined as developing an anti-HBs antibody level of ≥ 10 mIU/ml after receiving the HBV vaccine. Original research articles were searched using online databases and reference lists of shortlisted articles. HBV vaccine intervention studies reporting seroprotection rates in drug users and published in English language during or after 1989 were eligible. Out of 235 citations reviewed, 11 studies were included in this review. The reported seroprotection rates ranged from 54.5 – 97.1%. Combination vaccine (HAV and HBV) (Risk ratio 12.91, 95% CI 2.98-55.86, p = 0.003), measurement of anti-HBs with microparticle immunoassay (Risk ratio 3.46, 95% CI 1.11-10.81, p = 0.035) and anti-HBs antibody measurement at 2 months after the last HBV vaccine dose (RR 4.11, 95% CI 1.55-10.89, p = 0.009) were significantly associated with higher seroprotection rates. Although statistically nonsignificant, the variables mean age>30 years, higher prevalence of anti-HBc antibody and anti-HIV antibody in the sample population, and current drug use (not in drug rehabilitation treatment) were strongly associated with decreased seroprotection rates. Proportion of injecting drug users, vaccine dose and accelerated vaccine schedule were not predictors of heterogeneity across studies. Studies examined in this review were significantly heterogeneous (Q = 180.850, p = 0.000) and factors identified should be considered when comparing immune response across studies. The combination vaccine showed promising results; however, its effectiveness compared to standard HBV vaccine needs to be examined systematically. Immune response in DUs can possibly be improved by the use of bivalent vaccines, booster doses, and improving vaccine completion rates through integrated public programs and incentives.^

Die Bodentiergemeinschaft des Wattenmeeres ist von Frühjahr bis Herbst eines jeden Jahres durch extrem hohe Dichten von Jungtieren charakterisiert. Die Kenntnisse über die Ansiedlung von fplanktischen Larven im Wattenmeer, sowie die Dynamik postlarvaler Stadien sind aufgrund der üblicherweise verwendeten, großen Siebmaschenweiten gering. Gerade aber diesen Altersstadien kommt möglicherweise eine besondere Stellung im Energiefluß des Wattenmeeres zu. An 5 Stationen (von NWL bis HWL, B1-B5) im Rückseitenwatt der ostfriesischen Insel Borkum wurden 1986 Ansiedlung, räumliche Verteilung, Wachstum, Mortalität und Produktion der Altersklasse 0 von Macoma balthica, Mya arenaria und Cerastoderma edule untersucht. Um die Ansiedlung der planktotrophen Larven dieser Arten zu beschreiben, wurden ihre Dichten in Plankton und Bodenproben miteinander verglichen. Die Untersuchungen zur Dynamik der benthischen Stadien wurden mit zwei in der Probenfläche und der Siebmaschenweite unterschiedlichen Probenserien durchgeführt. Die Drift postlarvaler Stadien wurde durch bodennahe Planktonfänge innerhalb des Eulitorals nachgewiesen. Parallel zu den Untersuchungen an der Endofauna wurden das Vorkommen und die Größe epibenthischer Räuber im Untersuchungsgebiet erfaßt. Die Hauptansiedlung von M. balthica- und M. arenaria-Larven erfolgte nahezu gleichzeitig Ende Mai/Anfang Juni. Die meisten Larven beider Arten gingen an der prielnächsten (tiefsten) Station (B1) zum Bodenleben über, gefolgt von der nächst höher gelegenen Station B2. Während frühe Bodenstadien von M. arenaria nicht im oberen Bereich des Watts (B3,B4) gefunden wurden, ist eine geringfügige Erstansiedlung von M. balthica in diesem Gebiet nicht auszuschließen. Ein die Ansiedlung limitierender Einfluß der relativ dichten Mya arenaria-Siedlung an den Stationen B1 und B2 sowie der Alttiere von M. balthica konnte nicht festgestellt werden. Die Ähnlichkeit des Ansiedlungsprozesses bei beiden Arten, die sich im Zahlenverhältnis Larvenangebot zu Anzahl der ersten Bodenstadien widerspiegelt, kann ein Hinweis auf eine überwiegend passive Ansiedlung der Larven am Boden sein. Der Ort der Hauptansiedlung von C. edule wurde durch den Transekt nicht erfaßt. Die Station B2 war zwar durch ein Herzmuschelfeld charakterisiert, dieses war aber nach zwei Eiswintern nahezu vollständig eliminiert. Der Abundanz der planktischen Larven zufolge war der Hauptansiedlungszeitraum ebenfalls Ende Mai/Anfang Juni. Zu dieser Zeit wurden nur vereinzelt frühe Bodenstadien an den Stationen B1 und B2 gefunden, keine an den Stationen B3 und B4. Während die frühen postlarvalen Stadien von M. arenaria überwiegend am Ort der Ansiedlung blieben, verbreiteten sich die von M. balthica bis in den oberen Bereich des Untersuchungsgebietes (B3-B5). Analog zu der Besiedlung dieser Gebiete durch postlarvale M. balthica wurde die im Verlauf des Untersuchungsjahres stattfindende Kolonisierung der Station B1 durch C. edule ebenfalls postlarvalem Transport zugeschrieben. Demzufolge spielt bei beiden Muschelarten postlarvaler Transport eine wichtige Rolle bei der Besiedlung von Habitaten. Planktonfänge innerhalb der bodennahen Wasserschicht bestätigten, daß im Untersuchungsgebiet M. balthica die am stärksten verdriftende Muschelart war, gefolgt von C. edule. Mortalität, Wachstum, mittlere Biomasse, Produktion und P/B-Verhältnis wurden für M. balthica an den Stationen B1, B3 und B4 sowie für M. arenaria an der Station B1 bestimmt. Wachstum und damit auch Produktion beider Arten erwiesen sich hier - wie an den höher gelegen Stationen (nur M. balthica) - als durch größenselektiven Feinddruck beeinflußt. Der Effekt postlarvalen Transports auf Wachstum wird diskutiert. Übergreifend über die auf Artebene diskutierten Ergebnisse wird die Bedeutung der Dispersion postlarvaler Stadien und die Wirkung epibenthischen Feinddrucks im Wattenmeer erörtert. Der Vergleich postlarvalen Transportes mit der Dispersion planktischer Larvenstadien, der Dispersion von Meiofauna und der Mobilität adulter Stadien der Makrofauna verdeutlicht, daß es sich hierbei um eine Strategie handeln kann, innerhalb eines unvorhersagbaren Biotops freiwerdende Ressourcen zu nutzen und dadurch Konkurrenz zu vermeiden. Es wird die Hypothese aufgestellt, daß Initialansiedlung und Immigration einerseits sowie Feinddruck und Emigration andererseits einen Regelkreis darstellen, der in verschiedenen Teilbereichen des Watts mit unterschiedlicher Geschwindigkeit abläuft.

The genetic study of RNA viruses is greatly facilitated by the availability of infectious cDNA clones. However, their construction has often been difficult. While exploring ways to simplify the construction of infectious clones, we have successfully modified and applied the newly described technique of "long PCR" to the synthesis of a full-length DNA amplicon from the RNA of a cytopathogenic mutant (HM 175/24a) of the hepatitis A virus (HAV). Primers were synthesized to match the two extremities of the HAV genome. The antisense primer, homologous to the 3' end, was used in both the reverse transcription (RT) and the PCR steps. With these primers we reproducibly obtained a full-length amplicon of approximately 7.5 kb. Further, since we engineered a T7 promoter in the sense primer, RNA could be transcribed directly from the amplicon with T7 RNA polymerase. Following transfection of cultured fetal rhesus kidney cells with the transcription mixture containing both the HAV cDNA and the transcribed RNA, replicating HAV was detected by immunofluorescence microscopy and, following passage to other cell cultures, by focus formation. The recovered virus displayed the cytopathic effect and large plaque phenotype typical of the original virus; this result highlights the fidelity of the modified long reverse transcription-PCR procedure and demonstrates the potential of this method for providing cDNAs of viral genomes and simplifying the construction of infectious clones.

Com o objetivo de aumentar o lucro de plantas químicas, a Otimização em Tempo Real (RTO) é uma ferramenta que busca determinar as condições ótimas operacionais do processo em estado estacionário, respeitando as restrições operacionais estabelecidas. Neste trabalho foi realizada a implementação prática de um ciclo RTO em um processo de destilação por recompressão de vapor (VRD), propileno-propano, da Refinaria de Paulínia (Petrobras S.A.), a partir de dados históricos da planta. Foram consideradas as principais etapas de um ciclo clássico de RTO: identificação de estado estacionário, reconciliação de dados, estimação de parâmetros e otimização econômica. Essa unidade foi modelada, simulada e otimizada em EMSO (Environment for Modeling, Simulation and Optimization), um simulador de processos orientado a equações desenvolvido no Brasil. Foram analisados e comparados dois métodos de identificação de estado estacionário, um baseado no teste estatístico F e outro baseado em wavelets. Ambos os métodos tiveram resultados semelhantes e mostraram-se capazes de identificar os estados estacionários de forma satisfatória, embora seja necessário o ajuste de parâmetros na sua implementação. Foram identificados alguns pontos estacionários para serem submetidos ao ciclo RTO e foi possível verificar a importância de partir de um estado estacionário para a continuidade do ciclo, já que essa é uma premissa do método. A partir dos pontos analisados, os resultados deste estudo mostram que o RTO é capaz de aumentar o ganho econômico entre 2,5-24%, dependendo das condições iniciais consideradas, o que pode representar ganhos de até 18 milhões de dólares por ano. Além disso, para essa unidade, verificou-se que o compressor é um equipamento limitante no aumento de ganho econômico do processo.

The increasing economic competition drives the industry to implement tools that improve their processes efficiencies. The process automation is one of these tools, and the Real Time Optimization (RTO) is an automation methodology that considers economic aspects to update the process control in accordance with market prices and disturbances. Basically, RTO uses a steady-state phenomenological model to predict the process behavior, and then, optimizes an economic objective function subject to this model. Although largely implemented in industry, there is not a general agreement about the benefits of implementing RTO due to some limitations discussed in the present work: structural plant/model mismatch, identifiability issues and low frequency of set points update. Some alternative RTO approaches have been proposed in literature to handle the problem of structural plant/model mismatch. However, there is not a sensible comparison evaluating the scope and limitations of these RTO approaches under different aspects. For this reason, the classical two-step method is compared to more recently derivative-based methods (Modifier Adaptation, Integrated System Optimization and Parameter estimation, and Sufficient Conditions of Feasibility and Optimality) using a Monte Carlo methodology. The results of this comparison show that the classical RTO method is consistent, providing a model flexible enough to represent the process topology, a parameter estimation method appropriate to handle measurement noise characteristics and a method to improve the sample information quality. At each iteration, the RTO methodology updates some key parameter of the model, where it is possible to observe identifiability issues caused by lack of measurements and measurement noise, resulting in bad prediction ability. Therefore, four different parameter estimation approaches (Rotational Discrimination, Automatic Selection and Parameter estimation, Reparametrization via Differential Geometry and classical nonlinear Least Square) are evaluated with respect to their prediction accuracy, robustness and speed. The results show that the Rotational Discrimination method is the most suitable to be implemented in a RTO framework, since it requires less a priori information, it is simple to be implemented and avoid the overfitting caused by the Least Square method. The third RTO drawback discussed in the present thesis is the low frequency of set points update, this problem increases the period in which the process operates at suboptimum conditions. An alternative to handle this problem is proposed in this thesis, by integrating the classic RTO and Self-Optimizing control (SOC) using a new Model Predictive Control strategy. The new approach demonstrates that it is possible to reduce the problem of low frequency of set points updates, improving the economic performance. Finally, the practical aspects of the RTO implementation are carried out in an industrial case study, a Vapor Recompression Distillation (VRD) process located in Paulínea refinery from Petrobras. The conclusions of this study suggest that the model parameters are successfully estimated by the Rotational Discrimination method; the RTO is able to improve the process profit in about 3%, equivalent to 2 million dollars per year; and the integration of SOC and RTO may be an interesting control alternative for the VRD process.

Frutas e vegetais frescos, embora sejam essenciais para uma dieta equilibrada e nutritiva, estão frequentemente associados a surtos de origem alimentar, pois este tipo de produto é muitas vezes consumidos cru, sem qualquer processamento térmico que elimine ou reduza para níveis aceitáveis a carga microbiana patogénica. Desta forma, e para que o consumo deste tipo de produtos continue a aumentar, torna-se importante aumentar o controlo em termos de segurança alimentar em todas as fases de produção. O presente trabalho de investigação teve por objetivos estudar a ocorrência de cinco microrganismos patogénicos: Salmonella sp.; Escherichia coli; Staplylococcus aureus; Listeria monocytogenes; HAV (vírus da Hepatite A), em framboesas e alface à venda no mercado português, otimizando uma técnica simples e rápida para deteção desses microrganismos nesses produtos. Também neste trabalho foi feito um estudo sobre os hábitos de consumo de produtos minimamente processados, framboesas e alface através de um inquérito por questionário. A escolha dos produtos e microrganismos a pesquisar teve por base um levantamento de quais as contaminações e os hortofrutícolas mais afetados nos últimos anos. A investigação iniciou-se com a pesquisa do HAV em framboesas e alface, onde após uma revisão bibliográfica, foram testados quatro protocolos distintos. No entanto devido à complexidade das técnicas para pesquisa de vírus em alimentos, à falta de meios e controlos para o desenvolvimento das respetivas técnicas, este objetivo foi abandonado. Numa segunda fase foi otimizado um método de deteção (PCR) para pesquisa das bactérias patogénicas, igualmente em framboesas e alface. Após otimização das condições de PCR, foram testados dois métodos de preparação da amostra: um com um passo de rebentamento de células baseado na fervura; e outro onde apenas se recolheu algum volume de cultura de pré-enriquecimento, tendo-se usado a mesma diretamente para a reação de PCR. Concluiu-se que apenas o primeiro método é eficiente, mas apenas para bactérias Gram-negativas (Salmonella sp. e E. coli) uma vez que estas possuem uma parede celular fina, capaz de ser destruída pela fervura, ao contrário das Gram-positivas (S. aureus e L. monocytogenes). Das amostras de framboesas e alface à venda no mercado português analisadas, concluiu-se que, apesar do método ainda necessitar de alguns ajustes, apenas a amostra de alface fresca parece não apresentar qualquer contaminação com Salmonella sp. e E. coli. Para S. aureus e L. monocytogenes não foi possível obter conclusões definitivas quanto à sua presença nas amostras analisadas, já que vários testes de confirmação deverão ser repetidos. Relativamente aos resultados obtidos acerca do inquérito de consumo, tem-se que dos 186 inquiridos, a grande maioria consome produtos hortofrutícolas minimamente processados 1-3 vezes/semana, tendo, a grande maioria, o hábito de os lavar com água antes do seu consumo, prática esta que é aconselhada. Quanto ao consumo de alface, a frequência aumenta para 3-5 vezes/semana, sendo esta adquirida maioritariamente inteira fresca e em superfícies comerciais. Por fim o consumo de framboesas ainda é escasso, pois a grande maioria dos inquiridos alega consumir este produto esporadicamente, sendo estas adquiridas embaladas inteiras frescas em superfícies comerciais.
