The mammalian retromer protein complex, which consists of three proteins - Vps26, Vps29, and Vps35 - in association with members of the sorting nexin family of proteins, has been implicated in the trafficking of receptors and their ligands within the endosomal/lysosomal system of mammalian cells. A bioinformatic analysis of the mouse genome identified an additional transcribed paralog of the Vps26 retromer protein, which we termed Vps26B. No paralogs were identified for Vps29 and Vps35. Phylogenetic studies indicate that the two paralogs of Vps26 become evident after the evolution of the chordates. We propose that the chordate Vps26-like gene published previously be renamed Vps26A to differentiate it from Vps26B. As for Vps26A, biochemical characterization of Vps26B established that this novel 336 amino acid residue protein is a peripheral membrane protein. Vps26B co-precipitated with Vps35 from transfected cells and the direct interaction between these two proteins was confirmed by yeast 2-hybrid analysis, thereby establishing Vps26B as a subunit of the retromer complex. Within HeLa cells, Vps26B was found in the cytoplasm with low levels at the plasma membrane, while Vps26A was predominantly associated with endosomal membranes. Within A549 cells, both Vps26A and Vps26B co-localized with actin-rich lamellipodia at the cell surface. These structures also co-localized with Vps35. Total internal reflection fluorescence microscopy confirmed the association of Vps26B with the plasma membrane in a stable HEK293 cell line expressing cyan fluorescent protein (CFP)-Vps26B. Based on these observations, we propose that the mammalian retromer complex is located at both endosomes and the plasma membrane in some cell types.

The aim of this study was to investigate the effects of acute glyphosate (active ingredient) exposure on the oxidative stress biomarkers and antioxidant defenses of a hybrid surubim (Pseudoplatystoma sp). The fish were exposed to different herbicide concentrations for 96 h. The thiobarbituric acid-reactive substances (TBARS), protein carbonyls and antioxidant responses were verified. The 15 mg a.p L-1 of herbicide resulted in the death of 50% of the fish after 96 h. An increase in liver and muscle TBARS levels was observed when fish were exposed to the herbicide. The protein carbonyl content was also increased in the liver (4.5 mg a.p L-1 concentration) and brain (2.25 mg a.p L-1 concentration). The antioxidant activities decreased in the liver and brain after exposure to herbicide. Levels of ascorbic acid in the liver (2.25 mg a.p L-1 and 4.5 mg a.p L-1 concentrations) and brain (2.25 mg a.p L-1 concentration) were increased post-treatment. Levels of total thiols were increased in the liver and brain (2.25 mg L-1 and 7.5 mg a.p L-1, respectively). Glyphosate exposure, at the tested concentrations affects surubim health by promoting changes that can affect their survival in natural environment. Some parameters as TBARS and protein carbonyl could be early biomarkers for Roundup exposure in this fish species. (C) 2014 Elsevier Inc. All rights reserved.

This study addresses the questions of whether the frequency of generation and in vivo cross-reactivity of highly immunogenic tumor clones induced in a single parental murine fibrosarcoma cell line MCA-F is more closely related to the agent used to induce the Imm$\sp{+}$ clone or whether these characteristics are independent of the agents used. These questions were addressed by treating the parental tumor cell line MCA-F with UV-B radiation (UV-B), 1-methyl-3-nitro-1-nitrosoguanidine (MNNG), or 5-aza-2$\sp\prime$-deoxycytidine (5-azaCdR). The frequency of Imm$\sp{+}$ variant generation was similarly high for the three different agents, suggesting that the frequency of Imm$\sp{+}$ generation was related more closely to the cell line than to the inducing agent used. Cross-reactivity was tested with two Imm$\sp{+}$ clones from each treatment group in a modified immunoprotection assay that selectively engendered antivariant, but not antiparental immunity. Under these conditions each clone, except one, immunized against itself. The MNNG-induced clones engendered stronger antivariant immunity but a weaker variant cross-reactive immunity could also be detected.^ This study also characterized the lymphocyte populations responsible for antivariant and antiparental immunity in vivo. Using the local adoptive transfer assay (LATA) and antibody plus complement depletion of T-cell subsets, we showed that immunity induced by the Imm$\sp{+}$ variants against the parent MCA-F was transferred by the Thy1.2$\sp{+}$, L3T4a$\sp{+}$, Lyt2.1$\sp{-}$ (CD4$\sp{+}$) population, without an apparent contribution by Thy1.2$\sp{+}$, L3T4a$\sp{-}$, Lyt2.1$\sp{+}$ (CD8$\sp{+}$) cells. A role for Lyt2.1$\sp{+}$T lymphocytes in antivariant, but not antiparent immunity was supported by the results of LATA and CTL assays. Immunization with low numbers of viable Imm$\sp{+}$ cells, or with high numbers of non viable Imm$\sp{+}$ cells engendered only antivariant immunity without parental cross-protection. The associative recognition of parental antigens and variant neoantigens resulting in strong antiparent immunity was investigated using somatic cells hybrids of Imm$\sp{+}$ variants of MCA-F and an antigenically distinct tumor MCA-D. An unexpected result of these latter experiments was the expression of a unique tumor-specific antigen by the hybrid cells. These studies demonstrate that the parental tumor-specific antigen and the variant neoantigen must be coexpressed on the cell surface to engender parental cross-protective immunity. A considerable portion of Brazil's commercial eucalypt plantations is located in areas Subjected to periods of water deficit and grown in soils with low natural fertility, particularly poor In potassium. Potassium is influential in controlling water relations of plants. The objective of this study was to verify the influence of potassium fertilization and soil water potential (psi(w)) oil the dry matter production and oil water relations Of eucalypt seedlings grown under greenhouse conditions. the experimental units were arranged in 4x4x2 randomized blocks factorial design, as follow: four species of Eucalyptus (Eucalyptus grandis, Eucalyptus urophylla, Eucalyptus camaldulensis and hybrid Eucalyptus grandis x Eucalyptus urophylla), four dosages of K (0, 50, 100 and 200 mg dm(-3)) and two soil water potentials (-0.01 M Pa and -0.1 M Pa). Plastic containers with 15 cm diameter and 18 cm height, with Styrofoam base, containing 3.0 dm(3) of soil and two plants per container were used. Soil water potential was kept at -0.01 MPa for 40 days after seeding. Afterward, the experimental units were divided into two groups: in one group the potential was kept at 0.01 MPa, and in the other one, at -0.10 MPa. Sol I water potential was control led gravimetrically twice a day with water replacement until the desired potential was reestablished. A week before harvesting, the leaf water potential (psi), the photosynthetic rate (A), the stomatal conductance (gs) and the transpiration rate were evaluated. The last week before harvesting, the mass of the containers was recorded daily before watering to determine the consumption of water by the plants. After harvesting, total dry matter and leaf area were evaluated. the data were Submitted to analysis of variance, to Tukey's tests and regression analyses. The application of K influenced A, gs and the transpiration rate. Plants deficient in K showed lower A and higher Us and transpiration rates. There were no statistical differences in A, gs and transpiration rates ill plants with and Without water deficit. The addition of K reduced the consumption of water per unit of leaf area and, in general, plants submitted to water deficit presented a lower consumption of water.

Procurou-se conhecer a prevalência de enteroparasitoses na população urbana do 2.° subdistrito de Botucatu, SP (Brasil) através de exames coprológicos realizados pelos métodos de FAUST, HOFFMAN e processo de tamização. A prevalência de enteroparasitoses foi relacionada com atributos da população, tais como sexo, idade, cor e com fatores ligados ao meio ambiente. O processo de amostragem empregado foi o casual simples em duplo estágio, sendo o quarteirão a unidade primária do primeiro estágio e o domicílio a unidade do segundo estágio. Os resultados mostraram que 53,76% das 895 pessoas amostradas apresentavam-se infestadas por uma ou mais espécies de parasitas intestinais. As prevalências foram as seguintes: Ancylostomidae, 17,54%; T. trichiurus, 13,63%; A. lumbricoides, 10,69%; S. stercoralis, 6,03%; E. vermicularis, 3,69%; H. nana, 1,79%; Taenia sp, 1,22%; S. mansoni, 0,22%; E. coli, 15,53%; G. lamblia, 14,07%; E. nana, 2,35%; I. bütschlii, 1,01% e E. histolytica, 0,22%.

Estudo descritivo, feito por meio de dados oficiais, dos acidentes de motocicleta com vítimas (3.390), incluindo atropelamentos, ocorridos no Município de São Paulo, Brasil, em 1982, e que tiveram como conseqüência 4.480 vítimas, das quais, 166 faleceram dentro do período de até 180 dias após o evento. O coeficiente de mortalidade foi de aproximadamente 2/100.000 habitantes e a relação coeficiente masculino/feminino foi de 6:1. Na natureza das lesões verificou-se que os diagnósticos mais freqüentes foram as fraturas de crânio (27,96%), os traumatismos internos de tórax e de abdome (14,52%), e as fraturas dos membros inferiores (14,25%). Os óbitos no momento do acidente e nas primeiras 24 h perfizeram 62,35% do total. A morte ocorreu mais precocemente entre os motociclistas e passageiros do que entre os pedestres.

Human infections caused by a hantavirus were reported in different regions of the State of São Paulo (SP), Brazil during the first six months of 1998. Two cases of fatal pulmonary syndrome occurred in May of 1998 in the City of Guariba, located in the Northeastern Region of SP. Both patients worked in a corn storage barn infested by rodents. These patients, after 2 or 3 days of non-specific febrile illness, developed a severe interstitial pneumonia spreading widely in both lungs, causing respiratory failure and death. At autopsy both patients showed lung interstitial edema with immunoblast-like mononuclear cell infiltrates, consistent with a viral etiology. Hantavirus infection was diagnosed by ELISA in both cases and by RT-PCR in one of the patients. Aspects of the clinical presentation, physiopathology and differential diagnosis of Hantavirus Pulmonary Syndrome are discussed.

O presente trabalho mostrou freqüências diferentes de parasitoses intestinais (giardíase e helmintíases) entre crianças de pré-escolas e de escolas de 1 e 2 Graus, públicas e privadas. Pesquisou-se 199 indivíduos, sendo 96 pertencentes às instituições públicas e 103 pertencentes às instituições particulares. Nas instituições públicas as freqüências de giardíase foram maiores que nas instituições privadas. Para as helmintíases, as freqüências foram semelhantes entre as creches, porém maiores na escola pública em relação à particular. Indicadores do nível sócio-econômico, de escolaridade, de saneamento básico, como determinantes de parasitose intestinal, evidenciaram que as maiores freqüências de parasitoses ocorreram tanto entre as crianças de baixa renda quanto entre as que tinham pais com nível de escolaridade mais baixo. Também a origem das hortaliças foi um fator significativo para determinação destas parasitoses.

The Murcott tangor represent 20% of the tangerines trees in São Paulo State being the second more grown. Their fruits have good acceptance in the market cause of the good characteristics presented as: size, attractive internal and external color, transport resistance, high juice rate and industry potential. It is necessary to study the behavior of others varieties, in order to amplify the diversity of tangerine industry, which show suitable characteristics to the fresh fruit market and that make possible different harvest season. Many tangerine varieties, selected from the Citrus Germplasm Bank of the do Centro Avançado de Pesquisa Tecnológica do Agronegócio de Citros "Sylvio Moreira"/IAC, belong to trials carried out in 15 places in São Paulo State. The Capão Bonito area, south-west of the state, is one of this places where the Nova tangelo, the Ortanique and the Murcott tangors are showing quite good results about their fruit qualities. This paper had as an objective to compare the fruit characteristics of the Nova tangelo, the Murcott and the Ortanique tangors grafted on two rootstocks: Rangpur lime and Cleopatra mandarin. Accordingly to the gotten results, is possible to conclude that Nova and Ortanique had shown weight, width, fruit shape and juice percent, similar to the Murcott. In compliance with the harvest season, the Nova present suitable conditions to fresh fruit market in May and June. By the other hand the Murcott fruits can be harvested in July to August and the Ortanique in August to September. For this reason, is possible extend the harvest season of this mandarin-like, from two to five month, occurring inclusive in a period out of the crop at the north hemisphere.

El comportamiento agroforestal de 10 genotipos de J. regia y dos híbridos comerciales de Juglans, Ng23xRa y Mj209xRa, se ha evaluado desde 1995 a 2000 en dos localidades: Massanes (Girona) y Prades (Tarragona). Se establecen tres grupos de genotipos diferenciados de acuerdo a su tasa de crecimiento en altura: 1) crecimiento medio superior a 60 cm/año, que corresponde a los dos híbridos; 2) crecimiento medio de unos 50 cm/año, que corresponde a las progenies de J. regia con brotaciones anteriores al 20 de abril; y 3) crecimiento medio inferior a los 40 cm/año, que corresponde a los genotipos de J. regia con brotación posterior al 20 de abril. La evolución de las progenies en altura y diámetro está significativamente ligada a la localidad. El orden en la clasificación de las progenies por su crecimiento, en altura y en grosor, es prácticamente idéntico en ambas localidades. El diámetro a 2,50 m, al sexto período vegetativo, presenta una dependencia significativa de localidad y genotipo: en Massanes se alcanza un mayor crecimiento que en Prades y el diámetro a 2,50 m de los híbridos es el doble que el del mejor J. regia. La dominancia y la rectitud dependen de los genotipos. Sólo dos J. regia tienen una aptitud forestal aceptable, concretamente dos genotipos italianos, «Bleggiana» y «Boschi Maria Grazia». Los híbridos Ng23xRa y Mj209xRa presentan un mejor comportamiento agroforestal que los genotipos de J. regia estudiados: menor mortalidad en plantación, superiores crecimientos primario y secundario y buena dominancia apical y rectitud.

Previous studies have demonstrated that clinical pulpal pain can induce the expression of pro-inflammatory neuropeptides in the adjacent gingival crevice fluid (GCF). Vasoactive agents such as substance P (SP) are known to contribute to the inflammatory type of pain and are associated with increased blood flow. More recent animal studies have shown that application of capsaicin on alveolar mucosa provokes pain and neurogenic vasodilatation in the adjacent gingiva. Pain-associated inflammatory reactions may initiate expression of several pro- and anti-inflammatory mediators. Collagenase-2 (MMP-8) has been considered to be the major destructive protease, especially in the periodontitis-affected gingival crevice fluid (GCF). MMP-8 originates mostly from neutrophil leukocytes, the first line of defence cells that exist abundantly in GCF, especially in inflammation. With this background, we wished to clarify the spatial extensions and differences between tooth-pain stimulation and capsaicin-induced neurogenic vasodilatation in human gingiva. Experiments were carried out to study whether tooth stimulation and capsaicin stimulation of alveolar mucosa would induce changes in GCF MMP-8 levels and whether tooth stimulation would release neuropeptide SP in GCF. The experiments were carried out on healthy human volunteers. During the experiments, moderate and high intensity painful tooth stimulation was performed by a constant current tooth stimulator. Moderate tooth stimulation activates A-delta fibres, while high stimulation also activates C-fibres. Painful stimulation of the gingiva was achieved by topical application of capsaicin-moistened filter paper on the mucosal surface. Capsaicin is known to activate selectively nociceptive C-fibres of stimulated tissue. Pain-evoked vasoactive changes in gingivomucosal tissues were mapped by laser Doppler imaging (LDI), which is a sophisticated and non-invasive method for studying e.g. spatial and temporal characteristics of pain- and inflammation-evoked blood flow changes in gingivomucosal tissues. Pain-evoked release of MMP-8 in GCF samples was studied by immunofluorometric assay (IFMA) and Western immunoblotting. The SP levels in GCF were analysed by Enzyme immunoassay (EIA). During the experiments, subjective stimulus-evoked pain responses were determined by a visual analogue pain scale. Unilateral stimulation of alveolar mucosa and attached gingiva by capsaicin evoked a distinct neurogenic vasodilatation in the ipsilateral gingiva, which attenuated rapidly at the midline. Capsaicin stimulation of alveolar mucosa provoked clear inflammatory reactions. In contrast to capsaicin stimuli, tooth stimulation produced symmetrical vasodilatations bilaterally in the gingiva. The ipsilateral responses were significantly smaller during tooth stimulation than during capsaicin stimuli. The current finding – that tooth stimulation evokes bilateral vasodilatation while capsaicin stimulation of the gingiva mainly produces unilateral vasodilatation – emphasises the usefulness of LDI in clarifying spatial features of neurogenic vasoactive changes in the intra-oral tissues. Capsaicin stimulation of the alveolar mucosa induced significant elevations in MMP-8 levels and activation in GCF of the adjacent teeth. During the experiments, no marked changes occurred in MMP-8 levels in the GCF of distantly located teeth. Painful stimulation of the upper incisor provoked elevations in GCF MMP-8 and SP levels of the stimulated tooth. The GCF MMP-8 and SP levels of the non-stimulated teeth were not changed. These results suggest that capsaicin-induced inflammatory reactions in gingivomucosal tissues do not cross the midline in the anterior maxilla. The enhanced reaction found during stimulation of alveolar mucosa indicates that alveolar mucosa is more sensitive to chemical irritants than the attached gingiva. Analysis of these data suggests that capsaicin-evoked neurogenic inflammation in the gingiva can trigger the expression and activation of MMP-8 in GCF of the adjacent teeth. In this study, it is concluded that experimental tooth pain at C-fibre intensity can induce local elevations in MMP-8 and SP levels in GCF. Depending on the role of MMP-8 in inflammation, in addition to surrogated tissue destruction, the elevated MMP-8 in GCF may also reflect accelerated local defensive and anti-inflammatory reactions.

A solarização combinada com a prévia adição de material orgânico propicia controle efetivo de diversos fungos de solo que de outra forma são de difícil erradicação, permitindo, também, redução do tempo de tratamento. O objetivo do trabalho foi avaliar o controle do fungo Fusarium oxysporum f. sp. lycopersici Raça 2, por meio da incorporação da parte aérea de mandioca brava (Manihot utilisima) e mandioca mansa (Manihot dulcis) seguido de solarização do solo. Foi realizado experimento de campo, onde incorporou-se 3 Kg/m² da parte aérea de mandioca brava e mandioca mansa, separadamente. O experimento constou de seis tratamentos (solarização + mandioca brava; solarização + mandioca mansa; solo solarizado; solo; mandioca brava e mandioca mansa). A sobrevivência do fungo foi avaliado por meio das estruturas de resistência em meio semi-seletivo de Komada, aos 7, 14, 21 e 28 dias do início do experimento. As temperaturas do solo foram monitoradas através de um coletor de dados Tipo CR23X (Campbell Scientific) e a quantificação dos gases (CO2 e O2) foi feita pelo equipamento analisador de gases (Testo 325-1). Nos tratamentos solarizados, a temperatura do solo e aprisionamento de CO2 foram maiores em relação à testemunha (solo) e aos tratamentos apenas com a incorporação de material vegetal. A associação da incorporação de mandioca, tanto a "brava" quanto a "mansa", com a solarização, proporcionou a inativação de F. oxysporum f.sp. lycopersici Raça 2, podendo atuar no controle da doença murcha de fusarium causada por este patógeno. Os demais tratamentos foram inócuos, permitindo a sobrevivência do fungo.
