Twenty production blasts in two open pit mines were monitored, in rocks with medium to very high strength. Three different blasting agents (ANFO, watergel and emulsion blend) were used, with powder factors ranging between 0.88 and 1.45 kg/m3. Excavators were front loaders and rope shovels. Mechanical properties of the rock, blasting characteristics and mucking rates were carefully measured. A model for the calculation of the productivity of excavators is developed thereof, in which the production rate results as a product of an ideal, maximum, productivity rate times an operating efficiency. The maximum rate is a function of the dipper capacity and the efficiency is a function of rock density, strength, and explosive energy concentration in the rock. The model is statistically significant and explains up to 92 % of the variance of the production rate measurements.

The pendular motion of a giant censer (O Botafumeiro) that hangs in the transept of the cathedral of Santiago de Compostela, and is cyclically pumped by men who pull at the supporting rope, is analyzed. Maximum angular amplitude attainable, and number of cycles and time needed to attain it, are calculated; the results agree with observed values (~ 82°, ~ 17 cycles, ~ 80 seconds) to the few percent accuracy of both the analysis and the observations and parameter measurements. The energy gain in a pumping cycle is obtained for an arbitrary pumping procedure to two orders in the small fractional change of pendular length; the relevance of the ratio (characteristic radial acceleration during pumping)/g to the gain is discussed- Effects due to rope mass, air drag on both Censer and rope, and the fact that the Censer is not a point mass, are considered. If the pumping cycle is inverted once the maximum amplitude has been attained, the Censer could be swiftly brought to rest, avoiding the usual violent stop. Historically recorded accidents, rope shape, and the influence of relevant parameters on the motion are discussed.

Objective: To examine the association of breakfast consumption with objectively measured and self-reported physical activity, sedentary time and physical fitness. Design: The HELENA (Healthy Lifestyle in Europe by Nutrition in Adolescence) Cross-Sectional Study. Breakfast consumption was assessed by two non-consecutive 24 h recalls and by a ‘Food Choices and Preferences’ questionnaire. Physical activity, sedentary time and physical fitness components (cardiorespiratory fitness, muscular fitness and speed/agility) were measured and self-reported. Socio-economic status was assessed by questionnaire. Setting: Ten European cities. Subjects: Adolescents (n 2148; aged 12?5–17?5 years). Results: Breakfast consumption was not associated with measured or self-reported physical activity. However, 24 h recall breakfast consumption was related to measured sedentary time in males and females; although results were not confirmed when using other methods to assess breakfast patterns or sedentary time. Breakfast consumption was not related to muscular fitness and speed/agility in males and females. However, male breakfast consumers had higher cardiorespiratory fitness compared with occasional breakfast consumers and breakfast skippers, while no differences were observed in females. Overall, results were consistent using different methods to assess breakfast consumption or cardiorespiratory fitness (all P#0?005). In addition, both male and female breakfast skippers (assessed by 24 h recall) were less likely to have high measured cardiorespiratory fitness compared with breakfast consumers (OR50?33; 95% CI 0?18, 0?59 and OR50?56; 95 %CI 0?32, 0?98,respectively). Results persisted across methods. Conclusions: Skipping breakfast does not seem to be related to physical activity,sedentary time or muscular fitness and speed/agility as physical fitness components in European adolescents; yet it is associated with both measured and self-reported cardiorespiratory fitness, which extends previous findings.

RESUMEN Se han analizado veinte voladuras de producción en dos minas a cielo abierto, en rocas con resistencia entre media y muy alta. Se usaron tres agentes de voladura diferentes (ANFO, hidrogel y emulsión mezcla), con consumos específicos entre 0.88 y 1.45 kg/m3 . Las excavadoras eran cargadoras frontales y palas de cables. Se midió cuidadosamente las propiedades de la roca, las características de las voladuras y las velocidades de carga, a partir de las cuales se desarrolla un modelo para el cálculo de la productividad de las excavadoras en el que ésta es función de una productividad máxima o ideal multiplicada por una eficacia operativa. La producción máxima es función de la capacidad del cazo y la eficiencia es función de la densidad y resistencia de la roca, y de la concentración energética del explosivo en la roca. El modelo tiene significación estadística y explica un 92 % de la varianza de las medidas de velocidad.

Fuller explicitly connects his career with a childhood full of experiences marked by a merger between the biological and the cultural, constituting a major source of imagery, handled over his whole life. Childhood mobile games like the rope, kyte, swing, wheel, or balloon, all characterized by their dynamic condition, serve as narrative vehicles and provide a context to relate childhood, science and architectural design through an universal approach. We pose three elementary cathegories, in order to trace a genealogy of primary technical objects which operate under tension and mobility principles. Rescued experiences, notions and attitudes from Fuller?s childhood memories, refer to the experience of tension which, in clear contrast with traditional structural methods, which perpetuate the Euclidean, the stable, the massive and the compressive, promote multipolar, dynamic en the ephimeral.

The til-1 locus was identified as a common retroviral integration site in virus-accelerated lymphomas of CD2-myc transgenic mice. We now show that viral insertions at til-1 lead to transcriptional activation of PEBP2αA (CBFA1), a transcription factor related to the Drosophila segmentation gene product, Runt. Insertions are upstream and in the opposite orientation to the gene and appear to activate a variant promoter that is normally silent in T cells. Activity of this promoter was detected in rodent osteogenic sarcoma cells and primary osteoblasts, implicating bone as the normal site of promoter activity. The isoforms encoded by the activated gene all encompass the conserved runt DNA-binding domain and share a novel N terminus different from the previously reported PEBP2αA products. Minor products include isoforms with internal deletions due to exon skipping and a novel C-terminal domain unrelated to known runt domain factors. The major isoform expressed from the activated til-1 locus (G1) was found to account for virtually all of the core binding factor activity in nuclear extracts from its corresponding lymphoma cell line. Another member of this gene family, AML1(CBFA2), is well known for its involvement in human hemopoietic tumors. These results provide evidence of a direct oncogenic role for PEBP2αA and indicate that the Myc and Runt family genes can cooperate in oncogenesis.

Skipping of internal exons during removal of introns from pre-mRNA must be avoided for proper expression of most eukaryotic genes. Despite significant understanding of the mechanics of intron removal, mechanisms that ensure inclusion of internal exons in multi-intron pre-mRNAs remain mysterious. Using a natural two-intron yeast gene, we have identified distinct RNA–RNA complementarities within each intron that prevent exon skipping and ensure inclusion of internal exons. We show that these complementarities are positioned to act as intron identity elements, bringing together only the appropriate 5′ splice sites and branchpoints. Destroying either intron self-complementarity allows exon skipping to occur, and restoring the complementarity using compensatory mutations rescues exon inclusion, indicating that the elements act through formation of RNA secondary structure. Introducing new pairing potential between regions near the 5′ splice site of intron 1 and the branchpoint of intron 2 dramatically enhances exon skipping. Similar elements identified in single intron yeast genes contribute to splicing efficiency. Our results illustrate how intron secondary structure serves to coordinate splice site pairing and enforce exon inclusion. We suggest that similar elements in vertebrate genes could assist in the splicing of very large introns and in the evolution of alternative splicing.

SMN1 and SMN2 (survival motor neuron) encode identical proteins. A critical question is why only the homozygous loss of SMN1, and not SMN2, results in spinal muscular atrophy (SMA). Analysis of transcripts from SMN1/SMN2 hybrid genes and a new SMN1 mutation showed a direct relationship between presence of disease and exon 7 skipping. We have reported previously that the exon-skipped product SMNΔ7 is partially defective for self-association and SMN self-oligomerization correlated with clinical severity. To evaluate systematically which of the five nucleotides that differ between SMN1 and SMN2 effect alternative splicing of exon 7, a series of SMN minigenes was engineered and transfected into cultured cells, and their transcripts were characterized. Of these nucleotide differences, the exon 7 C-to-T transition at codon 280, a translationally silent variance, was necessary and sufficient to dictate exon 7 alternative splicing. Thus, the failure of SMN2 to fully compensate for SMN1 and protect from SMA is due to a nucleotide exchange (C/T) that attenuates activity of an exonic enhancer. These findings demonstrate the molecular genetic basis for the nature and pathogenesis of SMA and illustrate a novel disease mechanism. Because individuals with SMA retain the SMN2 allele, therapy targeted at preventing exon 7 skipping could modify clinical outcome.
