The structural stability of a peroxidase, a dimeric protein from royal palm tree (Roystonea regia) leaves, has been characterized by high-sensitivity differential scanning calorimetry, circular dichroism, steady-state tryptophan fluorescence and analytical ultracentifugation under different solvent conditions. It is shown that the thermal and chemical (using guanidine hydrochloride (Gdn-HCl)) folding/unfolding of royal palm tree peroxidase (RPTP) at pH 7 is a reversible process involving a highly cooperative transition between the folded dimer and unfolded monomers, with a free stabilization energy of about 23 kcal per mol of monomer at 25 degrees C. The structural stability of RPTP is pH-dependent. At pH 3, where ion pairs have disappeared due to protonation, the thermally induced denaturation of RPTP is irreversible and strongly dependent upon the scan rate, suggesting that this process is under kinetic control. Moreover, thermally induced transitions at this pH value are dependent on the protein concentration, allowing it to be concluded that in solution RPTP behaves as dimer, which undergoes thermal denaturation coupled with dissociation. Analysis of the kinetic parameters of RPTP denaturation at pH 3 was accomplished on the basis of the simple kinetic scheme N ->(k) D, where k is a first-order kinetic constant that changes with temperature, as given by the Arrhenius equation; N is the native state, and D is the denatured state, and thermodynamic information was obtained by extrapolation of the kinetic transition parameters to an infinite heating rate. Obtained in this way, the value of RPTP stability at 25 degrees C is ca. 8 kcal per mole of monomer lower than at pH 7. In all probability, this quantity reflects the contribution of ion pair interactions to the structural stability of RPTP. From a comparison of the stability of RPTP with other plant peroxidases it is proposed that one of the main factors responsible for the unusually high stability of RPTP which enhances its potential use for biotechnological purposes, is its dimerization. Two-photon absorption induced polymerization provides a powerful method for the fabrication of intricate three-dimensional microstructures. Recently, Lucirin TPO-L was shown to be a photoinitiator with several advantageous properties for two-photon induced polymerization. Here we measure the two-photon absorption cross-section spectrum of Lucirin TPO-L, which presents a maximum of 1.2 GM at 610 nm. Despite its small two-photon absorption cross-section, it is possible to fabricate excellent microstructures by two-photon polymerization due to the high polymerization quantum yield of Lucirin TPO-L. These results indicate that optimization of the two-photon absorption cross-section is not the only material parameter to be considered when searching for new photoinitiators for microfabrication via two-photon absorption.

Hydroxymethylnitrofurazone presents in vitro activity against Trypanosoma cruzi. The optimization of the synthesis of this compound was performed through a 3(2) factorial statistical design. Quadratic model produced the best response surface predicting a maximum yield (82%) close to the center design point with a seven hours reaction and a 1:1.5 (NF:K(2)CO(3)) ratio.

Gelatin is a cheap and abundant natural product with very good biodegradation properties and can be used to obtain acetic acid or LiClO(4)-based gel polymer electrolytes (GPEs) with high ionic conductivity and good stability. This article presents results of GPEs obtained by the plasticization of gelatin and addition of LiBF(4), where the optimization of the system was achieved by using a factorial design type 22 with two variables: glycerol and LiBF(4). From this analysis it was stated that the effect of glycerol as a plasticizer on the ionic conductivity results is much more important than the effect obtained by varying the lithium salt content or the effect of the interaction of both variables. Also all the samples were characterized by X-ray diffraction measurements, UV-vis-NIR spectroscopy and scanning electron microscopy (SEM) and impedance spectroscopy. The ionic conductivity results of all analyzed samples as a function of temperature obey predominantly an Arrhenius relationship and the samples are stable up to 160 degrees C. Good conductivity results combined with transparency and good adhesion to the electrodes have shown that gelatin-based GPEs are very promising materials to be used as solid electrolytes in electrochromic devices. The analysis of volatile compounds in Funchal, Madeira, Mateus and Perry Vidal cultivars of Annona cherimola Mill. (cherimoya) was carried out by headspace solid-phase microextraction (HS-SPME) combined with gas chromatography–quadrupole mass spectrometry detection (GC–qMSD). HS-SPME technique was optimized in terms of fibre selection, extraction time, extraction temperature and sample amount to reach the best extraction efficiency. The best result was obtained with 2 g of sample, using a divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fibre for 30 min at 30 °C under constant magnetic stirring (800 rpm). After optimization of the extraction methodology, all the cherimoya samples were analysed with the best conditions that allowed to identify about 60 volatile compounds. The major compounds identified in the four cherimoya cultivars were methyl butanoate, butyl butanoate, 3-methylbutyl butanoate, 3-methylbutyl 3-methylbutanoate and 5-hydroxymethyl-2-furfural. These compounds represent 69.08 ± 5.22%, 56.56 ± 15.36%, 56.69 ± 9.28% and 71.82 ± 1.29% of the total volatiles for Funchal, Madeira, Mateus and Perry Vidal cultivars, respectively. This study showed that each cherimoya cultivars have 40 common compounds, corresponding to different chemical families, namely terpenes, esters, alcohols, fatty acids and carbonyl compounds and using PCA, the volatile composition in terms of average peak areas, provided a suitable tool to differentiate among the cherimoya cultivars.

2-Aminothiazole covalently attached to a silica gel surface was prepared in order to obtain an adsorbent for Hg(II) ions having the following characteristics: good sorption capacity, chemical stability under conditions of use, and, especially, high selectivity. The accumulation voltammetry of mercury(II) was investigated at a carbon paste electrode chemically modified with silica gel functionalized with 2-aminothiazole (SIAMT-CPE). The repetitive cyclic voltammogram of mercury(II) solution in the potential range -0.2 to + 0.6 V versus Ag/AgCl (0.02 mol L-1 KNO3; V = 20 mV s(-1)) show two peaks one at about 0.1 V and other at 0.205 V. The anodic wave peak at 0.205 V is well defined and does not change during the cycles and it was therefore further investigated for analytical purposes using differential pulse anodic stripping voltammetry in differents supporting electrolytes. The mercury response was evaluated with respect to pH, electrode composition, preconcentration time, mercury concentration, cleaning solution, possible interferences and other variables. The precision for six determinations (n = 6) of 0.02 and 0.20 mg L-1 Hg(II) was 4.1 and 3.5% (relative standard deviation), respectively. The detection limit was estimated as 0.10 mu g L-1 mercury(II) by means of 3:1 current-to-noise ratio in connection with the optimization of the various parameters involved and using the highest-possible analyser sensitivity. A lagarta-do-cartucho, Spodoptera frugiperda (J.E. Smith), é uma das principais pragas do milho nas Américas. O estudo de sua distribuição espacial é fundamental para a utilização de estratégias de controle, otimização de técnicas de amostragens, determinação de danos econômicos e incorporação de um programa de agricultura de precisão. em uma área cultivada com milho foram realizadas amostragens com intervalo semanal, correspondendo ao estádio vegetativo que compreende desde a germinação até o pendoamento. Foram amostradas 10 plantas ao acaso por parcela, no total de 2000 plantas em cada amostragem. A produtividade foi obtida através da colheita de todas as parcelas que eram pesadas separadamente no campo e em cada parcela foram coletadas 15 espigas aleatoriamente para estimar o comprimento e o diâmetro médio. As análises espaciais, utilizando geoestatística, mostraram que o modelo esférico apresentou o melhor ajuste às lagartas pequenas. À medida que as lagartas foram se desenvolvendo sua distribuição foi tornando aleatória, representada por um modelo ajustado por uma reta, não tendo sido detectado nenhum tipo de dependência espacial nos pontos de amostragem. A produtividade e o diâmetro e comprimento da espiga foram descritos por modelos esféricos, indicando uma variabilidade espacial nos parâmetros de produtividade na área cultivada. A geoestatística mostrou-se promissora para a aplicação de métodos precisos no controle integrado de pragas.

O presente estudo foi desenvolvido na UNESP/Botucatu, São Paulo, Brasil, num solo identificado como Oxisol, onde foram plantadas mudas de limoeiro Siciliano (C. limon) enxertadas em laranjeira Azeda (C. aurantium) e em limoeiro Cravo (C. limonia). O experimento consistiu em três níveis de saturação por bases (50%, 70% e 90%) e três doses de boro (0,5; 1,5 e 4,5 mg dm-3) no plantio em esquema fatorial de 3x3x2, com quatro repetições. Houve diferentes comportamentos entre os porta-enxertos no que se refere à composição mineral de folhas de limoeiro Siciliano , bem como, no desenvolvimento do sistema radicular, sendo maior na laranjeira azeda em relação ao limoeiro cravo. Não houve nenhum efeito na interação de níveis de saturação por bases e doses de boro para nenhum dos parâmetros avaliados.

We evaluated the presence of the melatonin metabolite N-1-acetyl-N-2-formyl-5-methoxykynuramine (AFMK), in cerebrospinal fluid (CSF) of patients with viral meningitis (n = 20) and control samples (n = 8) and correlate AFMK levels with inflammatory markers such as cellularity, protein, tumor necrosis factor (TNF)-alpha, interleukin (IL)-8 and IL-1 beta levels. A portion of the CSF was extracted with dichloromethane (1:5) and analyzed by high-performance liquid chromatography (HPLC) under standardized conditions for AFMK. AFMK was detected in 16 of 20 CSF samples of patients with viral meningitis; the concentration of AFMK was found to be above the quantification limit (50 nmol/L) in six of these samples. AFMK was not detected in any of the eight control samples. The samples were classified into groups according to AFMK levels: undetectable (< 10 nmol/L, group I), detectable but below the quantification limit (< 50 nmol/L, group II), and quantified (> 50 nmol/L, group III). Group II presented the highest levels of proteins and IL-8, whereas group III showed the lowest levels of the inflammatory parameters. This study supports our hypothesis that inflammation favors the formation of AFMK and that this compound has immunomodulatory activity in vivo.

The mapping of the Wigner distribution function (WDF) for a given bound state onto a semiclassical distribution function (SDF) satisfying the Liouville equation introduced previously by us is applied to the ground state of the Morse oscillator. The purpose of the present work is to obtain values of the potential parameters represented by the number of levels in the case of the Morse oscillator, for which the SDF becomes a faithful approximation of the corresponding WDF. We find that for a Morse oscillator with one level only, the agreement between the WDF and the mapped SDF is very poor but for a Morse oscillator of ten levels it becomes satisfactory. We also discuss the limit h --> 0 for fixed potential parameters.

The dynamics of a nonconservative Gross-Pitaevskii equation for trapped atomic systems with attractive two-body interaction is numerically investigated, considering wide variations of the nonconservative parameters, related to atomic feeding and dissipation. We study the possible limitations of the mean-field description for an atomic condensate with attractive two-body interaction, by defining the parameter regions, where stable or unstable formation can be found. The present study is useful and timely considering the possibility of large variations of attractive two-body scattering lengths, which may be feasible in recent experiments.

The neutrino oscillation experiment KamLAND has provided us with the first evidence for e disappearance, coming from nuclear reactors. We have combined their data with all solar neutrino data, assuming two flavor neutrino mixing, and obtained allowed parameter regions which are compatible with the so-called large mixing angle MSW solution to the solar neutrino problem. The allowed regions in the plane of mixing angle and mass squared difference are now split into two islands at 99% C.L. We have speculated how these two islands can be distinguished in the near future. We have shown that a 50% reduction of the error on SNO neutral-current measurement can be important in establishing in each of these islands the true values of these parameters lie, We also have simulated KamLAND positron energy spectrum after I year of data taking, assuming the current best fitted values of the oscillation parameters, combined it the with current solar neutrino data and showed how these two split islands can be modified.
