Neopisinus gen. nov. é proposto com designação da espécie-tipo Neopisisnus fiapo sp. nov., com base em ambos os sexos, do Rio Grande do Sul, Brasil. Neopisinus distingue-se de todos os gêneros de Spintharinae pelo palpo do macho com enorme condutor trífido, com duas projeções afiladas e uma com ápice bifurcado; pela forma característica da apófise tegular de theridioideos com um lobo terminal e outro dorsal. Nas fêmeas, epígino com aberturas inconspícuas junto à fenda transversal no terço anterior e, internamente, por um espessamento mediano-longitudinal tubular, por onde correm os ductos de copulação em seu percurso inicial. Neopisinus urucu sp. nov. é descrita do norte do Brasil, com base em ambos os sexos. Sete espécies são transferidas de Episinus para Neopisinus: N. bigibbosus (O. P.-Cambridge, 1896), N. bruneoviridis (Mello-Leitão, 1948), N. cognatus (O. P.-Cambridge, 1893), N. gratiosus (Bryant, 1940), N. longipes (Keyserling, 1884), N. putus (O. P.-Cambridge, 1894) e N. recifensis (Levi, 1964). São descritos pela primeira vez o macho de N. longipes e a fêmea de N. recifensis. Novas ocorrências e ilustrações são apresentadas para N. bruneoviridis.

Três espécies de Cryptachaea Archer, 1946 são descritas, duas delas para o Brasil: C. amazonas sp. nov. da Reserva Florestal Adolpho Ducke, Manaus, Amazonas e Cryptachaea maldonado sp. nov. da Base de Operações Geólogo Pedro de Moura, Porto Urucu, rio Urucu, Coari, Amazonas, com base em machos. Uma nova associação de macho e fêmea é proposta para Achaearanea hieroglyphica (Mello-Leitão, 1940). O macho da Guiana Francesa, atribuído à última espécie, é considerado uma espécie nova de Cryptachaea, C. ingijonathorum. O macho de Achaearanea tingo Levi, 1963 é descrito pela primeira vez. Novas ocorrências são listadas para A. trapezoidalis (Taczanowski, 1873).

Hentziectypus tayrona sp. nov. é descrita com base em um macho de Santa Marta, Colômbia. A fêmea de H. rafaeli Buckup & Marques, 1991, é descrita pela primeira vez e a distribuição é estendida a Bolívia e ao sul do Brasil.

Com a finalidade de contribuir para o esclarecimento dos mecanismos da morte súbita na forma crônica da tripanossomose cruzi, foram analisados, de modo sistemático, os seguintes aspectos em 116 chagásicos falecidos subitamente: o sexo, a idade em que ocorreu o óbito, a profissão, o papel da emoção e do esforço físico no momento do desenlance fatal e a época da morte em relação às estações do ano. Todos estes tripanossomóticos, aparentemente, não manifestaram, em vida, sintomas e/ou sinais de insuficiência cardíaca. Para análise comparativa utilizou-se um grupo constituído por chagásicos crônicos falecidos com ou após manifestações de insuficiência cardíaca. Os resultados demonstram que o óbito acomete (tanto nos chagásicos falecidos subitamente como naqueles com insuficiência cardíaca) mais freqüentemente o homem que a mulher e que nos chagásicos do sexo masculino falecidos subitamente a morte ocorre mais cedo do que em tripanossomóticos cujo desenlace está asociado ou é precedido por quadro de insuficiência cardíaca. Finalmente, nossos dados sugerem que o esforço físico, os fatores emotivos e as variações sazonais não exercem papel essencial no desencadeamento do óbito do chagásico crônico.

Gastro-intestinal parasitism of 24 buffalo cows before parturition, and post-parturition, their infection and that of their respective calves during the following 30 weeks were studied. Willis, Hoffmann and whenever possible, the modified Gordon & Whitlock techniques were used for fecal examinations. Toxocara vitulorum eggs were the earliest forms encountered in calves feces, as follows: during the 1st week after birth, 58.33% of the calves were positive, and in the 4th week, 100% of these animals were positive. Eggs of Strongyloides sp were in the 1st week after birth in two of the calves and in the 5th week, all for them were positive. The next parasites to appear were the Coccidia of which oocysts were detected in the feces of two calves in the 2nd week after birth, and 58.33% of the calves were positive for these in the 3rd week, and in the 6th week, all calves shed oocysts in their feces. On the other hand, eggs of Strongylids were the last forms to appear in calves feces. However, despite their sporadic appearance in the feces, eggs of these parasites were observed continuously from the 11th week onwards, and at this point, the percentage of positive samples began to increase to reach its peak. Relatively to adult animals, eggs of T. vitulorum were observed in the feces of 11 cows, one or twice at most; eggs of Strongyloides sp were seen only once in the feces of four buffalo cows and eggs of Strongylids in 21 out of 24 cows. Oocysts of Coccidia were observed in 16 cows. Mechanisms of infestation of calves with these parasites are discussed.

Immunoglobulin G and M humoral response to recombinant protein B13 and glycoconjugate LPPG Trypanosoma cruzi defined antigens was evaluated by ELISA in 18 patients in the acute phase of Chagas disease, who were contaminated on the same occasion. LPPG showed 100% positivity detecting both IgM and IgG antibodies, while positivity of 55-65% was observed for B13. An epimastigote alkaline extract (EPI) also showed high sensitivity for acute IgM (100%) and IgG (90%) antibodies. However LPPG had better discriminatory reactivity since with EPI two patients showed negative IgG and several other sera presented OD values for IgG and IgM antibodies very close to the cutoff. Thus, it is suggested that detection of IgM antibodies by LPPG may be used for diagnosis of the acute phase of Chagas disease. An intense decline of IgG and IgM antibodies to the three antigens was observed in response to anti-T. cruzi chemotherapy in all acute phase patients. After treatment, six (30%) individuals maintained IgG positivity to EPI, LPPG, and B13 with lower reactivity than that measured at the acute phase. For comparison, serology of a group of 22 patients in the chronic phase of Chagas disease and also submitted to chemotherapy was determined. Positive IgM antibodies to EPI, LPPG and B13 were detected in only 5-9% cases. In all chronic-phase patients IgG antibodies highly reactive to the three antigens were present and no significant decrease resulted after benznidazole administration. These observations reinforce previous reports that treatment in the acute phase may reduce or eliminate the parasite.

Six hundred and eleven random-source dogs (338 male, 273 female) one year of age or older, from six sections of the city of Recife, Pernambuco, were examined antemortem for circulating microfilariae Dirofilaria immitis and Dipetalonema reconditum adult heartworm (D. immitis) antigen, and examined postmortem for adult heartworms. The prevalence of heartworm infection was 2.3% (14/611), as determined by necropsy for adult worms, and 1% (6/611) had circulating microfilariae of D. immitis; thus, 57.1% of the heartworm-infected dogs had occult infections. The results of serological testing indicated that 1.3% (8/611) of the dogs were positive for adult heartworm antigen. A total of 42 (6.9%) of the dogs had microfilariae of D. reconditum; 40 of these had only D. reconditum and two additional dogs had microfilariae of both species, D. immitis and D. reconditum.

Dengue virus type 3 was isolated for the first time in the country as an indigenous case from a 40 year-old woman presenting signs and symptoms of a classical dengue fever in the municipality of Nova Iguaçu, State of Rio de Janeiro. This serotype has been associated with dengue haemorrhagic epidemics and the information could be used to implement appropriate prevention and control measures. Virological surveillance was essential in order to detected this new serotype.

This study was carried out to investigate the immune response against 97 kDa (p97) molecular marker of Toxoplasma gondii that has been characterized as a cytosolic protein and a component of the excreted-secreted antigens from this parasite. A total of 60 serum samples from patients were analyzed by enzime-linked immunosorbent assay and Western blot for toxoplasmosis. These samples were organized in three groups, based on clinical symptoms and results of serological tests. Group I: 20 samples reactive to IgG and IgM (acute phase); group II: 20 non-reactive samples (control group); and group III: 20 samples reactive only to IgG (chronic phase). Western blot was performed with total antigenic extracts or with excreted and secreted antigen from T. gondii to identify the fraction correspondent to p97. It was observed that this cytosolic component from T. gondii stimulates the immunologic system to produce both IgM and IgG antibodies in the beginning of the acute infection and IgG throughout the chronic stage of the asymptomatic toxoplasmosis.

The increasing number of pertussis cases reported on the last twenty years and the existence of new acellular vaccines reinforce the need of research for experimental models to assure the quality of available pertussis vaccines. In this study, allotments of whole-cell and acellular pertussis vaccines were tested through the Intranasal Challenge Model (INM) using conventional NIH mice. The results have been compared to those achieved by the "Gold standard" Intracerebral Challenge Model (ICM). In contrast to ICM, INM results did not show intralaboratorial variations. Statistical analysis by Anova and Ancova tests revealed that the INM presented reproducibility and allowed identification and separation of different products, including three-component and four-component accellular pertussis vaccines. INM revealed differences between pertussis vaccines. INM provides lower distress to the mice allowing the reduction of mice number including the possibility of using conventional mice (less expensive) under non-aseptic environment. Thus, INM may be used as an alternative method of verifying the consistence of allotment production, including acellular pertussis vaccines.

In this study, IgA1 levels in the milk and serum of puerperae were compared and a correlation was established between the levels of this immunoglobulin and the occurrence of parasitism. Eighty-three paired milk and serum samples were obtained from puerperal and IgA1 levels were analyzed. In addition, the presence of intestinal parasites in stool samples from these puerperae was determined. Twelve puerperae tested positive for intestinal parasites and all their samples presented an IgA1 ELISA Index > 1. There was a correlation between serum and milk IgA1 levels and puerperae with any parasite in their stool (r = 0.6723; p = 0.0166). This finding may reinforce the importance of breast-feeding for the protection of neonates.

A peptide (SmB2LJ; r175-194) that belongs to a conserved domain from Schistosoma mansoni SmATPDase 2 and is shared with potato apyrase, as predicted by in silico analysis as antigenic, was synthesised and its immunostimulatory property was analysed. When inoculated in BALB/c mice, this peptide induced high levels of SmB2LJ-specific IgG1 and IgG2a subtypes, as detected by enzyme linked immunosorbent assay. In addition, dot blots were found to be positive for immune sera against potato apyrase and SmB2LJ. These results suggest that the conserved domain r175-194 from the S. mansoni SmATPDase 2 is antigenic. Western blots were performed and the anti-SmB2LJ antibody recognised in adult worm (soluble worm antigen preparation) or soluble egg antigen antigenic preparations two bands of approximately 63 and 55 kDa, molecular masses similar to those predicted for adult worm SmATPDase 2. This finding strongly suggests the expression of this same isoform in S. mansoni eggs. To assess localisation of SmATPDase 2, confocal fluorescence microscopy was performed using cryostat sections of infected mouse liver and polyclonal antiserum against SmB2LJ. Positive reactions were identified on the external surface from the miracidium in von Lichtenberg's envelope and, in the outer side of the egg-shell, showing that this soluble isoform is secreted from the S. mansoni eggs.
