Background Hippocampal neurogenesis has been suggested as a downstream event of antidepressants (AD) mechanism of action and might explain the lag time between AD administration and the therapeutic effect. Despite the widespread use of AD in the context of Major Depressive Disorder (MDD) there are no reliable biomarkers of treatment response phenotypes, and a significant proportion of patients display Treatment Resistant Depression (TRD). Fas/FasL system is one of the best-known death-receptor mediated cell signaling systems and is recognized to regulate cell proliferation and tumor cell growth. Recently this pathway has been described to be involved in neurogenesis and neuroplasticity. Methods Since FAS -670A>G and FASL -844T>C functional polymorphisms never been evaluated in the context of depression and antidepressant therapy, we genotyped FAS -670A>G and FASL -844T>C in a subset of 80 MDD patients to evaluate their role in antidepressant treatment response phenotypes. Results We found that the presence of FAS -670G allele was associated with antidepressant bad prognosis (relapse or TRD: OR=6.200; 95% CI: [1.875–20.499]; p=0.001), and we observed that patients carrying this allele have a higher risk to develop TRD (OR=10.895; 95% CI: [1.362–87.135]; p=0.008).Moreover, multivariate analysis adjusted to potentials confounders showed that patients carrying G allele have higher risk of early relapse (HR=3.827; 95% CI: [1.072–13.659]; p=0.039). FAS mRNA levels were down-regulated among G carriers, whose genotypes were more common in TRD patients. No association was found between FASL-844T>C genetic polymorphism and any treatment phenotypes. Limitations Small sample size. Patients used antidepressants with different mechanisms of action. Conclusion To the best of our knowledge this is the first study to evaluate the role of FAS functional polymorphism in the outcome of antidepressant therapy. This preliminary report associates FAS -670A>G genetic polymorphism with Treatment Resistant Depression and with time to relapse. The current results may possibly be given to the recent recognized role of Fas in neurogenesis and/or neuroplasticity.

This work aims to shed some light on longshore sediment transport (LST) in the highly energetic northwest coast of Portugal. Data achieved through a sand-tracer experiment are compared with data obtained from the original and the new re-evaluated longshore sediment transport formulas (USACE Waterways Experiment Station’s Coastal Engineering and Research Center, Kamphuis, and Bayram bulk formulas) to assess their performance. The field experiment with dyed sand was held at Ofir Beach during one tidal cycle under medium wave-energy conditions. Local hydrodynamic conditions and beach topography were recorded. The tracer was driven southward in response to the local swell and wind- and wave-induced currents (Hsb=0.75mHsb=0.75m, Tp=11.5sTp=11.5s, θb=8−12°θb=8−12°). The LST was estimated by using a linear sediment transport flux approach. The obtained value (2.3×10−3m3⋅s−12.3×10−3m3⋅s−1) approached the estimation provided by the original Bayram formula (2.5×10−3m3⋅s−12.5×10−3m3⋅s−1). The other formulas overestimated the transport, but the estimations resulting from the new re-evaluated formulas also yield approximate results. Therefore, the results of this work indicated that the Bayram formula may give satisfactory results for predicting the longshore sediment transport on Ofir Beach.

Several phlebotomine sand fly species have been regarded as putative or proven vectors of parasites of the genus Leishmania in Brazil, but data for the northeastern region remains incipient. In this study, a total of 600 phlebotomine sand flies were grouped in pools of 10 specimens each and tested by a Leishmania genus-specific PCR and by a PCR targeting Leishmania (Leishmania) infantum. Fourteen out of 60 pools were positive by the genus-specific PCR, being five pools of L. migonei, seven of L. complexa, one of L. sordellii and one of L. naftalekatzi, which correspond to a minimal infection rate of 2.3% (14/600). Our results, associated with their known anthropophily and their abundance, suggest the participation of L. migonei and L. complexa as vectors of Leishmania in northeastern Brazil. Remarkably, this is the first time in this country that the detection of Leishmania DNA in L. sordellii and L. naftalekatzi has been reported, but future studies are necessary to better understand the significance of these findings.

São estudados clínica e radiologicamente 566 cha¡sicos crônicos não selecionados, da área endêmica de Bambuí, Minas Gerais. Utilizou-se o método do tempo de trânsito esofágico, em abreugrafias de 70 mm, incidência perfil esquerdo ou oblíqua anterior direita. Verificou-se uma prevalência global de 8,83% de disperistalse esofágica, especialmente a partir da terceira década de vida sem diferença quanto aos sexos. Observou-se 72% de esofagopatias no grau I, 18% no grau II e 10% no grau III. Para 80 indivíduos soro-negativos verificou-se apenas um caso de disperistalse, no grau I. Para 115 pacientes com a fase aguda da tripanossomíase detectada há 27 anos, em média, a prevalência global da esofagopatia foi de 18,3%, sendo crescente a proporção de alterações com o tempo de evolução da doença. Verificou-se diferença significativa entre estes números e a prevalência encontrada nos pacientes sem forma aguda detectada, discutindo-se se a maior proporção de esofagopatias nos indivíduos com fase aguda mais intensa não se deveria à maior desnervação ocorrida nestes casos. Verificou-se a concomitância de cardiopatia crônica cha¡sica em pelo menos 50% dos casos de disperistalse de esôfago, em acordo com outros Autores. Aparentemente esta associação é mais evidente naqueles casos de esofagopatias de graus mais avançados. Ressalta-se a simplicidade do método empregado e seu baixo custo operacional quando utilizada a abreugrafia postal, adequado portanto à realidade social das áreas endêmicas de doença de Chagas no Brasil.

A maioria dos métodos utilizados na caracterização genética do HIV-1 baseia-se na análise de regiões específicas do genoma viral, fornecendo informação parcial sobre o mesmo e, por consequência, revelando-se inadequados para a identificação de vírus recombinantes. O único método que permite uma caracterização integral do genoma viral passa pela sua sequenciação completa. No entanto, este é um método dispendioso, laborioso e de difícil implementação quando se pretende a análise de elevados números de amostras. Como alternativa a este último, o conjunto de métodos genericamente designados de MHA (Multiple Region Hybridization Assay) baseiam-se na amplificação, por PCR em tempo-real, de várias regiões ao longo do genoma viral e na sua caracterização com sondas específicas (TaqMan). Tendo este modelo por base, o objectivo deste estudo foi o desenvolvimento de um ensaio de hibridação múltipla (MHABG0214) passível de ser aplicado ao estudo de um elevado número de amostras. Este método foi desenvolvido tendo como objectivo a genotipagem as estirpes circulantes dominantes na epidemia Portuguesa, nomeadamente os subtipos B, G e formas genéticas recombinantes CRF02_AG e CRF14_BG. Com base em alinhamentos de sequências de referência de genoma completo, delinearam-se primers universais e subtipo-específicos para a amplificação de diversas regiões codificantes distribuídas ao longo do genoma do HIV-1 (Gag, Protease, Transcriptase Reversa, Integrase, Rev, Gp120 e Gp41). A optimização foi efectuada, inicialmente, para um conjunto de amostras de referência e seguidamente avaliada num conjunto de 50 amostras clínicas. O MHABG0214 foi implementado numa estratégia de PCR em tempo-real, numa detecção dependente de SYBR® Green I para todas as regiões ou, como alternativa, usando sondas TaqMan (Gp41). Apresentamos ainda uma estratégia em que a análise de resultados se baseia, simplesmente, numa abordagem usando PCR/gel de agarose convencional. Estas abordagens constituem ferramentas úteis na identificação das estirpes de HIV-1 em Portugal.

RESUMO: O presente trabalho teve como objetivo avaliar, através de diferentes testes, a qualidade fisiológica de sementes de girassol na safrinha do Distrito Federal. Os testes foram realizados no Laboratório de Sementes da Faculdade de Agronomia e Veterinária ? FAV, da Universidade de Brasília - UnB, Campus Universitário Darcy Ribeiro, Brasília, DF, em 2014. Foram realizados testes de germinação padrão em areia e em papel, peso da matéria verde e da matéria seca, envelhecimento acelerado, condutividade elétrica e lixiviados de potássio. Foram encontradas diferenças significativas entre os genótipos para as todas as características avaliadas. Dentre os genótipos avaliados, os híbridos que se destacaram quanto a qualidades fisiológicas desejáveis pelos testes de germinação, condutividade elétrica e lixiviação de potássio foram BRS G42 e SYN 045. Materiais genéticos foram identificados no trabalho para possível exploração em programas de melhoramento. ABSTRACT: This study aims to evaluate, through different tests, the seed physiological quality of sunflower in the second crop in the Brazilian savannah. The tests were carried on in the Seed Laboratory of Veterinary and Agriculture Faculty of University of Brasilia, in 2014. The tests were seedling emergence in sand and paper, green and dry matter weight, accelerated aging, electrical conductivity and potassium leached. Related to the evaluated characteristics, significant differences were found between genotypes. Among them, the hybrids that stood out for the physiologic characteristics through the germination test, electric conductivity and potassium leaching were BRS G42 and SYN 045. Genetic materials were identified in this study for a possible exploration in breeding programs.

The distribution of sand flies and cases of tegumentary leishmaniasis in the area surrounding JB Alberd city, and the proximities of Catamarca province were studied, after an increase of reported cases from JB Alberdi, Tucumán province, in 2003. Of 14 confirmed cases, 57% were females and 57% were less than 15 years old, suggesting peridomestic transmission. However, 86% of them lived close to the Marapa river forest gallery and related wooded areas. Over 1,013 sand flies were collected; Lutzomyia neivai (Pinto, 1926) was prevalent at all the sites (92.3%), while Lutzomyia migonei (França, 1920) (6.7%) and Lu. cortelezzii (Brèthes, 1923) (1%) were also found. The spatial distribution of Lu. neivai overlapped that of the cases, with higher abundance in microfocal hot spots close to the river in stable vegetated habitats or modified habitats with shadow and animal blood sources. The cumulative outcome of anthropic, ecological and climatic factors could have contributed to the onset of the outbreak.

A total of 123 stool specimens collected in Teresina, Piauí between 1994 and 1996, from 0 to 2-year-old children with diarrhea, were used for this study. Molecular characterization of the G and P rotavirus genotypes was performed using the reverse transcriptase polymerase chain reaction. The following results were obtained for the P genotypes: P[8] (17. 1%), P[1] (4. 9%), P[4] (3. 3%), P[6, M37] (2. 4%) and mixtures (27. 6%). The P[1]+P[8] mixture was found in 19. 5% of the samples. For the G genotypes, the results were: G1 (25. 2%), G5 (13. 8%), G2 (2. 5%), G4 (2. 5%), G9 (0. 8%) and mixtures (41. 5%). G1+G5 was the mixture most frequently found (12. 1%). Our results showed unusual combinations such as P[1]G5 and P[1]+P[8]G5. The high percentage of mixtures and unusual combinations containing mixtures of human and animal rotavirus genotypes strongly suggests the possibility of gene reassortment and interspecies transmission.

INTRODUCTION: The work was conducted to study phlebotomine fauna (Diptera: Psychodidae) and aspects of American cutaneous leishmaniasis transmission in a forested area where Leishmania (Leishmania) amazonensis occurs, situated in the municipality of Bela Vista, State of Mato Grosso do Sul, Brazil. METHODS: The captures were conducted with modified Disney traps, using hamster (Mesocricetus auratus) as bait, from May 2004 to January 2006. RESULTS: Ten species of phlebotomine sandflies were captured: Brumptomyia avellari, Brumptomyia brumpti, Bichromomyia flaviscutellata, Evandromyia bourrouli, Evandromyia lenti, Lutzomyia longipalpis, Psathyromyia campograndensis, Psathyromyia punctigeniculata, Psathyromyia shannoni and Sciopemyia sordellii. The two predominant species were Ev bourrouli (57.3%) and Bi flaviscutellata (41.4%), present at all sampling sites. Two of the 36 hamsters used as bait presented natural infection with Leishmania. The parasite was identified as Leishmania (Leishmania) amazonensis. CONCLUSIONS: Analysis of the results revealed the efficiency of Disney traps for capturing Bichromomyia flaviscutellata and the simultaneous presence of both vector and the Leishmania species transmitted by the same can be considered a predictive factor of the occurrence of leishmaniasis outbreaks for the human population that occupies the location.

INTRODUCTION: Phlebotomine sand flies are small insects of great medical relevance. This article presents an updated list of the phlebotomine sand flies occurring in the State of Pernambuco, Brazil. METHODS: A review of literature published since the 1940s up to May 2010 was conducted and voucher material deposited in reference collections were studied. RESULTS: A total of 37 phlebotomine species have previously been reported as occurring in Pernambuco, but the record of six species needs confirmation. CONCLUSIONS: This work provides an updated list of phlebotomine sand flies of Pernambuco, with some notes on their taxonomy, ecology, distribution and epidemiological relevance.
