Trata-se de um estudo retrospectivo de 35 casos de ruptura de ligamento cruzado atendidos pelo Serviço de Cirurgia de Pequenos Animais da FMVZ-UNESP-Campus de Botucatu, no período janeiro de 1991 a junho de 1997. Os cães foram submetidos à técnica de PAATSAMA (1952), modificada para reconstituição do ligamento cruzado, que consistiu na passagem do retalho de fascia lata através da articulação do joelho e fixação no côndilo medial da tíbia e epicôndilo medial do fêmur. Pelos resultados obtidos, concluiu-se que esta técnica permite boa estabilização das superfícies articulares, com trans e pós operatório sem complicações, e permite recuperação completa da capacidade funcional do membro afetado.

A citometria de fluxo é um método emergente na medicina veterinária que permite a identificação e quantificação de células em suspensão. Apesar do custo elevado e da necessidade de técnicos especializados para realização da avaliação citofluorométrica, esta técnica tem uma ampla aplicação na hematologia veterinária, incluindo a avaliação de células-tronco hematopoiéticas, eritrócitos, leucócitos e plaquetas. O grande potencial de aplicação clínica da citometria de fluxo inclui a quantificação das células CD34+ como indicação da capacidade de reconstituição hematopoiética após o transplante de células-tronco e das subpopulações linfocitárias para avaliação da resposta imune frente aos transplantes e às doenças que acometem os animais. Projetos científicos sobre a avaliação citofluorométrica das células-tronco e subpopulações linfocitárias de cães têm sido desenvolvidos com sucesso por pesquisadores da área de hematologia veterinária na Universidade Estadual Paulista (UNESP), Campus de Jaboticabal. Portanto, no futuro, a citometria de fluxo deve se tornar uma técnica de rotina em laboratórios veterinários no Brasil.

As células-tronco hematopoéticas promovem a reconstituição hematopoética e de outros tecidos, estando presentes no embrião, sangue periférico, medula óssea e sangue do cordão umbilical. Os modelos experimentais de células-tronco em cães têm propiciado informações relevantes para transplantes de células-tronco em humanos. A capacidade de reconstituição hematopoética e da plasticidade das células-tronco de cães permite o emprego do modelo canino em várias propostas científicas e terapêuticas, que propiciam informações pré-clínicas ao homem. O objetivo desta revisão bibliográfica é relatar a importância das células-tronco hematopoéticas de cães, sendo que a sua principal aplicação clínica é o transplante das células-tronco.

Procurando-se reafirmar a importância da teoria marxista, este artigo propõe-se a redefinição da democracia enquanto método e a possibilidade de sua superação para democracia-condição social através da praxis intencional exercida no cotidiano. A compreensão da vida cotidiana requer a sua reconstituição ontológica pela via da conscientização dos valores éticos contidos na genericidade.

Mutants of each of the four divalent cation binding sites of chicken skeletal muscle troponin C (TnC) were constructed using site directed mutagenesis to convert Asp to Ala at the first coordinating position in each site. With a view to evaluating the importance of site-site interactions both within and between the N- and C-terminal domains, in this study the mutants are examined for their ability to associate with other components of the troponin-tropomyosin regulatory complex and to regulate thin filaments. The functional effects of each mutation in reconstitution assays are largely confined to the domain in which it occurs, where the unmutated site is unable to compensate for the defect, Thus the mutants of sites I and II bind to the regulatory complex but are impaired in ability to regulate tension and actomyosin ATPase activity, whereas the mutants of sites III and IV regulate activity but are unable to remain bound to thin filaments unless Ca2+ is present. When all four sites are intact, free Mg2+ causes a 50-60-fold increase in TnC's affinity for the other components of the regulatory complex, allowing it to attach firmly to thin filaments. Calcium can replace Mg2+ at a concentration ratio of 1:5000, and at this ratio the Ca2 . TnC complex is more tightly bound to the filaments than the Mg2 . TnC form, In the C-terminal mutants, higher concentrations of Ca2+ (above tension threshold) are required to effect this transformation than in the recombinant wild-type protein, suggesting that the mutants reveal an attachment mediated by Ca2+ in the N-domain sites.

Background: Models for the study of hematopoietic stem cells in dogs provide important information for bone marrow transplantation in humans. Recent studies have reported the importance of human umbilical cord blood (UCB) as an alternative to allogenic bone marrow for hematopoietic reconstitution. However, there are no studies on the UCB cells of dogs. Objective: the aim of this experiment was to characterize and quantify the blood cells of the umbilical cord of dogs. Methods: the blood of the umbilical cord of 20 neonatal dogs, delivered at term, with a median gestation time of 58 days, was collected with a 5-mL syringe containing EDTA. Total RBC, WBC, and platelet counts, HCT, hemoglobin (Hgb) concentration, and RBC indices were determined using an automatic cell counter. The differential leukocyte count was determined manually in blood smears stained with May-Grunwald-Giemsa. Reticulocyte percentages were determined on blood smears stained with brilliant cresyl blue and counterstained with May-Grunwald Giemsa. Results: the MCHC and numbers of RBCs, WBCs, neutrophils, and eosinophils in UCB were lower as compared with reference values for the peripheral blood of healthy neonatal and adult dogs; whereas, the MCV and reticulocyte percentages were higher. Conclusion: Erythrocyte macrocytosis and hypochromasia in UCB were consistent with marked reticulocytosis and indicative of high erythropoietic activity. The results of this study are an important first step in the characterization of UCB from neonatal dogs.

Uncoupling proteins (UCPs) are specialized mitochondrial transporter proteins that uncouple respiration from ATP synthesis. In this study, cDNA encoding maize uncoupling protein (ZmPUMP) was expressed in Escherichia coli and recombinant ZmPUMP reconstituted in liposomes. ZmPUMP activity was associated with a linoleic acid (LA)-mediated H+ efflux with Km of 56.36 ± 0.27 μM and Vmax of 66.9 μmol H+ min-1 (mg prot)-1. LA-mediated H+ fluxes were sensitive to ATP inhibition with Ki of 2.61 ± 0.36 mM (at pH 7.2), a value similar to those for dicot UCPs. ZmPUMP was also used to investigate the importance of a histidine pair present in the second matrix loop of mammalian UCP1 and absent in plant UCPs. ZmPUMP with introduced His pair (Lys155His and Ala157His) displayed a 1.55-fold increase in LA-affinity while its activity remained unchanged. Our data indicate conserved properties of plant UCPs and suggest an enhancing but not essential role of the histidine pair in proton transport mechanism. © 2006 Elsevier Inc. All rights reserved.

The aim of the present study was to determine the fluoride concentration in some brands of mother's milk substitutes and evaluate the possibility of developing dental fluorosis by consuming these products. The products, all powdered, were divided into 3 groups: infant formulae (group I, n = 7), milk-based (group M, n = 8) and soy-based (group 8, n = 3). Samples from 3 cans of different batches of each brand were reconstituted in deionized water and analyzed using the specific electrode method, after hexamethyldisiloxane (HMDS) facilitated diffusion. The fluoride content (mg F/L) of the products ranged from 0.044 to 0.326 (I), 0.014 to 0.045 (M) and 0.253 to 0.702 (S). There was significant difference in the fluoride content of cans from distinct batches (p < 0.05) in most of the brands. The reconstitution of all products in water with optimal fluoride concentration for consumption during the mineralization phase of the primary teeth could result in daily fluoride intake above 0.07 mg F/kg body weight/day. Therefore, the consumption of these products, especially when reconstituted with optimally fluoridated water, could increase the risk of developing dental fluorosis.

According to the environmental legislation enforced in Brazil and the process of marketing globalization, the commitment of the nations to the preservation of the environment is intensified. By reason of nature's negative responses to its intensive use, awareness then appears from enterprises and agencies about how the anthropic action over the environment needs to be minimized, becoming a challenge: development and sustainability. In this context, the present work made use of the Mechanical tillage of the soil, as a technique to apply, in a large scale, the strategies and methods to recover mined areas that were researched and developed experimentally by researchers on a theme project about the recovering of degraded areas. This work was conducted in the Amazon ecosystem, inside the Jamari National Forest - Rondônia (FLONA do Jamari), in deactivated cassiterite mines. The objectives of this work were to: Develop a computational program capable of managing a database and assist in the selection of machines and preparation methods to execute the operations of topographical reconstitution and tillage of surfaces in areas degraded by the mineral exploitation of cassiterite. Use the program that was developed in the planning of costs and operational development, for the operations required in the strategies for recovering the areas. Analyze the vegetable productivity in the mobilized areas and the quality of the superficial mobilization, making use of indicators and tillage methods. Evaluate, through biological indicators, the efficiency of the recovery strategies and techniques that were mechanized and applied on the location. The results showed that the developed computational program (SGMAD) served the methodological purposes (the analysis of costs and operational capacity) established for the planning and the selection of the tillage machines and methods in the areas of mineral exploitation of cassiterite. The applied methods and quality of the superficial mobilization were significant to the development of leguminous plants in the areas. The use of biological indicators (microbial biomass and enzymatic activity) in the evaluation of the adopted techniques and strategies revealed that the planting of leguminous plants and their posterior incorporation have been promoting gradually positive alterations in some of the analyzed soil/substract parameters. © 2010 WIT Press.
