This document, which is partly based on by the study Impacto de la Infraestructura de la Calidad en América Latina, is a second joint effort by the Economic Commission for Latin America and the Caribbean (ECLAC) and the German National Metrology Institute (PTB) to move towards a more detailed understanding of the role played by the Quality Infrastructure in opening up possibilities for the countries in the region to innovate and compete. Through methodological analysis and the realization of case studies at the national level in certain countries in the region, the document seeks to offer a more comprehensive picture of the impact of the Quality Infrastructure and its importance for the economic and social development of countries. The document analyzes various QI related aspects, with particular emphasis on a review of conceptual elements, the role of the QI in the innovation systems of countries and a brief analysis of a set of case studies in Latin American countries. It also identifies a series of challenges and limitations for carrying out impact studies. These elements are taken up again in the final conclusions of the book, where a number of policy recommendations are outlined.

A presente pesquisa explorou o tema gestão, trabalho e saúde docente. O objetivo deste estudo foi analisar as possíveis relações entre trabalho e saúde docente, no contexto das mudanças na gestão na Escola Bosque, no período de 1996 a 2006. Nesse sentido, a pesquisa objetivou, especificamente: a) analisar as relações estabelecidas entre os docentes e a gestão pedagógica; b) investigar se as mudanças na gestão da escola contribuíram para o adoecimento/afastamento dos docentes; c) analisar as políticas de atendimento à saúde docente existentes, implementada pela Secretaria Municipal de Educação. Para atingir tais objetivos, realizei um estudo de caso. A partir da pesquisa, a dissertação foi organizada em três partes. Na primeira, apresento o lócus da pesquisa, no qual procuramos identificar e delinear os principais acontecimentos que possibilitaram a concepção e a construção da Escola Bosque, assim como os fundamentos e o projeto político pedagógico pelos quais o trabalho docente é orientado, tendo como pano de fundo a gestão do governo do Partido da Frente Liberal (PFL). No segundo capitulo, ocupo-me dos diferentes programas, na área da educação, desenvolvidos pelos governos municipais, nas duas gestões do Partido dos Trabalhadores (1997 a 2004) e nos dois primeiros anos da gestão do Partido Trabalhista Brasileiro (PTB), buscando desvelar as repercussões no trabalho e na gestão da Escola Bosque, as relações preconizadas no interior da escola pesquisada, as concepções de trabalho docente e as conseqüências para a saúde desse trabalhador. No Capítulo 3, faço uma análise bibliográfica acerca do adoecimento docente, buscando interconexões com as reformas educacionais, nos anos 1990, e suas relações com as mudanças nas concepções de educação, gestão e trabalho docente, diante das reconfigurações das políticas econômicas e sociais. No quarto capítulo, analiso as entrevistas realizadas com docentes e coordenadores da instituição, buscando trazer à tona as relações existentes entre o trabalho realizado e a gestão. As conclusões, nesta pesquisa, permitem afirmar que pode existir uma correlação entre a gestão da educação e a forma como ela se apresenta na Escola Bosque, relação essa que se estende ao adoecimento e aos afastamentos dos docentes dessa instituição.

Este estudo analisa a incidência da política pública municipal de resíduo sólido sobre os catadores de materiais recicláveis no aterro sanitário do Aurá, no período de 2002 a 2008. A análise é pautada nas ações implementadas durante o governo do Partido dos Trabalhadores (PT) 1997-2004 e do Partido Trabalhista Brasileiro (PTB) 2005... Em termos metodológicos, trata-se de um estudo de caso baseado no exame das políticas públicas dirigidas a este segmento da população e suas formas organizativas. A situação dos catadores do aterro sanitário do Aurá, no município de Belém, constitui-se progressivamente como um problema complexo a ser enfrentado, sobretudo pelo poder público municipal por meio da implementação de políticas públicas. Assim, é objetivo deste trabalho analisar como a política pública de resíduo sólido implantada pela Prefeitura Municipal de Belém, através da Secretaria Municipal de Saneamento (SESAN), incidiu sobre a condição de vida (laboral e política) dos catadores do aterro sanitário do Aurá. Esta política, tem início com a implementação do Projeto de Biorremediação. Estruturado em dois eixos de atuação um físico e outro social. Este estudo se detém na análise deste último eixo, pois focalizou as ações para os catadores de materiais recicláveis, como representantes do mercado informal. A pesquisa revela as estratégias de sobrevivência, os níveis de empoderamento individual e coletivo dos catadores e a prática do aviamento mediando relações de trabalho e poder.

Em 2004 foram eleitos os vereadores que compunham a Câmara Municipal de Belém, tendo sido ocupadas às vagas deixadas por aqueles que não se reelegeram. Esse quadro da reeleição possibilitou a afirmação e fortalecimento de partidos políticos que, por vários mandatos, permanecem com representação política e assento no Parlamento Municipal. É o caso do Partido dos Trabalhadores (PT), do Partido do Movimento Democrático Brasileiro (PMDB), do Partido da Social Democracia Brasileira (PSDB), do Partido Trabalhista Brasileiro (PTB) entre outros.Esse grupo político tem estado representado no Parlamento municipal por um longo período ininterrupto. O campeão de renovação de mandato é o Vereador Pastor Paulo Queiroz da IEQ (Igreja do Evangelho Quadrangular) que está no seu quarto mandato. Em seguida, vem o Vereador Pastor Raul Batista da IURD (Igreja Universal do Reino de Deus) que está no seu terceiro mandato e o Vereador Iran Moraes da AD (Assembleia de Deus) que está no seu terceiro mandato. O ponto de partida da pesquisa, parte dos pressupostos sugeridos por Pereira e Rennó (2001) e sustentado por Downs (1958) de que, o objetivo central de um político é a busca de reeleição.Anthony Downs considera que o individuo dentro da sua racionalidade, tem a capacidade de interpretar as suas escolhas dentro de uma lógica de alternativas possíveis. Ele não se deixa ser seduzido pelos movimentos dos candidatos, mas, faz a sua escolha dentro das alternativas que se apresenta, mas mantém as escolhas racionais. Percebe-se que ao longo dos anos, os parlamentares evangélicos têm demonstrado maior força em algumas denominações em se manter no parlamento para vários mandatos. É o caso dos parlamentares da Igreja do Evangelho Quadrangular, Adventista do Sétimo Dia, Universal do reino de Deus, Assembleia de Deus. O processo de renovação contínua de mandato é baseado no apoio das lideranças e nos serviços prestados aos seguimentos internos das Igrejas.

Problem The most common DNA lesion generated by oxidative stress (OS) is 7, 8-dihydro-8-oxoguanine (8-oxoG) whose excision repair is performed by 8-oxoguanine glycosylase (OGG1). We investigated OGG1 expression changes in fetal membranes from spontaneous preterm birth (PTB) and preterm premature rupture of the membranes (pPROM) and its changes in vitro in normal fetal membranes exposed to OS inducer water-soluble cigarette smoke extract (CSE). Method of study DNA damage was determined in amnion cells treated with CSE by comet and FLARE assays. OGG1 mRNA expression and localization in fetal membranes from clinical specimens and in normal term membranes exposed to CSE were examined by QRT-PCR and by immunohistochemistry. Results DNA strand and base damage was seen in amnion cells exposed to CSE. OGG1 expression was 2.5-fold higher in PTB samples compared with pPROM (P=0.045). No significant difference was seen between term and pPROM or PTB and term. CSE treatment showed a nonsignificant decrease in OGG1. OGG1 was localized to both amnion and chorion with less intense staining in pPROM and CSE-treated membranes. Conclusion Increased OS-induced DNA damage predominated by 8-oxoG is likely to persist in fetal cells due to reduced availability of base excision repair enzyme OGG1. This can likely lead to fetal cell senescence associated with some adverse pregnancy outcome.

Seabather's eruption is a papulo-pruritic dermatitis caused by the nematocysts of the larvae of the jellyfish Linuche unguiculata retained in the clothing fibers. Previously reported in Brazil, this work describes fourteen cases that occurred in the State of Santa Catarina, in southern Brazil. The new cases observed over a short period of time (the first half of January, 2012), at the height of the summer season, should alert health teams to possible epidemics on the coast of the state of Santa Catarina.

Many adverse pregnancy outcomes (APOs), including spontaneous preterm birth (PTB), are associated with placental dysfunction. Recent clinical and experimental evidences suggest that premature aging of the placenta may be involved in these events. Although placental aging is a well-known concept, the mechanisms of aging during normal pregnancy and premature aging in APOs are still unclear. This review was conducted to assess the knowledge on placental aging related biochemical changes leading to placental dysfunction in PTB and/or preterm premature rupture of membranes (pPROM). We performed a systematic review of studies published over the last 50 years in two electronic databases (Pubmed and Embase) on placental aging and PTB or pPROM. The search yielded 554 citations, 30 relevant studies were selected for full-text review and three were included in the review. Only one study reported oxidative stress-related aging and degenerative changes in human placental membranes and telomere length reduction in fetal cells as part of PTB and/or pPROM mechanisms. Similarly, two animal studies reported findings of decidual senescence and referred to PTB mechanisms. Placental and fetal membrane oxidative damage and telomere reduction are linked to premature aging in PTB and pPROM but the risk factors and biomolecular pathways causing this phenomenon are not established in the literature. However, no biomarkers or clinical indicators of premature aging as a pathology of PTB and pPROM have been reported. We document major knowledge gaps and propose several areas for future research to improve our understanding of premature aging linked to placental dysfunction.

Abstract Background Smear-negative pulmonary tuberculosis (SNPTB) accounts for 30% of Pulmonary Tuberculosis (PTB) cases reported annually in developing nations. Polymerase chain reaction (PCR) may provide an alternative for the rapid detection of Mycobacterium tuberculosis (MTB); however little data are available regarding the clinical utility of PCR in SNPTB, in a setting with a high burden of TB/HIV co-infection. Methods To evaluate the performance of the PCR dot-blot in parallel with pretest probability (Clinical Suspicion) in patients suspected of having SNPTB, a prospective study of 213 individuals with clinical and radiological suspicion of SNPTB was carried out from May 2003 to May 2004, in a TB/HIV reference hospital. Respiratory specialists estimated the pretest probability of active disease into high, intermediate, low categories. Expectorated sputum was examined by direct microscopy (Ziehl-Neelsen staining), culture (Lowenstein Jensen) and PCR dot-blot. Gold standard was based on culture positivity combined with the clinical definition of PTB. Results In smear-negative and HIV subjects, active PTB was diagnosed in 28.4% (43/151) and 42.2% (19/45), respectively. In the high, intermediate and low pretest probability categories active PTB was diagnosed in 67.4% (31/46), 24% (6/25), 7.5% (6/80), respectively. PCR had sensitivity of 65% (CI 95%: 50%–78%) and specificity of 83% (CI 95%: 75%–89%). There was no difference in the sensitivity of PCR in relation to HIV status. PCR sensitivity and specificity among non-previously TB treated and those treated in the past were, respectively: 69%, 43%, 85% and 80%. The high pretest probability, when used as a diagnostic test, had sensitivity of 72% (CI 95%:57%–84%) and specificity of 86% (CI 95%:78%–92%). Using the PCR dot-blot in parallel with high pretest probability as a diagnostic test, sensitivity, specificity, positive and negative predictive values were: 90%, 71%, 75%, and 88%, respectively. Among non-previously TB treated and HIV subjects, this approach had sensitivity, specificity, positive and negative predictive values of 91%, 79%, 81%, 90%, and 90%, 65%, 72%, 88%, respectively. Conclusion PCR dot-blot associated with a high clinical suspicion may provide an important contribution to the diagnosis of SNPTB mainly in patients that have not been previously treated attended at a TB/HIV reference hospital.

Abstract Background Direct smear examination with Ziehl-Neelsen (ZN) staining for the diagnosis of pulmonary tuberculosis (PTB) is cheap and easy to use, but its low sensitivity is a major drawback, particularly in HIV seropositive patients. As such, new tools for laboratory diagnosis are urgently needed to improve the case detection rate, especially in regions with a high prevalence of TB and HIV. Objective To evaluate the performance of two in house PCR (Polymerase Chain Reaction): PCR dot-blot methodology (PCR dot-blot) and PCR agarose gel electrophoresis (PCR-AG) for the diagnosis of Pulmonary Tuberculosis (PTB) in HIV seropositive and HIV seronegative patients. Methods A prospective study was conducted (from May 2003 to May 2004) in a TB/HIV reference hospital. Sputum specimens from 277 PTB suspects were tested by Acid Fast Bacilli (AFB) smear, Culture and in house PCR assays (PCR dot-blot and PCR-AG) and their performances evaluated. Positive cultures combined with the definition of clinical pulmonary TB were employed as the gold standard. Results The overall prevalence of PTB was 46% (128/277); in HIV+, prevalence was 54.0% (40/74). The sensitivity and specificity of PCR dot-blot were 74% (CI 95%; 66.1%-81.2%) and 85% (CI 95%; 78.8%-90.3%); and of PCR-AG were 43% (CI 95%; 34.5%-51.6%) and 76% (CI 95%; 69.2%-82.8%), respectively. For HIV seropositive and HIV seronegative samples, sensitivities of PCR dot-blot (72% vs 75%; p = 0.46) and PCR-AG (42% vs 43%; p = 0.54) were similar. Among HIV seronegative patients and PTB suspects, ROC analysis presented the following values for the AFB smear (0.837), Culture (0.926), PCR dot-blot (0.801) and PCR-AG (0.599). In HIV seropositive patients, these area values were (0.713), (0.900), (0.789) and (0.595), respectively. Conclusion Results of this study demonstrate that the in house PCR dot blot may be an improvement for ruling out PTB diagnosis in PTB suspects assisted at hospitals with a high prevalence of TB/HIV.

Die Expression der humanen induzierbaren NO-Synthase (iNOS) wird sowohl über transkriptionelle als auch über post-transkriptionelle Mechanismen reguliert. Dabei spielt die Modulation der iNOS-mRNA-Stabilität durch RNA-bindende Proteine eine bedeutende Rolle. In dieser Arbeit konnte eine Beteiligung des p38-MAPK-Signaltransduktionsweges sowie der RNA-bindenden Proteine TTP, KSRP, HuR und PTB an der Regulation der iNOS-Expression dargestellt werden. Hemmung der p38-MAPK führte zu einer Reduktion der iNOS-mRNA-Expression, hatte aber keinen Effekt auf die iNOS-Promotoraktivität. Das RNA-bindende Protein Tristetraprolin (TTP) erhöhte die Stabilität der iNOS-mRNA nach Zytokin-Stimulation, ohne jedoch mit ihr zu interagieren. Die Proteinexpression von TTP war unter dem Einfluss von Zytokinen erhöht; Inhibition der p38-MAPK verursachte eine Verminderung der Zytokin-stimulierten TTP-Expression. Das „KH-type splicing regulatory protein" (KSRP) übte einen destabilisierenden Effekt auf die iNOS-mRNA aus. Der Abbau der mRNA wird dabei wahrscheinlich durch eine Zytokin-unabhängige Interaktion von KSRP mit dem Exosom vermittelt. Ebenso konnte zwischen KSRP und TTP eine Wechselwirkung beobachtet werden, die nach Induktion der iNOS-Expression mit Zytokinen verstärkt und durch p38-MAPK-Inhibitoren hemmbar war. Des Weiteren konnte gezeigt werden, dass die Bindung von KSRP an die iNOS-mRNA-3’-UTR für die Vermittlung des destabilisierenden Effekts essentiell ist. Eine genaue Lokalisierung der KSRP-Bindungsstelle ergab, dass KSRP ebenso wie HuR mit dem AU-reichen Element am 3’-Ende der 3’-UTR interagiert. KSRP und HuR sind in der Lage, um diese Bindungsstelle zu konkurrieren. Nach Zytokin-Stimulation war dementsprechend die endogene Bindung von KSRP an die iNOS-mRNA vermindert, während die endogene Bindung von HuR an die iNOS-mRNA verstärkt war. Die Stabilisierung der iNOS-mRNA nach Zytokin-Stimulation ergibt sich demnach aus einer Verminderung der Bindung des KSRP-Exosom-Komplexes an die iNOS-mRNA als Folge der verstärkten Interaktion von TTP und KSRP. Dies ermöglicht parallel eine vermehrte Bindung von HuR an die iNOS-3’-UTR und führt damit zu einer Stabilisierung der iNOS-mRNA und so letztendlich auch zu einer Erhöhung der iNOS-Expression. Außerdem konnte eine Beteiligung des Polypyrimidin-Trakt-bindenden Proteins (PTB) an der Regulation der humanen iNOS-Expression gezeigt werden. PTB erhöhte die Expression der iNOS und interagierte Zytokin-unabhängig mit KSRP. Zusammenfassend lässt sich schließen, dass ein Zusammenspiel verschiedener Proteine in einem komplexen Netzwerk für die fein abgestimmte Regulation der humanen iNOS-Expression auf post-transkriptioneller Ebene verantwortlich.

Light pseudoscalar bosons, such as the axion that was originally proposed as a solution of the strong CP problem, would cause a new spin-dependent short-range interaction. In this thesis, an experiment is presented to search for axion mediated short-range interaction between a nucleon and the spin of a polarized bound neutron. This interaction cause a shift in the precession frequency of nuclear spin-polarized gases in the presence of an unpolarized mass. To get rid of magnetic field drifts co-located, nuclear spin polarized 3He and 129Xe atoms were used. The free nuclear spin precession frequencies were measured in a homogeneous magnetic guiding field of about 350nT using LTc SQUID detectors. The whole setup was housed in a magnetically shielded room at the Physikalisch Technische Bundesanstalt (PTB) in Berlin. With this setup long nuclear spin-coherence times, respectively, transverse relaxation times of 5h for 129Xe and 53h for 3He could be achieved. The results of the last run in September 2010 are presented which give new upper limits on the scalar-pseudoscalar coupling of axion-like particles in the axion-mass window from 10^(-2) eV to 10^(-6) eV. The laboratory upper bounds were improved by up to 4 orders of magnitude.
