Zusammenfassung:Das Ziel dieser Arbeit ist ein besseres Verständnis von der Art und Weise wie sich Formregelungsgefüge entwicklen. Auf dieser Basis wird der Nutzen von Formregelungsgefügen für die Geologie evaluiert. Untersuchungsmethoden sind Geländearbeit und -auswertung, numerische Simulationen und Analogexperimente. Untersuchungen an Formregelungsgefügen in Gesteinen zeigen, daß ein Formregelungsgefüge nur zu einem begrenzten Grad als Anzeiger für die Stärke der Verformung benutzt werden kann. Der angenommene Grund hierfür ist der Einfluß des Verhältnisses von ursprünglicher zu rekristallisierter Korngröße auf die Gefügeentwicklung und von der Art und Weise wie dynamische Rekristallisation ein Gefüge verändert. Um diese Beobachtung zu evaluieren, wurden verschiedene numerische Simulationen von dynamischer Rekristallisation durchgeführt. Ein neuer Deformationsapparat, mit dem generelle Fließregime modelliert werden können, wurde entwickelt. Die rheologischen Eigenschaften von Materialien, die für solche Experimente benutzt werden, wurden untersucht und diskutiert. Ergebnisse von Analogexperimenten zeigen, daß die Intensität eines Formregelungsgefüges positiv mit der Abnahme der 'kinematic vorticity number' und einem nicht-Newtonianischen, 'power law' Verhalten des Materixmaterials korreliert ist. Experimente, in denen die Formveränderung von viskosen Einschlüssen während der progressiven Verformung modelliert werden, zeigen, daß verschiedene Viskositätskontraste zwischen Matrix- und Einschlußmaterial in charakteristische Formgefüge resultieren.

Mögliche Verformungsmechanismen, die zu den verschiedenen Glimmer- und Mineralfischen führen, sind: intrakristalline Verformung, Kristallrotation, Biegung und Faltung, Drucklösung in Kombination mit Ausfällung und dynamische Rekristallisation oder Mechanismen, die ein großes Mineral in mehrere kleine, fischförmige Kristalle aufspalten.Experimente mit ein neues Verformungsgerät und Objekten in zwei verschiedenen Matrixmaterialien werden beschrieben. Das eine ist PDMS, (Newtonianisch viskoses Polymer), und das andere Tapioca Perlen (Mohr-Couloumb Verhalten). Die Rotation von fischförmigen Objekten in PDMS stimmt mit der theoretischen Rotationsrate für ellipsenförmige Objekte in einem Newtonianischen Material überein. In einer Matrix von Tapioca Perlen nehmen die Objekte eine stabile Lage ein. Diese Orientierung ist vergleichbar mit der von Glimmerfischen. Die Verformung in der Matrix von Tapioca Perlen ist konzentriert auf dünne Scherzonen. Diese Ergebnisse implizieren, daß die Verformung in natürlichen Gesteinen auch in dünnen Scherzonen konzentriert ist.Computersimulationen werden beschrieben, mit denen der Einfluß der Eigenschaften einer Matrix auf die Rotation von Objekten und Verteilung von Deformation untersucht wird.Mit diesen Experimenten wird gezeigt, daß die Orientierung von Glimmerfischen nicht mit Verformung in einem nicht-linearen viskosen Material erklärt werden kann. Eine solche nicht-lineare Rheologie wird im Allgemeinen für die Erdkurste angenommen. Die stabile Orientierung eines Objektes kann mit weicheren Lagen in der Matrix erklärt werden.

In dieser Arbeit wurden wässrige Suspensionen ladungsstabilisierter kolloidaler Partikel bezüglich ihres Verhaltens unter dem Einfluss elektrischer Felder untersucht. Insbesondere wurde die elektrophoretische Mobilität µ über einen weiten Partikelkonzentrationsbereich studiert, um das individuelle Verhalten einzelner Partikel mit dem bisher nur wenig untersuchten kollektiven Verhalten von Partikelensembles (speziell von fluid oder kristallin geordneten Ensembles) zu vergleichen. Dazu wurde ein superheterodynes Dopplervelocimetrisches Lichtstreuexperiment mit integraler und lokaler Datenerfassung konzipiert, das es erlaubt, die Geschwindigkeit der Partikel in elektrischen Feldern zu studieren. Das Experiment wurde zunächst erfolgreich im Bereich nicht-ordnender und fluid geordneter Suspensionen getestet. Danach konnte mit diesem Gerät erstmals das elektrophoretische Verhalten von kristallin geordneten Suspensionen untersucht werden. Es wurde ein komplexes Fließverhalten beobachtet und ausführlich dokumentiert. Dabei wurden bisher in diesem Zusammenhang noch nicht beobachtete Effekte wie Blockfluss, Scherbandbildung, Scherschmelzen oder elastische Resonanzen gefunden. Andererseits machte dieses Verhalten die Entwicklung einer neuen Auswertungsroutine für µ im kristallinen Zustand notwendig, wozu die heterodyne Lichtstreutheorie auf den superheterodynen Fall mit Verscherung erweitert werden musste. Dies wurde zunächst für nicht geordnete Systeme durchgeführt. Diese genäherte Beschreibung genügte, um unter den gegebenen Versuchbedingungen auch das Lichtstreuverhalten gescherter kristalliner Systeme zu interpretieren. Damit konnte als weiteres wichtiges Resultat eine generelle Mobilitäts-Konzentrations-Kurve erhalten werden. Diese zeigt bei geringen Partikelkonzentrationen den bereits bekannten Anstieg und bei mittleren Konzentrationen ein Plateau. Bei hohen Konzentrationen sinkt die Mobilität wieder ab. Zur Interpretation dieses Verhaltens bzgl. Partikelladung stehen derzeit nur Theorien für nicht wechselwirkende Partikel zur Verfügung. Wendet man diese an, so findet man eine überraschend gute Übereinstimmung der elektrophoretisch bestimmten Partikelladung Z*µ mit numerisch bestimmten effektiven Partikelladungen Z*PBC.

This work presents a comprehensive methodology for the reduction of analytical or numerical stochastic models characterized by uncertain input parameters or boundary conditions. The technique, based on the Polynomial Chaos Expansion (PCE) theory, represents a versatile solution to solve direct or inverse problems related to propagation of uncertainty. The potentiality of the methodology is assessed investigating different applicative contexts related to groundwater flow and transport scenarios, such as global sensitivity analysis, risk analysis and model calibration. This is achieved by implementing a numerical code, developed in the MATLAB environment, presented here in its main features and tested with literature examples. The procedure has been conceived under flexibility and efficiency criteria in order to ensure its adaptability to different fields of engineering; it has been applied to different case studies related to flow and transport in porous media. Each application is associated with innovative elements such as (i) new analytical formulations describing motion and displacement of non-Newtonian fluids in porous media, (ii) application of global sensitivity analysis to a high-complexity numerical model inspired by a real case of risk of radionuclide migration in the subsurface environment, and (iii) development of a novel sensitivity-based strategy for parameter calibration and experiment design in laboratory scale tracer transport.

Surface tension induced convection in a liquid bridge held between two parallel, coaxial, solid disks is considered. The surface tension gradient is produced by a small temperature gradient parallel Co the undisturbed surface. The study is performed by using a mathematical regular perturbation approach based on a small parameter, e, which measures the deviation of the imposed temperature field from its mean value. The first order velocity field is given by a Stokes-type problem (viscous terms are dominant) with relatively simple boundary conditions. The first order temperature field is that imposed from the end disks on a liquid bridge immersed in a non-conductive fluid. Radiative effects are supposed to be negligible. The second order temperature field, which accounts for convective effects, is split into three components, one due to the bulk motion, and the other two to the distortion of the free surface. The relative importance of these components in terms of the heat transfer to or from the end disks is assessed

La fatiga superficial es uno de los principales problemas en las transmisiones mecánicas y es uno de los focos de atención de las investigaciones de los últimos anos en Tribología. La disminución de viscosidad de los lubricantes para la mejora de la eficiencia, el aumento de las potencias a transmitir, el aumento de la vida de los componentes o la mejora de su fiabilidad han supuesto que los fenómenos de fatiga superficial hayan cobrado especial relevancia, especialmente los fenómenos de pitting y micropitting en cajas multiplicadoras/reductoras de grandes potencias de aplicación, por ejemplo, en el sector eólico. Como todo fenómeno de fatiga, el pitting y micropitting son debidos a la aplicación de cargas ciclicas. Su aparición depende de las presiones y tensiones cortantes en el contacto entre dos superficies que al encontrarse en rodadura y deslizamiento varian con el tiempo. La principal consecuencia de la fatiga superficial es la aparición de hoyuelos de diferente magnitud segun la escala del fenómeno (pitting o micropitting) en la superficie del material. La aparición de estos hoyuelos provoca la perdida de material, induce vibraciones y sobrecargas en el elemento que finalmente acaba fallando. Debido a la influencia de la presión y tensión cortante en el contacto, la aparición de fatiga depende fuertemente del lubricante que se encuentre entre las dos superficies y de las condiciones de funcionamiento en las cuales este trabajando. Cuando el contacto trabaja en condiciones de lubricacion mixta-elastohidrodinamica tiende a aparecer micropitting debido a las altas tensiones localizadas en las proximidades de las asperezas, mientras que si el régimen es de lubricación completa el tipo de fatiga superficial suele ser pitting debido a las tensiones mas suavizadas y menos concentradas. En esta Tesis Doctoral se han analizado todos estos factores de influencia que controlan el pitting y el micropitting prestando especial atención al efecto del lubricante. Para ello, se ha dado un enfoque conjunto a ambos fenómenos resolviendo las ecuaciones involucradas en el contacto elastohidrodinamico no-Newtoniano (la ecuación de Reynolds, la deformación elástica de los sólidos y la reologia del lubricante) para conocer la presión y la tensión cortante en el contacto. Conocidas estas, se resuelve el campo de tensiones en el interior del material y, finalmente, se aplican criterios de fatiga multiaxial (Crossland, Dang Van y Liu-Mahadevan) para conocer si el material falla o no falla. Con la metodología desarrollada se ha analizado el efecto sobre las tensiones y la aparición de la fatiga superficial del coeficiente viscosidad-presion, de la compresibilidad, del espesor especifico de película y de la fricción así como de la influencia de las propiedades a fatiga del material y de las condiciones de funcionamiento (radios de contacto, velocidad, deslizamiento, carga y temperatura). Para la validación de los resultados se han utilizado resultados teóricos y experimentales de otros autores junto con normas internacionales de amplia utilización en el mundo industrial, entre otras, para el diseño y calculo de engranajes. A parte del trabajo realizado por simulación y cálculo de los diferentes modelos desarrollados, se ha realizado un importante trabajo experimental que ha servido no solo para validar la herramienta desarrollada sino que además ha permitido incorporar al estudio factores no considerados en los modelos, como los aditivos del lubricante. Se han realizado ensayos de medida del coeficiente de fricción en una maquina de ensayo puntual con la que se ha validado el cálculo del coeficiente de fricción y se ha desarrollado un proceso de mejora del coeficiente de fricción mediante texturizado superficial en contactos puntuales elastohidrodinamicos mediante fotolitografia y ataque quimico. Junto con los ensayos de medida de fricción en contacto puntual se han realizado ensayos de fricción y fatiga superficial en contacto lineal mediante una maquina de discos que ha permitido evaluar la influencia de diferentes aditivos (modificadores de fricción, antidesgaste y extrema-presion) en la aparición de fatiga superficial (pitting y micropitting) y la fricción en el contacto. Abstract Surface fatigue is one of the most important problems of mechanical transmissions and therefore has been one of the main research topics on Tribology during the last years. On the one hand, industrial demand on fuel economy has led to reduce lubricant viscosity in order to improve efficiency. On the other hand, the requirements of power and life of machine elements are continuously increasing, together with the improvements in reliability. As a consequence, surface fatigue phenomena have become critical in machinery, in particular pitting and micropitting in high power gearboxes of every kind of machines, e.g., wind turbines or cranes. In line with every fatigue phenomena, pitting and micropitting are caused by cyclic loads. Their appearance depends on the evolution of pressures and shear stresses with time, throughout the contact between surfaces under rolling and sliding conditions. The main consequence of surface fatigue is the appearance of pits on the surface. The size of the pits is related to the scale of the fatigue: pitting or micropitting. These pits cause material loss, vibrations and overloads until the final failure is reached. Due to the great influence of the pressures and shear stresses in surface fatigue, the appearance of pits depends directly on the lubricant and the operating conditions. When the contact works under mixed regime (or under elastohydrodynamic but close to mixed regime) the main fatigue failure is micropitting because of the high pressures located near the asperities. In contrast, when the contact works under elastohydrodynamic fully flooded conditions the typical fatigue failure is pitting. In this Ph.D. Thesis, the main factors with influence on pitting and micropitting phenomena are analyzed, with special attention to the effect of the lubricant. For this purpose, pitting and micropitting are studied together by solving the equations involved in the non-Newtonian elastohydrodynamic contact. Thus, pressure and shear stress distributions are found by taking into account Reynolds equation, elastic deflection of the solids and lubricant rheology. Subsequently, the stress field inside the material can be calculated and different multiaxial fatigue criteria (Crossland, Dang Van and Liu- Mahadevan) can be applied to predict whether fatigue failure is reached. The influences of the main parameters on pressure and surface fatigue have been studied, taking into account the lubricant compressibility and its viscosity-pressure coefficient, the specific film thickness, the friction coefficient and the fatigue properties of the contacting materials, together with the operating conditions (contact radius, mean velocity, sliding velocity, load and temperature). Several theoretical and experimental studies of different authors have been used to validate all the results obtained, together with international standards used worldwide in gear design industry. Moreover, an experimental stage has been carried out in order to validate the calculation methods and introduce additional influences not included previously, e.g., lubricant additives. The experimentation includes different friction tests in point contacts performed with a tribological equipment in order to validate the results given by the calculations. Furthermore, the reduction and optimization of the friction coefficient is analyzed by means of textured surfaces, obtained combining photolithography and chemical etching techniques. Besides the friction tests with point contact, friction and surface fatigue tests have also been performed with line contact in a tribological test rig. This equipment is also used to study the influence of different types of additives (friction modifiers, anti-wear and extreme-pressure additives) on surface fatigue (pitting and micropitting).

To “control” a system is to make it behave (hopefully) according to our “wishes,” in a way compatible with safety and ethics, at the least possible cost. The systems considered here are distributed—i.e., governed (modeled) by partial differential equations (PDEs) of evolution. Our “wish” is to drive the system in a given time, by an adequate choice of the controls, from a given initial state to a final given state, which is the target. If this can be achieved (respectively, if we can reach any “neighborhood” of the target) the system, with the controls at our disposal, is exactly (respectively, approximately) controllable. A very general (and fuzzy) idea is that the more a system is “unstable” (chaotic, turbulent) the “simplest,” or the “cheapest,” it is to achieve exact or approximate controllability. When the PDEs are the Navier–Stokes equations, it leads to conjectures, which are presented and explained. Recent results, reported in this expository paper, essentially prove the conjectures in two space dimensions. In three space dimensions, a large number of new questions arise, some new results support (without proving) the conjectures, such as generic controllability and cases of decrease of cost of control when the instability increases. Short comments are made on models arising in climatology, thermoelasticity, non-Newtonian fluids, and molecular chemistry. The Introduction of the paper and the first part of all sections are not technical. Many open questions are mentioned in the text.

A nistatina (NYS) é o fármaco de primeira escolha no tratamento da candidíase oral, que frequentemente acomete mais os indivíduos imunocomprometidos e pacientes com outras desordens (diabetes não tratada, neoplasias, imunodeficiências). No mercado brasileiro, a NYS é encontrada na forma de suspensão oral aquosa, onde o procedimento para sua administração consiste em bochechar o medicamento. Apesar de haver a indicação de que se mantenha o contato direto entre fármaco e a mucosa oral, na qual se encontra a Candida spp., o que aumentaria expressivamente o sucesso terapêutico, a suspensão não apresenta tal propriedade. Assim, a NYS que é fármaco com ação efetiva contra a candidíase oral, é considerada pertencente à Classe IV do Sistema de Classificação Biofarmacêutica, ou seja, apresenta baixa solubilidade e baixa permeabilidade. A baixa solubilidade pode comprometer sua disponibilidade na cavidade oral, e consequentemente, sua ação farmacológica. Diante desse quadro, o objetivo do presente trabalho foi o desenvolvimento de dispersões sólidas de NYS para o tratamento da candidíase oral, e sua posterior incorporação em gel mucoadesivo oral, favorecendo a formulação no local de ação. As dispersões sólidas são sistemas farmacêuticos, onde um fármaco pouco solúvel em água encontra-se dispersado em um carreador, no estado sólido. Os carreadores normalmente são hidrofílicos, o que permite que esses sistemas sejam empregados para aumentar a solubilidade aquosa do fármaco. Assim, foram desenvolvidas as dispersões sólidas de NYS, pelo método de eliminação do solvente, empregando como carreadores, lactose, HPMC, poloxamer 407 e poloxamer 188. Essas foram submetidas à caracterização por análise térmica, usando os ensaios de calorimetria exploratória diferencial (DSC) e termogravimetria/termogravimetria derivada (TG/DTG). Dentre essas dispersões sólidas, aquelas que se mostraram com comportamento térmico sugerindo a formação de um novo \"sistema\", foram analisadas por meio de ensaio de solubilidade. Dessa forma, a formulação NYS DS G2 (49) se destacou, pois apresentou maior solubilidade em água (4,484 mg/mL); em pH 5,5 (4,249 mg/mL) e em pH 7,0 (4,293 mg/mL), ou seja, houve um aumento de 1,426 vezes em água; 4,227 vezes em pH 5,5; e 2,743 vezes em pH 7,0. Essa formulação foi, por fim avaliada por difração de raio-X e espectroscopia de infravermelho com transformada de Fourier, técnicas que corroboraram com a análise térmica quanto à indicação de formação da dispersão sólida. Por sua vez, essa dispersão sólida foi incorporada em 4 bases de géis mucoadesivos de carbopol ® 934 PNF, alterando apenas a concentração do polímero (0,5; 1,0; 1,5; 2,0 %p/p). Foi observado que a liberação de NYS DS G2 (49) foi superior, quando comparada à liberação de NYS MP a partir do gel, e através do ensaio de mucoadesão, percebeu-se que os géis desenvolvidos apresentaram propriedades mucoadesivas compatíveis com relatos na literatura, independentemente da quantidade de carbopol ® empregada. As características reológicas foram distintas, e foi observado que as formulações Gel A e Gel B, que possuem menor quantidade de polímero, tiverem um indicativo de comportamento de fluido newtoniano, diferente dos demais, o que pode não ser desejado para esse tipo de forma farmacêutica tópica e semi-sólida. Ao final desse trabalho, pode-se concluir que foi possível desenvolver um sistema farmacêutico na forma de dispersão sólida com maior solubilidade que a NYS pura, e sua incorporação em uma forma farmacêutica mucoadesiva, e que a liberação da NYS na forma DS foi muito superior que o fármaco na forma \"convencional\", o que permite que a NYS esteja mais disponível na cavidade oral, e também junto à mucosa bucal, o que levaria a efeito farmacológico mais efetivo do antifúngico.

A argila bentonítica é amplamente utilizada em transporte de sólidos produzidos durante perfuração de poços. Teve por objetivo estudar o escoamento de misturas bentonita-água e determinar suas propriedades reológicas e parâmetros hidráulicos úteis nos projetos de instalações de recalque de misturas sólido-líquido. Foi montado um circuito fechado de tubulações para estudar dados de perda de carga e perfis de velocidade. Realizaram-se ensaios com misturas bentonita-água sob varias concentrações, algumas transportando areia. Observaram-se que a reologia da mistura bentonita-água é melhor descrita pela formulação de Herschell-Bulkley para fluidos não-Newtonianos. O coeficiente de atrito para descrever a perda de carga da mistura bentonita-água observada em tubulações no laboratório coloca-se entre as previsões de Tomita (1959) e Szilas et al (1981). A variação da velocidade da mistura na seção transversal do tubo é melhor aproximada pela equação de Bogue-Metzner (1963).

We consider the simplest relevant problem in the foaming of molten plastics, the growth of a single bubble in a sea of highly viscous Newtonian fluid, and without interference from other bubbles. This simplest problem has defied accurate solution from first principles. Despite plenty of research on foaming, classical approaches from first principles have neglected the temperature rise in the surrounding fluid, and we find that this oversimplification greatly accelerates bubble growth prediction. We use a transport phenomena approach to analyze the growth of a solitary bubble, expanding under its own pressure. We consider a bubble of ideal gas growing without the accelerating contribution from mass transfer into the bubble. We explore the roles of viscous forces, fluid inertia, and viscous dissipation. We find that bubble growth depends upon the nucleus radius and nucleus pressure. We begin with a detailed examination of the classical approaches (thermodynamics without viscous heating). Our failure to fit experimental data with these classical approaches, sets up the second part of our paper, a novel exploration of the essential decelerating role of viscous heating. We explore both isothermal and adiabatic bubble expansion, and also the decelerating role of surface tension. The adiabatic analysis accounts for the slight deceleration due to the cooling of the expanding gas, which depends on gas polyatomicity. We also explore the pressure profile, and the components of the extra stress tensor, in the fluid surrounding the growing bubble. These stresses can eventually be frozen into foamed plastics. We find that our new theory compares well with measured bubble behavior.

Subaerial debris flows, with water contents ranging from as little as 10 wt% up to no more than about 25 wt% (Pierson, 1986; Pierson and Costa, 1987), are non-Newtonian fluids that move as fairly coherent masses with yield strength (owing to bulk densities and viscosity that are much greater than those of clear water), which enables them to suspend and transport large clasts. Their flow behavior is thought to be predominantly laminar, although the relative importance of laminar and turbulent flow has not been established and is debatable. They leave deposits (debrites) that are characteristically poorly sorted with large clasts in their middle portions and commonly protruding from their tops. Although generally ungraded or normally graded in their upper portions, many have centimeter- to decimeter-thick inversely graded basal zones (Arguden and Rodolfo, 1990, doi:10.1130/0016-7606(1990)102<0865:SADDBH>2.3.CO;2).

The superplastic deformation behavior and superplastic forming ability of the Zr41.25Ti13.75Ni10Cu12.5Be22.5 (at.%) bulk metallic glass (BMG) in the supercooled liquid region were investigated. The isothermal tensile results indicate (hat the BMG exhibits a Newtonian behavior at low strain rates but a non-Newtonian behavior at hiqh-strain rates in the initial deformation stage. The maximum elongation reaches as high as 1624% at 656 K. and nanocrystallization was found to occur during the deformation process. Based cm the analysis on tensile deformation. a gear-like micropart is successfully die-forged via a superplastic forgings process. demonstrating that the BMG has excellent workability in the supercooled liquid region. The dynamics of drop formation and pinch-off have been investigated for a series of low viscosity elastic fluids possessing similar shear viscosities, but differing substantially in elastic properties. On initial approach to the pinch region, the viscoelastic fluids all exhibit the same global necking behavior that is observed for a Newtonian fluid of equivalent shear viscosity. For these low viscosity dilute polymer solutions, inertial and capillary forces form the dominant balance in this potential flow regime, with the viscous force being negligible. The approach to the pinch point, which corresponds to the point of rupture for a Newtonian fluid, is extremely rapid in such solutions, with the sudden increase in curvature producing very large extension rates at this location. In this region the polymer molecules are significantly extended, causing a localized increase in the elastic stresses, which grow to balance the capillary pressure. This prevents the necked fluid from breaking off, as would occur in the equivalent Newtonian fluid. Alternatively, a cylindrical filament forms in which elastic stresses and capillary pressure balance, and the radius decreases exponentially with time. A (0+1)-dimensional finitely extensible nonlinear elastic dumbbell theory incorporating inertial, capillary, and elastic stresses is able to capture the basic features of the experimental observations. Before the critical "pinch time" t(p), an inertial-capillary balance leads to the expected 2/3-power scaling of the minimum radius with time: R-min similar to(t(p)-t)(2/3). However, the diverging deformation rate results in large molecular deformations and rapid crossover to an elastocapillary balance for times t>t(p). In this region, the filament radius decreases exponentially with time R-min similar to exp[(t(p)-t)/lambda(1)], where lambda(1) is the characteristic time constant of the polymer molecules. Measurements of the relaxation times of polyethylene oxide solutions of varying concentrations and molecular weights obtained from high speed imaging of the rate of change of filament radius are significantly higher than the relaxation times estimated from Rouse-Zimm theory, even though the solutions are within the dilute concentration region as determined using intrinsic viscosity measurements. The effective relaxation times exhibit the expected scaling with molecular weight but with an additional dependence on the concentration of the polymer in solution. This is consistent with the expectation that the polymer molecules are in fact highly extended during the approach to the pinch region (i.e., prior to the elastocapillary filament thinning regime) and subsequently as the filament is formed they are further extended by filament stretching at a constant rate until full extension of the polymer coil is achieved. In this highly extended state, intermolecular interactions become significant, producing relaxation times far above theoretical predictions for dilute polymer solutions under equilibrium conditions. The aim of this study was to develop and characterize an intranasal delivery system for amantadine hydrochloride (AMT). Optimal formulations consisted of a thermosensitive polymer Pluronic® 127 and either carboxymethyl cellulose or chitosan which demonstrated gel transition at nasal cavity temperatures (34 ± 1°C). Rheologically, the loss tangent (Tan δ) confirmed a 3-stage gelation phenomena at 34 ± 1°C and non-Newtonian behavior. Storage of optimized formulation carboxymethyl cellulose and optimal formulation chitosan at 4°C for 8 weeks resulted in repeatable release profiles at 34°C when sampled, with a Fickian mechanism earlier on but moving toward anomalous transport by week 8. Polymers (Pluronic® 127, carboxymethyl cellulose, and chitosan) demonstrated no significant cellular toxicity to human nasal epithelial cells up to 4 mg/mL and up to 1 mM for AMT (IC50: 4.5 ± 0.05 mM). Optimized formulation carboxymethyl cellulose and optimal formulation chitosan demonstrated slower release across an in vitro human nasal airway model (43%-44% vs 79 ± 4.58% for AMT). Using a human nasal cast model, deposition into the olfactory regions (potential nose-to-brain) was demonstrated on nozzle insertion (5 mm), whereas tilting of the head forward (15°) resulted in greater deposition in the bulk of the nasal cavity.

Alginate is widely used as a viscosity enhancer in many different pharmaceutical formulations. The aim of this thesis is to quantitatively describe the functions of this polyelectrolyte in pharmaceutical systems. To do this the techniques used were Viscometry, Light Scattering, Continuous and Oscillatory Shear Rheometry, Numerical Analysis and Diffusion. Molecular characterization of the Alginate was carried out using Viscometry and Light Scattering to determine the molecular weight, the radius of gyration, the second virial coefficient and the Kuhn statistical segment length. The results showed good agreement with similar parameters obtained in previous studies. By blending Alginate with other polyelectrolytes, Xanthan Gum and 'Carbopol', in various proportions and with various methods of low and high shear preparation, a very wide range of dynamic rheological properties was found. Using oscillatory testing, the parameters often varied over several decades of magnitude. It was shown that the determination of the viscous and elastic components is particularly useful in describing the rheological 'profiles' of suspending agent blends and provides a step towards the non-empirical formulation of pharmaceutical disperse systems. Using numerical analysis of equations describing planar diffusion, it was shown that the analysis of drug release profiles alone does not provide unambiguous information about the mechanism of rate control. These principles were applied to the diffusion of Ibuprofen in Calcium Alginate gels. For diffusion in such non-Newtonian systems, emphasis was placed on the use of the elastic as well as the viscous component of viscoelasticity. It was found that the diffusion coefficients were relatively unaffected by increases in polymer concentration up to 5 per cent, yet the elasticities measured by oscillatory shear rheometry were increased. This was interpreted in the light of several theories of diffusion in gels.
