A Gram-negative, rod-shaped, non-spore-forming and nitrogen-fixing bacterium, designated ICB 89(T), was isolated from stems of a Brazilian sugar cane variety widely used in organic farming. 16S rRNA gene sequence analysis revealed that strain ICB 89(T) belonged to the genus Stenotrophomonas and was most closely related to Stenotrophomonas maltophilia LMG 958(T), Stenotrophomonas rhizophila LMG 22075(T), Stenotrophomonas nitritireducens L2(T), [Pseudomonas] geniculata ATCC 19374(T), [Pseudomonas] hibiscicola ATCC 19867(T) and [Pseudomonas] beteli ATCC 19861(T). DNA-DNA hybridization together with chemotaxonomic data and biochemical characteristics allowed the differentiation of strain ICB 89(T) from its nearest phylogenetic neighbours. Therefore, strain ICB 89(T) represents a novel species, for which the name Stenotrophomonas pavanii sp. nov. is proposed. The type strain is ICB 89(T) (=CBMAI 564(T) =LMG 25348(T)).

Acanthamoeba spp., known to cause keratitis and granulomatous encephalitis in humans, are frequently isolated from a variety of water sources. Here we report for the first time the characterization of an Acanthamoeba sp. (ACC01) isolated from tap water in Brazil. This organism is currently being maintained in an axenic growth medium. Phylogenetic analysis based on SSU rRNA gene sequences positioned the new isolate in genotype T4, closest to the keratitis-causing isolate, A. polyphaga ATCC 30461 (similar to 99% similarity). Acanthamoeba ACC01 and A. polyphaga 30461 both grew at 37 degrees C and were osmotically resistant, multiplying in hyperosmolar medium. Both isolates secreted comparable amounts of proteolytic enzymes, including serine peptidases that were optimally active at a near neutral/alkaline pH and resolved identically in gelatin gels. Incubation of gels at pH 4.0 with 2 mM DTT also indicated the secretion of similar cysteine peptidases. Altogether, the results point to the pathogenic potential of Acanthamoeba ACC01. Ureaplasma diversum infection in bulls may result in seminal vesiculitis, balanoposthitis and alterations in spermatozoids. In cows, it can cause placentitis, fetal alveolitis, abortion and the birth of weak calves. U. diversum ATCC 49782 (serogroups A), ATCC 49783 (serogroup C) and 34 field isolates were used for this study. These microorganisms were submitted to Polymerase Chain Reaction for 16S gene sequence determination using Tact High Fidelity and the products were purified and bi-directionally sequenced. Using the sequence obtained, a fragment containing four hypervariable regions was selected and nucleotide polymorphisms were identified based on their position within the 16S rRNA gene. Forty-four single nucleotide polymorphisms (SNP) were detected. The genotypic variability of the 16S rRNA gene of U. diversum isolates shows that the taxonomy classification of these organisms is likely much more complex than previously described and that 16S rRNA gene sequencing may be used to suggest an epidemiologic pattern of different origin strains. P>Although photodynamic therapy (PDT) has shown great promise for the inactivation of Candida species, its effectiveness against azole-resistant pathogens remains poorly documented. This in vitro study describes the association of Photogem (R) (Photogem, Moscow, Russia) with LED (light emitting diode) light for the photoinactivation of fluconazole-resistant (FR) and American Type Culture Collection (ATCC) strains of Candida albicans and Candida glabrata. Suspensions of each Candida strain were treated with five Photogem (R) concentrations and exposed to four LED light fluences (14, 24, 34 or 50 min of illumination). After incubation (48 h at 37 degrees C), colonies were counted (CFU ml-1). Single-species biofilms were generated on cellulose membrane filters, treated with 25.0 mg l-1 of Photogem (R) and illuminated at 37.5 J cm-2. The biofilms were then disrupted and the viable yeast cells present were determined. Planktonic suspensions of FR strains were effectively killed after PDT. It was observed that the fungicidal effect of PDT was strain-dependent. Significant decreases in biofilm viability were observed for three strains of C. albicans and for two strains of C. glabrata. The results of this investigation demonstrated that although PDT was effective against Candida species, fluconazole-resistant strains showed reduced sensitivity to PDT. Moreover, single-species biofilms were less susceptible to PDT than their planktonic counterparts.

The development of more efficient anti-tuberculosis drugs is of interest. Three oxovanadium(IV) and three cis-dioxovanadium(V) complexes with thiosemicarbazone derivatives bearing moieties with different lipophilicity have been prepared and had their inhibitory activity against Mycobacterium tuberculosis H(37)Rv ATCC 27294 evaluated. The analytical methods used by the complexes` characterization included IR, EPR, (1)H, (13)C and (51)V NMR spectroscopies, elemental analysis, cyclic voltammetry, magnetic susceptibility measurement and single crystal X-ray diffractometry. [VO(acac)(aptsc)], [VO(acac)(apmtsc)] and [VO(acac)(apptsc)] (acac = acetylacetonate; Haptsc = 2-acetylpyridinethiosemicarbazone; Hapmtsc = 2-acetylpyridine-N(4)-methyl-thiosemicarbazone and Happtsc = 2-acetylpyridine-N(4)-phenyl-thiosemicarbazone) are paramagnetic and their EPR spectra are consistent with the monoanionic N,N,S-tridentate coordination of the thiosemicarbazone ligands, resulting in octahedral structures of rhombic symmetry and with the oxidation state +IV for the vanadium atom. As result of oxidation of the vanadium(IV) complexes above, the diamagnetic cis-dioxovanadium(V) complexes [VO(2)(aptsc)[, [VO(2)(apmtsc)[ and [VO(2)(apptsc)] are formed. Their (1)H, (13)C and (51)V NMR spectra were acquired and support a distorted square pyramidal geometry for them, in accord with the solid state X-ray structures determined for [VO(2)(aptsc)] and [VO(2)(apmtsc)]. In general, the vanadium compounds show comparable or larger anti-M. tuberculosis activities than the free thiosemicarbazone ligands, with MIC values within 62.5-1.56 (mu g/mL). The reaction of cis-[RuCl2(dppb)(N-N)], dppb = 1,4-bis(diphenylphosphino)butane, complexes with the ligand HSpymMe(2), 4,6-dimethyl-2-mercaptopyrimidine, yielded the cationic complexes [Ru(SpymMe(2))(dppb)(N-N)]PF6, N-N = bipy (1) and Me-bipy (2), bipy = 2,2`-bipyridine and Me-bipy = 4,4`dimethyl-2,2`-bipyridine, which were characterized by spectroscopic and electrochemical techniques and X-ray crystallography and elemental analysis. Additionally, preliminary in vitro tests for antimycobacterial activity against Mycobacterium tuberculosis H37Rv ATCC 27264 and antitumor activity against the MDA-MB-231 human breast tumor cell line were carried out on the new complexes and also on the precursors cis-[RuCl2(dppb)(N-N)], N-N = bipy (3) and Me-bipy (4) and the free ligands dppb, bipy, Me-bipy and SpymMe(2). The minimal inhibitory concentration (MIC) of compounds needed to kill 90% of mycobacterial cells and the IC50 values for the antitumor activity were determined. Compounds 1-4 exhibited good in vitro activity against M. tuberculosis, with MIC values ranging between 0.78 and 6.25 mu g/mL, compared to the free ligands (MIC of 25 to >50 mu g/mL) and the drugs used to treat tuberculosis. Complexes I and 2 also showed promising antitumor activity, with IC50 values of 0.46 +/- 0.02 and 0.43 +/- 0.08 mu M, respectively, against MDA-MB-231 breast tumor cells. (C) 2008 Elsevier Inc. The chemical composition of the essential oil of Rollinia sericea (R.E.Fr.) R.E.Fr. leaves was determined by GC and GC/MS analysis. The analysis revealed the presence mainly of sesquiterpenes: beta-elemene (10%), beta-caryophyllene (10.0%), bicyclogermacrene (9.1%), germacrene-D (8.2%), bicycloelemene (6.2%) and (Z)-nerolidol (5.3%). Rollinia sericea oil was able to inhibit the growth of both fungi Aspergillus niger (16404) and Candida albicans (ATCC 10231) as well as the Gram-positive bacterium Staphyloccocus aureus (ATCC 6538) but it was inactive against the Gram-negative bacteria Escherichia coli (ATCC 8739) and Pseudomonas aeruginosa (ATCC 9027).

Although the production of patulin in apple fruits is mainly by Penicillium expansum, there is no information on the ability of heat resistant moulds that may survive pasteurization to produce this mycotoxin in juice packages during storage and distribution. In this study, the production of patulin by Byssochlamys spp (Byssochlamys nivea FRR 4421, B. nivea ATCC 24008 and Byssochlamys fulva IOC 4518) in cloudy and clarified apple juices packaged in laminated paperboard packages or in polyethylene terephthalate bottles (PET) and stored at both 21 degrees C and 30 degrees C, was investigated. The three Byssochlamys strains were able to produce patulin in both cloudy and clarified apple juices. Overall, the lower the storage temperature, the lower the patulin levels and mycelium dry weight in the apple juices (p<0.05). The greatest variations in pH and degrees Brix were observed in the juices from which the greatest mycelium dry weights were recovered. The maximum levels of patulin recovered from the juices were ca. 150 mu g/kg at 21 degrees C and 220 mu g/kg at 30 degrees C. HPLC-UV, HPCL-DAD and mass spectrometry analyses confirmed the ability of B. fulva IOC 4518 to produce patulin. Due to the heat resistance of B. nivea and B. fulva and their ability to produce patulin either in PET bottles or in laminated paperboard packages, the control of contamination and the incidence of these fungi should be a matter of concern for food safety. Control measures taken by juice industries must also focus on controlling the ascospores of heat resistant moulds. (C) 2010 Elsevier B.V. All rights reserved.

The chemical composition and the antimicrobial activity of the essential oil from Croton heterocalyx leaves were evaluated. The oil which was analyzed by GC and GUMS was found to contain germacrene D (12.5%), bicyclogermacrene (11.2%), delta-elemene (9.2%) beta-elemene (8.2%), spathulenol (6.9%), linalool (5.4%) and 1,8-cineole (3.7%) its major components. Croton. heterocalyx oil displayed a high inhibitory activity against the fungi Aspergillus niger (16404) and Candida albicans (ATCC 10231.) as well its the Gram-positive bacterium Staphylococcus aureus (ATCC 6538), hut a very weak activity was observed for the Gram-negative bacteria Escherichia coli (ATCC 8739) and Pseudomonas aeruginosa (ATCC 9027).

Soybean oil soapstock was utilized as an alternative carbon source for the production of rhamnolipids by Pseudomonas aeruginosa LBI strain. The chemical composition and properties of the rhamnolipid mixture obtained were determined to define its potential applications. The chemical characterization of the rhamnolipid has revealed the presence of ten different homologues. The monorhamnolipid RhaC(10)C(10) and the dirhamnolipid Rha(2)C(10)C(10) were the main components of the mixture that showed predominance of 44% and 29%, respectively, after 144-h of cultivation. The biosurfactant was able to form stable emulsions with several hydrocarbons and showed excellent emulsification for soybean oil and chicken fat (100%). The rhamnolipid removed 67% of crude oil present in sand samples and presented antimicrobial activity against Bacillus cereus and Mucor miehei at 64 mu g/mL and inhibition of Neurospora crassa, Staphylococcus aureus, and Micrococcus luteus at 256 mu g/mL. The results demonstrated that the rhamnolipid produced in soybean oil soapstock can be useful in environmental and food industry applications.

Actinobacillus pleuropneumoniae é o agente etiológico da pleuropneumonia suína, enfermidade amplamente distribuída no rebanho suíno mundial, responsável por prejuízos econômicos relevantes. Possui 12 sorotipos, determinados por técnicas de sorotipificação. Além disso é descrita a ocorrência de amostras não sorotipificáveis. O conhecimento do sorotipo prevalente nos surtos da enfermidade é necessário aos programas de profilaxia. Procurando contornar as dificuldades normalmente encontradas na sorotipificação de A. pleuropneumpnoae, a técnica de RAPD foi avaliada na genotipificação de amostras sorotipificáveis e não sorotipificáveis do agente. Foram utilizados amostras ATCC dos 12 sorotipos e amostras dos sorotipos, 1, 3, 5a, 5b, 7, 11 e 12 isolados no Brasil. Os primers OPG e OPG-19, utilizados individualmente nas reações, foram mais adequados para a diferenciação dos sorotipos. O primer OPG-19 detectou polimorfismos semelhantes entrer os sorotipos 1, 3, 4, 5 e 11; e sorotipos 7 e 12. O perfil de RAPD detectado pelo primier OPGF-10 diferenciou os isolados de campo dos sorotipos 1, 7, 11 e 12. Os sorotipos 3 e 5 apresentaram padrão de RAPD semelhantes, sendo diferenciados pelo perfil de exotoxinas característico, determinado previamente através de PCR. Este primer identificou quatro diferentes perfis de RAPD no sorotipo 3. Um destes foi semelhante ao obtido como sorotipo 11. Neste isolado, foi detecta a presença dos genes para ApxI e ApxII, características do sorotipo 11. As amostras do sorotipo 4 apresentaram perfil de RAPD semelhante ao identificado nos sorotipos 3 ou 5 com o primeir OPG-10, sendo identificada, por PCR, a presença dos genes para ApxI e ApxI, os quais não são característicos do sorotipo. Estas amostras foram isoladas em anos posteriores à amostras dos sorotipos 3 e 5 analisadas. Foi possível caracterizar 14 das 14 amostras não sorotipificáveis de A.pleuropneumpniae obtidas de suínos com sinais da doença. Entre as 4 amostras não sorotipificáveis isoladas de leitões sem sinais clínicos, apenas uma foi caracterizada através de RAPD. É possível que as demais amostras sejam outrtas bactérias NAD-dependente isoladas do trato respiratório de suínos. Amostras caracterizadas como A. minor e A. indolicus apresentaram perfis de RAPD divergentes dos identificados em isolados puros de A. pleuropneumoniae, comprovando a capacidade da técnica na caracterização do agente. Diferentes amostras do mesmo sorotipo de A. pleuropneumoniae apresentaram polimorfismos de RAPD idênticos, demonstrando reprodutividade da técnica. Os resultados comprovam a capacidade de tipificação de A. Pleuropneumoniae através de RAPD. A pesquisa de primers adequados para a diferenciação dos sorotipos 3, 4 e 5 aprimorar sua caracterização, o que pode vir a contribuir com as técnicas de sorotipificação tradicionalmente utilizados, ou permitir o uso como método de confirmação nas amostras cuja sorotipificação é problemática.

Com o objetivo de avaliar a sobrevivência ao congelamento de microrganismos potencialmente patogênicos, hambúrgueres de frango foram contaminados com Escherichia coli (ECHC), Staphylococcus aureus (SAFH) e Salmonella Enteritidis (SE86) e armazenados a -18ºC. Os mesmos microrganismos e ainda E. coli ATCC 25972, S. aureus ATCC 25923, and S. Enteritidis ATCC 13076 também foram inoculados em água peptonada 0,1% e congelados a -18ºC, a fim de avaliar um possível efeito protetor dos componentes do hambúrguer sobre os microrganismos. A quantificação dos microrganismos foi realizada nos intervalos de 0, 1, 2, 3, 4, 7, 14, 21 e 28 dias de congelamento. Com o propósito de estudar as alterações nos ácidos graxos das células microbianas expostas ao congelamento, foram extraídos os ácidos graxos de cada bactéria, congelada e não congelada, e estes foram analisados por cromatografia gasosa. Os resultados demonstraram que, de modo geral, houve uma redução média de menos de 1 unidade logarítmica (log10) no número de células artificialmente inoculadas em hambúrgueres de frango. As reduções obtidas para cada microrganismo em água peptonada 0,1% foram significativamente (P0,05) maiores do que as reduções observadas em hambúrguer de frango, sugerindo a existência de um efeito crioprotetor dos componentes do hambúrguer. Em todos os experimentos, as reduções mais expressivas foram observadas nas primeiras semanas de congelamento. Ocorreram alterações expressivas na composição de ácidos graxos de S. aureus (SAFH) e S. aureus ATCC 25923, o que pode indicar que estes microrganismos alteraram a composição dos seus ácidos graxos como resposta ao estresse causado pelo congelamento.

Motivado pelo cultivo de erva mate no Rio Grande do Sul, basicamente dentro de unidades de produção familiar, bem como pelo considerável volume de biomassa descartada, resultante do corte dos ervais, procurou-se identificar possível atividade antibacteriana em cambitos e folhas de Ilex paraguariensis St. Hill, com ênfase a zoonoses de interesse em saúde e produção animal, bem como em saúde coletiva. Definiu-se a verificação da Intensidade de Atividade de Inibição Bacteriana (IINIB) e Intensidade de Atividade de Inativação Bacteriana (IINAB) de extratos hídricos (decocto), alcoólicos (alcoolatura) e hidroalcoólicos (tintura) sobre inóculos padronizados de Staphylococcus aureus (ATCC 25923), Enterococcus faecalis (ATCC 19433), Samonella Enteritidis (ATCC 11076) e Escherichia coli (ATCC 11229). Os extratos vegetais alcoólicos foram preparados com folhas frescas, os hidroalcoólicos com os talos desidratados (cambitos) e os decoctos, tanto de folhas como de talos, com material desidratado. Todas as quatro formas de extração apresentaram capacidade de inativação e/ou inibição seletivas sobre os inóculos bacterianos, porém os extratos à base de álcool de cereais foram os que apresentaram os melhores resultados. Dentre as bactérias, a Salmonella Enteritidis demonstrou maior sensibilidade, seguida por Enterococcus faecalis. Posteriormente, estas duas amostras foram submetidas a testes sob tempo de exposição de 5, 15, 30 e 60 minutos, na ausência e na presença de matéria orgânica (soro bovino inativado), confrontando-as com alcoolatura de folhas e hidroalcoolatura de cambitos, por estes terem obtido maior ação antibacteriana sobre os agentes testados nos testes iniciais de IINIB e IINAB. Os decoctos de cambitos não corresponderam às expectativas iniciais, sendo que as alcoolaturas, tanto de folhas quanto de cambitos, apresentaram resultados eficazes. Tanto o tempo de exposição como a presença de matéria orgânica interferiram na sensibilidade apresentada por Salmonella Enteritidis e Enterococcus faecalis, positiva e negativamente, respectivamente.
