Acute respiratory distress syndrome (ARDS) is a devastating disorder characterized by increased alveolar permeability with no effective treatment beyond supportive care. Current mechanisms underlying ARDS focus on alveolar endothelial and epithelial injury caused by products of innate immune cells and platelets. However, the role of adaptive immune cells in ARDS remains largely unknown. In this study, we report that expansion of Ag-specific αβTh17 cells contributes to ARDS by local secretion of IL-17A, which in turn directly increases alveolar epithelial permeability. Mice with a highly restrictive defect in Ag-specific αβTh17 cells were protected from experimental ARDS induced by a single dose of endotracheal LPS. Loss of IL-17 receptor C or Ab blockade of IL-17A was similarly protective, further suggesting that IL-17A released by these cells was responsible for this effect. LPS induced a rapid and specific clonal expansion of αβTh17 cells in the lung, as determined by deep sequencing of the hypervariable CD3RβVJ region of the TCR. Our findings could be relevant to ARDS in humans, because we found significant elevation of IL-17A in bronchoalveolar lavage fluid from patients with ARDS, and rIL-17A directly increased permeability across cultured human alveolar epithelial monolayers. These results reveal a previously unexpected role for adaptive immune responses that increase alveolar permeability in ARDS and suggest that αβTh17 cells and IL-17A could be novel therapeutic targets for this currently untreatable disease.

Photocatalytic antibacterial low density polyethylene (LDPE)–TiO2 films are produced by an extrusion method and tested for photocatalytic oxidation activity, via the degradation of methylene blue (MB) and photocatalytic antibacterial activity, via the destruction of Escherichia coli. The MB test showed that extruded LDPE films with a TiO2 loading 30 wt.% were of optimum activity with no obvious decrease in film strength, although the activity was less than that exhibited by the commercial self-cleaning glass, Activ®. UVC pre-treatment (9.4 mW cm−2) of the latter film improved its activity, with the level of surface sites available for MB adsorption increasing linearly with UVC dose. Although the MB test revealed an optimum exposure time of ca. 60 min photocatalytic oxidation activity, only 30 min was used in the photocatalytic antibacterial tests in order to combine minimal reduction in film integrity with maximum film photocatalytic activity. The photocatalytic antibacterial activity of the latter film was over 10 times that of a non-UVC treated 30 wt.% TiO2 film, which, in turn was over 100 times more active than Activ®.

In the last 40 years a wide range of molecules, including neutrals, cations and anions, containing up to 13 atoms—in addition to detections of C<sub>60 and C<sub>70 — have been found in the harsh environment of the interstellar medium. The exquisite sensitivity and very high spectral and, more recently, spatial resolution, of modern telescopes has enabled the physics of star formation to be probed through rotational line emission. In this article, I review the basic properties of interstellar clouds and the processes that initiate the chemistry and generate chemical complexity, particularly in regions of star and planet formation. Our understanding of astrochemistry has evolved over the years. Before 1990, the general consensus was that molecules were formed in binary, gas-phase, or volume, reactions, most importantly ion-neutral reactions despite the very low ionization in clouds. Since then, observations have indicated unambiguously that there is also a contribution from surface processes, particularly on the icy mantles that form around refractory grain cores in cold, dense gas. The balance between these two processes depends on particular physical conditions and can vary during the life cycle of a particular volume of interstellar cloud.The complex chemistry that occurs in space is driven mostly through interaction of thegas with cosmic ray protons, a source of ionization that enables a rich ion-neutral chemistry. In addition, I show that the interaction between the gas and the dust in cold, dense regionsalso leads to additional chemical complexity through reactions that take place in ices at onlya few tens of degrees above absolute zero. Although densities are low compared to those in terrestrial environments, the extremely long life times of interstellar clouds and their enormous sizes, enable complex molecules to be synthesised and detected. I show that in some instances, particularly in reactions involving deuterium, the rotational populations of reactants, together with spin-selection rules, can determine the detailed abundances. Although the review is mainly focused on regions associated with star formation, I also consider chemistry in other interesting astronomical regions — in the early Universe and in the envelopes formed by mass loss during the final stages of stellar evolution.

We present optical and infrared monitoring data of SN 2012hn collectedby the Public European Southern Observatory Spectroscopic Survey forTransient Objects. We show that SN 2012hn has a faint peak magnitude(MR ˜ -15.65) and shows no hydrogen and no clearevidence for helium in its spectral evolution. Instead, we detectprominent Ca II lines at all epochs, which relates this transient topreviously described `Ca-rich' or `gap' transients. However, thephotospheric spectra (from -3 to +32 d with respect to peak) of SN2012hn show a series of absorption lines which are unique and a redcontinuum that is likely intrinsic rather than due to extinction. Linesof Ti II and Cr II are visible. This may be a temperature effect, whichcould also explain the red photospheric colour. A nebular spectrum at+150 d shows prominent Ca II, O I, C I and possibly Mg I lines whichappear similar in strength to those displayed by core-collapsesupernovae (SNe). To add to the puzzle, SN 2012hn is located at aprojected distance of 6 kpc from an E/S0 host and is not close to anyobvious star-forming region. Overall SN 2012hn resembles a group offaint H-poor SNe that have been discovered recently and for which aconvincing and consistent physical explanation is still missing. Theyall appear to explode preferentially in remote locations offset from amassive host galaxy with deep limits on any dwarf host galaxies,favouring old progenitor systems. SN 2012hn adds heterogeneity to thissample of objects. We discuss potential explosion channels includingHe-shell detonations and double detonations of white dwarfs as well aspeculiar core-collapse SNe.

As galactomananas das infusões de café apresentam atividade imunoestimuladora in vitro, sendo esta atividade semelhante à das mananas acetiladas extr­das de Aloe vera. As galactomananas presentes no resíduo de café também possuem atividade imunoestimuladora in vitro quando são parcialmente acetiladas. Como as galactomananas são o componente maioritário do resíduo de café e como o café é um produto de largo consumo a nível mundial, o reaproveitamento deste resíduo como fonte de galactomananas com atividade imunoestimuladora deve ser considerado. Esta dissert§Ã£o procura dar resposta a duas questões: 1. Quais são as estruturas das galactomananas responsáveis pela atividade imunoestimuladora destes polissacarídeos; e 2. Como é que as galactomananas podem ser extr­das quantitativamente do resíduo de café de modo a serem solúveis em água à temperatura ambiente e, assim, poderem ser utilizadas como ingredientes alimentares com atividade imunoestimuladora. A questão 1 foi respondida pela caracteriz§Ã£o estrutural de quatro galactomananas, de três origens: a) as galactomananas das infusões de café e do resíduo que apresentaram atividade imunoestimuladora; b) a galactomanana da goma de alfarroba (LBG), que não apresentou atividade imunoestimuladora; e c) a manana acetilada de Aloe vera, que apresentou atividade imunoestimuladora. Estes polissacarídeos foram submetidos à análise de §Ãºcares e de lig§Ãµes glicosídicas e a hidrólise por endo-β-D- (1→4)-mananase. Os fragmentos de oligossacarídeos mais pequenos foram ainda analisados por espetrometria de massa por ioniz§Ã£o de electrospray e espetrometria de massa tandem. As galactomananas das infusões de café, do resíduo de café e do Aloe vera apresentaram grau de ramificação e peso molecular semelhantes, enquanto as galactomananas da LBG apresentaram grau de ramificação e de polimeriz§Ã£o maiores. Todas as galactomananas apresentaram resíduos de arabinose como ramificação. O grau de acetil§Ã£o das galactomananas da LBG foi vestigial enquanto as galactomananas do Aloe vera apresentaram um grau de acetil§Ã£o de 2,08; para as galactomananas do resíduo de café o grau de acetil§Ã£o foi de 0,98 e para as infusões foi de 0,08. A localiz§Ã£o dos grupos acetilo foi irregular em todos os polímeros. Os resultados obtidos permitem inferir que baixos níveis de ramificação, cadeias pequenas e alguma acetil§Ã£o parecem promover a atividade imunoestimuladora atribuída às galactomananas. Para responder à questão 2, foi testada uma metodologia que envolveu a torra do resíduo de café a 160 ºC e a 220 ºC e a sua extr§Ã£o com água quente e com soluções de 4 M NaOH à temperatura de 20, 60 e 120 ºC. A torra do resíduo a 160 ºC e a extr§Ã£o sequencial permitiu extrair 56% das galactomananas presentes no resíduo de café e, simultaneamente, 54% das arabinogalactanas. As galactomananas mantiveram a sua estrutura caraterística de polissacarídeo acetilado composto por uma cadeira principal de resíduos de manose em lig§Ã£o β-(1→4) e resíduos de Gal e Ara nas cadeias laterais. A 220 ºC, as galactomananas foram parcialmente degradadas e o rendimento de extr§Ã£o foi muito menor do que a 160 ºC. No entanto, mesmo a esta temperatura as galactomananas apresentaram resíduos acetilados e a presença de pentoses nas cadeias laterais, o que permite inferir a elevada resistência destes polissacarídeos à temperatura e aos reagentes alcalinos. De forma a melhor compreender a estabilidade térmica das galactomananas do resíduo de café e a influência que a presença de arabinogalactanas pode ter na sua estabilidade, foi feita uma análise termogravimétrica aos polissacarídeos extr­dos do resíduo de café assim como a polissacarídeos relacionados estruturalmente com estes, como a celulose, a galactomanana de LBG e a goma arábica, uma arabinogalactana. As galactomananas são termicamente estáveis durante 3 h a 200 ºC, enquanto as arabinogalactanas são estáveis a 180 ºC. De acordo com os perfis dos termogramas obtidos, e pelo ¡lculo das energias de ativ§Ã£o da degrad§Ã£o térmica, o resíduo de café apresenta uma estabilidade térmica menor do que a galactomanana, possivelmente devido à presença de arabinogalactanas. Apesar de não se ter verificado alter§Ãµes no termograma da galactomanana do café submetida a um tratamento térmico de 200 ºC durante 3 h, verificam-se alter§Ãµes estruturais que envolvem a form§Ã£o de novas lig§Ãµes glicosídicas, nomeadamente, a form§Ã£o de resíduos de manose ligados em O-2 e em O-6, re§Ãµes de transglicosil§Ã£o, despolimeriz§Ã£o, form§Ã£o de resíduos de anidro-hexoses no terminal redutor e isomeriz§Ã£o manose-glucose. Estas alter§Ãµes promovem a solubiliz§Ã£o das galactomananas. Os resultados obtidos permitem propor que o resíduo de café possa ser submetido a uma torra seguida de extr§Ã£o com reagentes alcalinos a quente para obtenção das galactomananas com rendimentos elevados. Estes polissacarídeos podem tornar-se solúveis em água após tratamento térmico a 200 ºC, permitindo assim a sua utiliz§Ã£o em formul§Ãµes alimentares, nomeadamente, por prepar§Ã£o de compostos acetilados com baixos níveis de ramificação e cadeias pequenas de modo a promover a sua atividade imunoestimuladora.

En Suisse, les recommandations de fruits et légumes sont de 600 g par jour. En réalité, la consommation moyenne se situe à 60% des recommandations, c'est-à-dire à 380 g par jour. L'utilisation de produits tels que des shots de fruits et légumes pourrait réduire l'écart en facilitant la consommation de ces aliments. Cependant, ils ne peuvent pas remplacer à cent pour cent les aliments frais car leurs teneurs en fibres et vitamines C sont plus faibles, et ils interfèrent avec la physiologie de la prise alimentaire (satiété). Ces produits peuvent donc faire partie d'une alimentation variée en prenant la place d'une portion de fruits et légumes. Un autre aspect non négligeable en est le coût puisque si l'on consommait ces produits sur une semaine à la place d'aliments frais la différence serait d'environ CHF 30.- de plus. In Switzerland, fruits and vegetables recommendations are of 600 gr per day. In reality, the average intake is about 60% of these recommendations, it means 380 gr per day. The use of products like fruits and vegetables shots could reduce the gap by making drinking easier. However, they cannot replace one hundred percent fresh food for their content in fibers and vitamin C are lower and they interfere with the physiology of the food intake (satiety). These products can have their importance in a various supply by taking the place of one fruit or vegetable portion. Another non negligible aspect is the price as if we consume these products over one week instead of fresh food, the difference would be of about CHF 30.- more

Describe los principales resultados obtenidos en el Crucero evaluación de recursos pelágicos, efectuado a bordo del BIC/SNP-1 del 14 de noviembre de 1989, entre Chicama y Punta Doña María, constituyendo la segunda prospección planificada dentro del marco del proyecto Evaluación de los recursos: anchoveta, sardina, jurel y caballa, auspiciado por el Instituto del mar del Perú y la Comunidad económica europea.

Ligera proyección de Aguas Ecuatoriales Superficiales originaron anomalías de 0,6 °C a 2,7 °C al norte de los 6°S, mientras que anomalías negativas asociadas al afloramiento costero se ubicaron al sur de esta latitud, con valores entre -04° C a -2,6° C. La extensión sur de la Corriente de Cromwell se vio moderadamente fortalecida frente a Paita (100 m de profundidad), disminuyendo su espesor en Punta Falsa. La merluza se presentó entre Puerto Pizarro a Punta Falsa, en temperaturas de 14,2 °C a 17,2 °C, salinidades de 34,89‰ a 35,13‰ y oxígeno entre 0,29 a 3,0 mL/L.

The soil-inhabiting insect-pathogenic fungus Metarhizium robertsii also colonizes plant roots endophytically, thus showing potential as a plant symbiont. M robertsii is not randomly distributed in soils but preferentially associates with the plant rhizosphere when applied in agricultural settings. Root surface and endophytic colonization of switchgrass (Panicum virgatum) and haricot beans (Phaseolus vulgaris) by M robertsii were examined after inoculation with fungal conidia. Light and confocal microscopies were used to ascertain this rhizosphere association. Root lengths, root hair density and emergence of lateral roots were also measured. Initially, M robertsii conidia adhered to, germinated on, and colonized, roots. Furthermore, plant roots treated with Metarhizium grew faster and the density of plant root hairs increased when compared with control plants. The onset of plant root hair proliferation was initiated before germination of M robertsii on the root (within 1-2 days). Plants inoculated with M robertsii AMAD2 (plant adhesin gene) took significantly longer to show root hair proliferation than the wild type. Cell free extracts of M robertsii did not stimulate root hair proliferation. Longer term (60 days) associations showed that M robertsii endophytically colonized individual cortical cells within bean roots. Metarhizium appeared as an amorphous mycelial aggregate within root cortical cells as well as between the intercellular spaces with no apparent damage to the plant. These results suggested that not only is M robertsii rhizosphere competent but displays a beneficial endophytic association with plant roots that results in the proliferation of root hairs. The biocontrol of bean (Phaseolis vulgaris) root rot fungus Fusarium solani f. sp. phaseolis by Metarhizium robertsii was investigated in vitro and in vivo. Dual cultures on Petri dishes showed antagonism of M robertsii against F. solani. A relative inhibition of ca. 60% of F. solani growth was observed in these assays. Cell free culture filtrates of M robertsii inhibited the germination of F. solani conidia by 83% and the inhibitory metabolite was heat stable. Beans plants colonized by M robertsii then exposed to F. solani showed healthier plant profiles and lower disease indices compared to plants not colonized by M robertsii. These results suggested that the insect pathogenic/endophytic fungus M robertsii could also be utilized as a biocontrol agent against certain plant pathogens occurring in the rhizosphere.
