The aim with this Essay is to examine the two most read magazines in Sweden covering the areas of exercising, fitness, bodybuilding, diets and “wellness” –Fitness and Body. Fitness’s target group is predominantly woman, while Body is almost exclusively read by men. The analysis is first done quantitatively, by systematically categorising the contents of the magazines. Then a qualitative analysis is made. Using two different theories, Anja Hirdman’s gender concept along with her constructivistic media perspective and the theory of Symbolic Interactionism, i try to answer the following questions; Does the two magazines term of address and language differ from one and other? And if that is the case, in what way? With point of departure from contents and subject areas, how are the two magazines compounded? How can the underlying message in the different articles be interpreted?The analysis shows that both magazines followed their purpose of writing about exercising in general, diet, fitness and bodybuilding. However, the magazine Fitness writes more often then Body about matters not following the given purpose, and the language in Body is more informative and general in comparison with the language used in Fitness. Still, the messages sent out by the different articles are in majority of the respects similar. Both magazines are portraying body ideals who can be understand as extreme. In Body the message feels fairly straight, “Build bigger muscles and burn more fat”, whereas Fitness willingly use the concept of “wellness” as a cover for what the message really is, namely “get your self a rock hard body through dieting and hard training”!

The aim is first to identify concept of citizenship and the dimensions of the same, in the political science theory, which can be crucial, to that a citizenship may be found available for people who have a cognitive delay. Thereafter examine the prevalence and significance of these, in policy documents from two different organizational forms, through an analysis, in the form of a discussion, on the basis of political science theories in the field. The study's aim in a reflective way, highlighting key requirements for an accessible citizenship for the target audience, and thereby clarify the priorities in daily activities that can support an emancipatory citizenship process for people who have a cognitive delay.

Background: Home-management of malaria (HMM) strategy improves early access of anti-malarial medicines to high-risk groups in remote areas of sub-Saharan Africa. However, limited data are available on the effectiveness of using artemisinin-based combination therapy (ACT) within the HMM strategy. The aim of this study was to assess the effectiveness of artemether-lumefantrine (AL), presently the most favoured ACT in Africa, in under-five children with uncomplicated Plasmodium falciparum malaria in Tanzania, when provided by community health workers (CHWs) and administered unsupervised by parents or guardians at home. Methods: An open label, single arm prospective study was conducted in two rural villages with high malaria transmission in Kibaha District, Tanzania. Children presenting to CHWs with uncomplicated fever and a positive rapid malaria diagnostic test (RDT) were provisionally enrolled and provided AL for unsupervised treatment at home. Patients with microscopy confirmed P. falciparum parasitaemia were definitely enrolled and reviewed weekly by the CHWs during 42 days. Primary outcome measure was PCR corrected parasitological cure rate by day 42, as estimated by Kaplan-Meier survival analysis. This trial is registered with ClinicalTrials.gov, number NCT00454961. Results: A total of 244 febrile children were enrolled between March-August 2007. Two patients were lost to follow up on day 14, and one patient withdrew consent on day 21. Some 141/241 (58.5%) patients had recurrent infection during follow-up, of whom 14 had recrudescence. The PCR corrected cure rate by day 42 was 93.0% (95% CI 88.3%-95.9%). The median lumefantrine concentration was statistically significantly lower in patients with recrudescence (97 ng/mL [IQR 0-234]; n = 10) compared with reinfections (205 ng/mL [114-390]; n = 92), or no parasite reappearance (217 [121-374] ng/mL; n = 70; p <= 0.046). Conclusions: Provision of AL by CHWs for unsupervised malaria treatment at home was highly effective, which provides evidence base for scaling-up implementation of HMM with AL in Tanzania.

The human buccal micronucleus cytome assay (BMCyt) is one of the most widely used techniques to measure genetic damage in human population studies. Reducing protocol variability, assessing the role of confounders, and estimating a range of reference values are research priorities that will be addressed by the HUMNXL, collaborative study. The HUMNXL, project evaluates the impact of host factors, occupation, life-style, disease status, and protocol features on the occurrence of MN in exfoliated buccal cells. In addition, the study will provide a range of reference values for all cytome endpoints. A database of 5424 subjects with buccal MN values obtained from 30 laboratories worldwide was compiled and analyzed to investigate the influence of several conditions affecting MN frequency. Random effects models were mostly used to investigate MN predictors. The estimated spontaneous MN frequency was 0.74 parts per thousand (95% CI 0.52-1.05). Only staining among technical features influenced MN frequency, with an abnormal increase for non-DNA-specific stains. No effect of gender was evident, while the trend for age was highly significant (p < 0.001). Most occupational exposures and a diagnosis of cancer significantly increased MN and other endpoints frequencies. MN frequency increased in heavy smoking (>= 40 cig/day. FR = 1.37:95% CI 1.03-.82) and decreased with daily fruit consumption (FR = 0.68; 95% CI 0.50-0.91). The results of the HUMNXL, project identified priorities for validation studies, increased the basic knowledge of the assay, and contributed to the creation of a laboratory network which in perspective may allow the evaluation of disease risk associated with MN frequency. We report on the fabrication of novel lead-germanate glasses and fibers. We have characterized these glasses in terms of their thermal properties, Raman spectra and refractive indices (both linear and nonlinear) and present them as viable alternatives to tellurite glasses for applications requiring highly nonlinear optical fibers. © 2013 Optical Society of America.

Landscape fires show large variability in the amount of biomass or fuel consumed per unit area burned. Fuel consumption (FC) depends on the biomass available to burn and the fraction of the biomass that is actually combusted, and can be combined with estimates of area burned to assess emissions. While burned area can be detected from space and estimates are becoming more reliable due to improved algorithms and sensors, FC is usually modeled or taken selectively from the literature. We compiled the peerreviewed literature on FC for various biomes and fuel categories to understand FC and its variability better, and to provide a database that can be used to constrain biogeochemical models with fire modules. We compiled in total 77 studies covering 11 biomes including savanna (15 studies, average FC of 4.6 t DM (dry matter) ha 1 with a standard deviation of 2.2), tropical forest (n = 19, FC = 126 +/- 77), temperate forest (n = 12, FC = 58 +/- 72), boreal forest (n = 16, FC = 35 +/- 24), pasture (n = 4, FC = 28 +/- 9.3), shifting cultivation (n = 2, FC = 23, with a range of 4.0-43), crop residue (n = 4, FC = 6.5 +/- 9.0), chaparral (n = 3, FC = 27 +/- 19), tropical peatland (n = 4, FC = 314 +/- 196), boreal peatland (n = 2, FC = 42 [42-43]), and tundra (n = 1, FC = 40). Within biomes the regional variability in the number of measurements was sometimes large, with e. g. only three measurement locations in boreal Russia and 35 sites in North America. Substantial regional differences in FC were found within the defined biomes: for example, FC of temperate pine forests in the USA was 37% lower than Australian forests dominated by eucalypt trees. Besides showing the differences between biomes, FC estimates were also grouped into different fuel classes. Our results highlight the large variability in FC, not only between biomes but also within biomes and fuel classes. This implies that substantial uncertainties are associated with using biome-averaged values to represent FC for whole biomes. Comparing the compiled FC values with co-located Global Fire Emissions Database version 3 (GFED3) FC indicates that modeling studies that aim to represent variability in FC also within biomes, still require improvements as they have difficulty in representing the dynamics governing FC.

A specific separated-local-field NMR experiment, dubbed Dipolar-Chemical-Shift Correlation (DIPSHIFT) is frequently used to study molecular motions by probing reorientations through the changes in XH dipolar coupling and T-2. In systems where the coupling is weak or the reorientation angle is small, a recoupled variant of the DIPSHIFT experiment is applied, where the effective dipolar coupling is amplified by a REDOR-like pi-pulse train. However, a previously described constant-time variant of this experiment is not sensitive to the motion-induced T-2 effect, which precludes the observation of motions over a large range of rates ranging from hundreds of Hz to around a MHz. We present a DIPSHIFT implementation which amplifies the dipolar couplings and is still sensitive to T-2 effects. Spin dynamics simulations, analytical calculations and experiments demonstrate the sensitivity of the technique to molecular motions, and suggest the best experimental conditions to avoid imperfections. Furthermore, an in-depth theoretical analysis of the interplay of REDOR-like recoupling and proton decoupling based on Average-Hamiltonian Theory was performed, which allowed explaining the origin of many artifacts found in literature data. O artigo apresenta uma reflexão sobre a gestão do processo de projeto paramétrico, com superfícies complexas, baseado em duas experiências realizadas no Instituto de Arquitetura e Urbanismo, da USP de São Carlos, pelo grupo de pesquisa Nomads.usp. SLICE é o resultado parcial de um pavilhão temporário, com superfícies complexas, desenvolvido por meio de projeto paramétrico e técnicas de fabricação digital. Utilizou-se o procedimento de uma pesquisa-ação2 com alunos de graduação e recém-formados em arquitetura, arquitetos do Nomads.usp e a indústria. Os experimentos apontaram para reflexões sobre algumas mudanças na gestão destes processos, para facilitar a viabilidade técnico-construtiva e gerar conhecimento nas áreas de projeto paramétrico, fabricação digital e gestão. O artigo tem caráter exploratório e o desafio é entender e avaliar a gestão do projeto paramétrico em relação a restrição de material, custo de execução e tempo de produção.

This paper investigates the use of explicit structures of information in architectural design. Particularly, it approaches the use of diagrams related to cybernetics and information theory in experimental practices in the 1960’s and 1970’s. It analyses the diagram of cybernetic control proposed by the cybernetician Gordon Pask for the Fun Palace, the diagrams produced by the utopian architect Yona Friedman in the conceptual description of the Flatwriter program and Christopher Alexander’s diagrams and his theories of Synthesis of Form and Pattern Language. Finally it establishes a brief parallel between current domestication and use of dataflow programming with the cybernetic diagrams, highlighting differences in their complexity approach.

Protein disulfide isomerase (PDI) and its homologs are oxidoreductases facilitating protein folding in the ER. Endo-PDI (also termed ERp46) is highly expressed in endothelial cells. It belongs to the PDI family but its physiological function is largely unknown. We studied the role of Endo-PDI in endothelial angiogenic responses. Stimulation of human umbilical vein endothelial cells (with TNFα (10ng/ml) increased ERK1/2 phosphorylation. This effect was largely attenuated by Endo-PDI siRNA, whereas JNK and p38 MAP kinase phosphorylation was Endo-PDI independent. Similarly, TNFα-stimulated NF-κB signaling determined by IκBα degradation as well as TNFα-induced ICAM expression was unaffected by Endo-PDI siRNA. The action of Endo-PDI was not mediated by extracellular thiol exchange or cell surface PDI as demonstrated by nonpermeative inhibitors and PDI-neutralizing antibody. Moreover, exogenously added PDI failed to restore ERK1/2 activation after Endo-PDI knockdown. This suggests that Endo-PDI acts intracellularly potentially by maintaining the Ras/Raf/MEK/ERK pathway. Indeed, knockdown of Endo-PDI attenuated Ras activation measured by G-LISA and Raf phosphorylation. ERK activation influences gene expression by the transcriptional factor AP-1, which controls MMP-9 and cathepsin B, two proteases required for angiogenesis. TNFα-stimulated MMP-9 and cathepsin B induction was reduced by silencing of Endo-PDI. Accordingly, inhibition of cathepsin B or Endo-PDI siRNA blocked the TNFα-stimulated angiogenic response in the spheroid outgrowth assays. Moreover ex vivo tube formation and in vivo Matrigel angiogenesis in response to TNFα were attenuated by Endo-PDI siRNA. In conclusion, our study establishes Endo-PDI as a novel, important mediator of AP-1-driven gene expression and endothelial angiogenic function

Deutsch:In dieser Arbeit wurden Versuche zur funktionellen Expression von schwer ektopisch exprimierbaren nAChR in HEK-293/a1-Zellen durchgeführt: a7 nAChR und a6-enthaltenden nAChR. Die Probleme lagen dabei nicht auf dem Niveau der Transfektion, Transkription, Translation oder der Assemblierung, sondern beim Transport der Rezeptoren zur Zellmembran.Die Expression von a7 nAChR in der Plasmamembran von HEK-293/a1-Zellen konnte durch verbesserte Expressionsbedingungen (Koexpression des Faltungshelfers Calnexin oder weiterer nAChR-Untereinheiten, Erniedrigung der Expressionstemperatur, Expression in Gegenwart nikotinischer Antagonisten) nicht erreicht werden. Auch in anderen Zellinien mit neuronalem oder nicht-neuronalem Ursprung (QT6, GH4C1, S2 und PCC7-Mz1) war die EGFP-gekoppelte a7 nAChR-Untereinheit nur im Zellinneren lokalisiert.Eine intrazelluläre Lokalisation verhinderte auch eine funktionelle Expression homomerer a6 sowie heteromerer a6b2 und a6b3 nAChR in HEK-293/a1-Zellen. Im Gegensatz dazu führte eine Expression von stabil mit den nAChR-Untereinheiten a6 und b4 transfizierten HEK-293/a1-Zellen in Gegenwart von Calciumphosphat-Transfektionslösung und anschließend bei 30°C zu einem verbesserten Transport der Rezeptoren zur Zellmembran und damit zum erfolgreichen Expression funktioneller a6b4 nAChR. Die Wirkung der Transfektionslösung kann durch die erhöhte Calciumkonzentration erklärt werden, da in Ganzzellableitungen eine potenzierende Wirkung von Calciumionen auf den a6b4 nAChR bewiesen wurde. Somit konnte erstmalig der humane a6b4 nAChR in einer Säugerzellinie stabil und funktionell exprimiert werden.

Nach der Reinigung der Vinorin-Synthase ausHybrid-Zellkulturen von Rauvolfia serpentina/Rhazya strictamit den fünf chromatographischen TrennmethodenAnionenaustauschchromatographie an SOURCE 30Q, HydrophobeInteraktionen Chromatographie an SOURCE 15PHE,Chromatographie an MacroPrep Ceramic Hydroxyapatit,Anionenaustauschchromatographie an Mono Q undGrößenausschlußchromatographie an Superdex 75 konnte dieVinorin-Synthase aus 2 kg Zellkulturgewebe 991fachangereichert werden.Das nach der Reinigung angefertigte SDS-Gel ermöglichte eineklare Zuordnung der Protein-Bande als Vinorin-Synthase.Der Verdau der Enzymbande mit der Endoproteinase LysC unddie darauffolgende Sequenzierung der Spaltpeptide führte zuvier Peptidsequenzen. Der Datenbankvergleich (SwissProt)zeigte keinerlei Homologien zu Sequenzen bekannterPflanzenenzyme. Mit degenerierten Primern, abgeleitet voneinem der erhaltenen Peptidfragmente und einer konserviertenRegion bekannter Acetyltransferasen gelang es, ein erstescDNA-Fragment der Vinorin-Synthase zu amplifizieren. Mit derMethode der RACE-PCR wurde die Nukleoidsequenzvervollständigt, was zu einem cDNA-Vollängenklon mit einerGröße von 1263 bp führte, der für ein Protein mit 421Aminosäuren (46 kDa) codiert.Das Vinorin-Synthase-Gen wurde in den pQE2-Expressionsvektorligiert, der für einen N-terminalen 6-fachen His-tagcodiert. Anschließend wurde sie erstmals erfolgreich in E.coli im mg-Maßstab exprimiert und bis zur Homogenitätgereinigt. Durch die erfolgreiche Überexpression konnte dieVinorin-Synthase eingehend charakterisiert werden. DerKM-Wert für das Substrat Gardneral wurde mit 20 µM, bzw.41.2 µM bestimmt und Vmax betrug 1 pkat, bzw. 1.71 pkat.Nach erfolgreicher Abspaltung des His-tags wurden diekinetischen Parameter erneut bestimmt (KM- Wert 7.5 µM, bzw.27.52 µM, Vmax 0.7 pkat, bzw. 1.21 pkat). Das Co-Substratzeigt einen KM- Wert von 60.5 µM (Vmax 0.6 pkat). DieVinorin-Synthase besitzt ein Temperatur-Optimum von 35 °Cund ein pH-Optimum bei 7.8.Homologievergleiche mit anderen Enzymen zeigten, dass dieVinorin-Synthase zu einer noch kleinen Familie von bisher 10Acetyltransferasen gehört. Alle Enzyme der Familie haben einHxxxD und ein DFGWG-Motiv zu 100 % konserviert. Basierendauf diesen Homologievergleichen und Inhibitorstudien wurden11 in dieser Proteinfamilie konservierte Aminosäuren gegenAlanin ausgetauscht, um so die Aminosäuren einer in derLiteratur postulierten katalytischen Triade(Ser/Cys-His-Asp) zu identifizieren.Die Mutation aller vorhandenen konservierten Serine undCysteine resultierte in keiner Mutante, die zumvollständigen Aktivitätsverlust des Enzyms führte. Nur dieMutationen H160A und D164A resultierten in einemvollständigen Aktivitätsverlust des Enzyms. Dieses Ergebniswiderlegt die Theorie einer katalytischen Triade und zeigte,dass die Aminosäuren H160A und D164A exklusiv an derkatalytischen Reaktion beteiligt sind.Zur Überprüfung dieser Ergebnisse und zur vollständigenAufklärung des Reaktionsmechanismus wurde dieVinorin-Synthase kristallisiert. Die bis jetzt erhaltenenKristalle (Kristallgröße in µm x: 150, y: 200, z: 200)gehören der Raumgruppe P212121 (orthorhombisch primitiv) anund beugen bis 3.3 Å. Da es bis jetzt keine Kristallstruktureines zur Vinorin-Synthase homologen Proteins gibt, konntedie Struktur noch nicht vollständig aufgeklärt werden. ZurLösung des Phasenproblems wird mit der Methode der multiplenanomalen Dispersion (MAD) jetzt versucht, die ersteKristallstruktur in dieser Enzymfamilie aufzuklären.

An einer Vielzahl biogener Polyelektrolyte, wie z.B. den Nukleinsäuren DNA und RNA sowie Proteinen, ist die Ausbildung von Strukturhierarchien durch Selbstorganisation von Strukturelementen zu beobachten. Dabei wird das Strukturbildungsverhalten durch eine Kopplung von Wechselwirkungen auf verschiedenen Längenskalen, den kurzreichweitigen ausgeschlossenen Volumen und den langreichweitigen elektrostatischen Wechselwirkungen (Coulomb-Wechselwirkungen), die wiederum durch eine Vielzahl mikroskopische Parameter (z.B. Konformation) beeinflusst werden, bestimmt. Durch diese Komplexität ist es nicht möglich, den für die Strukturbildung hochgeladener Systeme bedeutsamen Beitrag der elektrostatischen Wechselwirkungen isoliert zu betrachten. Aus diesem Grund werden zur Aufklärung von Wechselwirkungs- und Strukturbildungsmechanismen vereinfachte Modell-Systeme herangezogen. Eine Möglichkeit besteht in der Verwendung synthetischer, kettensteifer Polyelektrolyte. Im Rahmen dieser Arbeit wurde das Aggregationsverhalten wässriger Lösungen dodecylsubstituierter Poly(para-phenylen)sulfonate (PPPS) sowie die Beeinflussung der Strukturbildung durch verschiedene Parameter charakterisiert. Als Einflussparameter wurden einerseits die Gegenion-Spezies und andererseits die Temperatur- und Konzentrations-Abhängigkeit untersucht. Hierzu wurden wässrige Lösungen der freien Säuren der PPPS mit Molekulargewichten zwischen MW = 18 kg/mol bis 58 kg/mol mittels Licht-, Röntgen- und Neutronenstreuung sowie durch Licht-, Polarisations-, Transmissionselektronen- und Rasterkraftmikroskopie in einem Konzentrationsbereich von 0,0008 < c < 1,1 g/L untersucht.
