La presente investigación se llevó a cabo en la Universidad Politécnica de Madrid (España) conjuntamente con la Universidad Nacional Experimental del Táchira (Venezuela). El estudio consistió en diseñar una cavidad interna dentro del perfil aerodinámico 2415-3s, el cual fue desarrollado en la Universidad Técnica Checa (Praga, República Checa). Se realizó un estudio computacional, mediante la técnica del CFD, de diferentes modelos de cavidades internas en este perfil, para seleccionar el diseño más adecuado, fabricando un prototipo en 3D; logrando de esta manera validar la simulación computacional con los datos experimentales obtenidos con los ensayos en el túnel de viento AF6109 de la Universidad Nacional Experimental del Táchira. También se aplicaron técnicas de visualización en el túnel de viento, como líneas de corriente de humo y películas de aceite sobre el perfil aerodinámico. Dicho procedimiento permitió corroborar la validación de la simulación computacional. El perfil aerodinámico seleccionado se denominó 2415-3s-TC, cuya característica principal consiste en tres canales independientes entre sí, alojados dentro de la cavidad interna, permitiendo que el flujo de aire forzado a través de la cavidad, cambiara de dirección, para desembocar lo más tangencialmente, así como, lo más perpendicularmente posible al escalón del perfil aerodinámico 2415-3s. Esta configuración de diseñó permitió elevar el coeficiente de sustentación para ángulos de ataque mayores a 8º, así como para ángulos cercanos al ángulo crítico. ABSTRACT This research was conducted at the Polytechnic University of Madrid (Spain) together with the National Experimental University of Táchira (Venezuela). The study was to design an internal cavity within the airfoil 2415-3s, which was developed in the Czech Technical University (Prague, Czech Republic). A computational study was performed using CFD technique, different models of internal cavities in the profile to select the most appropriate design, manufacturing a prototype 3D; thus achieving validate the computer simulation with experimental data obtained from the tests in the wind tunnel AF6109 of the National Experimental University of Táchira. Visualization techniques were also applied in the wind tunnel, as streamlines smoke and oil films on the airfoil. This procedure corroborated validation of computational simulation. The airfoil selected denominated 2415-3s-TC, whose main characteristic consists of three independent channels each other, housed within the inner cavity, allowing the forced air flow through the cavity, change direction, to lead as more tangentially and, as perpendicular as possible to the step 2415-3s aerofoil. This configuration designed allowed increasing the lift coefficient for higher angles of attack to 8º, and for angles near the critical angle.

We created a simulation based on experimental data from bacteriophage T7 that computes the developmental cycle of the wild-type phage and also of mutants that have an altered genome order. We used the simulation to compute the fitness of more than 105 mutants. We tested these computations by constructing and experimentally characterizing T7 mutants in which we repositioned gene 1, coding for T7 RNA polymerase. Computed protein synthesis rates for ectopic gene 1 strains were in moderate agreement with observed rates. Computed phage-doubling rates were close to observations for two of four strains, but significantly overestimated those of the other two. Computations indicate that the genome organization of wild-type T7 is nearly optimal for growth: only 2.8% of random genome permutations were computed to grow faster, the highest 31% faster, than wild type. Specific discrepancies between computations and observations suggest that a better understanding of the translation efficiency of individual mRNAs and the functions of qualitatively “nonessential” genes will be needed to improve the T7 simulation. In silico representations of biological systems can serve to assess and advance our understanding of the underlying biology. Iteration between computation, prediction, and observation should increase the rate at which biological hypotheses are formulated and tested.

Os contêineres metálicos foram desenvolvidos para a utilização no setor de logística e transporte, mas por sua escala adaptável à das edificações e pela mobilidade e praticidade de instalação, tiveram sua utilização apropriada também pelo setor da construção civil. Essas instalações possuem diversas qualidades ambientalmente amigáveis, mas seu aspecto térmico é extremamente insuficiente: sem isolamento térmico, demandam alta carga térmica de refrigeração e aquecimento, no verão e inverno, respectivamente e, consequentemente, um alto consumo energético. Tal característica foi crucial para que se determinassem como objetivos da presente pesquisa investigar o comportamento térmico dessas construções metálicas, avaliar seus parâmetros de desempenho, conforto e estresse térmicos, por meio de uma ampla coleta de dados experimentais. O experimento com duração de um ano - contou com três tipologias de contêiner em escala real, sendo o primeiro em aço Tipo X sem isolamento térmico, o segundo com um isolamento térmico para o fenômeno da condução e o terceiro com isolamento térmico para o fenômeno da radiação. Os diferentes tipos de tratamentos térmicos proporcionaram melhorias à envoltória dos contêineres, chegando a uma diferença nas temperaturas internas de até 9 °C. Constatou-se a extrema necessidade de adequação do tipo de isolamento térmico dos contêineres ao uso a que tais instalações se destinam escritório ou alojamento, no caso dos canteiros de obras para que as características da envoltória minimizem de fato a demanda ou mesmo atinjam a eliminação da necessidade de condicionamento artificial.

Este trabalho apresenta uma investigação experimental sobre o comportamento à fratura frágil de aços estruturais ferríticos, ASTM A285 Gr C e ASTM A515 Gr 65. Os resultados deste trabalho ampliam a base de dados de propriedades mecânicas utilizadas nas análises de integridade de estruturas pressurizadas tais como vasos de pressão e tanques de armazenamento construídos com esta classe de material. O trabalho tem por objetivo também avaliar a aplicabilidade de corpos de prova de dimensões reduzidas, PCVN, na determinação da temperatura de referência, T0, por meio da metodologia da Curva Mestra, a qual define a dependência da tenacidade à fratura do material em função da temperatura. Os ensaios de tenacidade à fratura foram conduzidos utilizando-se corpos de prova solicitados em flexão três pontos com geometria SE(B), PCVN e PCVN com entalhe lateral, extraídos de chapas laminadas. Os resultados dos ensaios foram obtidos em termos de integral J no momento da instabilidade, denotados por Jc. Dados adicionais de resistência à tração e de Impacto Charpy convencional também foram obtidos para caracterizar o comportamento mecânico dos aços utilizados. Os resultados mostraram uma forte influência da geometria dos corpos de prova sobre os valores de Jc, evidenciada pela grande variação nos valores de tenacidade à fratura.

Phase equilibrium data regression is an unavoidable task necessary to obtain the appropriate values for any model to be used in separation equipment design for chemical process simulation and optimization. The accuracy of this process depends on different factors such as the experimental data quality, the selected model and the calculation algorithm. The present paper summarizes the results and conclusions achieved in our research on the capabilities and limitations of the existing GE models and about strategies that can be included in the correlation algorithms to improve the convergence and avoid inconsistencies. The NRTL model has been selected as a representative local composition model. New capabilities of this model, but also several relevant limitations, have been identified and some examples of the application of a modified NRTL equation have been discussed. Furthermore, a regression algorithm has been developed that allows for the advisable simultaneous regression of all the condensed phase equilibrium regions that are present in ternary systems at constant T and P. It includes specific strategies designed to avoid some of the pitfalls frequently found in commercial regression tools for phase equilibrium calculations. Most of the proposed strategies are based on the geometrical interpretation of the lowest common tangent plane equilibrium criterion, which allows an unambiguous comprehension of the behavior of the mixtures. The paper aims to show all the work as a whole in order to reveal the necessary efforts that must be devoted to overcome the difficulties that still exist in the phase equilibrium data regression problem.

The (vapor + liquid), (liquid + liquid) and (vapor + liquid + liquid) equilibria of the ternary system (water + 1-butanol + p-xylene) have been determined. (Water + 1-butanol + p-xylene) is a type 2 heterogeneous ternary system with partially miscible (water + 1-butanol) and (water + p-xylene) pairs. By contrast, (1-butanol + p-xylene) is totally miscible under atmospheric conditions. This paper examines the (vapor + liquid) equilibrium in both heterogeneous and homogeneous regions at 101.3 kPa of pressure. (Liquid + liquid) equilibrium data at T = 313.15 K have also been determined, and for comparison, the obtained experimental data have been calculated by means of several thermodynamic models: UNIQUAC, UNIFAC and NRTL. Some discrepancies were found between the (vapor + liquid + liquid) correlations; however, the models reproduced the (liquid + liquid) equilibrium data well. The obtained data reveal a ternary heterogeneous azeotrope with mole fraction composition: 0.686 water, 0.146 1-butanol and 0.168 p-xylene.

Numerical modelling methodologies are important by their application to engineering and scientific problems, because there are processes where analytical mathematical expressions cannot be obtained to model them. When the only available information is a set of experimental values for the variables that determine the state of the system, the modelling problem is equivalent to determining the hyper-surface that best fits the data. This paper presents a methodology based on the Galerkin formulation of the finite elements method to obtain representations of relationships that are defined a priori, between a set of variables: y = z(x1, x2,...., xd). These representations are generated from the values of the variables in the experimental data. The approximation, piecewise, is an element of a Sobolev space and has derivatives defined in a general sense into this space. The using of this approach results in the need of inverting a linear system with a structure that allows a fast solver algorithm. The algorithm can be used in a variety of fields, being a multidisciplinary tool. The validity of the methodology is studied considering two real applications: a problem in hydrodynamics and a problem of engineering related to fluids, heat and transport in an energy generation plant. Also a test of the predictive capacity of the methodology is performed using a cross-validation method.

Observations of horizontal and vertical variations in piezometric head in a homogeneous, laboratory aquifer are presented and discussed. The observed fluctuations are induced by a simple harmonic oscillation in the clear water reservoir acting across a sloping boundary. The data qualitatively supports existing theories in that higher harmonics are generated in the active forcing zone and that a significant increase in the inland, asymptotic watertable over height (relative to that found for the vertical boundary case) is observed. The observed overheight is shown to be accurately reproduced by existing small-amplitude perturbation theory. Detailed measurements in the vicinity of the sloping boundary reveal that the signal of generated higher harmonics is strongest near the sand surface and that vertical flows are significant in this region. The aquifer is of finite-depth and is influenced by capillary effects, the experimental data therefore exposes limitations of theories which are based on the assumption of a shallow aquifer free of capillary effects. The dispersive properties of the measured pressure wave in the aquifer are comparable to those found from field observations and likewise do not agree with those predicted by the capillary free, shallow aquifer theory. Although some improvement is obtained, discrepancies between the data and theory persist even when a finite-depth aquifer and capillary effects are considered in the theoretical model. Further sand column experiments eliminate a truncated capillary fringe as a possible contributor to these discrepancies. A hydrogel intervertebral disc (lVD) model consisting of an inner nucleus core and an outer anulus ring was manufactured from 30 and 35% by weight Poly(vinyl alcohol) hydrogel (PVA-H) concentrations and subjected to axial compression in between saturated porous endplates at 200 N for 11 h, 30 min. Repeat experiments (n = 4) on different samples (N = 2) show good reproducibility of fluid loss and axial deformation. An axisymmetric nonlinear poroelastic finite element model with variable permeability was developed using commercial finite element software to compare axial deformation and predicted fluid loss with experimental data. The FE predictions indicate differential fluid loss similar to that of biological IVDs, with the nucleus losing more water than the anulus, and there is overall good agreement between experimental and finite element predicted fluid loss. The stress distribution pattern indicates important similarities with the biological lVD that includes stress transference from the nucleus to the anulus upon sustained loading and renders it suitable as a model that can be used in future studies to better understand the role of fluid and stress in biological IVDs. Analysis of variance (ANOVA) is the most efficient method available for the analysis of experimental data. Analysis of variance is a method of considerable complexity and subtlety, with many different variations, each of which applies in a particular experimental context. Hence, it is possible to apply the wrong type of ANOVA to data and, therefore, to draw an erroneous conclusion from an experiment. This article reviews the types of ANOVA most likely to arise in clinical experiments in optometry including the one-way ANOVA ('fixed' and 'random effect' models), two-way ANOVA in randomised blocks, three-way ANOVA, and factorial experimental designs (including the varieties known as 'split-plot' and 'repeated measures'). For each ANOVA, the appropriate experimental design is described, a statistical model is formulated, and the advantages and limitations of each type of design discussed. In addition, the problems of non-conformity to the statistical model and determination of the number of replications are considered. © 2002 The College of Optometrists.

An experimental testing system for the study of the dynamic behavior of fluid-loaded rectangular micromachined silicon plates is designed and presented in this paper. In this experimental system, the base-excitation technique combined with pseudo-random signal and cross-correlation analysis is applied to test fluid-loaded microstructures. Theoretical model is also derived to reveal the mechanism of such an experimental system in the application of testing fluid-loaded microstructures. The dynamic experiments cover a series of testings of various microplates with different boundary conditions and dimensions, both in air and immersed in water. This paper is the first that demonstrates the ability and performances of base excitation in the application of dynamic testing of microstructures that involves a natural fluid environment. Traditional modal analysis approaches are used to evaluate natural frequencies, modal damping and mode shapes from the experimental data. The obtained experimental results are discussed and compared with theoretical predictions. This research experimentally determines the dynamic characteristics of the fluid-loaded silicon microplates, which can contribute to the design of plate-based microsystems. The experimental system and testing approaches presented in this paper can be widely applied to the investigation of the dynamics of microstructures and nanostructures.

A fine control of the microstructured polymer fiber Bragg grating spectrum properties, such as maximum reflected power and 3-dB bandwidth, through acousto-optic modulation is presented. For simulation purposes, the device is modelled as a single structure, comprising a silica horn and a fiber Bragg grating. For similar sized structures a good correlation between the numerical results and the experimental data is obtained, allowing the strain field to be completely characterized along the whole structure. It is also shown that the microstructured polymer fiber Bragg grating requires less effort from the piezoelectric actuator to produce modification in the grating spectrum when compared with a silica fiber Bragg grating. This technique has potential to be applied on tunable optical filters and tunable cavities for photonic applications.

We report results of an experimental study, complemented by detailed statistical analysis of the experimental data, on the development of a more effective control method of drug delivery using a pH sensitive acrylic polymer. New copolymers based on acrylic acid and fatty acid are constructed from dodecyl castor oil and a tercopolymer based on methyl methacrylate, acrylic acid and acryl amide were prepared using this new approach. Water swelling characteristics of fatty acid, acrylic acid copolymer and tercopolymer respectively in acid and alkali solutions have been studied by a step-change method. The antibiotic drug cephalosporin and paracetamol have also been incorporated into the polymer blend through dissolution with the release of the antibiotic drug being evaluated in bacterial stain media and buffer solution. Our results show that the rate of release of paracetamol getss affected by the pH factor and also by the nature of polymer blend. Our experimental data have later been statistically analyzed to quantify the precise nature of polymer decay rates on the pH density of the relevant polymer solvents. The time evolution of the polymer decay rates indicate a marked transition from a linear to a strictly non-linear regime depending on the whether the chosen sample is a general copolymer (linear) or a tercopolymer (non-linear). Non-linear data extrapolation techniques have been used to make probabilistic predictions about the variation in weight percentages of retained polymers at all future times, thereby quantifying the degree of efficacy of the new method of drug delivery.
