Nucleotídeos extracelulares (ATP, ADP, AMP) e seu derivado adenosina são conhecidos sinalizadores do sistema cardiovascular podendo mediar vários processos fisiológicos entre eles a proliferação celular, agregação plaquetária, inflamação e o tônus vascular. Os níveis destas substâncias, localmente e na circulação sanguínea, são controlados pelas ecto-NTPDases em conjunto com a ecto-5’nucleotidase (ecto-5’-NT) que realizam a degradação completa do ATP até adenosina. Os hormônios tireoideanos tiroxina (T4) e triiodotironina (T3) e o hormônio esteróide sexual estradiol (E2) atuam ativamente no sistema vascular promovendo vasodilatação. Nosso objetivo foi investigar quais enzimas da família das NTPDases estão presente em células musculares lisas vasculares (CMLVs) e se a atividade destas enzimas pode ser influenciada pela ação desses hormônios, uma vez que seus substratos e produtos podem sinalizar processos de relaxamento/contração muscular. Para tanto, as CMLVs foram extraídas da artéria aorta de ratos Wistar adultos e cultivadas em meio de cultura DMEM. Após atingirem a confluência, as células foram tratadas com 50 nM de T3 ou T4 ou 1M de 17-estradiol por 72 horas. As atividades enzimáticas foram medidas pela liberação de fosfato inorgânico enquanto que a expressão das ectonucleotidases foi verificada por imunocitoquímica (proteína) e RT-PCR (RNAm). Os resultados deste trabalho mostram que as CMLVs expressam as NTPDases 1, 2, 3, 5 e 6 e a ecto-5’-NT, responsáveis pelo controle dos níveis de nucleotídeos e nucleosídeos extracelulares. O tratamento com os hormônios T3, T4 e E2 nestas células mostrou que a atividade da ecto-5’-NT foi aumentada pelos três hormônios. A análise do RT-PCR demonstrou que os tratamentos foram capazes de aumentar também a quantidade de RNAm da ecto-5’NT, indicando mecanismos de ação genômica dos hormônios estudados. Por outro lado, O tratamento com os hormônios tireoideos não alterou as atividades ATPásica e ADPásica, somente o estradiol foi capaz de aumentar a atividade ATPásica. Estes resultados também sugerem que, pela hidrólise aumentada do AMP, disponibilizem-se níveis maiores de adenosina, com importante potencial vasodilatador local. Entretanto, o fato de o estradiol ter aumentado a hidrólise de ATP, mas não a de ADP, nos permite pensar que o ADP, agregador plaquetário bem estabelecido, possa estar acumulando extracelularmente, contribuindo para o desenvolvimento de problemas circulatórios.

O desenvolvimento do mercado financeiro e, principalmente, a abertura para o capital externo impulsionaram o desenvolvimento das boas práticas de governança corporativa. Um de seus benefícios é reduzir o custo de captação da empresa e, consequentemente, gerar maior valor para a companhia. Com o novo cenário, o conselho de administração tem um papel fundamental na atividade de governança corporativa, supervisionando a diretoria executiva. O presente trabalho investiga se a adoção de melhores práticas de governança corporativa diminui o risco das empresas. Adicionalmente, analisa se um grau de escolaridade mais alto entre membros do conselho de administração e da diretoria executiva impacta no risco. Para atingir o objetivo, adotou-se o método dos mínimos quadrados para regredir o risco, variável dependente, contra as variáveis independentes nível de governança corporativa e grau de escolaridade. Para o cálculo do risco, utilizaremos a metodologia apresentada por Estrada (2007), o downside beta, ou seja, risco que considera apenas os retornos negativos. Os resultados do estudo sugeriram que um nível de governança corporativa mais alto está presente nas empresas que apresentam um maior risco visto pelo mercado, indicando que as empresas que necessitam de captação, isto é, empresas mais alavancadas, são as empresas que necessitam de um nível de governança corporativa mais alto. Constatou-se, também, que empresas com nível de escolaridade mais alto entre membros do conselho de administração e da diretoria executiva apresentam maiores riscos, pois as empresas que necessitam de pessoas com maior grau de escolaridade são empresas que querem se desenvolver e, portanto, mais arriscadas.

Este artigo busca compreender melhor a relação entre o beta e algumas variáveis fundamentais da empresa, de modo a verificar se estas possuem um impacto relevante no risco sistemático da companhia e contribuir para a literatura já existente sobre o tema. Foi realizada uma regressão linear múltipla utilizando dados em painel para a amostra de 1995 a 2012, além de duas sub amostras deste período, tendo como variável dependente o beta estimado de acordo com o CAPM para três horizontes de tempo (12,24 e 60 meses). Foram selecionadas variáveis que pertencessem a grupos das principais características da empresa, sendo estas o porte, a situação financeira, a rentabilidade, a percepção do mercado, a governança corporativa e a atividade. Verificou-se que a alavancagem, a liquidez e pertencer a setores regulados possuem impacto positivo no risco da empresa. Já o pagamento de dividendos, o nível de governança corporativa e a rentabilidade da companhia afetam negativamente o beta.

Two botryosphaerans, exopolysaccharides (EPS) secreted by the ascomyceteous fungus Botryosphaeria rhodina, when grown on sucrose and fructose as sole carbon sources, were structurally compared after their isolation from the culture medium. Both EPS were submitted to trypsin digestion, and eluted as a single peak on gel filtration. Total acid hydrolysis yielded only glucose, and data from methylation analysis and Smith degradation indicated that both EPS constituted a main chain of glucopyranosyl beta(1 -> 3) linkages substituted at O-6. The products obtained after partial acid hydrolysis demonstrated side chains consisting of glucosyl- and gentiobiosyl- linked beta(1 -> 6) residues. C-13-NMR spectroscopy studies showed that all glucosidic linkages were of the beta-configuration. The carbon source affected the side chain structures of botryosphaeran but not the main chain makeup. High protein content in the diet during childhood and adolescence has been associated to the onset insulin-dependent diabetes mellitus. We investigated the effect of interleukin-1 beta (IL-I beta) on insulin secretion, glucose metabolism, and nitrite formation by islets isolated from rats fed with normal protein (NP, 17%) or low protein (LP, 6%) after weaning. Pretreatment of islets with IL-1 beta for 1 h or 34 h inhibited the insulin secretion induced by glucose in both groups, but it was less marked in LP than in NP group. Islets from LP rats exhibited a decreased IL-1 beta -induced nitric oxide (NO) production, lower inhibition of D-[(UC)-C-14]-glucose oxidation to (CO2)-C-14, and less pronounced effect of IL-1 beta on alpha -ketoisocaproic acid-induced insulin secretion than NP islets. However, when the islets were stimulated by high concentrations of K+ the inhibitory effect of IL-1 beta on insulin secretion was not different between groups. In conclusion, protein restriction protects beta -cells of the deleterious effect of IL-1 beta, apparently, by decreasing NO production. The lower NO generation in islets from protein deprived rats may be due to increased free fatty acids oxidation and consequent alteration in Ca2+ homeostasis. (C) 2001 Elsevier B.V. Purpose: the objective of the present investigation was to determine implantation and pregnancy rates in patients undergoing ICSI and treated with beta(2)-adrenergic agonists, considering the uterine-relaxing action of these agents.Methods: A total of 225 women undergoing ICSI at the Center for Human Reproduction, Sinha Junqueira Maternity Foundation, entered the study. Patient participation in each group was random, by drawing lots, using a randomization table previously elaborated for the study (2:2:1). The group I (90 women) received 10 mg of terbutaline daily for 15 days starting on the day of oocyte retrieval; group II (90 women) received 20 mg of ritodrine daily during the same period of time as group I; group III (45 patients) received no treatment and was used as control. The evaluation was interrupted in 3 patients of group I and in 30 patients of group II because of a high incidence of side effects.Results: Pregnancy, implantation, and miscarriage rates were not significantly different (p>0.05) between the three groups: 29.88%, 13.25%, and 26.9% for group I; 33.33%, 17.5%, and 10.0% for group II; 28.88%, 15.07%, and 15.38% for group III, respectively.Conclusions: the results of this study do not support the routine use of beta(2)-adrenergic agonists during the peri-implantation period in assisted reproductive technology cycles.

Foram avaliados os coeficientes de digestibilidade aparente de MS, PB, energia bruta (EB) e EE de alimentos protéicos (farelo de soja e farinha de peixe) e energéticos (fubá de milho e farelo de trigo) para beta (Betta splendens). Fêmeas adultas foram alojadas em gaiolas e mantidas em dois aquários de fibra de vidro (30 peixes/aquário) para alimentação e dois para coleta de fezes, ambos de formato cônico e com capacidade para 30 L. Os resultados dos coeficientes de digestibilidade aparente de MS, PB, EB e EE foram, respectivamente, de 69,43; 72,52; 67,91 e 55,50% para farelo de soja; 60,67; 51,15; 75,55 e 58,26% para farinha de peixe; 63,88; 87,16; 77,61 e 50,40% para fubá de milho; e 61,06; 93,37; 58,17 e 65,51% para farelo de trigo. Os resultados obtidos neste estudo permitem otimizar a formulação de dietas práticas balanceadas, economicamente viáveis para a espécie.

In Experiments 1, 2, and 3, we evaluated the effects of temporary weaning (TW), equine chorionic gonadotropin (eCG), and follicle-stimulating hormone (FSH) treatments on results of it fixed-time artificial insemination (TAI) protocol in postpartum Bos indicus cows. In Experiment 1, treatment with 400 IU eCG or with TW for 48 h consistently improved pregnancy rates (PRs) at TAI, but, in Experiment 2, FSH treatment was less effective than eCG or TW. In Experiment 3, the inclusion of eCG treatment in cows subjected to TW did not improve PRs. We concluded that TW or 400 IU eCG should be included in the TAI protocol in postpartum Bos indicus cows to enhance fertility. In Experiment 4, we used records front heifers and cows treated with the proposed protocol during the 2006-2007 (n = 27,195) and 2007-2008 (n = 36,838) breeding seasons from multiple locations in Brazil to evaluate factors potentially affecting PRs. Overall PR at TAI was 49.6% (31,786 of 64,033). Pregnancy rate differed (P < 0.01) among farm within location (results ranging between 26.8% and 68.0%; P < 0.01), cow group within farm, by breed (Bos indicus, 48.3% [26,123 of 54,145]; Bos taurus, 61.7% [3652 of 5922]; and crossbred Bos indicus x Bos taurus, 50.7% [2011 of 3966]), category (nulliparous, 39.6% [2095 of 52901] suckled primiparous, 45.2% [3924 of 8677]; suckled multiparous, 51.8% [24,245 of 46,767]; and nonsuckled multiparous, 46.1% [1522 of 3299]), body condition score at TAI (<= 2.5 43.0% [3409 of 7923]; 3.0, 49.6% [18,958 of 38,229]; and >3.5, 52.7% [9419 of 17,881]). Days postpartum at beginning, of protocol did not affect PR (30 to 60 d, 47.6% [4228 of 8881]; 61 to 90 d, 51.7% 116,325 to 31,5721; and 91 to 150 d, 50.8% [7616 to 14,991]; P > 0.1). Pregnancy rate was also consistently affected (P < 0.01) by sire (results ranging from 7.2% to 77.3%) and artificial insemination technician (results ranging from 15.1% to 81.8%). Ovarian activity and plasma concentrations of progesterone and estradiol during pregnancy in jennies

The objectives of this study were to determine ovarian activity (with ultrasound) and plasma concentrations of progesterone and estradiol during pregnancy in jennies. There was considerable ovarian activity during the second month of pregnancy. Secondary corpora lutea (total of 2 to 7 per jenny) were formed (mainly by luteinization) starting on Day 41.8 +/- 1.0 (range Days 38 to 46; ovulation = Day 0). The echogenicity of the primary and secondary corpora lutea gradually decreased during pregnancy. Plasma progesterone concentrations increased between Days 0 and 10 (0.9 and 19.9 ng/mL, respectively), gradually decreased to Day 30 (12.1 ng/mL), increased between Days 30 and 40 (plateau, at approximately 17 ng/mL), gradually declined from Days 110 to 160 (nadir of approximately 6 ng/mL), and remained nearly constant until increasing again just before parturition. Plasma estradiol concentrations increased gradually from Day 65, peaked (1.2 ng/mL) on Day 165 (greater than or equal to 1 ng/mL on Days 150 to 210), and decreased thereafter, with very low concentrations during the last 20 d before parturition. Coupled bone turnover is directed by the expression of receptor-activated NF-kappa B ligand (RANKL) and its decoy receptor, osteoprotegerin (OPG). Proinflammatory cytokines, such as interleukin-1 beta (IL-1 beta) and tumor necrosis factor-alpha (TNF-alpha) induce RANKL expression in bone marrow stromal cells. Here, we report that IL-1 beta and TNF-alpha-induced RANKL requires p38 mitogen-activating protein kinase (MAPK) pathway activation for maximal expression. Real-time PCR was used to assess the p38 contribution toward IL-1 beta and TNF-alpha-induced RANKL mRNA expression. Steady-state RANKL RNA levels were increased approximately 17-fold by IL-1 beta treatment and subsequently reduced similar to 70%-90% when p38 MAPK was inhibited with SB203580. RANKL mRNA stability data indicated that p38 MAPK did not alter the rate of mRNA decay in IL-1 beta-induced cells. Using a RANKL-luciferase cell line receptor containing a 120-kB segment of the 5' flanking region of the RANKL gene, reporter expression was stimulated 4-5-fold by IL-1 beta or TNF-alpha treatment. IL-1 beta-induced RANKL reporter expression was completely blocked with specific p38 inhibitors as well as dominant negative mutant constructs of MAPK kinase-3 and -6. In addition, blocking p38 signaling in bone marrow stromal cells partially inhibited IL-1 beta and TNF-alpha-induced osteoclastogenesis in vitro. Results from these studies indicate that p38 MAPK is a major signaling pathway involved in IL-1 beta and TNF-alpha-induced RANKL expression in bone marrow stromal cells.

The objective of this study was to evaluate protocols for synchronizing ovulation in beef cattle. In Experiment 1, Nelore cows (Bos indicus) at random stages of the estrous cycle were assigned to 1 of the following treatments: Group GP controls (nonlactating, n=7) received GnRH agonist (Day 0) and PGF2 alpha (Day 7); while Groups GPG (nonlactating, n=8) and GPG-L (lactating, n=9) cows were given GnRH (Day 0), PGF2a (Day 7) and GnRH again (Day 8, 30 h after PGF2 alpha). A new follicular wave was observed 1.79+/-0.34 d after GnRH in 19/24 cows. After PGF2a, ovulation occurred in 19/24 cows (6/7 GP, 6/8 GPG, 7/9 GPG-L). Most cows (83.3%) exhibited a dominant follicle just before PGF2a, and 17/19 ovulatory follicles were from a new follicular wave. There was a more precise synchrony of ovulation (within 12 h) in cows that received a second dose of GnRH (GPG and GPG-L) than controls (GP, ovulation within 48 h; P<0.01). In Experiment 2, lactating Nelore cows with a visible corpus luteum (CL) by ultrasonography were allocated to 2 treatments: Group GPE (n=10) received GnRH agonist (Day 0), PGF2a (Day 7) and estradiol benzoate (EB; Day 8, 24 h after PGF2 alpha); while Group EPE (n=11), received EB (Day 0), PGF2a (Day 9) and EB (Day 10, 24 h after PGF2a). Emergence of a new follicular wave was observed 1.6+/-0.31 d after GnRH (Group GPE). After EB injection (Day 8) ovulation was observed at 45.38+/-2.03 h in 7/10 cows within 12 h. In Group EPE the emergence of a new follicular wave was observed later (4.36+/-0.31 d) than in Group GEP (1.6+/-0.31 d; P<0.001). After the second EB injection (Day 10) ovulation was observed at 44.16+/-2.21 h within 12 (7/11 cows) or 18 h (8/11 cows). All 3 treatments were effective in synchronizing ovulation in beef cows. However, GPE and, particularly EPE treatments offer a promising alternative to the GPG protocol in timed artificial insemination of beef cattle, due to the low cost of EB compared with GnRH agonists. Girolando (Gir x Holstein) is a very common dairy breed in Brazil because it combines the rusticity of Gir (Bos indicus) with the high milk yield of Holstein (Bos taurus). The ovarian follicular dynamics and hormonal treatments for synchronization of ovulation and timed artificial insemination were studied in Girolando heifers. The injection of a gonadotrophin-releasing hormone (GnRH) agonist was followed 6 or 7 days (d) later by prostaglandin F2a (PGF2a). Twenty-four hours after PGF2a injection either human chorionic gonadotropin (hCG, GPh-d6 and GPh-d7 groups) or estradiol benzoate (EB, GPE-d6 and GPE-d7 groups) was administered to synchronize ovulation and consequently allow timed artificial insemination (AI) 24 and 30 h after hCG and EB injection, respectively. Follicular dynamics in Girolando heifers was characterized by the predominance of three follicular waves (71.4%) with sizes of dominant follicles (10-13 mm) and corpus luteum (approximately 20 mm) similar to those for Bos indicus cattle. In the GnRH-PGF-hCG protocol, hCG administration induced earlier ovulation (67.4 h, P<0.01) compared to the control group (GnRH-PGF) and a better synchronization of ovulation, since most of it occurred within a period of 12 to 17 h. Pregnancy rate after timed AI was 42.8 (3/7, GPh-d6) to 50% (7/14, GPh-d7). In contrast, estradiol benzoate (GnRH-PGF-EB protocol) synchronized ovulation of only 5 of 11 heifers from the GPE-d7 group and of none (0/7) from the GPE-d6 group, which led to low pregnancy rates after timed AI (27.3 and 0%, respectively). However, since a small number of Girolando heifers was used to determine pregnancy rates in the present study, pregnancy rates should be confirmed with a larger number of animals.

A large number of functional foods, including those that contain P-glucan, have been shown to prevent the development of cancer and other chronic diseases. The aim of the present study was to elucidate its mechanism of action, as well as to understand its effects as an antigenotoxic, anticlastogenic agent, and to determine its capacity to preserve cell viability. The investigation was carried out in the CHO-k1 and CHO-xrs5 cell lines. The cytokinesis-blocked micronucleus assay indicated that the different doses of beta-glucan examined (5, 10, 20 and 40 mu g/ml) did not show clastogenic effects. In the CHO-k1 cell line, a chemopreventive effect could be observed in all the protocols tested: pre-treatment (% reduction of 35.0-57.3), simultaneous treatment (simple - 5 reduction of 19.7-55.6 and with pre-incubation - of 42.7-56.4) and post-treatment (% reduction of 17.9-37.6). This finding indicates mechanisms of action involving desmutagenesis and bio-antimutagenesis, albeit the latter having a lesser role. However, in the repair-deficient CHO-xrs5 cells, beta-glucan did not show a protective effect with post-treatment (% reduction of 2.96), thus supporting the involvement of bioantimutagenesis. The comet assay in CHO-k1 cells demonstrated that beta-glucan has neither a genotoxic nor an antigenotoxic effect. Cell viability tests indicated that beta-glucan preserves cell viability in both cell lines, preventing apoptotic events. These findings suggest that beta-glucan, when present in foods, could provide them with nutraceutical characteristics and act as a dietary supplement, or that P-glucan could be used in new drug development.
