Negócio de Artesão: Empreendendo no Artesanato Pessoal is a blog about entrepreneurship for craftspeople. The present work comes to help in the formation of the artisan, bringing material for study on business, experts and organizations who understand the subject show up to talk about several topics in a simple, concise and relaxed way. Moreover, we also want to tell the story of people who already undertake projects, and supplement their income by selling products they make, humanizing the testimonies. The overall goal is to generate interest for the entrepreneurial activity in people who do handcrafts or who want to start doing it. Thus, people who once used to handcraft just as a hobby can now find a way to supplement their income. The specific goals are: to encourage people to think of a business of their own; stimulate the creative industries through the blog; contribute to the formalization of the artisan; stimulate the exchange of knowledge among artisans; to spread the work of artisans. The methodology used for this project was exploratory research, using bibliographical and desk research, in addition to interviewing and direct and indirect documentary techniques. The method used was the deductive approach, as the methods of procedure used were the historical and comparative ones. As a result, we have the blog named Negócio de Artesão, which can be used both to inform the reader about how he can conceive their enterprise and to be able to make money from their work

From a diagnostic on the learning of written text, this research aimed to carry out educational activities aimed at textual production, performing operation and activity of reflection on texts written by students in a learning situation of parallel recovery, aiming to improve the standard textuality of his written production. These actions were guided from the conception of language as social practice and discourse and, later, a survey was made in the standard of textuality textual productions resulting from such actions. 5 students participated in the research of a 4th grade of elementary school, a School of Duartina peripheral zone integrating the local school system of that city. The textual productions written by these students were analyzed from three categories: common mistakes in the process of literacy, with subdivisions: phonetic transcription, dialetação, hollow hipossegmentação or vocabulary, or improper separation hiperssegmentação, overcorrection, exchange, omission or addition letters, phonemes homorganic exchange; factors textual and contextual, in which it reviewed the following factors: coherence, cohesion and story elements, clarity, in which category were analyzed: the segmentation of text into paragraphs, grammar errors of different nature, use of direct and indirect speech. The results of the intervention and ongoing analysis of data shows that there has been considered developments in the performance of the written production of the research subjects

Using the method of Social Networks Analysis, we’ll try to see if politicians swap votes between them seeking support for their projects, to this end, we’ll first map the behavior, activities and contacts of the Senators of the Brazilian Federative Republic within all the fixed committees which had some vote during the 52nd Legislature, then we can see the most popular and central Senators to each committee. After this analysis we wish to answer the following question: did logrolling exist in the Brazilian Senate? Previously the empirical analysis and response to the problems posed, we’ll present the necessary theoretical introduction understood by reviewing the literature on relevant subjects, whether they are purely theoretical with respect to the exchange of support, or analytical about the Brazilian political system.

To evaluate the cytotoxicity of dimethyl sulfoxide (DMSO) on the repair-related activity of cultured odontoblast-like MDPC-23 cells. Methods Solutions with different concentrations of DMSO (0.05, 0.1, 0.3, 0.5 and 1.0 mM), diluted in culture medium (DMEM), were placed in contact with MDPC-23 cells (5 × 104 cells/cm2) for 24 h. Eight replicates (n = 8) were prepared for each solutions for the following methods of analysis: violet crystal dye for cell adhesion (CA), quantification of total protein (TP), alizarin red for mineralization nodules formation (MN) and cell death by necrosis (flow cytometry); while twelve replicates (n = 12) were prepared for viable cell number (Trypan Blue) and cell viability (MTT assay). Data were analyzed by ANOVA and Tukey or Kruskal–Wallis and Mann–Whitney's tests (p < 0.05). Results Cell viability, adhesion and percentage of cell death by necrosis were not affected by DMSO at any concentration, with no statistical significant difference among the groups. A significant reduction in total protein production was observed for 0.5 and 1.0 mM of DMSO compared to the control while increased mineralized nodules formation was seen only for 1.0 mM DMSO. Significance: DMSO caused no or minor cytotoxic effects on the pulp tissue repair-related activity of odontoblast-like cells.

The aim of the present study was to evaluate the antibiofilm activity against Enterococcus faecalis, compressive strength. and radiopacity of Portland cement (PC) added to zirconium oxide (ZrO2), as radiopacifier, with or without nanoparticulated zinc oxide (ZnO).The following experimental materials were evaluated: PC, PC + ZrO2, PC + ZrO2 + ZnO (5%), and PC + ZrO2 + ZnO (10%). Antibiofilm activity was analyzed by using direct contact test (DCT) on Enterococcus faecalis biofilm, for 5 h or 15 h. The analysis was conducted by using the number of colony-forming units (CFU/mL). The compressive strength was performed in a mechanical testing machine. For the radiopacity tests, the specimens were radiographed together with an aluminium stepwedge. The results were submitted to ANOVA and Tukey tests, with level of significance at 5%. Work on Pt-Sn-C catalysts for ethanol oxidation showed that a thermal treatment at moderate temperatures leads to a significant increase in activity. The best activity was observed for Pt3Sn1 thermally treated at 200 degrees C and ascribed to a Pt3Sn1 phase plus a cleaning effect. However, electronic effects may be very important and these were not evaluated in the Pt3Sn1 phase. Therefore, in this work we investigated the effect of the degree of alloy on the electronic structure of Pt3Sn1 electrocatalysts by performing electrochemical in situ X-ray absorption (XAS) experiments in the Pt L-III XANES region. Overall, the results show that although the occupancy of the Pt 5d band depends on the degree of alloy other factors, such as the presence of tin oxides/hydroxides in the materials, have to be considered to understand the performance of the DEFC.

Experimental etch/leach of Carboniferous Limestone gravels on a laboratory time-scale has demonstrated that 234U 238U activity ratios (AR's) greater than the radiochemical equilibrium value may be generated on short time-scales. The molar U/Ca and Mg/Ca ratios show that both U and Mg are leached preferentially relative to Ca whereas the molar U/Mg ratio is only slightly greater than that of the rock matrix. The generation of enhanced AR's is attributed to a two-stage process in which the limestone surface is dissolved by zero-order etch and silicate minerals so released are subjected to first-order chemical leach of U and Mg. The implications of these results for the production of enhanced AR's in Carboniferous Limestone groundwater are discussed. It is suggested that chemical leaching or exchange of U between groundwater and its particulate load or at the aquifer fluid-solid interface is an important mechanism controlling AR changes as groundwater migrates beyond a redox boundary. AR's for dissolved U in groundwater are more probably related to chemical equilibria than to groundwater age. © 1993.

Due to its elevated cellulolytic activity, the filamentous fungus Trichoderma harzianum (T. harzianum) has considerable potential in biomass hydrolysis application. Cellulases from Trichoderma reesei have been widely used in studies of cellulose breakdown. However, cellulases from T. harzianum are less-studied enzymes that have not been characterized biophysically and biochemically as yet. Here, we examined the effects of pH and temperature on the secondary and tertiary structures, compactness, and enzymatic activity of cellobiohydrolase Cel7A from T. harzianum (Th Cel7A) using a number of biophysical and biochemical techniques. Our results show that pH and temperature perturbations affect Th Cel7A stability by two different mechanisms. Variations in pH modify protonation of the enzyme residues, directly affecting its activity, while leading to structural destabilization only at extreme pH limits. Temperature, on the other hand, has direct influence on mobility, fold, and compactness of the enzyme, causing unfolding of Th Cel7A just above the optimum temperature limit. Finally, we demonstrated that incubation with cellobiose, the product of the reaction and a competitive inhibitor, significantly increased the thermal stability of Th Cel7A. Our studies might provide insights into understanding, at a molecular level, the interplay between structure and activity of Th Cel7A at different pH and temperature conditions.

The effect of short-term creatine (Cr) supplementation upon content of skeletal muscle-derived-reactive oxygen species (ROS) was investigated. Wistar rats were supplemented with Cr (5 g/kg BW) or vehicle, by gavage, for 6 days. Soleus and extensor digitorum longus (EDL) muscles were removed and incubated for evaluation of ROS content using Amplex-UltraRed reagent. The analysis of expression and activity of antioxidant enzymes (superoxide dismutase 1 and 2, catalase and glutathione peroxidase) were performed. Direct scavenger action of Cr on superoxide radical and hydrogen peroxide was also investigated. Short-term Cr supplementation attenuated ROS content in both soleus and EDL muscles (by 41 and 33.7%, respectively). Cr supplementation did not change expression and activity of antioxidant enzymes. Basal TBARS content was not altered by Cr supplementation. In cell-free experiments, Cr showed a scavenger effect on superoxide radical in concentrations of 20 and 40 mM, but not on hydrogen peroxide. These results indicate that Cr supplementation decreases ROS content in skeletal muscle possibly due to a direct action of Cr molecule on superoxide radical.

Objective: To evaluate, in vitro, the antimicrobial activity and biofilm formation of three chlorhexidine varnishes in four Enterococcus faecalis strains: E. faecalis ATCC 29212, E. faecalis EF-D1 (from failed endodontic treatment), E. faecalis 072 (cheese) and E. faecalis U-1765 (nosocomial infection), and one Enterococcus durans strain (failed endodontic treatment). Study Design: The direct contact test was used to study the antimicrobial activity. Bacterial suspensions were exposed for one hour to EC40, Cervitec (CE) and Cervitec Plus (CEP) varnishes. "Eradication" was defined as 100% bacterial kill. The formation of enterococci biofilms was tested on the surface of the varnishes after 24 hours of incubation and expressed as percentage of biofilm reduction. Results: EC40 eradicated all strains except E. faecalis ATCC 29212, where 98.78% kill was achieved. CE and CEP showed antimicrobial activity against all the strains, but most clearly against E. durans and E. faecalis 072. EC40 completely inhibited the formation of biofilm of E. faecalis ATCC 29212, E. faecalis 072 and E. durans. CE and CEP led to over 92% of biofilm reduction, except in the case of E. faecalis U-1765 on CEP (76.42%). Conclusion: The three varnishes studied were seen to be effective in killing the tested strains of enterococci and in inhibiting the formation of biofilm, the best results being observed with EC40.
