Conventional programming techniques are not well suited for solving many highly combinatorial industrial problems, like scheduling, decision making, resource allocation or planning. Constraint Programming (CP), an emerging software technology, offers an original approach allowing for efficient and flexible solving of complex problems, through combined implementation of various constraint solvers and expert heuristics. Its applications are increasingly elded in various industries.

O estudo busca conhecer o trabalho desenvolvido por professores de diferentes componentes curriculares dos anos finais do Ensino Fundamental da rede pública de ensino da cidade de São Paulo de modo a identificar e compreender suas percepções a respeito dos saberes docentes no contexto do processo ensino e aprendizagem. Assim discute-se os saberes que os professores adquirem e/ou reelaboram na prática pedagógica e que são por eles vistos como possibilidades de mudanças no processo ensino e aprendizagem que contemple um ensino de qualidade. O referencial adotado pauta-se em estudos sobre saberes docentes e sua prática, o conhecimento sobre os processos de ensino e aprendizagem e a formação dos professores que atuam nos anos finais do Ensino de Fundamental, tendo como autores principais, Tardif, Garrido, Gatti e Luckesi. Para tanto, procede-se à análise de documentos oficiais e à aplicação de um questionário a doze professores dos anos finais do Ensino Fundamental com o objetivo de conhecer aspectos da vida profissional, bem como as articulações que eles fazem entre saberes docentes, práticas profissionais e processo ensino e aprendizagem. Os resultados demonstram os saberes desenvolvidos pelos docentes, e as práticas pedagógicas que construíram ao longo de seu exercício profissional frente as dificuldades evidenciadas em alguns grupos de alunos. Por fim, os dados revelam a necessidade de se garantir discussões sobre o currículo das turmas dos anos finais do Ensino Fundamental de forma a se perceber que a evolução que os educadores almejam com todos os envolvidos no processo ensino e aprendizagem se ressignificam na prática do conhecimento.

A transition as a function of increasing temperature from harmonic to anharmonic dynamics has been observed in globular proteins by using spectroscopic, scattering, and computer simulation techniques. We present here results of a dynamic neutron scattering analysis of the solvent dependence of the picosecond-time scale dynamic transition behavior of solutions of a simple single-subunit enzyme, xylanase. The protein is examined in powder form, in D2O, and in four two-component perdeuterated single-phase cryosolvents in which it is active and stable. The scattering profiles of the mixed solvent systems in the absence of protein are also determined. The general features of the dynamic transition behavior of the protein solutions follow those of the solvents. The dynamic transition in all of the mixed cryosolvent–protein systems is much more gradual than in pure D2O, consistent with a distribution of energy barriers. The differences between the dynamic behaviors of the various cryosolvent protein solutions themselves are remarkably small. The results are consistent with a picture in which the picosecond-time scale atomic dynamics respond strongly to melting of pure water solvent but are relatively invariant in cryosolvents of differing compositions and melting points.

The positional relationships among all of the visible organelles in a densely packed region of cytoplasm from an insulin secreting, cultured mammalian cell have been analyzed in three dimensions (3-D) at ≈6 nm resolution. Part of a fast frozen/freeze-substituted HIT-T15 cell that included a large portion of the Golgi ribbon was reconstructed in 3-D by electron tomography. The reconstructed volume (3.1 × 3.2 × 1.2 μm3) allowed sites of interaction between organelles, and between microtubules and organellar membranes, to be accurately defined in 3-D and quantitatively analyzed by spatial density analyses. Our data confirm that the Golgi in an interphase mammalian cell is a single, ribbon-like organelle composed of stacks of flattened cisternae punctuated by openings of various sizes [Rambourg, A., Clermont, Y., & Hermo, L. (1979) Am. J. Anat. 154, 455–476]. The data also show that the endoplasmic reticulum (ER) is a single continuous compartment that forms close contacts with mitochondria, multiple trans Golgi cisternae, and compartments of the endo-lysosomal system. This ER traverses the Golgi ribbon from one side to the other via cisternal openings. Microtubules form close, non-random associations with the cis Golgi, the ER, and endo-lysosomal compartments. Despite the dense packing of organelles in this Golgi region, ≈66% of the reconstructed volume is calculated to represent cytoplasmic matrix. We relate the intimacy of structural associations between organelles in the Golgi region, as quantified by spatial density analyses, to biochemical mechanisms for membrane trafficking and organellar communication in mammalian cells.

The Sma and Mad related (Smad) family proteins are critical mediators of the transforming growth factor-β (TGF-β) superfamily signaling. After TGF-β-mediated phosphorylation and association with Smad4, Smad2 moves to the nucleus and activates expression of specific genes through cooperative interactions with DNA-binding proteins, including members of the winged-helix family of transcription factors, forkhead activin signal transducer (FAST)-1 and FAST2. TGF-β has also been described to activate other signaling pathways, such as the c-Jun N-terminal Kinase (JNK) pathway. Here, we show that activation of JNK cascade blocked the ability of Smad2 to mediate TGF-β-dependent activation of the FAST proteins. This inhibitory activity is mediated through the transcriptional factor c-Jun, which enhances the association of Smad2 with the nuclear transcriptional corepressor TG-interacting factor (TGIF), thereby interfering with the assembly of Smad2 and the coactivator p300 in response to TGF-β signaling. Interestingly, c-Jun directly binds to the nuclear transcriptional corepressor TGIF and is required for TGIF-mediated repression of Smad2 transcriptional activity. These studies thus reveal a mechanism for suppression of Smad2 signaling pathway by JNK cascade through transcriptional repression.

This layer is a georeferenced raster image of the historic paper map entitled: OKI regional land use : 1975. It was published by OKI Regional Planning Authority in 1975. Scale [ca. 1:5,000]. Covers Cincinnati Region, Ohio including Butler, Clermont, Hamilton, Warren counties, Ohio; Boone, Campbell, and Kenton counties, Kentucky; and Dearborn and Ohio counties, Indiana. The image inside the map neatline is georeferenced to the surface of the earth and fit to the Ohio South State Plane NAD 1983 coordinate system (in Feet) (Fipszone 3402). All map collar and inset information is also available as part of the raster image, including any inset maps, profiles, statistical tables, directories, text, illustrations, index maps, legends, or other information associated with the principal map. This map is colored to show land use categories: Urban residential ; Suburban residential ; Commercial ; Institutional/Service ; Utilities ; Industrial ; Resource extraction ; Recreational/Open space ; Cropland ; Grassland ; Woodland ; Water. It also shows features as major roads, drainage, administrative and political boundaries, and more. This layer is part of a selection of digitally scanned and georeferenced historic maps from The Harvard Map Collection as part of the Imaging the Urban Environment project. Maps selected for this project represent major urban areas and cities of the world, at various time periods. These maps typically portray both natural and manmade features at a large scale. The selection represents a range of regions, originators, ground condition dates, scales, and purposes.
