We analyze how unemployment, job finding and job separation rates react to neutral and investment-specific technology shocks. Neutral shocks increase unemployment and explain a substantial portion of unemployment volatility; investment-specific shocks expand employment and hours worked and mostly contribute to hours worked volatility. Movements in the job separation rates are responsible for the impact response of unemployment while job finding rates for movements along its adjustment path. Our evidence qualifies the conclusions by Hall (2005) and Shimer (2007) and warns against using search models with exogenous separation rates to analyze the effects of technology shocks.

Background: In insects, like in most invertebrates, olfaction is the principal sensory modality, which provides animals with essential information for survival and reproduction. Odorant receptors are involved in this response, mediating interactions between an individual and its environment, as well as between individuals of the same or different species. The adaptive importance of odorant receptors renders them good candidates for having their variation shaped by natural selection. Methodology/Principal Findings: We analyzed nucleotide variation in a subset of eight Or genes located on the 3L chromosomal arm of Drosophila melanogaster in a derived population of this species and also in a population of Drosophila pseudoobscura. Some heterogeneity in the silent polymorphism to divergence ratio was detected in the D. melanogaster/D. simulans comparison, with a single gene (Or67b) contributing ~37% to the test statistic. However, no other signals of a very recent selective event were detected at this gene. In contrast, at the speciation timescale, the MK test uncovered the footprint of positive selection driving the evolution of two of the encoded proteins in both D. melanogaster ¿OR65c and OR67a ¿and D. pseudoobscura ¿OR65b1 and OR67c. Conclusions: The powerful polymorphism/divergence approach provided evidence for adaptive evolution at a rather high proportion of the Or genes studied after relatively recent speciation events. It did not provide, however, clear evidence for very recent selective events in either D. melanogaster or D. pseudoobscura.

Com a primera part de la monitorització, mitjançant el líquens com a indicadors, del Parc Nacional d'Aigüestortes i Estany de Sant Maurici, hem seleccionat, marcat i caracteritzat 20 estacions de seguiment. Les estacions han quedat establertes en els següents ambients: rouredes amb boix, bosc caducifoli mixt, fagedes, fagedes amb avet, pinedes de pi roig, avetosa, avetosa amb neret, pinedes de pi negre amb nereto amb ginebró, i roquissars, tant en roques carbonatades com àcides. Donem, com a exemple de l'estudi ¿aquestes estacions, el cas ¿una d'avetosa amb neret. El seguiment de l'evolució, en el temps, de les diferents comunitats, ens haurà de permetre correlacionar les modificacions observades en les estacions amb els canvis produïts en la qualitat de l'aire o a causa de l'impacte humà.

El artículo propone hacer un retrato de determinados barrios de la ciudad de Barcelona: "Las fronteras de este territorio son muy claras: al sur limita con la Travessera de Gràcia, al norte con el Monte Carmelo y el Parque Güell, al este con la plaza Lesseps y Gran de Gràcia y al oeste con el Guinar³" a través de la mirada de las novelas de Juan Marsé, durante los oscuros años que corresponden a los años cuarenta y cincuenta del siglo XX y de la forma que sobrevivían unos actores, que si bien eran de ficción, estaban muy cercanos a lo que podría corresponder a personas de carne y hueso, como podremos comprobar por las fuentes históricas que los avalan; insertas en un marco que, llegado a través de la capacidad evocadora de un autor de novelas, siempre están enmarcados en situaciones y espacios muy cercanos a lo real.

Résumé : La sécrétion de l'insuline en réponse au glucose circulant dans le sang est la fonction principale de la cellule β. La perte de cette fonction est une des caractéristiques du diabète de type 2. L'exocytose est une fonction cellulaire indispensable au renouvellement des composants lipidiques et protéiques de la membrane cellulaire, à la communication entre les cellules et au maintien d'un environnement a©quat. On peut distinguer deux types d'exocytose : l'exocytose constitutive et l'exocytose régulée. Cette dernière est ©clenchée par des stimuli externes. L'exocytose régulée est contrôlée au niveau de la fusion des vésicules de sécrétion avec la membrane plasmique. Certains composants moléculaires impliqués dans ce processus font partie de la famille des GTPases Rab. Les deux membres de cette famille impliqués sont Rab3 et Rab27. Nous avons étudié le rôle de la GTPase Rab27 dans les cellules INS-1E, une lignée cellulaire pancréatique β qui sécrète de l'insuline de façon régulée. Nous avons trouvé que la diminution d'expression de la protéine en utilisant le technique de « RNA interference » diminue la sécrétion stimulée, mais que la distribution des granules n'est nullement affectées par ce changement d'activité intrinsèque. Un des effecteurs identifiés de cette GTPase est Slac2c/MyRIP. Cette protéine possède plusieurs domaines fonctionnels dont un qui lui permet de se lier à l'actine, constituant du cytosquelette cellulaire. L'ensemble de nos résultats suggèrent que Rab27 et MyRIP font partie d'un complexe permettant l'interaction de la granule de sécrétion avec le cytosquelette d'actine corticale et participent à la régulation des dernières étapes de l'exocytose d'insuline. Ensuite, nous avons étudié les phosphoinositides (PI). Les phosphoinositides sont d'importantes molécules impliquées dans le régulation du trafic vésiculaire. Nous avons trouvé que le phosphatidylinosito1-4-phosphate (PI4P) et le phosphatidylinositol-4,5-biphosphate (PI(4,5)P2) augmentent la sécrétion sous l'action de 10µM de Ca2+ dans les cellules INS-1E perméabilisées avec la streptolysine-O. En plus, nous avons ©montré que l'exocytose est diminuée dans les cellules intactes exprimant une protéine qui séquestre le PI(4,5)P2. Une diminution similaire est observée en diminuant l'expression de deux enzymes impliquées dans la production du PI(4,5)P2, la PI4Kinase β type III et la PIP5Kinase γ type I. Pour clarifier le mécanisme d'action des PI, nous avons investigué l'implication de trois cibles potentielles des PI, la PLD1, CAPS1 et Mint1. Pour ce faire, nous avons réduit le niveau d'expression endogène de ces protéines, ce qui inhibe la libération d'hormones provoquée par le glucose. Tout ceci indique donc que la production du PI(4,5)P2 est nécessaire pour le contrôle de la sécrétion et suggère qu'une partie de l'effet du PI sur la sécrétion pourrait être exercé par l'activation de la PLD1, CAPS1 et Mint1. Abstract Insulin release from pancreatic β-cells plays an essential role in the achievement of blood glucose homeostasis and defects in the regulation of this process lead to profound metabolic disorders and hyperglycaemia (eg. type 2 diabetes). Almost every cell in our organism releases proteins and other biological compounds using a fundamental cellular process known as constitutive exocytosis. In exocrine and endocrine glands, the cells are endowed with an additional and more refined release mechanism directly tuned by extracellular signals. This process, referred to as regulated exocytosis, ensures the timely delivery of molecules such as peptide hormones and digestive enzymes to match the moment¬-to-moment requirements of the organism. Some of the molecular components involved in this process have been identified, including Rab3 and Rab27, two GTPases that regulate the final steps of secretion in many cells. We investigated the involvement of Rab27 GTPase in the secretory process of the insulin-secreting cell line INS-1E. We found that selective reduction of Rab27 expression by RNA interference did not alter granule distribution but impaired exocytosis triggered by insulin secretagogues. Screening for potential effectors revealed that Slac2c/MyRIP is associated with granules and attenuation of Slac2c expression severely impaired hormone release. This protein contains several functional domains, including, a binding domain for the cellular cytoskeleton constituent actin. Taken together our data suggest the Rab27 and MyRIP are part of a complex mediating the interaction of secretory granules with cortical cytoskeleton and participate to the regulation of the final steps in insulin exoctytosis. In the second part of the thesis, we studied phosphoinositides (PI). Phosphoinositides are important molecules involved in the regulation of vesicular trafficking. We found that phosphatidylinosito1-4-phosphate (PI4P) and phosphatidylinosito1-4,5-biphosphate (PI(4,5)P2) increase the secretory response triggered by 10µM Ca2+ in streptolysin-O permeabilized insulin-secreting INS-1E cells. In addition, nutrient-induced exocytosis was diminished in intact cells expressing constructs that sequester PI(4,5)P2. A similar decrease was observed after silencing of two enzymes involved in PI(4,5)P2 production, type III PI4Kinase β and type I PIP5Kinase γ, by RNA interference. To clarify the mechanism of action of PI, we investigated the involvement in the regulation of exocytosis of three potential PI targets, PLD1, CAPS1 and Mint1. Transfection of cells with silencers capable of reducing the endogenous levels of these proteins inhibited hormone release elicited by glucose. Our data indicate that the production PI(4,5)P2 is necessary for proper control of p-cell secretion and suggest that at least part of the effects of PI on insulin exocytosis could be exerted through the activation of PLD1, CAPS1 and Mint1.

O trabalho objetivou determinar a composição corporal de cordeiros Santa Inês e estimar suas exigências de magnésio, potássio e sódio, para ganho de peso. Foram conduzidos dois experimentos com 18 cordeiros machos em cada um, com peso médio inicial de 25 e 15 kg no primeiro e no segundo experimentos, respectivamente. Em cada experimento, seis animais foram abatidos, para determinação das quantidades de cada mineral retido no corpo, servindo como animais-referência para a técnica do abate comparativo. Os doze animais remanescentes em cada experimento foram divididos em dois grupos: seis animais receberam alimentação ad libitum e seis receberam alimentação restrita. Os cordeiros do grupo ad libitum e restrito foram abatidos quando os do grupo ad libitum atingiram 35 e 25 kg de peso vivo no primeiro e segundo experimentos, respectivamente. A composição corporal em Ca, P, Mg, K e Na foi estimada a partir de equações de regressão do logaritmo da quantidade desses minerais presentes no corpo vazio dos animais, em razão do peso corporal vazio. As exigências líquidas desses minerais por kg de ganho de peso vivo, obtidas a partir da derivação das equações de predição da composição corporal foram: 0,47 e 0,41 g de Mg, 2,32 e 2,05 g de K e 1,33 e 0,55 g de Na, respectivamente, em animais com 15 e 35 kg.

El nostre estudi consisteix a facilitar el desenvolupament, des d'una visió integral, d'un pla de ciutadania i immigració per a localitats de menys de 4.000 habitants amb un percentatge de població estrangera superior al 10 %. La nostra recerca es fa a partir d'un estudi de cas. En aquest projecte hem intentat aprofundir en la realitat de Sant Quintí de Mediona, fent un diagnòstic de tipus participatiu sobre els canvis experimentats pels processos migratoris en aquesta població i establint una priorització de les necessitats observades. Hem estudiat especialment la xarxa d'entitats que hi ha a la població i la seva obertura real que hi participin persones immigrades. A partir d'aquest estudi, s'ha elaborat i negociat un programa orientat al desenvolupament d'una ciutadania intercultural, basada en la convivència com a eix fonamental.

Dois experimentos foram conduzidos com o objetivo de determinar a composição corporal e estimar as exigências de Ca e P, para ganho em peso, de cordeiros Santa Inês em crescimento. Em cada experimento foram usados 18 animais com 25 a 35 kg de peso vivo (PV), no primeiro, e com 15 a 25 kg no segundo experimento. Seis animais foram abatidos no início de cada experimento para avaliar o conteúdo de Ca e P corporal, servindo como animais referência para o método do abate comparativo, seis animais receberam alimentação ad libitum e seis, alimentação restrita. As exigências líquidas de Ca e P para o ganho em peso foram estimadas a partir da derivação de equações de regressão do logaritmo da quantidade desses minerais presentes no corpo vazio, em função do peso do corpo vazio. As exigências líquidas por quilo de ganho de PV para animais com 15, 25 e 35 kg de PV foram, respectivamente: 11,63, 10,52 e 9,82 g de Ca e 5,82, 4,99 e 4,28 g de P.
