O objetivo deste trabalho foi avaliar os efeitos de pré-tratamentos osmóticos sobre a cinética de secagem de fatias de abóbora (Cucurbita moschata) e sobre características sensoriais do produto seco, utilizando sacarose e NaCl como solutos. Condições de pré-tratamento foram selecionadas com base na relação perda de água/ganho de solutos e na limitação de conteúdo de sal no produto final. Amostras tratadas nas condições selecionadas (50% de sacarose; 47% de sacarose e 3% de NaCl; 40% de sacarose; 37% de sacarose e 3% de NaCl, p/p, 1 hora) e amostras não tratadas foram secas com ar aquecido a 70 °C, 2 m/s. A cinética de secagem foi descrita pela Segunda Lei de Fick. Coeficientes de difusão efetivos de água foram determinados considerando-se a espessura inicial e a espessura média das fatias durante a secagem. As difusividades determinadas em amostras tratadas e não tratadas resultaram próximas quando a espessura média foi utilizada. Amostras tratadas apresentaram taxas de secagem maiores que amostras sem tratamento, mostrando que os pré-tratamentos melhoraram a eficiência do processo. Os tratamentos também melhoram a cor e a aparência geral das fatias secas, em relação às amostras sem tratamento.

Os fungos filamentosos termorresistentes são capazes de sobreviver aos processos de pasteurização aplicados aos produtos de frutas. Este estudo visou isolar e identificar fungos termorresistentes durante o processamento de néctar de maçã (pH 3,4 e 11,6 °Brix), de forma a selecionar o bolor mais termorresistente. O isolamento foi obtido após choque térmico de 70 °C/2 horas e incubação a 30 °C em PDA (3% de ágar) acrescido de rosa de bengala. A identificação foi baseada na morfologia e nas estruturas micro e macroscópicas observadas após incubação das cepas em três diferentes meios (G25N, CYA e MEA) e temperaturas. O fungo mais termorresistente foi selecionado pela aplicação de choques térmicos (de 80 °C/20 minutos até 97 °C/15 minutos). Foram detectadas 11 linhagens de bolores, sendo 5 termorresistentes. Dentre estas, três cepas, isoladas da matéria-prima e do concentrado de maçã, foram identificadas como Neosartorya fischeri; uma cepa isolada da matéria-prima foi identificada como Byssochlamys fulva, e uma cepa isolada do produto após a primeira pasteurização foi identificada como Eupenicillium sp. As cepas mais resistentes foram as de N. fischeri e B. fulva, que sobreviveram ao tratamento de 95 °C/20 minutos. Dentre as seis cepas restantes, uma foi identificada como Aspergillus sp., entretanto, nenhuma destas apresentou termorresistência.

Abstract Fish consumption has increased in recent years. However, fish meat is highly perishable, which demonstrates the need for technologies to preserve its quality. Edible coatings (EC) might provide an alternative to extend the shelf life of fish. The goal of this study was to evaluate the effect of EC of chitosan (C) in combination with carvacrol (CAR) on the physical and microbiological changes of tilapia fillets. Fillets were submerged for two minutes in different treatments (T1: control; T2: C 2%; T3: C 2% + 0.125% CAR; T 4: C 2% + 0.25% CAR). At the end of storage, T1 and T2 showed the lowest values of total volatile bases (TVB). The color parameters L*, a* and b* varied from each treatment. The texture decreased and the different treatments reduced the microbial population in relation to the control; T3 and T4 were the most effective. These results show that the use of C with CAR might be an alternative method to preserve the quality and safety of tilapia fillets.

Abstract The aim of this study was to analyze the microbiological quality of meals served in nursing homes in the city of Ponta Grossa, Paraná, before and after the training of food handlers. The first stage was to perform a checklist conforming with current legislation. The second consisted of the collection of 4 food samples from each location and a microbiological investigation in accordance with the relevant legislation. The third was the training of food handlers in relation to good food handling practices. The fourth was a further microbiological analysis of new samples. The application of a checklist showed that the locations met the requirements of current legislation. Of the 40 samples analyzed, 17.5% (7 samples) were unfit for consumption. Among the unfit samples 15% (6 samples) had coliforms at 45 °C, 2.5% (1 sample) had coagulase-positive staphylococci, 2.5% (1 sample) had Bacillus cereus and 2.5% (1 sample) had Salmonella sp. The results of this study show the importance of controlling the quality of food served to an age group that is prone to health risks.

Objetivou-se com esse trabalho avaliar a germinação de sementes de três espécies de Stryphnodendron Mart. (Stryphnodendron adstringens (Mart.) Coville, S. obovatum Benth. e S. polyphyllum Mart.) osmocondicionadas em diferentes agentes osmóticos, nos potenciais de -1.0 MPa e -0.5 MPa. Sementes escarificadas em ácido sulfúrico durante 40 minutos foram condicionadas nos seguintes tratamentos: 1) PEG 6000; 2) PEG 6000 + KNO3, em partes iguais; 3) KNO3; 4) Água e 5) Testemunha, e incubadas em câmara de germinação (BOD) na temperatura de 20 ºC (± 2 ºC) durante 0 h (controle), 6h, 12h e 24 horas, com luz contínua. Para cada espécie, o delineamento estatístico foi o inteiramente casualizado em esquema fatorial 5 (tratamentos de osmocondicionamento) x 4 (tempos de condicionamento) com quatro repetições de 20 sementes. Foram avaliados a porcentagem de germinação (%G), índice de velocidade de germinação (IVG), comprimento médio de raiz (CMR) e de parte aérea (CMPA), e massa fresca (MF) e seca (MS) das plântulas condicionadas a -1,0 MPa e, a %G e IVG daquelas condicionadas a -0,5 MPa. O osmocondicionamento no potencial de -1,0 MPa não alterou significativamente o desempenho fisiológico das sementes de S. obovatum e S. polyphyllum, mas foi prejudicial ao desenvolvimento de S. adstringens. O condicionamento das sementes de S. adstringens e S. polyphyllum em potencial de -0,5 MPa promove maior %G e IVG.

The objective in this study was to verify the efficiency of different procedures for evaluating the physiological potential of bell pepper seed and identify its relationship with germination at different temperatures and with seedling emergence. Five seed lots each of the Reinger and Sentinel hybrids were used. Seed physiological potential was evaluated by germination, saturated salt accelerated aging (48 h/41 °C), seedling emergence (percentage and speed), and tetrazolium tests (preconditioning at 45 °C/3 h and seed staining at 45 °C/2 h). Germination (percentage and speed) on a thermogradient table at 15 °C, 18 °C, 21 °C, 25 °C and 30 °C was also evaluated the saturated salt accelerated aging and tetrazolium tests are suitable for access the physiological potential of bell pepper seeds. It was also confirmed that vigorous seed lots perform better when exposed to different temperatures during germination.

A new synthetic pathway to analogues of the aglucones of naturally occurring cyclic hydroxamic acids (2,4-dihydroxy-l,4-benzoxazin-3-ones) has been developed. The new pathway involves the coupling of substituted nitrophenols wdth /-propyl-abromo- O-methoxymethylglycolate. These materials were reductively cyclised to reveal the hydroxamic acid functionality. Removal of the C-2 0-methoxymethyl protecting group was achieved chemoselectively using boron trichloride. The analogue 7-methoxy-2,4-dihydroxy-l,4-benzoxazin-3-one (DIMBOA) was assayed with papain and a semilog plot of activity of papain in the presence of excess DIMBOA was found to be linear. A single exponential equation was suggested as the model for kinetic analysis. '^ Nuclear magnetic resonance (NMR) spectra of a couple of hydroxamates were acquired as reference standards for future mechanistic studies of these compounds as thiol protease inhibitors. A 10% '^-labeled sample ofDIMBOA was also prepared for future mechanistic studies using NMR techniques.

To further understand in vivo localization and trafficking of a-tocopherol (a-Toe), the most biologically active form of vitamin E, between lipid environments, tocopherols are required that can be followed by teclu1iques such as confocal microscopy and fluorescence resonance energy transfer (FRET) assays. To this end, sixteen fluorescent analogues of a-tocopherol (la-d [(1)anthroy loxy -a-tocopherols, A O-a-Toes], 2a-d [w-nitro benzoxadiazole-a-tocopherols, NBD-aToes], 3a-d [w-dansyl-a-tocopherols, DAN-a-Toes], and 4a-d [w-N-methylanthranilamide-atocopherols, NMA-a-TocsD were prepared by substituting fluorescent labels at the terminus of w-functionalized alkyl chains extending from C-2 of the chroman ring while retaining key binding features of the natural ligand. These compounds were prepared starting from (S)-Trolox® acid VIa esterification, protection, and reduction producing the silyl-protected (S)-Trolox aldehyde that was coupled using Wittig chemistry to different w-hydroxyalkylphosphonium bromides. Reduction of the alkene generated the w-hydroxy functionalized 2-n-alkyl intermediates 9a-d having the necessary 2R stereochemistry. A series of functional group manipulations including mesylation, substitution with azide, and hydride reduction provided w-amino functionalized intermediates 12a-d as well. Coupling intermediates 9a-d and 12a-d with the selected fluorophores (9- anthracene carboxylic acid, 4-chloro-7-nitrobenz-2-oxa-l,3-diazole, 5- dimethylaminonapthalene-l-sulfonyl chloride, and I-methyl-2H-3,1-benzoxazine-2,4(1H)dione), followed by deprotection of the phenolic silyl group, gave the desired fluorescent ligands la-d, 2a-d, 3a-d and 4a-d in good yield. Assessment of their binding affinities with recombinant human a-tocopherol transfer protein (ha-TTP) utilizing fluorescent titration binding assays identified competent ligands for further use in protein studies. Compounds Id (C9-AO-a-Toc) and 2d (C9-NBD-a-Toc) both having nonyl alkyl chain extensions between the chromanol and fluorophore were shown to bind specifically to ha-TTP with dissociation constants (KdS) of approximately 280 nM and 55 nM respectively, as compared to 25 nM for the natural ligand 2R,4'R,^'R-a-tocophQxoL.

Madagascar periwinkle (Catharanthus roseus) produces the well known and remarkably complex dimeric anticancer alkaloids vinblastine and vincristine that are derived by coupling vindoline and catharanthine monomers. This thesis describes the novel application of carborundum abrasion (CA) technique as a tool for large scale isolation of leaf epidermis enriched proteins. This technique was used to facilitate the purification to apparent homogeneity of 16-hydroxytabersonine-16-0-methyltransferse (l60MT) that catalyses the second step in the 6 step pathway that converts tabersonine into vindoline. This versatile tool was also used to harvest leaf epidermis enriched mRNAs that facilitated the molecular cloning of the 160MT. Functional expression and biochemical characterization of recombinant 160MT enzyme showed that it had a very narrow substrate specificity and high affinity for 16-hydroxytabersonine, since other closely related monoterpene indole alkaloids (MIAs) did not act as substrates. In addition to allowing the cloning of this gene, CA technique clearly showed that 160MT is predominantly expressed in Catharanthus leaf epidermis, in contrast to several other OMTs that appear to be expressed in other Catharanthus tissues. The results provide compelling evidence that most of the pathway for vindoline biosynthesis including the 0- methylation of 16-hydroxytabersonine occurs exclusively in leaf epidermis, with subsequent steps occurring in other leaf cell types. Small molecule O-methyltransferases (OMTs) (E.C. 2.1.1.6.x) catalyze the transfer of the reactive methyl group of S-adenosyl-L-methionine (SAM) to free hydroxyl groups of acceptor molecules. Plant OMTs, unlike their monomeric mammalian homologues, exist as functional homodimers. While the biological advantages for dimer fonnation with plant OMTs remain to be established, studies with OMTs from the benzylisoquinoline producing plant, Thalictrum tuberosum, showed that co-expression of 2 recombinant OMTs produced novel substrate specificities not found when each rOMT was expressed individually (Frick, Kutchan, 1999) . These results suggest that OMTs can fonn heterodimers that confer novel substrate specificities not possible with the homodimer alone. The present study describes a 160MT model based strategy attempting to modify the substrate specificity by site-specific mutagenesis. Our failure to generate altered substrate acceptance profiles in our 160MT mutants has lead us to study the biochemical properties ofhomodimers and heterodimers. Experimental evidence is provided to show that active sites found on OMT dimers function independently and that bifunctional heterodimeric OMTs may be fonned in vivo to produce a broader and more diverse range of natural products in plants.

Several inorganic substances (e.g., C£ , Mg , Ca , H ) are potent negative modulators of hemoglobin-oxygen affinity. To evaluate the possibility that potentially adaptive changes in the red cell ionic environment of hemoglobin may take place during acclimation of fishes to increased environmental temperature, hematological status (hemoglobin, hematocrit, red cell numbers, mean erythrocytic volume and hemoglobin content), plasma + + 2+ 2+ and packed red cell electrolyte levels (Na , K , Ca , Mg , C£ ) were evaluated in summer and winter populations of the stenothermal rainbow trout, Salmo gairdneri, following acclimation to 2°, 10°, 18°C, and in a spring population of eurythermal carp, Cyprinus carpio, held at 2°, 16° and 30°C. From these data cell ion concentrations and ion:hemoglobin ratios were estimated. In view of the role of red cell carbonic anhydrase in the reductions of blood C02 tensions and the recruitment of Na and C£~ lost by fishes, a preliminary investigation of thermoacclimatory changes in the activity of this system in rainbow trout erythrocytes was conducted. Few changes in hematological status were encountered following acclimation. There was, however, some evidence of weight-specific differential hematological response in carp. This lead to markedly greater increases in hemoglobin, hematocrit and red cell numbers in smaller rather than in larger specimens at higher temperatures; variations which were 2+ well correlated with changes in plasma Ca . Plasma composition in summer trout was not altered by acclimation. In winter trout plasma Na and K increased at higher temperatures. Carp were characterized by increases in plasma calcium, and reductions in sodium and magnesium under these conditions. Several significant seasonal differences in plasma ion levels were observed in the trout. (n) In trout, only erythrocytic K and K :Hb were altered by acclimation, rising at higher temperatures. In carp Na , Na :Hb, C£~ and C£~:Hb in- 2+ 2+ creased with temperature, while Mg and Mg :Hb declined. Changes in overall ionic composition in carp red cells were consistent with increases in H content. In both species significant reciprocal variations in C£~ 2+ - + and Mg were found. In mammalian systems increases in C£ and H reduce hemoglobin-oxygen affinity by interaction with hemoglobin. Reduction in 2+ 2+ Mg maximizes organophosphate modulator availability by decreasing ATP»Mg complex formation. Thus, the changes observed may be of adaptive value in reducing hemoglobin-oxygen affinity, and facilitating oxygen release to cells at higher temperatures. Trout appear to maintain a high chloridelow magnesium state over the entire thermal tolerance zone. Carp, however, achieved this state only at higher temperatures. In both species mean erythrocytic volume was decreased at higher temperatures and this may facilitate branchial oxygen loading. Since mean erythrocytic volume was inversely related to red cell ion content, it is hypothesized that reductions in cell volume are achieved by export of some unidentified solute or solutes. Variations in the carbonic anhydrase activity that could be attributed to the thermoacclimatory process were quite modest. On the other hand, assays performed at the temperature of acclimation showed a large temperature effect where under in vivo conditions of temperature fish acclimated to higher temperatures might be expected to have higher activities. Furthermore, since hematocrit increased with temperature in these fish, while carbonic anhydrase is present only in the erythrocyte, the whole blood levels of this enzyme are expected to increase and further augment the temperature effect. This, in turn, could aid in the reduction of C02 (111) tension and increase the production of H and HC0~~ used in the active uptake of Na and C£ at higher temperatures.

Dans cet ouvrage sera décrite la synthèse de nouveaux analogues du sialyl Lewis X (sLex). A cet effet, nous avons préparé une librairie d’analogues synthétisée à partir d’une approche mettant en jeu un «espaceur» acyclique permettant d’avoir un biais conformationnel que nous avons défini comme la stratégie ATC-B. Nous avions déjà démontré que certains analogues portant un groupe benzoate en C-2 et en C-4 du galactose présentent une activité 50 fois supérieure à celle du sLex. Nous avions par ailleurs démontré qu’en l’absence du benzoate en C-2, l’activité devient alors trois fois plus faible. A présent, il paraissait interessant de synthétiser des analogues ayant seulement un groupe benzoate en C-4 pour evaluer l’impact de ce groupement sur la puissance de nos analogues. Par le passé, nous avions également mis en évidence le rôle des esters sur l’activité des analogues portant un «espaceur» acyclique dans le cadre de la stratégie ATC-B. Nous effectuerons donc des variations à ce niveau pour en évaluer l’impact. Enfin, nous avons préparé une nouvelle famille d’analogues de type dimère. Ceux-ci seront constitués de 2 unités des composés monomériques synthétisés précédemment. La synthèse de ces dimères fera l’emploi de la «Click Chemistry». Cette étude nous mènera a vous présenter la synthèse de ces composés et la méthodologie employée.
