We evaluated the effect of gamma irradiation doses (0, 125, 250, and 500 Gy) in control of psychrotrophic bacteria in different strains of Agaricus bisporus (ABI-07/06, ABI-05/03, and PB-1) during storage, cultivated in composts based on oat straw (Avena sativa) and Brachiaria spp. The experimental design was completely randomized in a factorial scheme 4  2  3 (irradiation doses  composts  strains), with 24 treatments, each consisting of 2 replicates, totaling 48 experimental units (samples of mushrooms). The mushrooms collected from all culture conditions were packaged in plastic polypropylene with 200 g each and subjected to Cobalt-60 irradiator, type Gammacell 220, and dose rate 0.740 kGy h–1 , according to the treatments. Subsequently, the control (nonirradiated) and other treatments were maintained at 4 ± 1°C and 90% relative humidity (RH) in a climatic chamber to perform the microbiological analysis of mushrooms on the 1st and 14th day of storage. According to the results, it was found that the highest mean colony psychotrophic count, after 14 days of storage, was observed in strain ABI-07/06 1.30 × 108 g -1 most probable number (MPN) in nonirradiated mushrooms, coming from Brachiaria grass-based compost, and this same strain under the same storage conditions, coming from the same type of compost that underwent a dose of 500 Gy, obtained a significant reduction in mean colonies of psychrotrophic bacteria (2.25 × 104 g –1 MPN). Thus, the irradiation doses tested favored reducing the number of colonies of psychrotrophic bacteria, regardless of the type of compound and strain of A. bisporus.

The aim of the work was to evaluate the productivity, leaf nutrient content and soil nutrient concentration in maize (Zea mays L.) grown in sequence with black oats (Avena strigosa Schreb.) under Leucaena diversifolia alley cropping agroforestry system (AFS) and traditional management system/sole crop (without trees-TS), after two years of cultivation following a randomized block design. The experiment was carried out in the Brazilian Association of Biodynamic Agriculture, in Botucatu—S?o Paulo, Brazil. Treatments were: control (C), chemical fertilizer application (F), biomass of L. diversifolia alley cropping application (B), biomass of L. diversifolia alley cropping + chemical fertilizer application (B + F). In the second year of management it was observed that black oat yield was higher in treatments B + F and F with significant difference in relation to the others treatments in both systems, followed by treatment B. Between systems, only treatment B showed significant difference, with higher yield value corresponding to AFS, reflecting the efficiency of AFS to promote soil fertility. Maize production presented the second year of cultivation an increasing trend in all treatments in both production systems. This result may be due to the cumulative effect of mineralization and maize straw and oats, along the experiment. How productivity was higher in the AFS system, could also be occurring effect of biological nitrogen fixation, water retention and reduction of extreme microclimate through the rows of L. diversifolia. Comparing the AFS and TS, it was observed that the concentration of N in leaf tissue was higher in the AFS treatments, probably due to nitrogen fixation performed through the rows of L. diversifolia, that is a nitrogen fixing tree species. After two years, carbon stocked in soil show higher values in the treatments biomass + fertilizer and biomass application, in both systems, AFS and TS.

The objective of this research work was to evaluate the effects of doses and time of application of N on the C/N ratio of the straw cover and on the growth and productivity of maize plants growing in a no tillage system. The experiment was carried out at the Experimental Farm of the College of Agriculture of the São Paulo State University (UNESP) on its campus of Botucatu, state of São Paulo, Brazil. The treatments were distributed in the field according to a randomized complete block design in a split plot arrangement. The treatments consisted of four doses of N (0, 20, 40, and 60 kg ha-1 ) applied to oat crop and N doses (60, 80, 100, and 120 kg ha-1 ) sidedressed to corn. The development and productivity of the maize crop in a no-tillage system were found to be dependent of the C/N ratio and the straw cover. The response of the maize plants to the early application of N is dependent on doses and time of application.

This study was conducted at University of Marília, São Paulo State, Brazil, aiming to determine the phase of intake transition between exclusively liquid diet (milk) and diet based on milk, grass and/or ration by lambs by δ13C technique. Six Suffolk crossbreeding lambs were distributed in a totally random design (3x2), with two experimental groups and three replications. At the first one, lambs were penned and sucked milk from dams fed with C4 diet. besides, they had access to C3 ration in creep feeding, since their birth. At the second group, lambs were kept on a paddock with Tifton – 85 grass (C4 photosynthetic way) as well as their dams and had access to C3 ration in creep feeding, since their birth. The C3 ration contained soybean meal, ground oat grains, lucerne hay and also mineral salt. The penned lambs’ ration consumption overcame the milk intake at 30-35 days after birth; however, grazing lambs ingested more milk and pasture than ration until 40 days after birth. The δ 13C technique was considered effective on studies of lambs feeding behavior.

The present study was aimed to evaluate different (semi-solid) media for the production of Metarhizium anisopliae and Beauveria bassiana propagules, and to evaluate the tolerance of these propagules to ultraviolet radiation and temperature. The experiments were performed at the Biological Control Laboratory of the Instituto Biológico at Campinas, São Paulo, Brazil. For both fungi, 6 repetitions were performed for each of the 17 treatments: corn starch, full rice, parboiled rice, type-1 rice, type-2 rice, oat flakes, canjiquinha [grits], wheat flour, raw cassava flour, yellow corn flour, special wheat flour, corn flour, corn in grains, cassava starch, soy in grains, crushed wheat, and turf. The viability analysis was done in plastic plates containing BDA. For the bioassays involving exposure to ultraviolet light and temperature, BDA was also used for viability analysis, and each treatment was exposed to the UV radiation for 0, 25 and 50 seconds, the temperature exposure being at 20, 25, 30 and 35º C. Using a Potter tower, 2 mL of fungus suspension from each treatment was inoculated to the Diatraea saccharalis caterpillars. Regarding the sporulation, the largest concentrations of M. anisopliae and B. bassiana were found for the treatments with parboiled rice, type-1 rice, type-2 rice, yellow corn flour, corn flour and crushed wheat. The viability of all treatments was superior to 94.00%. Also, the longer the duration of the exposition to the UV, the smaller the number of fertile conidia. At 35o C, a significant loss of conidia viability was observed, and all the treatments presented some level of virulence.

Beyond the physiological and behavioural, differences in appendage morphology between the workers and queens of Apis mellifera are pre-eminent. The hind legs of workers, which are highly specialized pollinators, deserve special attention. The hind tibia of worker has an expanded bristle-free region used for carrying pollen and propolis, the corbicula. In queens this structure is absent. Although the morphological differences are well characterized, the genetic inputs driving the development of this alternative morphology remain unknown. Leg phenotype determination takes place between the fourth and fifth larval instar and herein we show that the morphogenesis is completed at brown-eyed pupa. Using results from the hybridization of whole genome-based oligonucleotide arrays with RNA samples from hind leg imaginal discs of pre-pupal honeybees of both castes we present a list of 200 differentially expressed genes. Notably, there are castes preferentially expressed cuticular protein genes and members of the P450 family. We also provide results of qPCR analyses determining the developmental transcription profiles of eight selected genes, including abdominal-A, distal-less and ultrabithorax (Ubx), whose roles in leg development have been previously demonstrated in other insect models. Ubx expression in workers hind leg is approximately 25 times higher than in queens. Finally, immunohistochemistry assays show that Ubx localization during hind leg development resembles the bristles localization in the tibia/basitarsus of the adult legs in both castes. Our data strongly indicate that the development of the hind legs diphenism characteristic of this corbiculate species is driven by a set of caste-preferentially expressed genes, such as those encoding cuticular protein genes, P450 and Hox proteins, in response to the naturally different diets offered to honeybees during the larval period.

Nitrosyl ruthenium complexes are promising NO donor agents with numerous advantages for the biologic applications of NO. We have characterized the NO release from the nitrosyl ruthenium complex [Ru(NO2)(bpy)(2)(4-pic)](+) (I) and the reactive oxygen/nitrogen species (ROS/RNS)-mediated NO actions on isolated rat liver mitochondria. The results indicated that oxidation of mitochondrial NADH promotes NO release from (I) in a manner mediated by NO2 formation (at neutral pH) as in mammalian cells, followed by an oxygen atom transfer mechanism (OAT). The NO released from (I) uncoupled mitochondria at low concentrations/incubation times and inhibited the respiratory chain at high concentrations/incubation times. In the presence of ROS generated by mitochondria NO gave rise to peroxynitrite, which, in turn, inhibited the respiratory chain and oxidized membrane protein-thiols to elicit a Ca2+-independent mitochondrial permeability transition; this process was only partially inhibited by cyclosporine-A, almost fully inhibited by the thiol reagent N-ethylmaleimide (NEM) and fully inhibited by the NO scavenger 2-(4-carboxyphenyl)-4,45,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO). These actions correlated with the release of cytochrome c from isolated mitochondria as detected by Western blotting analysis. These events, typically involved in cell necrosis and/or apoptosis denote a potential specific action of (I) and analogs against tumor cells via mitochondria-mediated processes. The role of the delta-ornithine amino transferase (OAT) pathway in proline synthesis is still controversial and was assessed in leaves of cashew plants subjected to salinity. The activities of enzymes and the concentrations of metabolites involved in proline synthesis were examined in parallel with the capacity of exogenous ornithine and glutamate to induce proline accumulation. Proline accumulation was best correlated with OAT activity, which increased 4-fold and was paralleled by NADH oxidation coupled to the activities of OAT and Delta(1)-pyrroline-5-carboxylate reductase (P5CR), demonstrating the potential of proline synthesis via OAT/P5C. Overall, the activities of GS. GOGAT and aminating GDH remained practically unchanged under salinity. The activity of P5CR did not respond to NaCl whereas Delta(1)-pyrroline-5-carboxylate dehydrogenase was sharply repressed by salinity. We suggest that if the export of P5C from the mitochondria to the cytosol is possible, its subsequent conversion to proline by P5CR may be important. In a time-course experiment, proline accumulation was associated with disturbances in amino acid metabolism as indicated by large increases in the concentrations of ammonia, free amino acids, glutamine, arginine and ornithine. Conversely, glutamate concentrations increased moderately and only within the first 24 h. Exogenous feeding of ornithine as a precursor was very effective in inducing proline accumulation in intact plants and leaf discs, in which proline concentrations were several times higher than glutamate-fed or salt-treated plants. Our data suggest that proline accumulation might be a consequence of salt-induced increase in N recycling, resulting in increased levels of ornithine and other metabolites involved with proline synthesis and OAT activity. Under these metabolic circumstances the OAT pathway might contribute significantly to proline accumulation in salt-stressed cashew leaves. (C) 2011 Elsevier GmbH. All rights reserved.

A armazenagem de água no solo é muito variável no tempo e no espaço devido à influência de vários fatores ambientais e topográficos. Objetivou-se neste estudo: quantificar a armazenagem de água no solo; avaliar sua estabilidade temporal e sua variabilidade espacial em um local próximo e outro distante do sistema radicular numa sucessão feijão/aveia-preta; e constatar sua variabilidade espacial em função do relevo. Em área experimental de 1.500 m², situada em Piracicaba-SP (latitude de 22° 42' 30" S, longitude de 47° 38' 00" W e 546 m de altitude), estabeleceram-se 60 pontos de amostragem, distanciados entre si de 5 m, numa grade de 10 por 6 pontos (50 x 30 m). Os valores da armazenagem de água no solo apresentaram comportamento-padrão para o solo estudado, com valores maiores em profundidade do que na camada mais superficial. Houve maior estabilidade temporal da armazenagem de água no solo durante o período chuvoso, porém na fase de secagem do solo a estabilidade temporal também foi constatada, mas com valores de coeficiente de correlação mais elevados na camada de 0,0-0,80 m, mostrando que essa estabilidade foi claramente devida à posição topográfica desses pontos, os quais estão localizados na porção mais baixa do relevo. Assim, o ponto 52 foi escolhido como representativo da média na fase de recarga de água no solo em ambas as camadas estudadas, e o ponto 46, na camada superior durante a fase de secagem do solo. A dependência espacial da variabilidade da armazenagem de água no solo foi detectada em ambas as camadas de solo, porém com maior alcance na camada de 0,0-0,40 m, quando houve ocorrência de precipitação. Quando as chuvas cessaram, o alcance foi maior na camada de 0,0-0,80 m. Durante o período chuvoso, o padrão de variabilidade espacial foi muito semelhante em ambas as camadas de solo, com armazenagens maiores na faixa de maior inclinação e depressões do terreno.

This study evaluated the effectiveness of different sealants applied to a nanofiller composite resin. Forty specimens of Filtek Z-350 were obtained after inserting the material in a 6x3 mm stainless steel mold followed by light activation for 20 s. The groups were divided (n=10) according to the surface treatment applied: Control group (no surface treatment), Fortify, Fortify Plus and Biscover LV. The specimens were subjected to simulated toothbrushing using a 200 g load and 250 strokes/min to simulate 1 week, 1, 3 and 6 months and 1 and 3 years in the mouth, considering 10,000 cycles equivalent to 1 year of toothbrushing. Oral-B soft-bristle-tip toothbrush heads and Colgate Total dentifrice at a 1:2 water-dilution were used. After each simulated time, surface roughness was assessed in random triplicate readings. The data were submitted to two-way ANOVA and Tukey's test at a 95% confidence level. The specimens were observed under scanning electron microscopy (SEM) after each toothbrushing cycle. The control group was not significantly different (p>0.05) from the other groups, except for Fortify Plus (p<0.05), which was rougher. No significant differences (p>0.05) were observed at the 1-month assessment between the experimental and control groups. Fortify and Fortify Plus presented a rougher surface over time, differing from the baseline (p<0.05). Biscover LV did not differ (p>0.05) from the baseline at any time. None of the experimental groups showed a significantly better performance (p>0.05) than the control group at any time. SEM confirmed the differences found during the roughness testing. Surface penetrating sealants did not improve the roughness of nanofiller composite resin.

Aspergillus phoenicis is an interesting heat tolerant fungus that can synthesize enzymes with several applications in the food industry due to its great hydrolytic potential. In this work, the fungus produced high enzymatic levels when cultivated on inexpensive culture media consisting of flakes from different origins such as cassava flour, wheat fibre, crushed soybean, agro-industrial wastes, starch, glucose or maltose. Several enzymatic systems were produced from these carbon sources, but amylase was the most evident, followed by pectinase and xylanase. Traces of CMCases, avicelase, lipase, β-xylosidase, β-glucosidase and α-glucosidase activities were also detected. Amylases were produced on rye flakes, starch, oat flakes, corn flakes, cassava flour and wheat fibre. Significant amylolytic levels were produced in the culture medium with glucose or when this sugar was exhausted, suggesting an enzyme in the constitutive form. Cassava flour, rye, oats, barley and corn flakes were also used as substrates in the hydrolytic reactions, aiming to verify the liberation potential of reducing sugars. Corn flakes induced greater liberation of reducing sugars as compared to the others. Thin layer chromatography of the reaction end products showed that the hydrolysis of cassava flour liberated maltooligosaccharides, but cassava flour and corn, rye, oats and barley flakes were hydrolyzed to glucose. These results suggested the presence of glucoamylase and α-amylase as part of the enzymatic pool of A. phoencis.

Fifty-five yearling crossbred steers and 3C cow-calf pairs were used in a forage-based beef production system demonstration project at the Armstrong Outlying Research Farm. From May 11 to June 13, steers rotationally grazed a 41-acre grass pasture that was divided into eight paddocks. From June 13 to August 24, steers were placed in a drylot and fed berseem clover/oat soilage from a strip-intercropping system. Beginning June 5, 36 cow-calf pairs were allowed to rotationally graze the 41-acre pasture until September 18. Calf weight gains for the 110 days were 1.57 pounds per day, and total production from the pasture was 151 pounds per acre. No cow weight change or condition score change was measured. Total steer production was 29 and 580 pounds per acre or average daily gains were .67 and 2.23 pounds while grazing pasture and being fed in a drylot.

BACKGROUND Osteochondral autograft transfer (OAT) aims at restoring normal articular cartilage surface geometry and articular contact mechanics. To date, no studies have evaluated the contact mechanics of the canine stifle following OAT. Additionally, there are no studies that evaluated the role of the meniscus in contact mechanics following OAT in human or canine femorotibial joints. The objective of this study was to measure the changes in femorotibial contact areas (CA), mean contact pressure (MCP) and peak contact pressure (PCP) before and after osteochondral autograft transplantation (OAT) of a simulated lateral femoral condylar cartilage defect with an intact lateral meniscus and following lateral meniscectomy. RESULTS With an intact lateral meniscus, creation of an osteochondral defect caused a decrease in MCP and PCP by 11% and 30%, respectively, compared to the intact stifle (p < 0.01). With an intact meniscus, implanting an osteochondral graft restored MCP and PCP to 96% (p = 0.56) and 92% (p = 0.41) of the control values. Lateral meniscectomy with grafting decreased CA by 54% and increased PCP by 79% compared to the intact stifle (p < 0.01). CONCLUSIONS OAT restored contact pressures in stifles with a simulated lateral condylar defect when the meniscus was intact. The lateral meniscus has a significant role in maintaining normal contact pressures in both stifles with a defect or following OAT. Meniscectomy should be avoided when a femoral condylar defect is present and when performing OAT.

Berseem clover and oats were incorporated into a corncorn- oat/berseem clover rotation in 1994 and 1995. Two cuttings of oat-berseem clover hay were harvested during the summer before forage was allowed to stockpile for winter grazing. In 1995, a brown midrib sorghum x sudangrass hybrid was seeded into a field adjacent to a corn field. After corn grain harvest in 1994 and 1995, Charolais x Angus x Simmental cows in midgestation were allotted to replicated fields containing corn crop residues with no complementary forages at 2.5 acres/cow, or corn crop residues and stockpiled berseem clover (2:1) at 2.5 acres/cow to simultaneously graze, or to a drylot. In 1995, cows were allotted to fields containing corn crop residues and brown midrib sorghum x sudangrass (7:3) at 2.5 acres/cow. Berseem clover had greater concentrations of digestible organic matter and crude protein than corn crop residues at the initiation of grazing, but had a more rapid decrease in digestible organic matter concentration than corn crop residues. Brown midrib sorghum x sudangrass forage also had a higher initial concentration of digestible organic matter, but an equal rate of decrease in digestible organic matter concentration to corn crop residues in ungrazed areas of the field. Cows grazing berseem clover with corn crop residues had greater body condition score increases during the first half of the grazing season than cows grazing corn crop residues without complementary forages. Cows grazing corn crop residues without complementary forages required 2,786 and 1,412 less lb hay per cow than cows maintained in a drylot in 1994 and 1995. In 1994, simultaneous grazing of berseem clover with corn crop residues did not reduce hay feeding more than feeding corn crop residues alone. However, in 1995, grazing berseem clover or brown midrib sorghum x sudangrass with corn crop residues reduced the amount of hay required to maintain cows by 358 and 376 lb hay per cow compared with grazing corn crop residues without complementary forage.
