The pathogenesis of hepatic encephalopathy is multifactorial, involving gut-derived toxins such as ammonia, which has been demonstrated to induce oxidative stress. Therefore, a primary hepatic encephalopathy treatment target is reducing ammonia production in the gastrointestinal tract. AST-120, an oral adsorbent of engineered activated carbon microspheres with surface areas exceeding 1600 m(2) /g, acts as a sink for neurotoxins and hepatotoxins present in the gut. We evaluated the capacity of AST-120 to adsorb ammonia in vitro and to lower blood ammonia, oxidative stress and brain edema in cirrhotic rats. Cirrhosis was induced in rats by bile duct ligation for 6 weeks. AST-120 was administered by gavage preventively for 6 weeks (0.1, 1, and 4 g/kg/day). In addition, AST-120 was evaluated as a short-term treatment for 2 weeks and 3 days (1 g/kg/day) and as a sink to adsorb intravenously infused ammonium acetate. In vitro, AST-120 efficiently adsorbed ammonia. Ammonia levels significantly decreased in a dose-dependent manner for all AST-120-treated bile duct-ligated rats (nontreated: 177.3 ± 30.8 μM; AST-120, 0.1 g/kg/day: 121.9 ± 13.8 μM; AST-120, 1 g/kg/day: 80.9 ± 30.0 μM; AST-120, 4 g/kg/day: 48.8 ± 19.6 μM) and significantly correlated with doses of AST-120 (r = -0.6603). Brain water content and locomotor activity normalized after AST-120 treatments, whereas arterial reactive oxygen species levels remained unchanged. Furthermore, AST-120 significantly attenuated a rise in arterial ammonia after ammonium acetate administration (intravenously). Conclusion:AST-120 treatment decreased arterial ammonia levels, normalized brain water content and locomotor activity but did not demonstrate an effect on systemic oxidative stress. Also, AST-120 acts as an ammonia sink, efficiently removing blood-derived ammonia. Additional studies are warranted to evaluate the effects of AST-120 on hepatic encephalopathy in patients with advanced liver disease. (HEPATOLOGY 2011;).

L’hypertension essentielle étant un facteur majeur de morbidité, la compréhension de son l’étiologie est prépondérante. Ainsi, la découverte de nouvelles composantes ou mécanismes de régulation de la PA par l’identification de QTL et l’étude de leurs interactions s’avère une approche prometteuse. L’utilisation de souches congéniques de rats pour l’étude de l’hypertension est une stratégie payante puisqu’elle permet de masquer les effets de l’environnement, tout en gardant le caractère polygénique de la PA. Longtemps conçu comme un trait issu de l’accumulation des effets minimes des QTL, la PA est régulée par une architecture basée sur l’existence d’interactions épistatiques. L’analyse par paires de QTL individuels a permis d’établir une modularité dans l’organisation des QTL chez le rat Dahl Salt-sensitive en fonction de la présence ou de l’absence d’une interaction épistatique entre eux. Ainsi, deux modules épistatiques ont été établis; EM1 et EM2 où tous les QTL appartenant à EM1 sont épistatiques entre eux et agissent de façon additive avec les membres de EM2. Des hiérarchies dans la régulation peuvent alors être révélées si les QTL d’un même EM ont des effets opposés. L’identification de la nature moléculaire des candidats C18QTL4/Hdhd2 et C18QTL3/Tcof1, membres du EM1, et de l’interaction épistatique entre ces deux QTL, a permis, en plus, d’élucider une régulation séquentielle au sein du module. Hdhd2 pourrait agir en amont de Tcof1 et réguler ce dernier par une modification post-traductionnelle. Cette interaction est la première évidence expérimentale de la prédiction des relations entre QTL, phénomène établi par leur modularisation. Le dévoilement du fonctionnement de l’architecture génétique à la base du contrôle de la PA et la découverte des gènes responsables des QTL permettrait d’élargir les cibles thérapeutiques et donc de développer des traitements antihypertenseurs plus efficaces.

The study deals with the generation of variability for salt tolerance in rice using tissue culture techniques. Rice is the staple food of more than half of the world’s population. The management of drought, salinity and acidity in soils are all energy intensive agricultural practices. The Genetic variability is the basis of crop improvement. Somaclonal and androclonal variation can be effectively used for this purpose. In the present study, eight isozymes were studied and esterase and isocitric dehydrogenase was found to have varietal specific, developmental stage specific and stress specific banding pattern in rice. Under salt stress thickness of bands and enzyme activity showed changes. Pokkali, a moderately salt tolerant variety, had a specific band 7, which was present only in this variety and showed slight changes under stress. This band was faint in tillering and flowering stage .Based on the results obtained in the present study it is suggested that esterase could possibly be used as an isozyme marker for salt tolerance in rice. Varietal differences and stage specific variations could be detected using esterase and isocitric dehydrogenase . Moreover somaclonal and androclonal variation could be effectively detected using isozyme markers.

The data have been rationalized by considering the structure breaking effects of the ions of the salts, the localised hydrolysis model, the internal pressure theory and Symons' theory of water structure.

Solubilities of 4-nitrobenzoic acid at 25°, 35° and 42°C have been determined in water and in the presence of several concentrations of electrolytes. The free energies, enthalpies and entropies of transfer are also reported. The data have been rationalized by considering the structure-breaking effects of the ions of the salts and the requirement of the localized hydrolysis model. The theory of Symons is not satisfactory to rationalise the experimental data.

The Setschenow parameters of solubility in salt solutions and the thermodynamic parameters (25·C) of transfer from aqueous solution to aqueous salt solutions for 2-nitrobenzoic acid and 3-nitrobenzoic acid have been reported. The data have been rationalized on the basis of the localized hydrolysis model and the structure breaking action of ions of the electrolytes.

The thesis mainly discussed the isolation and identification of a probiotic Lactobacillus plantarum, fermentative production of exopolysaccharide by the strain, its purification, structural characterisation and possible applications in food industry and therapeutics. The studies on the probiotic characterization explored the tolerance of the isolated LAB cultures to acid, bile, phenol, salt and mucin binding. These are some of the key factors that could satisfy the criteria for probiotic strains . The important factors required for a high EPS production in submerged fermentation was investigated with a collection of statistical and mathematical approach. Chapter 5 of the thesis explains the structural elucidation of EPS employing spectroscopic and chromatographic techniques. The studies helped in the exploration of the hetero-polysaccharide sequence from L. plantarum MTCC 9510. The thesis also explored the bioactivities of EPS from L. plantarum. As majority of chemical compounds identified as anti-cancerous are toxic to normal cells, the discovery and identification of new safe drugs has become an important goal of research in the biomedical sciences. The thesis has explored the anti-oxidant, anti-tumour and immunomodulating properties of EPS purified from Lactobacillus plantarum. The presence of (1, 3) linkages and its molecular weight presented the EPS with anti-oxidant, anti-tumour and immunomodulating properties under in vitro conditions.

Prevalence of faecal indicator bacteria, Escherichia coli and pathogenic bacteria, Vibrio cholerae, Vibrio parahaemolyticus and Salmonella were analysed in Vembanadu lake (98350N 768250E), along south west coast of India for a period of one year from ten stations on the southern and northern sides of a salt water regulator constructed in Vembanadu Lake in order to prevent incursion of seawater during certain periods of the year. While the northern side of the lake has a connection to the sea, the southern side is enclosed when the salt water regulator is closed. The results revealed the water body is polluted with high faecal coliform bacteria with mean MPN value ranging from 1718-7706/100 ml. E. coli, V. cholerae, V. parahaemolyticus and Salmonella serotypes such as S. paratyphi A, B, C and S. newport were isolated and this is the first report on the isolation of these Salmonella serovars from this lake. E. coli showed highest percentage of incidence (85.6–86.7%) followed by Salmonella (42–57%), V. choleare (40–45%) and V. parahaemolyticus (31.5–32%). The increased prevalence of indicator and pathogenic bacteria in the enclosed southern part of Vembanadu Lake may be resulting from the altered flow patterns due to the salt water regulator.

Contamination of environmental water by pathogenic microorganisms and subsequent infections originated from such sources during different contact and non- contact recreational activities are a major public health problem worldwide particularly in developing countries. The main pathogen frequently associated with enteric infection in developing countries are Salmonella enterica serovar typhi and paratyphi. Although the natural habitat of Salmonella is the gastrointestinal tract of animals, it find its way into natural water through faecal contamination and are frequently identified from various aquatic environments (Baudart et al., 2000; Dionisio et al., 2000; Martinez -Urtaza et al., 2004., Abhirosh et al., 2008). Typhoid fever caused by S. enterica serotype typhi and paratyphi are a common infectious disease occurring in all the parts of the world with its highest endemicity in certain parts of Asia, Africa, Latin America and in the Indian subcontinent with an estimated incidence of 33 million cases each year with significant morbidity and mortality (Threlfall, 2002). In most cases the disease is transmitted by polluted water (Girard et al., 2006) because of the poor hygienic conditions, inadequate clean water supplies and sewage treatment facilities. However in developed countries the disease is mainly associated with food (Bell et al., 2002) especially shellfish (Heinitz et al., 2000

Halobacteria, members of the domain Archaea that live under extremely halophilic conditions, are often considered as dependable source for deriving novel enzymes, novel genes, bioactive compounds and other industrially important molecules. Protein antibiotics have potential for application as preserving agents in food industry, leather industry and in control of infectious bacteria. Halocins are proteinaceous antibiotics synthesized and released into the environment by extreme halophiles, a universal characteristic of halophilic bacteria. Herein, we report the production of halocin (SH10) by an extremely halophilic archeon Natrinema sp. BTSH10 isolated from salt pan of Kanyakumari, Tamilnadu, India and optimization of medium for enhanced production of halocin. It was found that the optimal conditions for maximal halocin production were 42 C, pH 8.0, and 104 h of incubation at 200 rpm with 2% (V/V) inoculum concentration in Zobell’s medium containing 3 M NaCl, Galactose, beef extract, and calcium chloride as additional supplements. Results indicated scope for fermentation production of halocin for probable applications using halophilic archeon Natrinema sp. BTSH10
